Cyclin-dependent kinase 8 (CDK8) has been implicated in various tumors, with its role differing across tumor types. However, the association between CDK8 polymorphisms and bladder cancer (BC) remains unclear. This study investigated the association between CDK8 polymorphisms and BC susceptibility and prognosis. This case-control study included 271 patients with BC and 381 healthy controls. Two-tag single-nucleotide polymorphisms in the CDK8 gene (rs17083838 and rs7992670) were genotyped using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Background: Cervical cancer (CC) is the most prevalent gynecological tumor among women. Cyclin-dependent kinase 8 (CDK8), which plays a crucial role in cellular transcriptional processes and various signaling pathways, has been identified as a key oncogenic factor in numerous cancers. However, limited data exists on the correlation between CDK8 and CC. The objective of our study was to investigate whether there is an association between CDK8 gene polymorphisms and the development of CC in Han women from Southwest China. Materials and methods: A total of 300 unrelated CC patients and 335 healthy controls from Southwest China were included in the study. The polymerase chain reaction-restriction fragment length polymorphism analysis was used to genotype the two tag single nucleotide polymorphisms (SNPs) of CDK8 gene (rs17083838 and rs7992670), and the relationship between the two tag SNPs and CC incidence was analyzed by SNPstats software. Multifactor dimensionality reduction (MDR) was used to analyze the interaction of multiple polymorphisms of the CDK8 gene. The false-positive report probability (FPRP) was used to verify the effective correlation. Results: The frequency of the A allele of CDK8 rs17083838 in the CC group was significantly higher than that in the control group (25% vs. 12%, p < 0.0001, odds ratio (OR): 0.42, 95% confidence intervals [CI]: 0.31-0.58). The frequency of the A allele at rs7992670 was higher in the CC group than that in the control group (52% vs. 45%, p = 0.026, OR: 0.78, 95% CI: 0.63-0.97). MDR analysis showed that rs17083838 and rs7992670 as the overall model was the best model, the detection accuracy was 0.6157, and the cross-validation consistency was 10/10 (p < 0.0001). In addition, 22 valid FRPR values were verified by using the FPRP detection method. Conclusion: The two SNPs of the CDK8 gene may be associated with the increased risk of CC in the Han population in Southwest China.
Identification of body fluid types at crime scenes is a critical step in forensic science, providing essential context for criminal investigations and the interpretation of evidence. Epigenetic markers, particularly small non-coding RNAs (sncRNAs), have attracted increasing attention in forensic body fluid identification, with various small RNA species demonstrating potential as biomarkers for distinguishing different body fluid types. A novel class of sncRNAs, tRNA-derived small RNAs (tsRNAs), has been detected in various biological samples, yet their potential application in forensic body fluid identification remains unexplored. In this study, we identified differentially expressed tsRNAs in saliva and vaginal secretions, two epithelial cell-derived body fluids. Using stem-loop reverse transcription followed by SYBR Green quantitative polymerase chain reaction (RT-qPCR), we measured tsRNA abundance with U6-snRNA as the reference control. After implementing quality control measures to ensure detection accuracy and adjusting Cq values, we identified 10 tsRNAs that were differentially expressed between saliva and vaginal secretions. To further investigate the potential of tsRNAs in forensic body fluid identification, we extended our analysis to include semen, peripheral blood, and menstrual blood. Our results revealed 18 tsRNAs with significant differential expression patterns across these five body fluids. By integrating quantitative expression data with a logistic regression machine learning model, a panel of 9 tsRNAs achieved 98 % accuracy in leave-one-out cross-validation. While our findings demonstrate the promising potential of tsRNAs as biomarkers for forensic body fluid identification, additional body fluid-specific tsRNAs would likely enhance prediction accuracy while potentially reducing the number of required markers. This preliminary validation establishes tsRNAs as valuable candidates for forensic body fluid identification applications.
Cervical cancer (CC) ranks as the fourth most common cancer and the fourth leading cause of cancer-related deaths among women globally, with declining incidence and mortality rates in recent decades. Previous studies have suggested that the transforming growth factor-beta (TGF-β) activated kinase 1 (TAK1) binding protein 2 (TAB2) can influence the stemness characteristics of squamous CC cells. However, the specific genetic impact of the TAB2 gene on CC remains unclear. This study aimed to evaluate the relationship between TAB2 genetic polymorphisms and susceptibility to CC using a hospital-based retrospective analysis. A total of 306 CC patients and 309 healthy controls were included in this study. Genetic analysis involved genotyping of subjects using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. 目的 回顾性分析2012~2021 年我院血培养检出非伤寒沙门菌(NTS)儿童患者的临床资料和耐药性,为临床早期诊疗提供依据.方法 对我院2012 年1 月至2021 年12 月血培养检出NTS患儿临床资料进行回顾性分析,使用VITEK MS质谱细菌鉴定仪、血清凝集试验和VITEK2 COMPACT全自动药敏分析系统对NTS进行鉴定和药敏试验.结果 共25 例儿童患者血液中分离出了NTS,主要为6 岁以下儿童80%(20/25),主要检出月份为7~9 月(60.0%).患者主要临床表现为发热 25 例(100.0%),腹泻 9 例(36.0%),腹痛 3 例(12.0%),血便 4 例(16.0%),呕吐 4 例(16.0%).实验室检查示白细胞升高14 例(56.0%),CRP升高22 例(88.0%),PCT升高(100.0%,10/10).NTS血清型以D群和B群为主,分别为15 株(60.0%)和 6 株(24.0%).药敏结果显示对头孢唑林耐药率 100%,氨苄西林耐药率60%,头孢曲松、头孢他啶和头孢吡肟敏感率均>95%,对亚胺培南和美罗培南均100%敏感.结论 NTS菌血症多发生于6 岁以下儿童,多发生于夏季,血清型以D群和B群为主,临床表现多样,以发热为主,对三、四代头孢和碳青霉烯类敏感率高,临床医生应根据实验室药敏结果来选择适合的抗生素.
Aim: Ovarian cancer (OC) is a gynecological malignancy with a challenging judgment of prognosis due to complicated etiology and high recurrence rate. The oncostatin M receptor (OSMR) from members of the IL-6 receptor family is associated with tumor development. This study aims to explore the correlations between OSMR gene polymorphisms (rs2278329 [G/A, missense, Asp553Asn], rs2292016 [G/T, promoter, -100G/T]) and OC. Methods: This study enrolled 160 OC patients and 397 healthy controls. Genotypes of two single-nucleotide polymorphisms were distinguished using TaqMan SNP Genotyping Assay, and statistical analysis was performed using SPSS software. Results: A significantly decreased overall survival rate was found in serous OC patients carrying rs2278329 GA/AA genotypes. Meanwhile, TT genotype carriers of rs2292016 had an improved relapse rate, and the GT genotype showed a definitive correlation with a lower relapse rate. Conclusion: OSMR gene polymorphisms may be related to recurrence and overall survival of serous OC patients.
ObjectiveTo conduct preliminary investigation into the correlation between transforming growth factor beta-activated protein kinase 1-binding protein 2 ( TAB2) gene and the incidence of cryptorchidism in Han Chinese population in Southwest China.MethodsA total of 259 patients with cryptorchidism and 355 healthy controls from Southwest China were enrolled for the study. Polymerase chain reaction-restriction fragment length polymorphism method was used to analyze the genotype of the 3 tag single nucleotide polymorphisms (SNPs) of TAB2 gene, i.e., rs237028, rs521845 and rs652921. The Chi-square test was used to analyze the relationship between the genotype frequency of the three tag SNPs and the incidence of cryptorchidism.ResultsThe distribution of the 3 tag SNPs' alleles and genotypes were in agreement with the Hardy-Weinberg equilibrium, and the genotype results of polymerase chain reaction-restriction fragment length polymorphism assay were consistent with those of Sanger sequencing. The frequency of the G allele at TAB 2 rs237028 was significantly higher in the cryptorchidism group than that in the control group (30.9% vs. 25.6%, P=0.04, OR=1.31, 95% CI: 1.01-1.70). In the dominant model, the risk of cryptorchidism was significantly higher in AG/GG genotype carriers ( P=0.006, OR=1.57, 95% CI: 1.14-2.17). In the cryptorchidism group, the TC/CC genotype frequency of the rs652921 locus were significantly higher than that of the control group (75.3% vs. 67.0%, P=0.03, OR=1.50, 95% CI: 1.05-2.14). Correlation between rs521845 and susceptibility to cryptorchidism was not observed in the Han Chinese population.ConclusionThe AG/GG genotype of rs237028 locus and the TC/CC genotype of rs652921 locus of the TAB2 gene may be associated with increased risks of cryptorchidism in Han Chinese population in southwest China.
成像式光电容积描记法(imaging Photoplethysmography,iPPG)是一种利用视频信号实现非接触式生理参数检测的方法,但该方法的性能易受外界条件影响,尤其对环境光变化及受试者头部运动等噪声敏感.为了消除噪声影响,根据噪声信号和iPPG信号的时频特性,提出了一种基于变分模态分解(Variational Mode Decomposition,VMD)的相关能量熵阈值自适应去噪与方差表征序列(Variance Characterization Series,VCS)的抗干扰非接触式生理参数信号获取方法.利用分解算法将由人脸视频中获取到的血容量脉冲信号分解为频率由高频到低频且具有一定带宽的模态分量.分别计算各个模态分量子区间的噪声能量熵从而获得该分量的阈值,再经阈值处理得到重构的去噪信号.利用VCS方法检测重构后的信号的质量从而自适应地选择信号分析方法,实现心率和呼吸率的生理参数的精确估计.在公开数据集和自采数据集上进行验证实验,实验结果表明,提出的方法对环境光与头部运动干扰有着较强的稳健性,相比目前已有的方法能够得到更精确的心率与呼吸率估计值.
Interleukin-32 (IL-32) as a pro-inflammatory cytokine participates in the progression of inflammation and cancer. Ovarian cancer (OC) accounts for a considerable mortality rate, but research on IL-32 and OC is almost nil. Our study aims to explore the association between IL-32 and the progression as well as prognosis of OC initially. This hospital-based case-control study enrolled 147 OC patients and 337 healthy controls, and we used the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method to distribute the genotypes. The results showed that the homozygous genotype (TT) of rs28372698 SNP was significantly higher in patients compared to controls (12.9% vs. 6.2%, P = 0.018, OR (95% CI) = 2.23 (1.16-4.29)). This revealed that TT genotype might be a risk factor in OC progression. This present study indicates that IL-32 gene polymorphism relates to an increased OC susceptibility, and IL-32 may be a marker for OC progression.
目的 了解2011-2017年该院儿童患者分离出金黄色葡萄球菌的分布情况及耐药变迁,为临床治疗提供依据.方法 回顾性分析该院2011年1月至2017年12月从儿童患者送检标本中分离出的金黄色葡萄球菌的分布及耐药性,使用VITEK2 Compact全自动细菌鉴定药敏仪对所分离出的金黄色葡萄球菌进行鉴定药敏试验.结果 总共分离出2 016株金黄色葡萄球菌,患儿年龄段主要集中在小于1个月和1个月至小于1岁,分别占27.8%和48.6%,2 016株金黄色葡萄球菌分布来源前5位的是门诊、新生儿科、心血管儿科、消化儿科及儿科重症监护病房,分别占26.3%、19.8%、16.7%、13.9%及13.2%.菌株主要分离自痰液标本(78.6%),其次为全血、咽拭子、脐部分泌物和脓液.2 016株金黄色葡萄球菌中检出耐甲氧西林金黄色葡萄球菌(MRSA)518株,检出率为25.7%.耐药率较高的抗菌药物包括青霉素(94.7%)、红霉素(63.0%)、克林霉素(61.6%),对其他的药物则敏感性较高,该2 016株金黄色葡萄球菌中未检出对万古霉素、替加环素和利奈唑胺耐药的菌株.其中MRSA对青霉素、克林霉素、利福平、四环素、红霉素的耐药率高于甲氧西林敏感金黄色葡萄球菌(MSSA),对庆大霉素和复方磺胺甲嗯唑的耐药率低于MSSA.结论 儿童患者分离出的金黄色葡萄球菌以MSSA为主,总体耐药率较低,但金黄色葡萄球菌分离率和MRSA检出率有上升趋势,应予以重视.
临床上常采用肾动态图联合Gates分析法来获得肾小球滤过率(GFR)并用于肾功能评价,但该方法需要手动勾画肾脏及背景感兴趣区(ROI)进行定量计算,其结果易受到图像质量、医生经验等因素的影响,且耗时耗力.为了快速且客观准确地获取GFR值,本研究提出了先验信息约束的正则化水平集肾脏感兴趣区自动提取方法.该方法以正则化水平集算法为基础,融入肾动态图先验信息进行水平集演化约束,实现肾脏ROI自动提取.由临床数据验证,该自动方法能够准确且稳定地提取肾脏及背景ROI,实现准确的GFR计算,尤其有利于肾功能严重受损时的GFR值估计.
Objective To analyze the medical expenses of inpatients and its influencing factors in 2014-2016 with acute appendicitis taken as an example, and to put forward policy suggestions for the public hospital reform and the cost control optimization. Methods A total of 4 hospitals were chosen from Zhuhai of Guangdong and Zunyi of Guizhou. 1 114 cases of acute appendicitis were selected to analyze the medical expenses. The influencing factors of hospitalization expenses were analyzed by grey correlation analysis. Results The effect of the cost control of surgical patients was better than non-surgical patients. The total cost of consumables increased obviously. The proportion of non-surgical patients′ drug expenses were 39. 5% , 41. 2% and 41. 7% in 2014 -2016 respectively. The total hospitalization expenses were greatly affected by the treatment expense, diagnostic expense, drug expense, comprehensive medical service expense. The level of medical institutions, age and operation had significant impacts on medical expenses. Conclusions The hospitalization expenses of inpatients with acute appendicitis are influenced by many factors. In order to effectively control the irrational growth of medical expenses, the government is recommended to further strengthen the control of consumables, strengthen the fine management of hospitals and targeted cost control, improve the mechanism of price compensation and deepen the medical insurance payment methods reform.
The model-free adaptive algorithm is a data-driven method, which does not require an accurate model of the system, has small calculation cost and is easy to implement. The existing model-free adaptive control algorithm often adopts the cut-and-trial method or uses a fixed constant when selecting the penalty factor, which is difficult to achieve a satisfactory control performance. To solve this problem, this paper proposes a method to online optimize the penalty factor of the control law and the pseudo partial derivative. By using the idea of iterative optimization in the steepest descent method, the penalty factor is optimized, the convergence speed is increased significantly, and better system performance parameters are obtained. Hence, the optimal performance of the system can be achieved. On this basis, the stability of the closed-loop system is strictly proved. Finally, Matlab simulation results show that the proposed method has better control quality and stronger anti-disturbance abilities than the existing model-free adaptive control method.
In gas metal arc welding,it is difficult to establish an accurate model of arc length nonlinear systems.Moreover,the system has various uncertainties and is vulnerable to the problem of internal and external disturbance.In this paper,an improved model-free adaptive control method was proposed to address the above issues.First of all,a dynamic linear generalized model with measurable and unmeasurable disturbances and un-certain factors was designed.A parameter estimation algorithm was used to estimate pseudo partial derivative online.Then,the disturbance observ-er was introduced to compensate the internal and external disturbances and uncertainties in real time. Furthermore,the control law of arc length system was designed by constructing a criterion function.On this basis,a model-free adaptive control scheme was developed.Finally,the robust sta-bility of the method was validated.The simulation results showed that the method had better tracking performance and faster response speed than the existing model-free adaptive control schemes.It also had a stronger robustness when the unmeasured disturbances and uncertain factors are en-countered in the welding process,which ensuring the stability of the arc length in welding process and the better control of quality.
Considering the complex characteristics of quad-rotor aerial vehicles,such as multiple variables,strong coupling and the constraints,we proposed a controlling method based on explicit model prediction.According to the dynamic characteristics of the quad-rotor flight system,a nine-state space model was built,which was decoupled into two subsystems.Then,the explicit optimal controlling law of the quad-rotor flight system is calculated offline by multi-parameter quadratic programming,and the controlled quantity corresponding to the current state was searched online.Numerical simulation and experiment results show that the proposed method has the advantages of short adjusting time,small overshoot and certain antidisturbance ability,as well as meeting the constraint requirements,and the stability control of the attitude is successfully achieved.
Objective: To analyze the clinical characteristics and perinatal outcomes of listeriosis during pregnancy. Methods: From July 2010 to April 2017, 70 131 women delivered in West China Second University Hospital. Nineteen cases were confirmed as listeriosis. The clinical symptoms, laboratory results, pathogens, placenta pathology and perinatal outcomes were analyzed retrospectively. Results: The median age of the 19 cases was 29.7 (19.0-42.0) years old. The median time before diagnosis was 4.8(0.5-19.0) days. The main clinical symptoms at first visits were high fever (17/19), increased white blood cells (18/19), abdominal pain (12/19). Listeria was found in samples of mother's blood (11/19), vaginal secretions (15/19), placenta (1/19), neonatal blood (4/19), neonatal phlegm (5/19) and neonatal ear secretions (1/19), respectively. Inflammation of placenta was identified in all 19 cases. Among the 19 cases, 1 was grade Ⅰ chorioamnionitis, 4 was grade Ⅱ, 5 was grade Ⅲ and 9 was grade Ⅵ. Only 4 newborn survived after therapy, and others suffered perinatal death, including 8 cases of intrauterine death, 3 cases of miscarriage and 6 cases of treatment failure. Conclusions: Listeriosis has characteristics of acute onset, quick development and high morbidity during pregnancy. The empiric use of antibiotics might not cover listeria. The understanding of listeriosis should be improved.
The direct torque control can improve the motor speed control performance,because it can control torque instantly.This paper presents a novel flux linkage self-control direct torque control base on Extended Kalman Filter algorithm,this method overcomes some defects of traditional direct torque control,such as mechanical sensors may decrease the robustness of system,flux linkage is calculated difficultly because of the variable voltage of turn-off phase,and the torque is difficult to obtain.This method has no need of flux linkage observer,uses line voltage and phase current as input,estimates the real-time torque and rotation speed by Extend Kalman Filter algorithm.The simulation based on MATLAB shows that the proposed scheme estimates rotation speed and rotation angle precisely,and obtains the torque without flux linkage calculation,in the situation of without mechanical sensors,achieves the goal of the direct torque control speed regulation system of brushless DC motor.The control optimization is verified by simulation.

In the traditional method of regularized super-resolution,the parameter of regularization is,in the rebuilding process, a constant value,which can not adjust itself automatically according to local image smoothing texture features.The fixed para-meter causes the edge and texture features of the reconstructed image can not be reserved well.A super-resolution algorithm based on adaptive weighted L1 norm total variation regularization and ADMM was proposed.The proposed method made the regularization weight automatically according to local structure of the target image information,and the super-resolution was transformed to an image denoising problem using the fast decoupled method.The optimal solution was got through the ADMM and fast Fourier transform.The experimental and visual results verify the superiority of the proposed method over the traditional method on preserving the texture features of the reconstructed image and improving peak signal-to-noise ratio (PSNR)and struc-tural similarity (SSIM).
目的调查新生儿科无乳链球菌的分离情况,探讨耐药情况及临床特点。方法对送检的细菌培养标本进行无乳链球菌的培养,对分离出的无乳链球菌进行鉴定药敏试验;对分离出无乳链球菌的患儿的临床资料和治疗结果进行回顾性分析。结果共从新生儿科16例患儿的临床标本中离出20株无乳链球菌,对青霉素、氨苄西林、头孢噻肟及万古霉素等均敏感,对四环素耐药率90.0%,对克林霉素耐药率70.0%;16例患儿中临床表现肺炎12例(其中早发感染8例),败血症8例,发热5例,颅内感染3例(均为晚发型感染)。结论新生儿无乳链球菌早发感染以呼吸道表现为主,晚发感染常合并颅内感染,治疗首选青霉素、头孢类抗生素,应加强对孕产妇的无乳链球菌筛查及预防性抗生素使用。
