目的 探究S100B抑制剂Pentamidine对内侧颞叶癫痫(MTLE)不同时期COX-2、IL-1β蛋白表达的影响及其可能的作用机制.方法 160只小鼠随机分为对照组和模型组,模型组小鼠右背海马区给予注射50 nL海人酸-PBS溶液(1μg/μL),对照组小鼠则注射等体积生理盐水.分别于建模前1 d和建模6 h后向各组小鼠尾静脉注射Pentamidine(5mg/kg)或等体积的PBS,每天1次,连续7 d.MTLE成功诱导后12 h(急性期)、7 d(潜伏期)、40 d(慢性期)取小鼠海马组织行后续实验.采用RT-PCR检测小鼠海马组织S100B mRNA表达水平;采用尼氏染色检测小鼠海马组织尼氏小体和神经元丢失;采用Western blot检测小鼠海马组织COX-2和IL-1β蛋白表达.结果 与对照-PBS组比较,急性期、潜伏期和慢性期-PBS组海马组织中S100B mRNA表达不同程度增加(P<0.05);与各PBS组比较,不同时期Pentamidine组S100B mRNA的表达下降(P<0.05).尼氏染色显示,与对照-PBS组比较,急性期、潜伏期和慢性期-PBS组神经元结构不同程度受损,尼氏小体数量下降(P<0.05);与各PBS组比较,不同时期Pentamidine组的神经元受损程度减轻,尼氏小体数量明显增加(P<0.05).Western blot显示,与对照-PBS组比较,急性期、潜伏期和慢性期-PBS组海马组织COX-2、IL-1β和phospho-NF-κB的蛋白表达增加(P<0.05);与各PBS-组比较,不同时期Pentamidine组COX-2、IL-1β和phospho-NF-κB蛋白表达下调(P<0.05).结论 S100B抑制剂Pentamidine可通过下调COX-2和IL-1β的表达降低MTLE小鼠海马炎症反应和神经损伤,为MTLE的临床治疗提供重要的参考价值.
目的 探究内侧颞叶癫痫(MTLE)不同时期S100B、RAGE和COX-2的动态表达变化及其临床意义.方法 120只SPF级雄性C57BL/6小鼠随机分为对照组和模型组,模型组小鼠右背海马注射50nl海人酸(KA,1μg·μL-1),对照组小鼠则注射等体积的PBS溶液.分别于癫痫诱发成功后6h(急性期)、7d(潜伏期)、40d(慢性期)取各组鼠脑行后续实验.采用免疫组化检测个各组小鼠不同时期海马组织中RAGE和S100B的表达水平;采用Western blot检测各组小鼠不同时期海马组织中COX-2的表达水平.结果 对照组小鼠未见任何癫痫发作表现.模型组小鼠KA注射1h后出现面部肌肉抽搐并逐渐加重至全身痉挛,脑电图(EEG)监测到明显的癫痫样放电;7d后癫痫发作基本消失,进入潜伏期;30d后小鼠开始出现慢性、反复性癫痫发作.免疫组化显示,急性期小鼠海马组织中RAGE和S100B蛋白表达较对照组显著增加(P<0.05);与急性期比较,潜伏期和慢性期小鼠RAGE和S100B表达逐渐降低,差异有统计学意义(P<0.05).Western blot结果显示,急性期小鼠海马组织中COX-2的蛋白表达较对照组显著增加(P<0.05);潜伏期和慢性期小鼠海马组织中COX-2的表达较急性期显著下降,差异有统计学意义(P<0.05).结论 RAGE、S100B和COX-2的表达在MTLE小鼠急性期、慢性期和潜伏期显著增加,有望为MTLE的临床治疗提供新的靶点.
目的 探讨蒙古族、汉族人群多发性硬化(MS)与人类白细胞抗原(HLA) DQB1、DPB1等位基因多态性的相关性.方法 选取2013年3月-2017年3月在内蒙古自治区人民医院及合作医院就诊的蒙古族MS病人26例(蒙古族MS组)与汉族MS病人43例(汉族MS组),另选43名健康体检者为对照组.采用高分辨分型方法SBT法进行HLA-DQB1、HLA-DPB1等住基因检测,并比较3组等位基因的多态性.结果 检测到13个DQB1等位基因片段,汉族MS组HLA-DQB1* 0302基因频率明显低于对照组(P<0.05),HLA-DQB1* 0602基因频率明显高于对照组(P<0.05),但蒙古族MS组与汉族MS组比较差异无统计学意义(P>0.05).检测到12个DPB1等位基因片段,MS组HLA-DPB1* 0501基因频率明显高于对照组(P<0.05),但蒙古族MS组与汉族MS组比较差异无统计学意义(P>0.05).结论 蒙古族、汉族人群多发性硬化与人类白细胞抗原HLA-DQB1* 0302、HLA-DQB1* 0602和HLA-DPB1* 0501有关.
目的 探讨阿司匹林联合氯吡格雷治疗对青年急性脑梗死患者神经功能及血小板及凝血功能的影响.方法 将青年急性脑梗死患者69例随机分为对照组和观察组,对照组行阿司匹林(100 mg/d)单药治疗,观察组行阿司匹林(100 mg/d)与氯吡格雷(75mg/d)联合治疗,均治疗2周.于治疗前、治疗2周后采用美国国立卫生研究院卒中量表(NIHSS)对两组患者的神经功能缺损程度进行评分并比较,同时比较血小板及凝血功能指标的影响.结果 治疗2周后,两组NIHSS评分显著低于同组治疗前(P<0.01),且观察组的NIHSS评分显著低于对照组(P<0.01).治疗2周后,观察组PA降低显著(P<0.01),PLT、PT、APTT、FIB无明显差异(P>0.05).结论 阿司匹林联合氯吡格雷治疗对青年急性脑梗死治疗效果显著,可有效改善患者神经功能,并对凝血功能及血小板无明显变化,出血风险小.
Objective To investigate the influence of icari n (ICA)on the expression of cyclooxygramse 2 (COX 2)in inflammational rat model hippocampus with Alzheimer’s disease(AD)induced by lipopolysaccharides(LPS). Methods Thirty six rats were al oca-ted into 4 groups:control group,model group,low dose ICA group and high dose ICA group.The rat models of AD were induced by being injected LPS into the lateral cerebral ventricle. After two weeks of intragastric administration,brain tissue of rats was removed. The expressions of COX 2 protein and mRNA in hippocampus of the rats were detected by immunohistochemistry(SP method)and SYBR GREEN fluorescence quantitative RT PCR. Results The expressions of COX 2 protein and mRNA in high dose ICA group were significantly lower than those in model group (P<0.01). Conclusion The high dose ICA can reduce the expression of COX 2 in inflammational rat model hippocampus of AD and can inhibit the inflammatory reaction of nerve cel s injury.
目的 用脂多糖(lipopolysaccharide,LPS)建立阿尔茨海默病(Alzheimer's disease,AD)炎症模型大鼠,观察淫羊藿苷(Icariin ICA)对该模型大鼠脑海马组织内炎症因子:如GFAPmRNA、TNF-amRNA表达的影响并探讨其可能的作用机制.方法 经八臂迷宫试验后45只雄性Wistar大鼠随机分5组,分别为模型组、对照组、ICA低剂量组、ICA高剂量组、双氯芬酸组,采用侧脑室注射LPS制作实验性AD炎症模型,制模后对照组和模型组给予灌胃蒸馏水,双氯芬酸组给予双氯芬酸钠,高、低剂量组给予相应剂量的ICA混悬液后对上述动物进行为期15 d的八臂迷宫测验并记录其数据.迷宫测验完成后断头取脑海马组织,提取RNA,应用SYBR GREEN工荧光定量PCR检测实验性AD模型大鼠大脑海马组织内GFAPmRNA、TNF-amRNA表达.用SPSS统计学软件进行统计学分析.结果 SYBR GREEN工荧光定量PCR检测显示:与模型组比较,高剂量组和双氯芬酸组大鼠海马内GFAPmRNA、TNF-amRNA的表达明显减低(P=0.000).结论 高剂量ICA显著降低GFAPmRNA、TNF-amRNA表达.低剂量ICA未产生明显影响,其作用机制可能是通过抑制炎症实现的.
目的 探究多发性硬化患者外周血中IL-17A、IL-23、TGF-β1的表达.方法 选择2015年1月-2016年6月之间在我院就诊的复发-缓解型多发性硬化患者31例以及健康体检患者30例作为本次实验的研究对象.记录多发性硬化患者复发期、缓解期以及健康体检患者外周血IL-17A、IL-23、TGF-β1的水平情况.结果 MS患者外周血IL-17A、IL-23、TGF-β1水平明显高于健康组,MS复发期明显高于缓解期,差异显著(P<0.05).结论 外周血IL-17A、IL-23、TGF-β1测定对于多发性硬化的诊断以及病情的发展有重要的提示作用.
目的:用脂多糖(lipopolysaccharide,LPS)建立阿尔茨海默病(Alzheimer's disease,AD)炎症模型大鼠,观察淫羊藿苷 (Icariin ICA)对该模型大鼠学习记忆及超微结构的影响.方法:经八臂迷宫训练成功的45只雄性Wistar大鼠随机分为对照组、模型组、淫羊藿苷30mg/kg组、淫羊藿苷120mg/kg组、双氯芬酸组10mg/kg组,采用侧脑室注射LPS(10μl)制作实验性AD炎症模型,并八臂迷宫测验并记录其数据.迷宫测验完成后断头取脑海马组织.应用电镜观察大鼠海马组织CA1区神经细胞超微结构变化,用SPSS统计学软件进行统计学分析.结果:(1)侧脑室注射LPS(10μl)后对大鼠一般情况观察出现了行为异常.(2)八臂迷宫测试系统分析结果:制模后与对照组相比,其它各组大鼠总记忆错误(TE)、参考记忆错误(RME)、工作记忆错误(WME)明显增多,用淫羊藿苷治疗后与模型组比较,淫羊藿苷120mg/kg组和双氯芬酸组10mg/kg组大鼠的TE、RME、WME均明显减少,淫羊藿苷30mg/kg组错误选择次数无统计学意义.超微结构观察示:对照组、海马组织线粒体比较规则,淫羊藿苷120mg/kg组、双氯芬酸组线粒体轻度受损,模型组海马神经细胞核固缩,核周隙扩张,甚至核膜结构不清,核形态极不规则.结论:侧脑室注射LPS(10μl)能成功复制AD炎症动物模型;淫羊藿苷120mg/kg组显著改善AD模型大鼠的学习记忆能力,淫羊藿苷对炎症模型大鼠海马超微结构有保护作用.
目的 观察淫羊藿苷(ICA)对脂多糖诱导的阿尔茨海默病炎症模型大鼠记忆能力的影响,并探讨其作用机制.方法 大鼠八臂迷宫训练成功后,36只Wistar大鼠随机分为对照组、模型组、ICA低剂量组、ICA高剂量组,侧脑室注射脂多糖建立AD炎症模型,八臂迷宫实验检测大鼠的空间记忆障碍,免疫组化法(SP法)检测大鼠海马内胶质纤维酸性蛋白(GFAP)、肿瘤坏死因子α(TNF-α)、白细胞介素6 (IL-6)的表达.结果 治疗结束后,与模型组相比,高剂量组大鼠的TE 、RME、WME均明显减少(P<0.01),低剂量组大鼠的错误选择次数无统计学意义(P>0.05).与模型组相比,大鼠海马GFAP、TNF-α、IL-6的表达在高剂量组明显降低(P<0.01),低剂量组无统计学意义(P>0.05).结论 淫羊藿苷能显著改善AD模型大鼠的学习记忆能力,其作用机制可能是通过抑制胶质细胞的增生活化、抑制炎症对神经细胞的损伤实现的.
Objective:To investigate the Therapeutic effect of health education on patients with hemiplegia.Methods:108 patients with hemiplegia were divided into teatment group and control group.the patient in both groups received the similar rehabilitation.and the patient in the treatment group also accepted the health education.they were assessed with the Fugl-Meyer assessment(FMA)and modified Barthel index(MBI) and Record length of stay.Result: The scores of FMA and MBI in the both groupswere improved after treatment(P<0.001),and the result of the treatment group was better than that of the control group(P<0.05).hospital stay of Treatment group was significantly shorter than control group.Conclusion: Health education can improve the recovery of motor function and activity of daily living in patients with hemiplegia by knowing more about the disease.Health education establishment of a good doctor-patient relationship,and improve the treatment results 、improving the quality of life.
目的:探讨15例多发性硬化MS病人周围神经电生理检查异常改变,从中证实MS存在周围神经的阶段性脱髓鞘病损。方法:本组从60例MS中经肌电图检查,发现15例MS病人存在周围神经的异常电生理改变。结果:肌电图显示运动传导速度(MCV)减慢的10例,感觉传导速度减慢的8例,波形离散的3例,3例测定了F波,潜伏期均延长,15例均有不同程度的纤颤电位。结论:MS患者不但出现CNS的脱髓鞘病损,而且部分病人同时存在PNS的脱髓鞘病损。
Objective:To investigate the clinical effect of Dianxianning pian to patients with epilepsy.Method:In this double-blind,randomized clinical trial,69 patients with generalized tonic-clonic seizures were selected from clinic or department of neurology in Inner Mongolia Hospital from April 2007 to April 2008.They had been made a definite diagnosis by Electroencephalogram and had at least two seizures in previous three months.They were treated with Dianxianning pian or placebo for 3 months,based on previous anti-epileptic drug.We recorded and observed clinical symptoms each month.Data was analyzed and safety evaluation was gived by SPSS 13.0 statistic software at last.Result:46 subjects received Dianxianning pian and 23 received placebo.After 3 months therapy.the seizure frequency of experiment group was significantly decreased compared with control group(P0.01).The severity of seizures total scores were significantly decreased compared with control group(P0.01).Therapeutic effect of seizure frequency and therapeutic effect of symptom had significant difference befween the two groups(P0.01).Safety index such as ECG,blood routine,urine routine,blood biochemical change had no statistically difference between the two groups(P0.05).Conclusion:Add-on Dianxianning pian can raise therapeutic effect in patients with epilepsy of generalized t tonic-clonic seizures.And Dianxianning pian treatmentEpileptic seizures is safe and well tolerated.
Objective:To establish inflammational rat model with Alzheimer's disease(AD)by injecting lipopolysaccharides(LPS) into the lateral cerebral ventricle.Methods:18 rats were allocated into 2 groups:model group and control group.After successfully trained in the 8-arm maze,the model group rats were injected LPS into the lateral cerebral ventricle,and the control group rats were injected nomal sodium.The spatial memory dysfunction of the rats were detected by 8-arm maze test after two weeks.The changes of cellmorphology were detected by HE stain.The expression of glial fibrillary acidic protein(GFAP)ang tumor necrosis factor alpha(TNF-α)in hippocampus of the rats was measured with immunohistochemical stain(SP method).Results:The rats' memory dysfunction show up after being injected LPS and TE/RME/WME were significantly increased in the model group compared with those in the control group(P0.01).Under the light microscope,hippocampus exhibited noticeable neuron loss and damage in model group.Expressions of GFAP、TNF-α in model group were significantly higher than those in control grroup(P0.01).Conclusion:Inflammational rat model with Alzheimer's disease could be induced by being injected LPS into the lateral cerebral ventricle.
