OBJECTIVE:To observe the effect of penetrative needling of "Baihui" (GV 20) to "Qubin" (GB 7) on neurologic functions and expression of tumor necrosis factor (TNF)-α, interleukin (IL)-6 and toll-like receptor 4 (TLR-4, involving in inflammatory reactions) in the tissue around the local cerebral hematoma in rats with intracerebral hemorrhage (ICH), so as to provide evidence for clinical treatment of ICH.METHODS:Fifty-four male Wistar rats were randomly divided into sham control, mo-del and acupuncture groups, and then further divided into three time-point subgroups(1,3,7 days after modeling, n=6/subgroup). The ICH model was established by injection of the rat's autoblood (50 μL) into the putaman region (P:0.2 mm, R:3.5 mm) in a stereotaxic apparatus and confirmed by Berderson's neurologic examination grading system (0-3 points). The neurologic function was assessed by using Longa's scoring (5-points) and footfault asymmetry testing[footfault index=(contra faults-ipsi faults)/total steps in 2 min]. For penetrative needling, an acupuncture needle was inserted into GV 20 and controlled to advance to GB 7 on the affected side and retained for 30 min, once daily. The expression of TNF-α, IL-6 and TLR-4 in the cerebral tissue around the putaman was detected by immunohistochemistry.RESULTS:After penetrative needling stimulation, the increased Longa's score and footfault asymmetry score in ICH rats were significantly decreased on day 1, 3 and 7 after modeling (P<0.01), suggesting an improvement of neurologic function after the treatment. Immunohistochemical staining outcomes of the cerebral tissue surrounding the autoblood injection site showed that the expression levels of TNF-α, IL-6 and TLR-4 proteins on day 1, 3 and 7 were considerably higher in the model group than in the control group (P<0.01), and markedly lower in the acupuncture group than in the model group (P<0.01), suggesting a suppression of the proinflammatory factors and TLR-4 levels around the locus of the brain after needling intervention. A positive correlation existed between the expression levels of TLR-4 and IL-6/TNF-α.CONCLUSIONS:Penetrative needling stimulation of GV 20 to GB 7 can reduce the levels of proinflammatory factors TNF-α and IL-6, and TLR-4 in the ICH tissues in rats with cerebral hemorrhage, which may contribute to its effect in improving neurological function.
目的:探讨针刺“百会”透“曲鬓”对脑出血大鼠脑组织中PI3K和p-AKT蛋白表达的影响.方法:选取清洁级健康Wistar雄性大鼠72只随机分为假手术组、模型组和针刺组3组,每组再随机分为6h、1天、3天、7天共4个亚组,每个亚组6只大鼠,制备大鼠脑出血模型.采用蛋白免疫印迹法(west-em blot)检测PI3K和p-AKT的蛋白表达.结果:模型组大鼠术后6h即出现PI3K、p-AKT蛋白表达明显增多,二者均于1天达到峰值,3天减弱,7天仍有表达.模型组、针刺组各时间点PI3K、p-AKT蛋白表达与假手术组比较差异均有统计学意义(P<0.05);针刺组各时间点PI3K、p-AKT蛋白表达均高于模型组,差异均有统计学意义(P<0.05).结论:针刺“百会”透“曲鬓”能够提高脑出血模型大鼠脑组织中PI3K和p-AKT的蛋白表达水平,从而起到保护脑出血灶周神经元、减少继发性损伤的作用.
ObjectiveTo investigate the effect of acupuncture on motor function and serum expressions of TNF-aand IL-6 contents in cerebral hemorrhage rats.MethodSeventy-two male Wistar rats were randomized into sham operation, model and acupuncture groups. A rat model of cerebral hemorrhage was made by intracerebral injection of autoblood. Baihui-to-Qubin acupuncture was performed after successful model making. The TNF-aand IL-6 contents of rat serum were measured by enzyme linked immunosorbent assay at three time points: one, three and seven days.ResultBaihui-to-Qubin acupuncture markedly increased the neurological function score and reduced neurologic deficits in cerebral hemorrhage rats. At the same time point, serum TNF-aand IL-6 contents were significantly lower in the acupuncture group of rats thanin the model and acupuncture group; there were statistically significant differences (P<0.01).ConclusionBaihui-to-Qubin acupuncture can reduce neurologic deficits in cerebral hemorrhage rats. It may be related to decreases in serum TNF-aand IL-6 contents after acupuncture.
目的:探讨中药白芨胶[1]对动物移植性肝癌治疗前后影像学比较。方法:选择新西兰大白兔40只,介入技术及动物分组处理参照Lindell等的肝动脉插管方法,将40只荷瘤兔随机分为四组,每组10只,分别为生理盐水加碘油组注射剂量1.5ml/kg;5-FU加碘油组注射剂量0.35mg/kg;白芨胶加碘油热灌注组注射剂量10 mg/kg;白芨提取液加碘油热灌注组注射剂量10 mg/kg。将四组家兔治疗前、后CT影像进行比较。结果:白芨胶加碘油组肝动脉化疗栓塞术能抑制兔VX2肝移植瘤的生长,对肿瘤的抑制作用更明显。结论:白芨具有良好的血管栓塞作用,使用安全,简易,对荷瘤兔肿瘤生长有抑制作用;白芨能抑制肿瘤增殖,使瘤体缩小,是一种理想的栓塞剂。
>脑膜瘤为颅内常见的肿瘤,多数属良性,生长缓慢。一些患者常因瘤周水肿而较早出现临床症状或体征。体积较小的脑膜瘤出现重度脑水肿的病例资料较少见。本文报道一例合体细胞型脑膜瘤于围手术期发生重度脑水肿的患者,并结合文献对脑膜瘤合并重度脑水肿的原因及治疗进行探讨。患者男性,56岁,于2011年5月因渐进加重性头痛、右侧肢体麻木到吉林油田总医院就诊,影像学检查示颅内占位性病变,于2011年5月31日收入院。入院时体检:神清语利,右侧肢体痛、温觉减退,肌力正常,无病理征。先后行头颅CT及MRI检查,示左颞顶部40 mm×26 mm基底病变,周围
