Cisplatin (CP) is a platinum-based anticancer drug used to treat many different solid tumors. Although CP has strong anticancer properties, its clinical use is limited due to side effects such as ototoxicity, neurotoxicity, myelosuppression and nephrotoxicity. Taxifolin (Tax) is reported to exhibit various possess effects such as anti-inflammatory, antioxidant, antimicrobial, antiviral and anticancer. In this study, we aimed to investigate the possible effects of Tax on CP-induced nephrotoxicity. This study consisted of Control (C), Taxifolin (Tax), Cisplatin (CP) and Cisplatin + Taxifolin (CP + Tax) groups, and there were 6 rats in each group. CP was administered to rats intraperitoneally (i.p.) in a single dose of 7 mg/kg, and Tax was administered orally at a dose of 50 mg/kg for 7 consecutive days. In this case report, primary pulmonary mycobacteriosis in a domestic cat was assessed using histopathological and immunohistochemical methods. The cat was admitted to a private veterinary clinic and presented with hyporexia, lethargy, and dyspnea with an abdominal component. For diagnostic purposes, thorax radiography was performed. Radiographic lung imaging showed the presence of opaque foci that were widely distributed throughout the lung. The cat’s condition gradually deteriorated, and the cat died 2 hours after being brought to the clinic. Following owner consent, gross post-mortem examination was performed immediately after death at the same clinic. White foci measuring 2–10 mm in diameter covered the surface of the lungs and were present throughout the parenchyma, mostly multifocally and occasionally coalescing. No other macroscopical pathological findings were observed in other organs or on the skin. Some of the lung tissue with macroscopic lesions underwent further histopathological and immunohistochemical examination. Multifocal to coalescing areas of caseous necrosis (without a fibrous capsule) with, karyorrhectic nuclei, and numerous macrophages were observed. Ziehl-Neelsen staining revealed many acid-fast bacteria. Immunohistochemical staining revealed positive immunostaining for Mycobacterium spp. This case study highlights the importance of considering zoonotic risks in cats diagnosed with primary pulmonary mycobacteriosis. Primarna pljučna mikobakterioza pri mački: imunohistokemična in histopatološka ocena Izvleček: V tem poročilu o primeru je bila ocenjena primarna pljučna mikobakterioza pri domači mački z uporabo histopatoloških in imunohistokemičnih metod. Mačka je bila sprejeta v zasebno veterinarsko kliniko zaradi simptomov hiporeksije, letargije in dispneje z abdominalno komponento. Za diagnostične namene je bila opravljena rentgenska preiskava prsnega koša, ki je pokazala prisotnost motnih žarišč, široko razporejenih po pljučih. Stanje mačke se je postopoma slabšalo, umrla je dve uri po prihodu v kliniko. Po privolitvi lastnika je bila takoj po smrti opravljena raztelesba. Bela žarišča, premera od 2 do 10 mm, so prekrivala površino pljuč. Prisotna so bila po celotnem parenhimu večinoma multifokalno in mestoma zlivajoče. V drugih organih in na koži ni bilo opaznih drugih makroskopskih patoloških sprememb. Nekatere dele pljučnega tkiva z makroskopskimi spremembami smo dodatno histopatološko in imunohistokemično pregledali. Opazili smo multifokalna do zlivajoča se območja kazeozne nekroze (brez fibrozne kapsule) s karioznimi jedri in številnimi makrofagi. Ziehl-Neelsenovo barvanje je razkrilo številne acidofilne bakterije. This study presents the first report of congenital tuberculosis in an 8 -month -old male British Shorthair cat. The case was examined using histopathological and immunohistochemical methods. The cat was referred to a private veterinary clinic with general respiratory system problems and subsequent deterioration, leading to death. The cat owner granted permission for the cat necropsy and pathological examinations at Department of Pathology, Faculty of Veterinary Medicine, Selcuk University, Konya, Turkiye. During systemic necropsy, white round foci with diameters ranging from 3.00 to 5.00 mm were observed in the lung and spleen. Tissue samples were collected from the lung, spleen, liver, heart, kidney, mediastinal lymph nodes and brain for histopathological examinations. The tissues were subjected to routine histological tissue processing and sections were stained with Hematoxylin and Eosin and Ziehl-Neelsen. Additionally, Mycobacterium spp. antibodies were used for immunohistochemical staining. Microscopic examination revealed exudative tuberculosis lesions, areas of necrosis without a fibrous capsule and karyorrhectic cells only in the lung and spleen. Acid -resistant bacteria observed by ZN staining in the lesioned areas of the lung and spleen were identified as Mycobacterium spp. using immunohistochemical staining. No positive staining was observed in other organs using ZN and immunohistochemical methods. As a result, congenital tuberculosis was diagnosed in a cat for the first time, especially in relation to lesions in the spleen.
White muscle disease (WMD) is also known as Stiff Lamb Disease or Nutritional Muscular Dystrophy. Selenium and/or Vitamin E deficiency constitutes the etiology of the disease. This study aimed to immunohistochemically evaluate local protein expressions of Nuclear factor kappa B (NF-kB), Insulin-like growth factor-1 (IGF-1) and Microtubule-related protein 1A/1B-light chain 3 beta (LC3B) in WMD. The material of the study consisted of 15 WMD, and 6 healthy lamb heart samples. The heart tissues of the autopsied lambs were subjected to routine tissue processing and paraffin blocks were obtained. Then, it was stained with Hematoxylin-Eosin and immunohistochemical methods. Control group lambs had normal macroscopic appearance. Macroscopically, hyaline degeneration and zenker’s necrosis, calcification areas were observed in WMD tissues. In this study, it was aimed to determine Bordetella bronchiseptica, Mycoplasma felis, Staphylococcus aureus and Chlamydia felis, which cause bronchopneumonia in cats, by Real-time PCR and to compare the pathological findings of the identified agents. The material of the study was constituted of paraffin blocks belonging to the lungs, of which 21 bronchopneumonia were detected in microscopic examination (with Hematoxylin Eosin (HE)) from a total of 78 cats samples brought to Selcuk University, Faculty of Veterinary Medicine, Department of Pathology for pathological diagnosis. Histopathologically, polymorphonuclear leukocytes (PMNL) and mononuclear cell infiltration (MCI) in the bronchi and bronchiolar lumens, desquamation in the bronchi and bronchiolar epithelium, PMNL infiltration with oedama in alveolar lumens and desquamated alveolar epithelium, PMNL infiltration in the interstitium, and peribronchi and peribronchiolar MCI, and pleuritis were detected. Real-time PCR analysis revealed Bordetella bronchiseptica in 3 (14.29%) cases, Mycoplasma felis in 3 (14.29%), Staphylococcus aureus in 5 (23.8%), and Chlamydia felis in 5 (23.8%). Morever, Mycoplasma felis and Staphylococcus aureus infection was detected in 1 case, and Staphylococcus aureus and Chlamydia felis mixed infection was observed in 1 case. Our results show that relevant agents can frequently be isolated in cases of feline bronchopneumonia.
In this study, a case of plasmacytoma with widespread metastasis in a nine-year-old male Rottweiler dog was described by histopathological and immunohistochemical methods. Macroscopic examination revealed multifocal white or pink-red colored masses ranging in size from 0,5 to 3 cm in the lung, liver, mediastinal and mesenterial lymph nodes, spleen, heart, pancreas, intestine and kidneys. In microscopic examination, tumor foci consisting mostly of atypical plasma cells were observed in the lung, liver, spleen, pancreas, mediastinal and mesenteric lymph nodes, intestine, and kidneys. Tumor-type giant cells were observed in tumor foci in the lung, mitotic figures in tumor foci in other organs, including the lung, and abundant tumor cells in capillary vessels. In Methyl Green Pyronin staining of lung, liver, spleen, pancreas, mediastinal and mesenterial lymph nodes, intestine and kidney sections, the cytoplasm of plasma cells was observed to be stained pink. Congo Red staining applied to liver and spleen sections showed amyloid deposits stained in brick red color. In addition, in the immunohistochemical staining of lung, liver, spleen, pancreas, mediastinal and mesenterial lymph nodes, intestine and kidney sections with Proliferating Cell Nuclear Antigen (PCNA) antibody, severe immunoreactivity was detected in tumor cells. As a result, due to the presence of rarely reported widespread metastases in the lung, liver, spleen, pancreas, mediastinal and mesenterial lymph nodes, intestine and kidneys of the diagnosed plasmacytoma case, it was deemed appropriate to publish the case in order to contribute to the literature by attracting the attention of veterinarians.
Helicobacter species such as H. heilmannii, H. pylori, H. felis, H. bizzozeronii and H. salomonis have been identified in cats and dogs, and research suggesting that these species may be zoonotic agents and has been studied intensified in recent years. The aim of this study was to reveal the presence, comparing the histopathological findings and Real-time PCR results of H. felis, H. heilmannii, and H. pylori in the stomach and liver tissues taken during the necropsies of owned, stray or shelter dogs. The material of the study consisted of stomach and liver tissues taken from 35 dogs that died for different reasons and were brought for necropsy. DNA copies of H. heilmannii were detected by Real-time PCR in the liver samples of 30 dogs using H. heilmannii-specific primers. In the case of gastric samples, Real-time PCR detected H. heilmannii in 13 cases, H. pylori in 3 cases, both H. heilmannii and H. pylori in 13 cases, and H. felis, H. heilmannii and H. pylori in 3 cases. Microscopically, neutrophil leukocyte infiltration, epithelial degeneration, fibrosis and oedema in the lamina propia, and lymphoplasmacytic cell infiltration were determined in the stomachs. In the Hemotoxylin Eosin staining of the sections, 5 cases and 14 cases in the Warthin–Starry staining were found positive for Helicobacter-like microorganisms. Microscopically, dissociation of the remark cords and hydropic degeneration in hepatocytes, and focal mononuclear cell infiltrations in some sections were determined in the livers. In conclusion, with this study, it was understood that Real-time PCR analyzes are very useful in the diagnosis of H. felis, H. heilmannii, and H. pylori. However, histopathological examinations are necessary to associate the presence of bacteria with the development of the disease.
There are many bacterial factors that cause respiratory diseases in dogs. Bacterial bronchopneumonia is a lung disease caused by bacteria in the lower respiratory tract and lung parenchyma. In this study, it was aimed to determine Mycoplasma spp., Streptococcus spp., Bordetella bronchiseptica, Klebsiella spp., in dogs with bronchopneumonia by Real-time PCR and to compare the histopathological findings determined according to the agents. The material of the study consisted of lung paraffin blocks of 37 dogs that died due to different reasons and were found to have bronchopneumonia in microscopic examination. Microscopically, edema in the alveolar lumens, shedding of the alveolar epithelium, shedding of the bronch and bronchial epithelium and bronchiectasis, mononuclear cell infiltration (MCI) and Polymorphonuclear leukocytes (PMNL) infiltration in the bronchial lumens, peribronchiolar MCI infiltration, Bacterial clusters localized to the bronch epithelium, PMNL infiltration in the interstitium, multifocal necrosis areas, bleeding and pleuritis were observed. Real-time PCR analysis revealed Bordetella bronchiseptica in 18 (48.64%) cases, Mycoplasma spp. in 9 (24.32%) cases, Streptococcus spp. in 10 (27.02%) cases, and Klebsiella spp. in 2 (5.4%) cases. In conclusion, this study showed that the causative agent can be determined in bacterial bronchopneumonias of dogs with Real-time PCR even in tissues without culture opportunity. In addition, this study indicates that polymicrobial lower respiratory tract infections can also be seen in dogs and reveals that more than one bacterial species should be investigated for diagnosis.
Bakır zehirlenmesi, bakırın vücuda alınımı ve atılımı arasındaki dengesizlikten kaynaklanan patolojik bir durumdur. Akut zehirlenme genellikle antihelmintik ilaçlar, mineral karışımları veya aşırı miktarda çözünür bakır tuzlarının uygulanmasından sonra görülür. Sunulan bu çalışmanın materyalini 900 toklu oluşturmaktadır. Hasta hayvanlarda abdominal sancı, ishal, hemoglobinüri, ikterus ve ölüm bulunmaktaydı. Yapılan nekropsiler sonucu deri altı yağ dokusu, serozalar ve organların etrafındaki yağ dokuda şiddetli sarılık belirlendi. Karaciğerin koyu kahve ya da sarımtırak renkte ve kolay parçalandığı görüldü. Safra keseleri dolgun, safra koyu renkte ve granüler yapıdaydı. Böbreklerin oldukça şişkin ve koyu siyah renkte olduğu görüldü. İdrar keseleri hemoglobinüriye bağlı olarak koyu kırmızı renkte idrar içermekteydi. Histopatolojik muayenede hepatositlerde hidropik dejenerasyon, vakuoler dejenerasyon, yağlanma, multifokal nekrozlar ve dissosiasyon, hepatositlerin sitoplazmasında sarı kahve renkte granüllerin yanı sıra hepatosit sitoplazmaları ve safra kanalcıklarında sarımsı-yeşil renkte safra pigmentlerine rastlandı. Portal alanlarda mononükleer hücre infiltrasyonları ile safra kanalı artışı belirlendi. Karaciğer kesitlerine Rodaninle yapılan boyamalarda hepatositlerin sitoplazmalarında kırmızımsı renkte pozitif boyanmalar tespit edildi, Böbreklerde tubul epitellerinde şişkinlik ve vakuoler dejenerasyon, bilurubin pigmenti dikkati çekti. Sunulan bu çalışmada koyunlarda görülen ve yem katkısından kaynaklandığı belirlenen akut bakır zehirlenmesinin klinik, toksikolojik ve patolojik bulgularının ortaya konulması ve son yıllarda oldukça sık karşılaşılan yem katkısı kaynaklı zehirlenme olaylarına dikkat çekilmesi amaçlanmıştır.
Infectious bursal disease (IBD) is a viral disease that causes significant economic losses in young chickens, characterized by lymphoid depletion and inflammation in the bursa Fabricius (BF). The incidence of the disease shows an increasing trend all over the world. Active and passive immunization is very important as well as strict hygiene measures in combating outbreaks. However, the fact that live-attenuated vaccines (mild, intermediate, hot) used for this purpose cause immunosuppression because of bursal damage is seen as an important limitation. In this study, it was aimed to histopathologically investigate the effects of commercial IBD vaccines originating from WF2512 (intermediate plus/hot, orally with drinking water) on BF under routine broiler rearing conditions. For this, BFs of 55 Ross 308 hybrid breed chickens (50 test, 5 controls) from five different broiler farms were used. In addition to standard vaccines, the IBD vaccine was given on day 15, and five samples from each farm were obtained 10 days later (25th day). After the first sampling, the second BF sampling was performed at the age of 38 days. Histopathological bursal lesion score was applied to evaluate the effectiveness of the vaccine. Accordingly, it was determined that the bursal lesion score, which increased slightly to moderately in the first samples, decreased in the second samples (27-61%). This was accepted as an indication that the bursal damage, which increased with IBD vaccine administration, diminishes over time and that histological regeneration was increased.
