目的 建立河北省妊娠期胎儿脐带血血常规红细胞参数的参考范围,并探讨孕周、性别与胎儿红细胞参数参考范围变化的关系.方法 收集河北省人民医院 2010 年 5 月至 2021 年 7 月因各种指征行产前诊断随访结果均正常的 3069 例孕 18~38 周的胎儿脐带血,进行脐带血血常规检测,根据不同孕周对血常规红细胞计数、血红蛋白浓度、红细胞压积、平均红细胞体积、平均红细胞血红蛋白量、平均红细胞血红蛋白浓度的检测结果进行汇总分析.结果 胎儿的血常规红细胞各参数结果呈非正态分布.随着孕周增加红细胞计数、血红蛋白、红细胞压积、平均血红蛋白浓度逐渐升高,红细胞平均体积、平均血红蛋白含量逐渐降低.不同性别胎儿红细胞计数、血红蛋白、平均血红蛋白浓度、红细胞平均体积、平均血红蛋白含量的总体差异具有统计学意义(P<0.05).结论 河北省 22~38 周健康胎儿脐带血血细胞参数在不同孕周及性别之间存在差异,应该独立制订河北省妊娠期胎儿的血细胞参数参考范围,有助于胎儿产前和早产儿疾病的诊断.
This paper reported the genetic analysis of a pedigree in which three affected fetuses with short limbs were revealed by first-trimester ultrasonography in three consecutive pregnancies. Tissues of the second aborted fetus were collected and analyzed by chromosome karyotype analysis and whole exome sequencing. The results indicated compound heterozygous mutations of EX64-EX83 Del and c.8190G>T in the DYNC2H1 gene. Real-time fluorescence quantitative polymerase chain reaction and Sanger sequencing further confirmed that the two variants were inherited from the father and the mother with normal phenotypes, respectively. EX64-EX83 Del was a likely pathogenic variant and c.8190G>T was a variant of uncertain significance. Based on the above results and the medical history, it was highly suspected that the fetus had autosomal recessive short rib polydactyly syndrome type Ⅲ caused by compound heterozygous variants. Real-time fluorescent quantitative polymerase chain reaction and Sanger sequencing results of the third aborted fetus were consistent with the second fetus. Given the same phenotypes of fetuses in the second and third pregnancy, it was strongly suggested that the heterozygous variations of EX64-EX83 Del and c.8190G>T in the DYNC2H1 gene were the pathogenic variants in this pedigree.
目的 基于网络药理学研究栀子-川芎药对抗抑郁物质基础及作用机制,并使用ICR(Institute of Cancer Research)小鼠进行栀子-川芎药对抗抑郁物质基础的初步测试.方法 通过 web of science,CNKI,sciencedirect等数据库,收集栀子-川芎药对治疗抑郁症的活性成分并根据类药性原则进行筛选.使用Swiss Target Prediction database及GeneCards进行栀子-川芎药对抗抑郁作用靶点预测及筛选.通过String数据库对筛选出的靶点进行蛋白相互作用分析,并采用cluego软件对关键抗抑郁作用靶点进行信号通路的富集分析.在此基础上构建栀子-川芎药对抗抑郁活性成分-作用靶点-信号通路网络,并通过Cytoscape 3.7.0软件对其进行拓扑分析.根据分析结果 ,对栀子-川芎抗抑郁靶标网络中筛选出的排名前四位的化合物进行 ICR小鼠抗抑郁活性初步测试.结果 基于Cytoscape 3.7.0软件的分析结果 ,发现栀子-川芎药对抗抑郁的活性成分主要为4-methoxyl-phenethyl-butyl ether,jasminoside J,Gardenal-I和丁基苯酞,主要作用靶点为COMT、HTR1A、DRD2、SLC6A4、SLC6A3、MAOA、GRM5、DRD4、DRD3、HTR2A、HTR3A、GRIA1、APP、HTR1B 和 GRM1 等,主要信号通路为 Serotonergic synapse 和Dopaminergic synapse.动物实验表明,与空白组相比,阳性对照组、丁基苯酞及Gardenal-I中、高剂量组均能显著性减少不动时间,4-methoxyl-phenethyl-butyl ether高剂量组能减少不动时间,进一步验证了丁基苯酞、Gardenal-I,4-methoxyl-phenethyl-butyl ether具有一定的抗抑郁活性,而jasminoside J组未能明显减少不动时间,显示无抗抑郁活性.结论 基于网络药理学预测及动物实验初步验证,栀子-川芎药对抗抑郁的物质基础为丁基苯酞、Gardenal-I,4-methoxyl-phenethyl-butyl ether,此实验为进一步研究栀子-川芎药对抗抑郁的作用机制和筛选抗抑郁新药奠定基础.
目的 比较右美托咪定和咪达唑仑用于小儿麻醉中的临床效果.方法 在2017年1月至2018年5月进行手术治疗的患儿中挑选70例进行此次实验研究,根据患儿在手术中采取的静脉麻醉药物不同,将患儿平均分为对照组和观察组,每组35例.对照组患儿采用咪达唑仑进行静脉麻醉;观察组患儿采用右美托咪定进行静脉麻醉.观察2组患儿给药后不同时间段的心率、呼吸及血氧饱和度,同时对2组患儿的镇静效果、疼痛程度、镇静满意率、面罩及静脉穿刺接受率进行比较,对2组患儿入睡时间、苏醒时间及术后躁动发生率、不良反应发生率等进行观察比较.结果 2组患儿给药后心率、呼吸及血氧饱和度都有明显下降,但2组患儿以上指标在不同时间段差异无统计学意义(P>0.05);观察组患儿镇静效果评分、镇静满意度、面罩及静脉穿刺接受率明显高于对照组,疼痛评分显著低于对照组(P<0.05);2组患儿苏醒时间差异无统计学意义(P>0.05),观察组患儿入睡时间明显早于对照组,术后躁动发生率及不良反应发生率明显低于对照组,差异有统计学意义(P<0.05).结论 用右美托咪定进行小儿静脉麻醉的临床效果比咪达唑仑更加显著,对患儿镇静效果更强,而且患儿术后躁动率和不良反应发生率明显降低,因此,在临床中可以大力推荐这种静脉麻醉的药物.
目的 建立快速检测实验大鼠冠状病毒和仙台病毒的双重PCR方法.方法 根据大鼠冠状病毒N基因、仙台病毒L基因设计特异性引物;经过双重PCR优化,特异性和敏感性的检测,建立双重PCR体系.应用该PCR体系检测人工感染仙台病毒组织DNA样本和实验动物组织样本,并与ELISA方法比对.结果 双重PCR扩增出大鼠冠状病毒(168 bp)和仙台病毒(262 bp)目的条带,PCR扩增产物测序结果利用核酸BLAST功能进行同源序列对比,仙台病毒和大鼠冠状病毒同源性分别为100%和99%.仙台病毒和大鼠冠状病毒的检测下限为1.56×102 copies/μL.特异性检测对小鼠肝炎病毒扩增,产生片段大小近似大鼠冠状病毒产物.应用建立的双重PCR体系检测人工感染仙台病毒组织DNA样本,30份DNA标本均被检出;检测94份实验动物肺组织样本,结果均阴性.结论 建立的双重PCR方法操作简单、快速、特异性强、灵敏度高,能够实现对实验动物仙台病毒和大鼠冠状病毒病原体的快速检测.
目的 探讨危重气管支气管异物患儿硬质气管镜检查术中适宜的气道管理及麻醉方案.方法 选取134例危重气管支气管异物患儿,患儿进入手术室后给予面罩无创机械通气或经气管插管加压给氧,静脉注射盐酸戊乙奎醚0.03 mg/kg,地塞米松0.4~0.5 mg/kg.呼吸窘迫不明显的119例患儿采用不保留自主呼吸的静脉麻醉方式,已出现呼吸窘迫的13例患儿采取保留自主呼吸的静脉麻醉方式;呼吸窘迫严重导致紫绀、意识不清的2例患儿由耳鼻喉科医师紧急置入硬质气管镜.术中根据血氧情况间断给予高频喷射呼吸机喷射给氧,高频通气频率60~80次/min,吸呼比1:1.5,驱动压不超过60 kPa.术毕观察患儿呼吸及神志恢复情况,待自主呼吸恢复,刺激有反应后转回耳鼻喉科术后监护室或儿科ICU进行进一步治疗.结果 134例危重气管支气管异物患儿均一次手术成功取出异物,无严重喉痉挛及支气管痉挛情况,无因屏气呛咳被迫退镜情况发生,无麻醉意外发生.1例患儿术前缺氧时间过长,术后8h因多脏器功能衰竭抢救无效死亡;其余患儿术后缺氧改善,自主呼吸平稳,经治疗3~8d后痊愈出院.结论 硬质气管镜检查术中麻醉医师需根据呼吸困难程度采用不同的麻醉方案,保证充分氧供,避免胃内容物误吸、气道痉挛,协助耳鼻喉医师尽快解除气道梗阻.
目的 研究罗库溴铵应用于可以保证通气的小儿支气管异物取出术麻醉的最佳剂量.方法 选择年龄10~18个月,ASAⅠ~Ⅱ级,需行气管异物取出术的患儿90例,随机分为3组,每组30例.按罗库溴铵剂量分为3组:Ⅰ组:0.3 mg/kg,Ⅱ组:0.4 mg/kg,Ⅲ组:0.5 mg/kg.术中连续监测生命体征,记录诱导后(T0)、置镜(T1)、钳取异物(T2)、术毕(T3)等时间点的HR、MAP及SPO2,起效时间、临床肌松时间、丙泊酚总量及麻醉满意度.结果 与Ⅱ组、Ⅲ组比较,T1、T2时间点Ⅰ组患儿HR、MAP增高(P<0.05);与Ⅰ组比较,Ⅱ组与Ⅲ组起效时间明显增快(P<0.05);与Ⅱ组比较,Ⅲ组起效时间增快(P<0.05).与Ⅰ组及Ⅱ组比较,Ⅲ组临床肌松时间明显延长(P<0.05);与Ⅰ组比较,Ⅱ组及Ⅲ组丙泊酚总量减少,镜检满意度增高(P<0.05).结论 小剂量罗库溴铵0.4 mg/kg应用于可以保证通气的小儿支气管异物取出术,配合高频呼吸机控制通气,具有麻醉深度适合、不良反应较少、苏醒迅速及满意度高的特点,是较为理想的麻醉方式.
目的 探讨不同剂量米库氯铵在不同年龄儿童气管异物取出术麻醉中的应用.方法 选择拟行气管异物取出术儿童100例,依据其年龄和米库氯铵的诱导剂量随机分为4组各25例.Ⅰa、Ⅰb组(幼儿组,1~3岁)分别于麻醉诱导时静脉注射米库氯铵0.15mg/kg、0.20 mg/kg;Ⅱa、Ⅱb组(年长儿组,>3~5岁)分别于麻醉诱导时静脉注射米库氯铵0.15 mg/kg,0.20 mg/kg.术中观察并记录肌松时效指标,包括起效时间,临床作用时间,RI、TOFr75和TOFr90的时间.观察并记录置镜前后MAP、HR、SpO2的变化,置镜满意率和不良反应的发生情况.结果 不同剂量米库氯铵起效时间比较:与Ⅰa、Ⅱa组比较,Ⅰb、Ⅱb组ThD75、ThD90和ThDmax明显缩短,差异均有统计学意义(P<0.05).不同年龄米库氯铵起效时间比较:与Ⅰa、Ⅱa组比较,Ⅱa、Ⅱb组ThDmax明显延长,差异有统计学意义(P<0.05).各组临床作用时间,RI、TOFr 75、TOFr 90的时间差异均无统计学意义(P>0.05).4组置镜满意率分别为96.0%、100.0%、88.0%和96.0%,其差异无统计学意义(P>0.05).MAP、HR、SpO2在组间、时点间、组间·时点间交互作用差异均无统计学意义(P>0.05).4组颈前及胸前皮肤潮红发生率差异无统计学意义(P>0.05),4组均未发生血压下降和支气管痉挛.结论 在应用同等剂量米库氯铵进行麻醉诱导时,幼儿的起效时间显著短于年长儿,但对肌松的恢复无影响;0.15 mg/kg米库氯铵即可为行气管异物取出术儿童提供满意的肌松效果.
目的 探讨危重气管、支气管异物患儿硬质气管镜检查术中的气道管理及麻醉方法.方法 回顾性分析2012年1月至2016年6月共收治的34例危重气管、支气管异物患儿的诊治资料,包括入室(T1)、麻醉诱导后(T2)、支气管镜进入声门后(T3)及退镜后(T4)的HR、SpO2,观察术中有无肢动、呛咳、喉痉挛及呕吐误吸等情况,并记录术前及术后2 h血气分析结果.结果 T2、T3、T4与T1比较患儿HR减慢,差异有统计学意义(P<0.05),T4与T1比较SpO2升高,差异有统计学意义(P<0.05).术后2h PaO2、SaO2较术前升高,差异有统计学意义(P<0.05);术后2hPaCO2较术前下降,差异有统计学意义(P<0.05).2例(5.9%)患儿麻醉诱导时出现呕吐,4(11.8%)例患儿在术中出现肢动及呛咳,全部患儿一次手术成功取出异物,无严重喉及气道痉挛发生,无麻醉意外情况发生.结论 硬质气管镜检查术中麻醉医师需要根据呼吸困难程度采用不同的麻醉方案,保证充分氧供,避免胃内容物误吸、气道痉挛,协助耳鼻喉医师尽快解除气道梗阻,是抢救成功、降低的病死率的关键.
Objective To investigate the effects of sevoflurane combined with remifentanil on agitation at stage of analepsia of anesthesia in children patients. Methods Eighty children patients who were ready to undergo tonsillectomy and adenoidectomy in our hospital were enrolled in the study. According to random number table method , these patients were divided into control group (n=40) and experimental group (n=40). The patients in control group were treated by sevoflurane anesthesia,however,the patients in experimental group were treated by sevoflurane combined with remifentanil anesthesia. The palinesthesia time,,length of operation,the proportion of patients with restlessness,the proportion of patients with agitation>15 minutes and the restlessness scores were observed and compared between the two groups. Results There were no significant differences in palinesthesia time and length of operation between the two groups (P>0. 05). Moreover the restlessness rate at stage of analepsia and average agitation scores in experimental group were superior to those in control group (P <0. 05). Conclusion The combined anesthesia of sevoflurane with remifentanil for children patients undergoing tonsillectomy and adenoidectomy can significantly reduce the incidence of agitation of patients at stage of analepsia,which is helpful to improve clinical curative effects.
Objective The purpose of this study was to determine blood biochemical parameters of HFJ and MIJ rats in different weeks,observe their characteristics of performance,and provide a basic reference data for further research in MIJ and HFJ rats.Method Blood samples were collected from 4,8,12 weeks MIJ and HFJ rats (including MIJ male sterility rats) of different gender,20 in each group.The biochemical parameters were determined including glutamic-pyruvic transaminase (ALT),aspartate aminotransferase (AST),alkaline phosphate (ALP),total protein (TP),serum albumin (ALB),serum globulin (GLO),glutamyl transferase (GGT),cretonnes (CRE),serum urea nitrogen (BUN),blood sugar (GLU),uric acid (UA),triglyceride (TG),total cholesterol (TC),high-density lipoprotein cholesterol (HDLC) and low density lipoprotein cholesterol (LDLC).Calculated the AST/ALT,A/G.Result Comparison of biochemical parameters in female rats of HFJ and MIJ showed:alkaline phosphate (ALP) low-density lipoprotein cholesterol (LDLC) indicators at 4,8,12 weeks of the three groups were extremely significant difference (P < 0.01);HFJ male rats and MIJ males were compared in normal rats:low density lipoprotein cholesterol (LDLC) indexes were significantly different in the three groups of 4,8 and 12 weeks (P < 0.01).HFJ male rats compared with MIJ male infertility in rats:blood glucose (GLU) indicators at 4,8,12 weeks of three groups were extremely significant difference (P <0.01),low density lipoprotein cholesterol (LDLC) index in just 12 weeks of age difference was significant (P < 0.01);The ALP index of HFJ male rats was consistently higher than HFJ female rats at three time points and the difference was significant (P < 0.01).Conclusion Blood biochemical parameters of MIJ and HFJ rats were different because of the difference of species and strains of animals,age and gender.Blood biochemical parameters of MIJ male sterility rats did not show obvious defects.These blood biochemical parameters data have important reference significance for the further research and application in MIJ and HFJ rants.
Objective Aiming at detecting Staphylococcus aureus、Pseudomonas aeruginosa and Klebsiella pneumoniae in laboratory animals,the paper provides a rapid,sensitive and simple test method.Methods According to Staphylococcus aureus nuc gene,Pseudomonas aeruginosa LasI gene,Klebsiella pneumonia PhoE gene and general 16S rRNA gene, designed specific primers;Through the optimization of multiplex PCR primer concentrations and annealing temperature, the specificity and sensitivity of detection, establishing multiplex PCR system.Application of the PCR system test specimens of artificial infections and experiment animal feces is compared with traditional test method.Results Multiplex PCR amplification of Staphylococcus aureus (153 bp), Pseudomonas aeruginosa (600 bp) with Klebsiella pneumoniae (368 bp) and general (520 bp).The multiplex sensitivity for the purpose of 10pg, specificity of detection was not detected from other pathogens.Application of establishing multiplex PCR system to detect the artificial positive samples, and detect 1 Pseudomonas aeruginosa positive case in 76 fecals.Conclusions This paper established the multiplex PCR method which has the advantages of specific,sensitive,simple and rapid, and provides a reliable way for rapid test in laboratory animals microbiology.
The aim of this study is to establish a multiplex polymerase chain raction (PCR) to identify of four kinds of laboratory animal pathogens: Pasteurella multocida, Bordetella bronchiseptica, Mycoplasma pneumoniae and Klebsiella pneumoniae.Methods Specific primers were designed based on GenBank data.The multiplex PCR system was established through optimization of multiple PCR and detection of its specificity and sensitivity.This technique was used to test artificially infected samples and tracheal secretions of experimental animals (rat, mouse, guinea pig, rabbit, hamster), and comparing the detection results by this method and traditional detection test.Results Target bands of Pasteurella multocida (356 bp), Bordetella bronchiseptica (237 bp), Mycoplasma pneumoniae (266 bp), and Klebsiella pneumoniae (142 bp) were obtained, with a detection sensitivity of Klebsiella pneumoniae of 10 pg, and that of Pasteurella multocida, Bordetella bronchiseptica and Mycoplasma pneumoniae of 1 pg by this newly developed multiplex PCR assay.No target bands were observed from the non-specific pathogens of artificially infected samples.The tracheal secretions taken from 45 experimental animals (mice and rabbits) were tested with this new PCR assay, among which 15 cases of Klebsiella pneumonia and 9 cases of Pasteurella multocida were detected as positive, while all the results of traditional method and serological test were negative.Conclusions A simple, rapid, specific and highly sensitive multiplex PCR system has been successfully established.It is valuable for detection of Pasteurella multocida, Bordetella bronchiseptica, Mycoplasma pneumoniae, and Klebsiella pneumoniae in laboratory animals.
Clostridium piliforme,Mycoplasma pulmonic and Rabbit hemorrhagic disease virus are important pathogenic organism to laboratory animals which cause respiratory system disease and digestive system disease and badly affected to product and animal laboratory. To obtain the better diagnostic ways with high specificity and sensibility,thus the methodology of detecting these pathogenic organisms that national testing standards demands has been summarized.The study suggests that two steps should be taken:the first test in traditional methods and the second test in PCR methods to improve veracity and reliability of testing results.
目的 探讨染色体数目、结构和多态性异常与不孕不育临床表现的关系.方法 按照常规G显带方法制备外周血淋巴细胞染色体,并对其核型进行数目和结构的分析,必要的做C显带和Y染色体微缺失.结果 7658例不孕不育病例中251染色体核型异常,异常率为3.28%,其中数目异常93例,结构异常158例,比例分别为1.22%、2.06%,染色体多态性630例,比例为8.23%.结论 细胞学遗传学对于不孕不育患者在优生优育中具有指导意义.
Insufficient matrix metalloproteinase (MMP)-9 and MMP-2 is considered to be a contributor of extracellular matrix (ECM) accumulation in diabetic nephropathy (DN). C-peptide can reverse fibrosis, thus exerting a beneficial effect on DN. Whether C-peptide induces MMP-9 and MMP-2 to reverse ECM accumulation is not clear. In the present study, in order to determine ECM metabolism, rat mesangial cells were treated with high glucose (HG) and C-peptide intervention, then the early and late effects of C-peptide on HG-affected MMP-9 and MMP-2 were evaluated. Firstly, it was confirmed that HG mainly suppressed MMP-9 expression levels. Furthermore, C-peptide treatment induced MMP-9 expression at 6 h and suppressed it at 24 h, revealing the early dual effects of C-peptide on MMP-9 expression. Subsequently, significant increase in MMP-9 expression at 72, 96 and 120 h C-peptide treatment was observed. These changes in MMP-9 protein content confirmed its expression changes following late C-peptide treatment. Furthermore, at 96 and 120 h C-peptide treatment reversed the HG-inhibited MMP-9 secretion, further indicating the late induction effect of C-peptide on MMP-9. The present results demonstrated that C-peptide exerted a late induction effect on MMP-9 in HG-stimulated rat mesangial cells, which may be associated with the underlying mechanism of C-peptide's reversal effects on DN.
Objective To investigate the effects of different doses of dexmedetomidine on perioperative inflammatory response of infants with congenital heart disease(CHD)undergoing intracardiac operation under direct vision. Methods Sixty infant patients with ventricular septal defect at NYHA cardiac function gradeⅠor Ⅱ grade,including 28 males and 32 females,who underwent repair of ventricular septal defect by means of cardopulmonary bypass(CPB)were enrolled in the study. The patients were randomly divided into three groups according to numeration table:control group(group C),low-dose dexmedetomidine group(group L)and high - dose dexmedetomidine group(group H),with 20 patients in each group. The patients in group L were given dexmedetomidine intravenously and infused continuously at 0. 3μg·kg - 1 ·h - 1 until end of the operation, and the patients in group H were given dexmedetomidine intravenously and infused continuously at 0. 6μg·kg - 1 ·h - 1 until end of the operation,however,the patients in group C were given equal volume of 0. 9% sodium chloride solution instead. The patients’venous blood specimens were collected after anesthesia induction,at baseline(T0), 10min after CPB(T1),4h after CPB(T2),24h after CPB(T3),then the serum levels of TNF-α and IL-6 were detected by ELISA. Results As compared with those at T0,the serum levels of TNF-α and IL-6 were significantly increased at T1 ~ T3 in the three groups( P < 0. 05). As compared with those in group C,the serum levels of TNF-α and IL-6 were significantly decreased at T1 ~ T3 in group H( P < 0. 05),but there were no significant differences in these indexes in group L( P >0. 05). Conclusion The intraoperative continuous infusion of dexmedetomidine at a rate of 0. 6μg·kg - 1 ·h - 1 can obviously inhibit the release of inflammatory mediators(TNF-αand IL-6)in infants patients after cardiopulmonary bypass.
目的:评价右美托咪定在小儿纤维支气管镜检查术中的麻醉效果。方法60例接受纤维支气管镜检查的患儿,ASAⅠ或Ⅱ级,1~3岁,随机分为右美托咪定组( D组)和对照组( C组),每组30例。术前30 min静脉注射阿托品0.01 mg/kg,于麻醉诱导前约10 min由家长陪同进入麻醉准备室,D组5 min内泵注右美托咪定0.5μg/kg,C组给予同等容量的0.9%氯化钠溶液。2组采用丙泊酚2 mg/kg、芬太尼1μg/kg诱导后,开始镜检操作,术中均保留自主呼吸。记录2组镜检时间、苏醒时间、清醒时躁动发生率;对2组麻醉效果进行评级;观察2组各时间点MAP、HR变化。结果 D组清醒时躁动发生率低于C组( P <0.05)。 D组镇痛效果优于C组( P <0.05)。与T0时比较,T1时2组平均动脉压(MAP)、心率(HR)均明显降低( P <0.05),T2~T4时C组MAP明显升高、D组HR明显降低(P<0.05);与C组比较,T1时D组HR明显降低,T2~T4时D组MAP、HR均明显降低( P <0.05)。结论右美托咪定复合丙泊酚、芬太尼用于小儿纤维支气管镜检查术,麻醉效果满意。
目的:比较右美托咪定和咪达唑仑抑制患儿气管插管反应的效果。方法拟行斜视矫正术的患儿60例,ASAⅠ或Ⅱ级,性别不限,年龄4~8岁,随机分为2组:右美托咪定组( D组)和咪达唑仑组( M组),麻醉诱导:2组患儿给予丙泊酚,入睡后D组依次静脉泵注右美托咪定0 g.5 g/kg,芬太尼2 g/kg、罗库溴铵1 mg/kg;M组以相同方法给予咪达唑仑0.1 mg/kg,肌松起效后行气管插管。记录麻醉诱导前(T0)、气管插管时(T1)、插管后1 min(T2)、插管后3 min(T3)、插管后5 min(T4)的心率(HR)、血压(MAP)和肾上腺素(E)、去甲肾上腺素(NE)的浓度。结果 T1~T4时M组MAP、HR显著高于T0时和D组( P <0.05);T3~T4时M组E和NE的浓度显著高于T0时和D组( P <0.05)。结论右美托咪定抑制小儿气管插管时的应激反应的效果优于咪达唑仑。
OBJECTIVE:To explore the effect of high-fat diet and food restriction on energy metabolism in obesity-prone (OP) and obesity-resistant (OR) rats.METHODS:Sixty male Sprague-Dawley (SD) rats were divided into OP, OR and control groups according to their body weight gain after fed with high-fat diet for 3 wk. OP and OR groups were fed with high-fat diet in the following 12 wk to promote the development of obesity. Then one-half of the rats of each group began to food restriction and were allowed access to 50% of their individual baseline mean daily food intake each day, while the other half were maintained on ad libitum food for 2 wk. Basal metabolic rate (BMR), resting metabolic rate (RMR) of each group were measured by indirect calorimetry during the high-fat diet feeding and food restriction conditions. After the rats were sacrificed, body fat content was measured.RESULTS:OR rats had significantly higher BMR and RMR than the other two groups during high-fat diet feeding condition. There was no significant difference between OP and control group. Food restriction led to a reduction in BMR and RMR in all groups. OR rats showed a significantly greater reduction. OP group showed a significant decrease in body fat weight and fat content during the food restriction period, while there was no significant differences in OR rats.CONCLUSION:There are significant differences between OP and OR rats in BMR and RMR either in high-fat diet feeding condition or food restricted state. OR rat has the ability to sense and respond to energy imbalance more accurately than OP rat.
