Cirsium setidens Nakai contains bioactive compounds that exert biological activities. Method validation for analysis of the pectolinarin content in Cirsium setidens extracts (CSE) and radical scavenging-linked antiobesity activities using 3T3-L1 cells and C57BL/6 mice were performed. The pectolinarin content of CSE was 2.81±0.01 mg/g with a high degree of linearity in calibration curves ( R 2 =0.9999). CSE exhibited free radical-scavenging activities and a reducing power. CSE and pectolinarin inhibited lipid accumulation during adipogenesis of 3T3-L1 cells via down-regulation of adipogenic transcription factors. CSE supplementation suppressed body weight in C57BL/6 mice fed a high fat diet and reduced plasma total cholesterol, triglyceride, insulin, and glucose levels. Pectolinarin-enriched CSE can be considered as a good source of natural antioxidants and antiobesity ingredients.
The physicochemical properties of Jerusalem artichoke (Helianthus tuberosus L.) tea infusions produced using drying, roasting, puffing, and extrusion methods were investigated. The surfaces of puffed and extruded artichokes were more porous than the surfaces of dried and roasted artichokes. The water solubility index was highest for tea infusions of extruded and puffed artichokes. The total phenolic contents and antioxidant activities of artichoke tea infusions were in the order of puffed > extruded > roasted > hot air dried. Moderate and fine particle sizes used for tea infusions had the highest browning degrees and antioxidant activities, similar to total phenolic contents. Basic data are provided with implications for further development of processed food products, including tea bags using Jerusalem artichoke.
십자화과 채소인 콜라비 및 무에는 glucosinolate 성분이 함유되어 있으며, 특히 새싹 채소의 경우 성숙한 채소보다 glucosinolate의 함량이 더 높은 것으로 알려져 있다. 이와 같은 이유로 최근 새싹채소에 대한 관심은 높으나, 아직 새싹채소에 대한 연구는 미비한 편이다. 따라서 본 연구에서는 십자화과 채소인 콜라비 새싹 추출물(KSE) 및 무순 추출물(RSE)의 total carotenoid 및 glucosinolate의 함량을 분석하였고 이들 추출물의 항산화 및 anti-adipogenic 활성을 평가하였다. KSE의 total carotenoid 및 glucosinolate 함량은 각각 $39.50{\pm}0.67$ 및 $2.65{\pm}0.02mg/g$이며, RSE의 total carotenoid 및 glucosinolate 함량은 각각 $76.73{\pm}2.75$ 및 $8.13{\pm}0.54mg/g$으로 KSE보다 높았다. 다양한 항산화 평가 모델(DPPH, ABTS, FRAP, reducing power)을 통하여 항산화 활성을 측정한 결과, KSE 및 RSE는 농도 의존적으로 항산화 활성이 증가하는 경향을 나타내었다. 또한, 지방세포의 분화과정에서 KSE 및 RSE의 처리는 지방세포의 지방축적 및 ROS 생성을 억제하였으며, KSE 및 RSE의 주요 성분으로 알려진 ${\beta}$-carotene, glucoraphanin 및 glucoraphenin의 처리는 지방 축적 억제 및 ROS 생성 저감 활성을 나타내었다. 이상의 결과로부터, ${\beta}$-carotene, glucoraphanin 및 glucoraphenin이 풍부한 RSE 및 KSE는 항산화 활성 및 지방세포 분화억제 효능을 갖으며, 천연물 유래 항비만 소재로서 활용 가능성이 높은 것으로 기대된다. Common cruciferous vegetables, kohlabi (Brassica oleracea) and radish (Raphanus sativus), contain phytochemicals such as glucosinolates and carotenoids. Therefore, this study investigated the antioxidant and anti-adipogenic effects of kohlrabi sprout extract (KSE) and radish sprout extract (RSE). The total carotenoid and glucosinolate contents of KSE and RSE were $39.50{\pm}0.67$ and $76.73{\pm}2.75mg/g$, respectively. The total glucosinolate contents of KSE and RSE were $2.65{\pm}0.02$ and $8.13{\pm}0.54mg/g$, respectively. The in vitro-antioxidative activities of KSE and RSE were significantly increased in a dose-dependent manner. Bisphenol A (BPA), an environmental endocrine disrupting chemical, is contained in polycarbonate plastic products. BPA plays as a mixed agonist‐antagonist with respect to steroid hormones such as estrogens. Recent studies have suggested that BPA affects adipocytes diffenetiation and lipid accumulation by regulation of adipogenic transcription factors. In this study, we evaluated the influence of BPA on oxidative stress during the differentiation of 3T3‐L1 preadipocytes. We also evaluated that inhibitory effects of Brassicaceae spp. spout extracts such as Raphanus sativus L, Brassica oleracea var gongyloides, Brassica rapa var. glabra Regel on BPA‐induced oxidative stress during the differentiation of 3T3‐L1 preadipocytes. Our results showed that BPA treatment significantly increased ROS production in 3T3‐L1 cells compared with control by down‐regulating expression of antioxidant enzyme such as manganese superoxide dismutase (Mn‐SOD) and glutathione reductase (GR). However, Brassicaceae spp. spout extracts significantly inhibited BPA‐induced ROS production. In addition, Brassicaceae spp. spout extracts increased the level of the expression of antioxidant enzyme such as Mn‐SOD and GR. These results indicated that Brassicaceae spp. spout extracts consider as inhibitors that attenuate BPA‐induced adipocytes dysfunction.
As previously reported, high temperature- and high pressure-treated red ginseng (HRG) contain higher contents of phenolic compounds and protect C2C12 muscle cells and 3T3-L1 adipocytes against oxidative stress. This study investigated the effect of HRG on oxidative stress using a mouse model. Our results show that the levels of glutamic oxaloacetic transaminase and glutamic pyruvic transaminase, hepatic malondialdehyde in the HRG group were significantly lower than those of the exercise groups supplemented with commercial red ginseng (CRG) or not supplemented. The muscular glycogen level, glucose-6-phosphate dehydrogenase and lactate dehydrogenase activities of the HGR group were higher than that of the CGR group. Furthermore, the HRG treatment group displayed upregulated mRNA expression of Cu/Zn-SOD and muscle regulatory factor 4. These results indicate that HRG may protect oxidative stress induced by exercise as well as improve exercise performance capacity.
In this study, we investigated the antioxidant and anti-adipogenic activities of aqueous extracts of mugwort and pine needles fermented by Leuconostoc mesenteroides 1076. We also evaluated the phenolic composition, total phenolic content and flavonoid contents of both the fermented mugwort extract (FME) and fermented pine needle extract (FPE). The antioxidant potentials of the FME and FPE were assessed using various antioxidant assays, including the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical assay, the 2,2-azino-bis-(3-ethylenebenzothiozoline-6-sulfonic acid) (ABTS) radical scavenging assay, the ferric-reducing antioxidant power (FRAP) assay, and the reducing power assay. In addition, the anti-adipogenic effects of the FME and FPE were investigated during the adipogenesis of 3T3-L1 cells. Our results showed that the total phenolic contents of the FME and FPE were 182 +/- 4 and 116 +/- 5 mg GAE/g, respectively. Among the phenolic compounds, chlorogenic acid was found to be the major phenolic acid in the FPE, whereas hesperetin was found to be the major phenolic acid in the FME. The antioxidant activities of the FME and FPE increased in a dose-dependent manner. The FME exhibited higher antioxidant activities than the FPE. Furthermore, both the FME and FPE inhibited lipid accumulation and reactive oxygen species (ROS) production, which involved the down-regulation of peroxisome proliferator-activated receptor gamma (PPAR gamma), adipocyte protein 2 (aP2) and NADPH oxidase 4 (NOX4) mRNA expression. These results indicated that the FPE and FME could be useful in the treatment of obesity and as natural sources of antioxidants.
고려엉겅퀴는 예로부터 구황식품으로 사용되어 식용뿐만 아니라 한방 약재로도 사용되어 왔으며, 최근에는 건강기능식품 원료로서도 각광받고 있다. 본 총설에서는 고려엉겅퀴에 대한 주요 생리활성을 중심으로 요약하였으며, 고려엉겅퀴의 항산화 활성, 간 보호작용, 항염증 활성 및 지표성분 pectolinarin의 다양한 생리활성도 소개하고자 기술하였다. 최근 합성 항산화제를 대체하기 위한 천연항산화제의 필요성이 점차 증가하고 있음에도 불구하고, 효능이 우수한 천연항산화제가 상업화되기 위해서는 천연항산화제에 대한 안전성 및 경제성 등이 확보되어야 할 것으로 사료된다. 고려엉겅퀴는 수천 년 동안 식품으로 사용되어 왔고, 최근에는 다양한 생리활성을 나타내는 바 건강기능식품의 원료로서 사용도가 점차 증가하고 있으며 고려엉겅퀴 추출물의 독성평가가 이루어진다면 추후 개별인정형 건강기능식품으로도 개발될 수 있을 것으로 사료된다.
Brassica spp. vegetables have been known to have biological activities such as anti-cancer and anti-inflammatory activities. In this study, we investigated the in vitro physicochemical characteristics and antioxidant activities of commonly used Brassica spp. sprout vegetables such as kohlabi (Brassica oleracea var. gongyloides), red radish (RaphanussativusL. var. sativus), broccoli (B. oleraceavar. italica), cabbage (B. rapavar. glabra Regel), rape (B. napus), radish (R. sativus), and tatsoi (B. campestris var. narinosa) sprouts. Our results showed that the vegetables with the highest total phenolics contents were the radish sprout (24.40±1.24 mg TAE/g) and kohlabi sprout extracts (23.97±0.46 mg TAE/g). Furthermore, the vegetable with the highest total flavonoid content was the radish sprout extract (15.30±1.35 mg CE/g). However, the kohlabi sprout extract showed the highest DPPH radical scavenging value (IC50= 1.95 mg/mL) and ORAC value (79.03 mM TE/g). In addition,the six kinds of Brassica spp. sprout vegetable extracts, except tatsoi, significantly inhibited the reactive oxygen species (ROS) production and showed thatintracellular oxidative stress is closely related tothe accumulation of differentiated adipocytes and fat during the adipogenesis of 3T3-L1 preadipocytes. These results suggest that Brassica spp. sprout vegetables, especially kohlabi and radish sprout extracts, can be used to develop natural antioxidants.
The aim of this study was to determine the total phenol, total flavonoids, and proanthocyanidin contents of fermented black rice and its fractions, as well as to assess the antioxidant activities. Antioxidative activities were assessed in various in vitro models using 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2"-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), reducing power, ferric ion reducing antioxidant power (FRAP), and nitrite scavenging activity (Griess reagent assay). Our results show that the antioxidant activity was significantly higher in the low-molecular fraction of fermented black rice than in the other samples (p<0.05). Among the fermented black rice and its fractions, the low-molecular fraction had the highest total phenol (109.2±2.9 ㎎ GAE, gallic acid equivalent/g), total flavonoids (39.4±0.8 ㎎ RE, rutin equivalent/g), and proanthocyanidin (32.9±1.4 ㎎ CE, catechin equivalent/g) contents, which correlated strongly with its antioxidative activity. Considering the high consumer demand due to the beneficial health effects, fermented black rice and its fractions can be utilized to develop functional food, as well as health-promoting and pharmaceutical agents.
This study was to evaluate the phenolic content and composition of Carthamus tinctorius L. seed extract (CSE) and to further assess its antioxidant and anti-adipogenic activities using various radical scavenging systems and 3T3-L1 cells. Our results show that the total phenolic and flavonoid contents of CSE were 126.0 ± 2.4 mg GAE/g and 62.2 ± 1.9 mg QE/g, respectively. The major phenolic compounds in CSE was (−)-epigallocatechin (109.62 mg/g), with a 4-hydroxy benzhydrazide derivative and gallocatechin present at 18.28 mg/g and 17.02 mg/g, respectively. CSE exhibited remarkable radical scavenging activities, FRAP (ferric reducing antioxidant power) and reducing power in a dose-dependent manner. Moreover, the oxygen radical absorbance capacity (ORAC) value of CSE (0.1 mg/mL) was 62.9 ± 4.7 μM TE (trolox equivalent)/g. During adipogenesis, CSE significantly inhibited fat accumulation in 3T3-L1 cells compared with control cells. Overall, these results indicate that CSE might be a valuable source of bioactive compounds that impart functional food and natural antioxidant properties.
Wild herbs, which are edible plants that grow in mountainous areas, have diverse biological effects such as anti-obesity and anti-cancer activities. The aim of this study was to evaluate the total phenolic and flavonoid contents as well as the antioxidant activity of methanol extracts of Aster scaber, Ligularia fischeri, Kalopanax pictus, Codonopsis lanceolata, and Cirsium setidens and to assess their effects on lipid accumulation and reactive oxygen species (ROS) production during adipogenesis of 3T3-L1 cells. The results revealed that among the five studied wild herb extracts, Ligularia fischeri showed the highest total phenolic contents (215.8 ± 14.2 mg GAE/g) and Aster scaber showed the highest total flavonoid content (103.9 ± 3.4 mg RE/g). Furthermore, Aster scaber and Ligularia fischeri extracts showed higher antioxidant activity than the other wild herbs. Regarding anti-adipogenic activity, the Cirsium setidens extract significantly inhibited lipid accumulation (~80%) and ROS production (~50%) during adipogenesis of 3T3-L1 cells compared with control cells. These results suggest that wild herbs could be used for the development of functional foods as well as health promoting and pharmaceutical agents.
본 연구에서는 강내한성 강원양파의 건조방법을 달리하여 건조방법에 따른 이화학적 특성을 비교하였다. FD, HD 40 및 HD 60의 세 가지 분말에 대해 일반성분 분석, 색도, 갈색도, 총 페놀 함량 및 DPPH free radical 소거능을 측정하였다. 일반성분 분석의 결과 FD의 조단백질 및 조지방의 함량이 각각 10.99%, 0.84%로 HD 40 및 HD 60 보다 유의적으로 높게 나타났다. 색도의 경우, Hunter's value로 L값(lightness), a값(redness), b값(yellowness)으로 나타내었다. FD의 L값이 $77.19{\pm}0.01$로 가장 높게 나왔으며, 나머지 a값 및 b값은 각각 $6.09{\pm}0.01$과 $6.09{\pm}0.01$으로 HD 60이 높게 나왔다. 갈변도의 경우에도 HD 60이 0.200으로 제일 높은 값으로 나타나 건조 온도가 높아질수록 양파의 갈변정도가 커진 다는 것으로 사료된다. 총 페놀 함량의 경우에 HD 40과 HD 60이 FD 보다 더 높은 함량을 나타내었다. DPPH radical 소거능은 시료의 농도를 0.1, 0.5, 1 mg/mL로 처리하여 측정한 결과 농도에 따라 유의적으로 항산화 활성이 증가되었으며, FD 보다 HD 40 및 HD 60에서 유의적으로 더 높은 항산화 활성을 나타내었다. This study was performed to provide the basic data for the prediction of the usefulness of onion with cold tolerance cultivated in Kangwon. The physicochemical properties and antioxidant activity of freeze-dried and hot-air-dried (40 and $60^{\circ}C$) onions were investigated. The moisture content of the raw onion was 90.55%. The crude protein and crude fat contents of the freeze-dried onions were slightly higher than those of the hot-air-dried onions ($40^{\circ}C$ and $60^{\circ}C$). As for the color values, the freeze-dried onion powder was highest in lightness (77.19), and the $60^{\circ}C$ hot air-dried onion was highest in redness (6.09) and yellowness (24.60). Moreover, the color difference (${\Delta}E$) between the freeze-dried and hot-air-dried ($40^{\circ}C$ and $60^{\circ}C$) onion powders was significant. The brown index was lower in the freeze-dried onion than in both hot-air-dried onions. The total phenol content and the 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) scavenging activity of both hot-air-dried onions were higher than those of the freeze-dried onion. These results indicate that the freeze-drying methods protected the physicochemical properties of the onion powder, whereas the hot-air-drying method enhanced the antioxidant activity and the total phenol content of the onion powder.
Buckwheat sprouts contain various bioactive compounds including rutin which have a number of biological activities. We have previously shown that buckwheat sprouts (TBWE) treated with methyl jasmonate (MeJA) significantly increased the amount of phenolics and the antioxidant activity. The aim of this study was to demonstrate the effect of TBWE on anti-adipogenesis and pro-oxidant enzyme in 3T3-L1 adipocytes. We also evaluated the anti-oxidative activity of TBWE in adipocytes by using the nitroblue tetrazolium assay. Our data showed that TBWE markedly inhibited adipocyte differentiation and ROS production in 3T3-L1 cells compared with control groups. Moreover, TBWE has strongly shown the inhibition of adipogenic transcription factor as well as pro-oxidant enzymes. Together, we demonstrate that the MeJA treatment significantly increased the amount of phenolic compound, resulting in the suppression of adipogenesis and ROS production in the 3T3-L1 cells. These findings indicate that TBWE has the potential for anti-adipogenesis activity with anti-oxidative properties.
In this study, the antioxidant potential of commercial beverages against a variety of radicals was determined using various antioxidant activity analytical methods. The physicochemical properties (pH value and °Brix), total phenolic content and antioxidant activities were assessed. Our results showed that the pH value and sugar content (°Brix) of commonly consumed beverages ranged from 2.4 to 3.9 and from 3.8 to 18.5, respectively. The DPPH radical scavenging activity and nitrite scavenging ability were highest in No. 45 vitamin beverage (87.5% and 86.0%, respectively). However, no clear correlation appeared between the total phenolic content and the DPPH radical scavenging activity (R=0.2565). The total phenolic content and oxygen radical absorbance capacity (ORAC) values were highest in No. 55 pomegranate beverage (183.3mg GAE/100mL and 1824.4μMTE/mL, respectively). In particular, a high correlation was shown between the total phenolic content and the ORAC value (R=0.7954). Based on the results of various antioxidant activity methods, the greatest preventative antioxidant capacity of consumed beverages in Korea was found in No. 55 pomegranate. These results will enable further research on the daily phenolic compound intake as well as on the development of healthy beverages.
소나무 껍질 추출물은 높은 항산화 활성을 가지는 것으로 많은 연구 결과를 통해 조사되어 왔으며, 해외에서는 이미 프랑스 해송 껍질 추출물인 피크노제놀(pycnogenol)에 관한 연구가 많이 이루어진 반면, 우리나라 적송 껍질 추출물(PineXol$^{(R)}$)의 생리활성 효과에 대해서는 아직 연구가 많이 이루어지지 못했다. 따라서 본 연구에서는 우리나라 적송 껍질 추출물인 PineXol$^{(R)}$의 항산화 활성 및 anti-adipogenic 활성을 평가하였다. PineXol$^{(R)}$의 총 페놀 및 플라보노이드 함량은 각각 $717.40{\pm}6.86$ GAE mg/g 및 $54.44{\pm}0.01$ RE mg/g으로 측정되었다. 또한 다양한 항산화 평가 모델(DPPH, ABTS, FRAP, 환원력)을 통하여 PineXol$^{(R)}$의 항산화 활성을 측정한 결과, 농도가 증가함에 따라 항산화 활성이 유의적으로 증가하였으며, 대조군으로 사용한 동일한 농도의 아스코르빈산과 유사한 항산화 활성을 나타내었다. 또한 ORAC value는 $693.97{\pm}14.13{\mu}M$ TE/g으로 측정되었고, 1.0 mg/mL의 농도에서 55.39%의 아질산염소거능을 나타내었다. PineXol$^{(R)}$은 3T3-L1 지방세포에서 세포독성을 나타내지 않았으며, 분화과정동안 50, 100 및 200 ${\mu}g/mL$의 농도에서의 지방축적량은 각각 $66.85{\pm}5.87$, $44.59{\pm}5.71$ 및 $20.85{\pm}2.78%$의 농도가 증가함에 따른 유의적인 억제효과를 보였다. 이상의 결과로부터, PineXol$^{(R)}$은 항산화 활성 및 지방세포 분화억제 효능을 갖으며, 천연물 유래 항산화제로써 활용 가능성이 높은 것으로 기대된다. Pine bark extract is made from the bark of Pinus densiflora which naturally contains occurring phytochemicals such as phenolic compounds. PineXol$^{(R)}$ from products of pine bark extract is sold under the brand name. The aim of this study was to evaluate the total phenol, total flavonoids contents and antioxidant activity of the PineXol$^{(R)}$ as well as to assess the lipid accumulation during adipogenesis of 3T3-L1 cells. Our results demonstrate that the total phenolic and flavonoids contents of the PineXol$^{(R)}$ were $717.40{\pm}6.86$ GAE mg/mL and $54.44{\pm}0.01$ RE mg/mL, respectively. In addition, the PineXol$^{(R)}$ inhibited the adipocyte differentiation of 3T3-L1 preadipocytes. Exposure to 200 ${\mu}g/mL$, PineXol$^{(R)}$ significantly reduced lipid accumulation (~80%) in 3T3-L1 cells compared to control cells.
Cirsium setidens Nakai, a wild perennial herb, grows mainly in Kangwon province, Korea, and it has been reported to contain bioactive ingredients with various medicinal activities including treat edema, bleeding and hemoptysis. However, the potentiality of anti‐obesity effects of C. setidens Nakai has not been fully investigated. Thus, we investigated the anti‐obesity effect of C. setidens Nakai extract (CNE) on lipid accumulation in 3T3‐L1 cells during the differentiation and obesity in high‐fat diet (HFD)‐ induced obese C57BL/6 mice. Furthermore, evaluate the concentrations of serum lipids, including total cholesterol, triglycerides, HDL cholesterol, and LDL cholesterol, in mice fed a high‐fat diet. Our results show that CNE inhibited adipocyte differentiation of 3T3‐L1 preadipocytes. Exposure to 100 ¥ìg/mL, CNE significantly reduced lipid accumulation (~30%) in 3T3‐L1 cells compared to control cells by downregulating gene expression of adipogenesis‐related mRNA such as peroxisome proliferator‐activated receptor‐¥ã (PPAR‐¥ã). In addition, CNE supplementation in mice fed a HFD revealed that significantly decreased HFD‐induced increases in body and organs weights as compared to the HFD group. These results suggest that CNE can be useful in the treatment of obesity as non‐toxic natural resources.

The aim of this study was to evaluate the effect of BPA on adipogenesis and oxidative stress during the differentiation of 3T3‐ L1 preadipocytes. We also demonstrate that effect of a combination of BPA plus beta‐carotene on lipid accumulation and ROS production in 3T3‐L1 cells. Our results show that BPA promotes the lipid accumulation and ROS production in 3T3‐L1 cells. Especially, BPA significantly increase ROS production (~40%) in 3T3‐L1 cells compared to control cells by up‐regulating gene expression of pro‐oxidant enzyme. Nevertheless, BPA in combination with beta‐carotene was markedly inhibited lipid accumulation and ROS production compared with control groups that treated with BPA alone. These results indicate that beta‐carotene consider inhibitor that acceleration of adipocyte differentiation by BPA
본 연구는 홍화씨의 식품학적 성분 및 항균활성을 분석하였다. 홍화씨의 수분함량은 $5.58{\pm}0.09%$, 조단백질 $37.16{\pm}0.85%$, 조회분 $3.52{\pm}0.04%$, 조지방 $13.69{\pm}8.98%$, 탄수화물 40.05%이었다. 홍화씨의 총 아미노산은 391.99 mg%이고, 이중 필수아미노산의 함량은 122.97 mg%이었다. 홍화씨의 아미노산 중 alanine (75.43 mg%), arginine (37.93 mg%) 및 aspartic acid (33.24mg%)을 많이 함유하고 있었다. 홍화씨의 주요 유리당은 fructose가 3.29%이었고, linoleic acid (C18:2)는 약 79.46%의 비율을 차지하여 홍화씨의 조지방 중 가장 많은 것으로 나타났다. 또한, Oleic acid (C18:1)도 약 10.42%를 함유하여 불포화지방산의 함유량도 매우 높게 나타났다. 홍화씨의 무기질 함량 측정 결과, K 및 Ca의 함량이 1,528 및 1,005 mg%으로 가장 많이 함유되어 있었다. 총 페놀 및 총 플라보노이드 함량은 $55.52{\pm}0.99$ mg GAE/g, $78.69{\pm}0.91$ mg QE/g으로 나타났다. 홍화씨의 항균활성은 농도가 증가함에 따라 Staphylococcus aureus, Salmonella typhimurium, Escheria coli, Candida albicans, Bacillus cereus, Listeria monocytogenes, Vibrio parahaemolyticus 및 Clustridium perfringens 균에 대한 항균 효과가 높아지는 것으로 나타났다. This study was conducted to investigate the physicochemical characteristic of Carthamus tinctorius L. seed and to assess its total phenol content, total flavonoids content and antimicrobial activity. The moisture, crude protein, crude fat, crude ash and carbohydrates of the Carthamus tinctorius L. seed were 5.58, 37.16, 13.69, 3.52, and 40.05%, respectively. Total amino acid in Carthamus tinctorius L. seed was 391.99 mg%. The major free sugar of Carthamus tinctorius L. seed were fructose(3.29%) and sucrose(1.74%). Linoleic acid(79.46%) was a major fatty acids in the crude fat of Carthamus tinctorius L. seed. The K and Ca contents were the highest in Carthamus tinctorius L. seed. Total phenol and total flavonoids contents of the ethanolic extract were $55.52{\pm}0.99$ mg GAE/g and $78.69{\pm}0.91$ mg QE/g, respectively. The extract from Carthamus tinctorius L. seed showed growth inhibitory effect on Staphylococcus aureus, Salmonella typhimurium, Escheria coli, Candida albicans, Bacillus cereus, Listeria monocytogenes, Vibrio parahaemolyticus, and Clustridium perfringens. These results indicate that the Carthamus tinctorius L. seed extract can inhibit food pathogen associated with total phenol and total flavonoids contents.
Rhodiola species have anti‐fatigue, anti‐stress, antioxidant and immune enhancing effects. Skeletal muscle generates reactive oxygen species, which contributes to muscle fatigue. This study investigated the inhibitory effects of the fermented mixture of Rhodiola rosea and red ginseng on H2O2 induced oxidative damage in C2C12 myotubes by analyzing morphological changes, cell viability, and gene expression level of antioxidant enzyme responses. C2C12 myoblasts were treated with fermented mixture for up to 5 days. Our data showed that the fermented mixture of Rhodiola rosea and red ginseng significantly protected oxidative damage during the myogenesis compared to H2O2 treated C2C12 cells in a dose dependent manner. The fermented mixture of Rhodiola rosea and red ginseng also markedly enhanced mRNA expression of Cu/Zn‐superoxide dismutase (SOD) and glutathione peroxidase (GPx) as well as MyoD, a muscle‐specific transcription factor. Our results suggest that the inhibitory activity of the fermented mixture of Rhodiola rosea and red ginseng on oxidative damage may involve coordinated changes of cellular antioxidant enzymes in C2C12 cells.
