A suitable medium,95% wheat bran +5% maize flour + 5% soybean powder,was chosen for mass production of Beauveria bassiana for solid-state fermentation,and the spore count of primary product was up to 244.7×10~(8) conidia per gram.The optimal conditions for solid-state fermentation in a 25m~(3) solid fermentator were as followings.Initial water content of the medium was 60%~65%.The best aeration ratio was: 0 in first 12h,1:2 in 12~48h and 1∶1 after 48h.In order to emit heat released by substantial growth of mycelium,it was necessary to control the temperature from 12h to 48±8h.As to the desiccating process,the optimal condition was with dried air at normal temperature to keep conidia viable.
In the present paper, the high quality liquid seed and technique of semi-solid cultivation for producing Beauveria bassiana was studied by using flat plate and plastic mesh plate as carriers. The results showed that the high quality liquid seed could be obtained after cultivating the thalli for 36 h in 25℃, and suitable technique for the thalli to grow included 10% amount inoculation, about 0.25 cm in thickness of medium, about 90%-95% of environment humidity and 9 days for spores^cultivation. As a result, the output of spores was 1.2×10~(11)-2.0×10~(11) conidia·g~(-1) (water content 9.05%-11.36%) and the germination rate was more than 98%.
The resource of entomogenous fungi from Urochela distincta Distant in Huangshan Scenic Area was investigated.They were Beauveria bassiana (Bals.) Vuill,Paecilomyces farinosus (Holm.ex Gray.) Brown Smith,Verticillum lecanii (Zimmermann) Viegas,Cladosporium cladosporioides(Fres.)de Vries and Acremonium sp..Among them,Beauveria bassiana was the dominant species,accounting for 92% of the total.Bioassays of 10 Beauveria bassiana strains against Urochela distincta adults were made,the results showed that the virulence of Bb357 was the highest,and could be used in biocontrol.
Wheat bran medium was shosen to make non-woven fabric bands impregnated with B.bassiana according to the amount of the spores produced on the bands and the economic factor. The optimal conditions for making bads were: inoculationat normal temperature with the ration of 1:3.5 and suitable inoculation temperature at 25℃ with an optimal humidity of 90%-95% for 6-7 days. The amount of spore produced on the bands could be 1.2×10~(8)/cm~(2)in average.
The control efficiency of ultra low volume spraying technique of oil solution of B.bassiana and deltamethrin against Dendrolimus punctatus was studied.The mortality ranged from 28 1% to 94 4% as the dosages of B.bassiana +deltamethrin ranged from 1 500 mL+1 5 mL to 6 000 mL+12 mL per hm 2.The muscardine rate was correlated with the dosage of B.bassiana,and the rate ranged from 7 5% to 31 5%.The mixture of B.bassiana and deltamethrin was an effective and sustainable control measure against D.punctatus.
To evaluate the control efficacy of different formulations of Beauveria bassiana against Masson′s pine caterpillar, the following factors were investigated, the population density of overwintering larvae, emergence rate, population density, pupa infected and parasitic rates of first generation larvae. Based on the data, the grey correlation coefficients of different treatment areas, mixed dust formulation, oil formulation, high conidia dust and the check, were calculated as 0 7995, 0 4898, 0 2794 and 0 2004. The results indicated that the mixed dust formulation had the highest control efficiency. The formulated conidia had better control efficiency than unformulated ones. The findings also showed that the parasitic rates by natural enemies were not affected by applying different formulations of Beauveria bassiana.
