Objective:To investigate the effects of 5-bromodeoxyuridine (BrdU) tracing in vivo on the biological status of bone marrow stromal stem cells(BMSCs).Methods:Healthy adult beagle dog bone marrow was extracted,then cultured and indentified by BrdU markers,the cell cycle,apoptosis rate,cell viability were determined in vitro.The stem cells were given implantation of autogenous femoral head defect by BMSCs labeled BrdU in vivo,the other side were given implantation of autogenous bone as the control,the bone mass and molecular markers expression were recorded.Results:The BrdU labeling rate of BMSCs in vitro was 85.2%.The apoptosis rate of BrdU group was 3.62+ 1.33%,the unlabeled group was 3.52+ 1.08%.The cell survival rate of BrdU group and unlabeled group were 96.31 + 1.39% and 95.20±+ 2.10%(P>0.05).Immunohistochemical staining of BrdU labeled BMSCs showed positive BrdU staining,the positive rate was 81.6%.The expression quantity and the intensity of osteocalcin and collagen in the transplantation bone side were higher than those of the control side.The bone mass in the transplantation bone side was 17.46± 2.12%,which was 9.06+ 1.24% in the control (P<0.05).Conclusion:BrdU labeled BMSCs could effectively reflect the biological state of cells in vitro,it showed that the BMSCs transplantation could survive in vivo,it could promote the bone formation and repair of necrotic bone.
Objective To investigate the methods and effects of the establishment and identification of rabbit bone marrow stromal stem cells.Methods 6 New Zealand rabbits of SPF grade were selected and sacrificed,and the bilateral femoral bone were separated and the bone marrow was given to aspiration,the rabbit bone marrow stromal stem cells were separated and purified by adherent culture method,after several passages,the cell surface antigen was detected by flow cytometry.And the BrdU was used to label bone marrow stromal stem cells.And the bone marrow stromal stem cells were injected into the rabbit brain,1 and 3 weeks after the brain for paraffin sections and immunofluorescence staining.Results The purified bone marrow stromal cells could be obtained by adherent culture method.Immunocytochemical staining showed that the CD44+ cell rate in bone marrow stromal stem cells was over 90%,and the nuclei were not stained.The CD90+ cell rate and CD34+ cell rate in cultured bone marrow stromal cells by flow cytometry were 94.5% and 0.24%.BrdU markers showed that bone marrow stromal cells could be synthesized into DNA,and the percentage of labeled cells was above 40%.Conclusion The cultured rabbit bone marrow cells are polygonal,juvenile short spindle,it can obtain purified bone marrow stromal stem cells,BrdU can be used as one of the ideal markers of bone marrow stromal stem cells in vivo.
ObjectiveTo briefly research culture, identification and mark of rabbit bone marrow stroma stem cells.MethodsBone marrow cells of young rabbits were cultured by adhesion method for purification by repeated passage, and cell surface antigen was detected by flow cytometry. Bone marrow stroma stem cells marked by 5-bromodeoxyuridine (BrdU) was injected into rabbit brain for immunofluorescent staining of brain paraffin section after 2 weeks.ResultsBone marrow cells cultured by adhesion method provided purified cells suitable for long-term culture in vitro. CD31 had negative rate as 5.67%, CD34 had negative rate as 4.31%, CD45 had negative rate as 4.42%, CD44 had positive rate as 98.82%. CD71 had positive rate as 99.17%. CD90 had positive rate as 98.23%. CD31, CD34 and CD45 showed negative outcome, and CD44, CD71 and CD90 showed positive outcome. BrdU was suitable to be taken by bone marrow stroma stem cells in DNA synthesis.Conclusion Adhesion method provides bone marrow stroma stem cells with high purification, and BrdU can be used as ideal marker for in vivo tracking bone marrow stroma stem cells.
目的:探讨支气管哮喘患儿血清25羟维生素D3[25-(OH)D3]水平对病情严重程度及临床转归的影响.方法:选取144例支气管哮喘患儿为研究对象,依据病情严重程度分为轻度间歇组、轻度持续组、中度持续组及重度持续组,选取60例健康儿童作为对照组,检测血清25-(OH)D3水平;在治疗4周后应用儿童哮喘控制测试评分系统(C-ACT)对患儿哮喘控制情况给予评价,将所有患儿分为未控制组、部分控制组及完全控制组,比较各组血清25-(OH)D3水平的差异.结果:对照组、轻度间歇组、轻度持续组、中度持续组、重度持续组5组之间血清25-(OH)D3水平存在显著的统计学差异(P<0.05),从对照组到重度持续组血清25-(OH)D3水平逐渐降低(P<0.05);完全控制组、部分控制组及未控制组三组血清25-(OH)D3水平亦存在显著的统计学差异(P<0.05),从完全控制组到未控制组血清25-(OH)D3水平亦呈现逐渐降低趋势(P<0.05);血清25-(OH)D3水平与病情严重程度呈负相关(rs =-0.489,P<0.05),血清25-(OH)D3水平与C-ACT评分亦呈负相关(rs =-0.470,P<0.05);随着血清25-(OH)D3水平下降程度的加重,患儿发生重度持续发作、出现C-ACT评分<19分的风险值(OR)则逐渐增大.结论:血清25-(OH)D3水平与哮喘患儿病情严重程度及病情控制情况密切相关,早期血清维生素D水平对于哮喘患儿病情及近期预后具有一定意义.
目的 探讨轻度胃肠炎伴婴幼儿良性惊厥临床特点及预后.方法 对符合轻度胃肠炎伴婴幼儿良性惊厥诊断的30例患儿的临床资料进行回顾性分析,并随访1年以上.结果 BICE患儿惊厥常发生在病程前3d内,呈全面性强直或强直-阵挛性发作,每次惊厥发作持续时间短,在1次病程中可发作多次,无神经系统阳性体征.绝大多数患儿血常规、脑脊液、CT/MRI、脑电图等无明显异常.BICE患儿经补液处理后惊厥容易控制,预后好.结论 轻度胃肠炎伴良性婴幼儿惊厥为婴幼儿常见惊厥发作疾病,治疗以控制反复发作为原则,预后良好.
目的:探讨胰岛素样生长因子-1 (IGF-1)、转化生长因子-β1 (TGF-β1)水平变化在过敏性紫癜患儿肾损害发病机制中作用.方法:选取HSP患儿30例、HSPN患儿30例和30例健康儿童为对照组,分别采血,用化学发光法检测各组标本IGF-1浓度,采用双抗体一步夹心法酶联免疫吸附试验检测各组TGF-β1浓度;同时用全自动生化仪检测各组患儿血清Cys C、BUN、SCr水平.结果:血IGF-1、TGF-β1的水平HSPN组、HSP组与对照组患儿比较,差异均有统计学意义(P<0.05).血清Cys C水平对照组、HSP组与HSPN组患儿比较,差异有统计学意义(P<0.05).采用直线相关分析HSPN组的血清CysC水平与血IGF-1、TGF-β1水平比较,存在正相关(r=0.759、r=0.802,均P<0.05),血IGF-1与TGF-β1也存在正相关(r=0.850,P<0.05).结论:血IGF-1、TGF-β1共同参与紫癜性肾炎发病,其水平变化与病理损害程度相关.
Objective:To understand the clinical features,imaging features,complications of children with severe mycoplasma pneumonia(MPP) and sum up the treatment.Method: To analyse the 40 cases of children with severe mycoplasma pneumonia .Result: 40 cases of children have fever,cough performance,combined with pleural effusion in 15 cases,12 cases of atelectasis;two cases of pulmonary hemorrhage occurred;21 cases with extrapulmonary complications.Conclusion: The MPP in children is often associated with pleural effusion,atelectasis,the common symptoms of extrapulmonary are cardiac,liver dysfunction and neurological damage.If the anti-inflammatory effect of azithromycin is poor,rifampicin,methylprednisolone,gamma globulin combination therapy are often required.
Objective:To investigate the clinical effect of Dianxianning pian to patients with epilepsy.Method:In this double-blind,randomized clinical trial,69 patients with generalized tonic-clonic seizures were selected from clinic or department of neurology in Inner Mongolia Hospital from April 2007 to April 2008.They had been made a definite diagnosis by Electroencephalogram and had at least two seizures in previous three months.They were treated with Dianxianning pian or placebo for 3 months,based on previous anti-epileptic drug.We recorded and observed clinical symptoms each month.Data was analyzed and safety evaluation was gived by SPSS 13.0 statistic software at last.Result:46 subjects received Dianxianning pian and 23 received placebo.After 3 months therapy.the seizure frequency of experiment group was significantly decreased compared with control group(P0.01).The severity of seizures total scores were significantly decreased compared with control group(P0.01).Therapeutic effect of seizure frequency and therapeutic effect of symptom had significant difference befween the two groups(P0.01).Safety index such as ECG,blood routine,urine routine,blood biochemical change had no statistically difference between the two groups(P0.05).Conclusion:Add-on Dianxianning pian can raise therapeutic effect in patients with epilepsy of generalized t tonic-clonic seizures.And Dianxianning pian treatmentEpileptic seizures is safe and well tolerated.
Objective:To discuss the galanin content changes and clinical significance,of children with seizure in the epileptic.Method:To determine the galanin contents in plasma by enzyme-linked immunosorbent assay(ELISA),It divide three groups,33 cases of epilepsy group、15 cases with status epilepticus group and 30 cases of normal control group.Results: Plasma of Children with epilepsy group were significantly higher than that galanin status epilepticus group and the normal control group(F=205.911,P0.05);Status epilepticus group and normal control group,no significant difference(t=0.475,P0.05).Conclusion: The relationship is closely between galanin and epilepsy,so galanin may play an anticonvulsant role.

Objective:To study the effect of continuous intravenous infusion midazolam in treatment of status epilepticus.Method:40 cases of children with SE were randomly divided into two groups: treatment group 21 cases were treated by midazolam continuous intravenous infusion;control group 19 cases were treated by valium intravenous which is the first-line anti-epileptic agent;to observe the onset time,the effect of the two groups,heart rate,blood pressure before and after the treatment of them.Results: There was no significant different on the onset time between the two groups,the effect of the treatment group was better than the controls(P0.05).Conclusion: It's a safe and effective way to treat SE with continuous intravenous infusion midazolam.

Objective:To find out characteristic of Coxsackie B virus(CBV) from Viral myocarditis in the area of Inner Mongolia.Methods:The ELISA test was used.Results: The total positive rate of CBV 1-6IgM antibodies was 52.5%(72/137),CBVB3 type was as high as 40.3%(29/72),the others were B2、B4、B5 BI、B6 of 26.4%(19/72)、20.8%(15/72)、6.9%(5/72)、2.8%(2/72),2.8%(2/72)respectively.Meanwhile only 13.6%(19/140)CBV IgM positive cases could be detected in the control group.The result showed that the positive rate of CBV 1-6 IgM antibodies in clinically diagnosed myocarditis was significantly higher than in the control group.Conclusions:The CBV may be the main agents causing VMC in the area of Inner Mongolia.
