To explore the mechanism through which moxibustion promotes endometrial repair in rats with in thin endometrium (TE). Female SD rats were randomized into control group, 95% anhydrous ethanol-induced TE model group and moxibustion (at "Guan Yuan") group. High-throughput sequencing was used to identify the target genes of TE, and the targeting relationship between miR-223-3p and NLRP3 was verified using a dual luciferase assay. Histopathological of rat uterus was observed with HE staining, and expressions of miR-223-3p and NLRP3 were detected using RT-qPCR; serum levels of IL-1β and IL-18 of the rats were detected using ELISA, and protein expressions of NLRP3, ASC, caspase-1 and GSDMD in the uterus were detected with Western blotting. The pregnancies of the rats after treatment were counted. Enrichment analysis of the differential genes suggested up-regulated inflammatory response in TE, and dual luciferase assay verified targeted inhibition of NLRP3 expression by miR-223-3p. The rat models of TE had significantly decreased endometrial thickness and reduced endometrial glands and blood vessels with enhanced mRNA expression of NLRP3, increased serum levels of IL-1β and IL-18, up-regulated protein expressions of NLRP3, ASC, caspase-1 and GSDMD, lowered pregnancy rates on both the affected and unaffected sides and the overall number of pregnancies. Treatment of the rat models with mo-xibustion obviously increased the endometrial thickness and the density of glands and blood vessels, up-regulated miR-223-3p expression, lowered serum IL-1β and IL-18 levels and the protein expressions of NLRP3, ASC, caspase-1 and GSDMD, and significantly increased the number of pregnancies. Moxibustion at "Guan Yuan" acupoint up-regulates the expression of miR-223-3p, which results in targeted inhibition of NLRP3 to suppress pyroptosis and promote endometrial repair in rat models of TE.
BACKGROUND:Endometriosis (EMS), a well-recognized chronic inflammatory disorder, characterized by significant immune dysregulation, in which myeloid-derived suppressor cells (MDSCs) are essential for facilitating immunosuppression and driving to disease progression. Cryptotanshinone (CTS) is an active compound capable of modulating MDSC-mediated immunosuppression; however, its therapeutic effects and mechanisms in the treatment of EMS remain unclear. PURPOSE:This study aims to investigate the therapeutic potential of CTS in modulating MDSCs through JAK2/STAT3 signaling pathway and to evaluate its effects on immune microenvironment and endometriotic lesion growth in EMS. METHODS:Transcriptomic data (GSE141549) and single-cell RNA sequencing data (GSE213216) were analyzed to compare immune cell populations in control endometrium (CE), eutopic endometrium (EuE) and ectopic endometrium (EcE) of patients with EMS. Network pharmacology analysis, surface plasmon resonance (SPR) and cellular thermal shift assay (CETSA) were utilized to explore the molecular mechanism of CTS's effects on MDSCs. A C57BL/6J EMS mice model was established to evaluate CTS's influence on MDSC-mediated immune response in vivo. Flow cytometry and immunofluorescence were used to analyze the immune cell populations, particularly MDSCs and CD8+ T cells. Ex vivo bone marrow (BM)-derived MDSCs were prepared to investigate the modulatory activities of CTS on the frequency and function of MDSCs. The impacts of CTS on JAK2/STAT3 pathway were further examined by western blot. RESULTS:Bioinformatic analysis revealed that, among the three progression stages (CE, EuE, and EcE), the EcE stage exhibited a relatively elevated level of MDSCs and a reduced level of CD8+ T cells. Network pharmacological analysis, along with SPR and CETSA identified that CTS potentially modulates MDSCs in EMS by targeting the JAK2/STAT3 pathway. In vivo studies demonstrated that a relatively high dose of CTS treatment (60mg/kg) effectively inhibited lesion growth, reduced the population of MDSCs, and enhanced CD8+ T cell infiltration. Ex vivo experiments showed that CTS decreased the BM-derived MDSC frequency and rescued the suppressive ability of MDSC upon CD8+ T cells in a dose-dependent manner. Further mechanism analysis confirmed that CTS modulates the expression of immunosuppressive genes and proteins associated with MDSCs through JAK2/STAT3 pathway. CONCLUSION:This study is the first to demonstrate that CTS is a promising natural compound for EMS treatment by inhibiting MDSC accumulation and modulating MDSC-mediated immune responses. Its therapeutic efficacy is linked to the modulation of the JAK2/STAT3 signaling pathway.

Endometrium plays a key role in embryo implantation and maintenance of pregnancy. However, to repair endometrial injury is still a challenge. In recent years, hydrogel materials have been widely used as effective support matrices to prevent intrauterine adhesions after endometrial injury. They can also be used as preparation scaffolds for encapsulating MSCs and certain therapeutic drugs. This study aimed to develop a preparation scaffold with high tissue affinity, high viscoelasticity and controlled release for repair of endometrial injury. The scaffold utilized heparin poloxamer (HP) as the matrix material and ε-polylysine (EPL) as the functional excipient to prepare a hydrogel that is suitable for endometrial adhesion and further encapsulate BMSCs. Furthermore, a strategy of the thermo-sensitive EPL-HP hydrogel-encapsulated BMSCs were used for better homing of BMSC after transplantation into the rat endometrial injury model, so as to exert the potential of endometrial regeneration by activating Nrf2 to regulate SDF-1/CXCR4 axis.
Objectives: This study offers valuable insights into the research hotspots and trends related to ovarian hyperstimulation syndrome (OHSS). By systematically analyzing articles published on the subject up to July 15, 2024, it provides significant perspectives for future research endeavors. Methods: A literature search was executed utilizing the Web of Science core database to retrieve relevant records, with case reports subsequently reassigned to a separate file. During the analysis phase, countries, institutions, authors, journals, references, and keywords underwent thorough examination using tools such as VOSviewer v1.6.10, CiteSpace, R package, and Microsoft Excel 2021. Results: From the inception of the database to July 15, 2024, a total of 1125 articles were retrieved, including 255 case reports, with a citation count of 23,768. The top 5 countries with the highest article counts were China, the USA, Ireland, the United Kingdom, and Spain. In terms of institutions, Tel Aviv University, Zhengzhou University, Aristotle University of Thessaloniki, Cairo University, and Free University of Brussels were the leading contributors, with Tel Aviv University having the highest number of published articles. Keywords were systematically classified into 4 clusters within the coverage keyword network: diagnosis and clinical manifestations; risk factors and pathogenesis; prevention and treatment; and prognosis and complications. The case reports primarily focused on patients with clinical manifestations of pleural and abdominal fluid accumulation, venous thrombosis, and, in some cases, fatal outcomes. Conclusion: The present study conducts a bibliometric analysis of OHSS case reports to understand its prevalence and trends. The study highlights the shift towards advanced assisted reproduction technologies and prophylactic medication as research hotspots, offering valuable guidance for future OHSS research.
ObjectivesThe objective of this study was to investigate the glycolytic activity of adenomyosis, which is characterized by malignant biological behaviors including abnormal cell proliferation, migration, invasion, cell regulation, and epithelial-mesenchymal transition.MethodsFrom January 2021 to August 2022, a total of 15 patients who underwent total hysterectomy for adenomyosis and 14 patients who had non-endometrial diseases, specifically with cervical squamous intraepithelial neoplasia and uterine myoma, were included in this study. Myometrium with ectopic endometrium from patients with adenomyosis while normal myometrium from patients in the control group were collected. All samples were confirmed by a histopathological examination. The samples were analyzed by liquid chromatography-mass spectrometry (LC-MS), real-time quantitative PCR, NAD+/NADH assay kit as well as the glucose and lactate assay kits.ResultsEndometrial stroma and glands could be observed within the myometrium of patients in the adenomyosis group. We found that the mRNA expressions of HK1, PFKFB3, glyceraldehyde-3-phospate dehydrogenase (GAPDH), PKM2, and PDHA as well as the protein expressions of PFKFB3 were elevated in ectopic endometrial tissues of the adenomyosis group as compared to normal myometrium of the control group. The level of fructose 1,6-diphosphate was increased while NAD + and NAD+/NADH ratio were decreased compared with the control group. Besides, increased glucose consumption and lactate production were observed in myometrium with ectopic endometrium.ConclusionsWe concluded that altered glycolytic phenotype of the myometrium with ectopic endometrium in women with adenomyosis may contribute the development of adenomyosis.
背景:金丝桃苷具有抗炎、抗氧化等多种药理作用,但将其应用于修复子宫内膜损伤的研究并不多见.目的:制备金丝桃苷纳米粒,利用泊洛沙姆407搭载金丝桃苷纳米粒,研究其对大鼠子宫内膜损伤的修复作用.方法:分别制备金丝桃苷纳米粒、泊洛沙姆407水凝胶与泊洛沙姆407水凝胶搭载金丝桃苷纳米粒.取24只雌性SD大鼠,随机分4组,每组6只:模型组、单纯水凝胶组、载药水凝胶组以搔刮法建立子宫内膜损伤模型,分别向子宫内注射PBS、泊洛沙姆407水凝胶、泊洛沙姆407水凝胶搭载金丝桃苷纳米粒,空白对照组仅行开腹不进行造模.造模7 d后取材,苏木精-伊红染色观察子宫内膜形态学变化,采用ELISA法检测子宫内膜组织中白细胞介素1β、白细胞介素8、肿瘤坏死因子α水平,免疫荧光染色检测宫内膜组织中血管内皮生长因子、角蛋白、波形蛋白的表达.结果 与结论:①苏木精-伊红染色显示,与空白对照组比较,模型组大鼠子宫内膜层变薄,内膜结构不完整,血管及腺体及明显减少稀疏;与模型组比较,单纯水凝胶组大鼠子宫内膜增厚、内膜结构较完整,载药水凝胶组大鼠子宫内膜增厚、内膜结构较完整、腺体较丰富;②与空白对照组比较,模型组的白细胞介素1β、白细胞介素8、肿瘤坏死因子α水平均升高(P<0.01);与模型组比较,单纯水凝胶组、载药水凝胶组的白细胞介素1β、白细胞介素8、肿瘤坏死因子α水平均下降(P<0.01);③免疫荧光染色显示,与空白对照组比较,模型组的血管内皮生长因子、角蛋白、波形蛋白表达均降低(P<0.01);与模型组比较,载药水凝胶组血管内皮生长因子、角蛋白、波形蛋白表达均升高(P<0.01),单纯水凝胶组血管内皮生长因子表达升高(P<0.01);④结果表明,水凝胶搭载金丝桃苷纳米粒对大鼠子宫内膜损伤有修复作用.
目的 运用超高效液相色谱-四极杆-静电场轨道阱串联质谱(UHPLC-QE-MS)技术分析橘荔散结丸的药物成分,围绕核因子-κB(NF-κB)信号通路探讨橘荔散结丸诱导子宫肌瘤细胞凋亡的炎症机制.方法 ①采用UHPLC-QE-MS技术分析橘荔散结丸化学成分.②培养原代人子宫肌瘤细胞,免疫组化法进行鉴定.实验分为 5 组,空白组常规培养,基质组加入0.05%DMSO培养,米非司酮组加入1×10-5mol/L米非司酮含药液培养,橘荔散结丸高、低浓度组分别加入2 mg/mL和1 mg/mL橘荔散结丸含药液培养.采用流式细胞术检测子宫肌瘤细胞凋亡率,采用Western blot法检测子宫肌瘤细胞中IKKα、IKBα、p-IKBα、p65、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)蛋白表达情况,采用qPCR技术检测子宫肌瘤细胞中IKKα、IKBα、p65、TNF-α、IL-6 mRNA表达量.结果 鉴别橘荔散结丸主要化学成分 91 个,冻干粉与中成药间共有22 个.米非司酮组和橘荔散结丸高、低浓度组子宫肌瘤细胞早期凋亡率和晚期凋亡率均明显高于空白组和基质组(P均<0.05),且米非司酮组、橘荔散结丸高浓度组早期凋亡率和晚期凋亡率均明显高于橘荔散结丸低浓度组(P均<0.05),橘荔散结丸高浓度组早期凋亡率明显高于米非司酮组(P<0.05).橘荔散结丸高浓度组和米非司酮组子宫肌瘤细胞中IKKα、p-IKBα、p65、TNF-α蛋白相对表达量均明显高于空白组和橘荔散结丸低浓度组(P均<0.05),且橘荔散结丸高浓度组和米非司酮组子宫肌瘤细胞中IL-6 蛋白相对表达量均明显高于空白组(P均<0.05);橘荔散结丸高浓度组子宫肌瘤细胞中p-IKBα、p65、IL-6 蛋白相对表达量均明显低于米非司酮组(P均<0.05).米非司酮组、橘荔散结丸高浓度组、橘荔散结丸低浓度组子宫肌瘤细胞中 IKKα、IKBα、p65、IL-6 mRNA相对表达量和米非司酮组、橘荔散结丸高浓度组TNF-α mRNA相对表达量均明显高于空白组(P均<0.05);橘荔散结丸高浓度组和米非司酮组IKBα、p65、IL-6 mRNA相对表达量均明显高于橘荔散结丸低浓度组(P均<0.05).结论 橘荔散结丸化学成分具有抗炎免疫调节作用,其治疗子宫肌瘤分子机制与干预肌瘤细胞微环境炎症反应密切相关.
目的 基于网络药理学和分子对接技术探讨黄芩-白术治疗先兆流产的潜在机制.方法 利用TCMSP数据库筛选黄芩-白术的有效成分,并预测潜在靶基因,从GeneCards、NCBI、OMIM、DrugBank、DisGeNET数据库获取与先兆流产相关的靶基因,将两者取交集得到黄芩-白术治疗先兆流产的潜在靶基因.通过Cytoscape3.7.2软件构建主要活性成分-潜在治疗靶基因网络;利用STRING数据库构建蛋白互做网络;运用在线网站对作用靶基因进行GO功能和KEGG通路富集分析,同时使用SYBYL软件V2.0进行分子对接验证.结果 黄芩-白术活性成分-先兆流产疾病靶点网络包含43个化合物,46个靶点和20条通路,主要化合物包括黄芩中的黄连素、汉黄芩、黄芩素等化合物,白术中的α-香树脂醇,关键靶点涉及IL6、VGFR、MMP9、FOS、MAPK14、ESR1等;共同靶点基因功能偏向蛋白质磷酸化的积极调节、炎症反应、凋亡信号通路、血管形态、血管发育等;重要的KEGG通路涉及IL-17信号通路、MAPK信号通路、凋亡通路、TNF信号通路、PI3K/Akt信号通路、雌激素信号通路.分子对接结果显示,黄芩新素、二甲氧基黄酮、麝香黄酮、三甲氧基黄酮、豆甾醇、红花素、黄芩素等15种主要活性成分与HSP90AA1、PPARG、PTGS2结合较好.结论 黄芩-白术可能通过多成分、多靶点和多通路协同作用发挥抗感染、营养支持、调节激素水平作用,从而治疗先兆流产.
背景:水凝胶作为一类组织工程的支架材料,为移植细胞在体内增殖分化提供了良好的物理支持.目的:探讨水凝胶搭载骨髓间充质干细胞对大鼠受损子宫内膜的修复效果.方法:将分离培养的SD大鼠骨髓间充质干细胞进行CM-Dil标记,然后制备成泊洛沙姆407-骨髓间充质干细胞复合物.取30只8周龄SD雌性大鼠随机分为5组:假手术组、模型组、水凝胶组、干细胞组、复合物组,除假手术组外,其余4组建立子宫内膜机械损伤模型,然后每侧宫角分别注射0.2 mL PBS、泊洛沙姆407水凝胶、骨髓间充质干细胞悬液或泊洛沙姆407水凝胶-骨髓间充质干细胞悬液.观察7 d后取出子宫组织,进行苏木精-伊红染色、免疫荧光染色和酶联免疫吸附检测.结果 与结论:①与假手术组相比,模型组、水凝胶组、干细胞组与复合物组内膜厚度变薄(P<0.05),其中水凝胶组、干细胞组和复合物组内膜厚度较模型组均有明显改善(P<0.05),复合物组改善最明显,但水凝胶组与干细胞组间差异无显著性意义(P>0.05);②与干细胞组相比,复合物组CM-Dil标记的骨髓间充质干细胞迁移至子宫内膜数量更多(P<0.01),并且两组细胞均具有向子宫内膜间质细胞分化潜能;③与模型组相比,水凝胶组、干细胞组和复合物组子宫内膜组织中白细胞介素1β、白细胞介素6和和肿瘤坏死因子α水平明显下降(P<0.01),其中,干细胞组和复合物组白细胞介素1β、白细胞介素6和肿瘤坏死因子α水平显著低于水凝胶组(P<0.05),干细胞组白细胞介素6和肿瘤坏死因子α水平低于复合物组(P<0.01),但两组白细胞介素1β水平无显著差异(P>0.05);④结果表明,泊洛沙姆407水凝胶搭载骨髓间充质干细胞治疗子宫内膜损伤具有可行性,为修复受损子宫内膜提供新思路.
Background: Acupuncture is widely used in treatment of postoperative ileus (POI), but the safety and efficacy of acupuncture in POI after gynecological surgery still lack of evidence-based basis. Methods: PubMed, CINAHL, EMBASE, Web of science, Google Scholar, Wangfang database, Chinese Biomedical Literature Database (SinoMed), Chinese Science and Technology Periodical Database, and China National Knowledge Infrastructure database will be searched until December 31, 2020. Two independent investigators will screen the relevant randomized controlled trials from Data one by one by using prespecified criteria. The relevant data from included studies will be extracted and analyzed by using RevMan V.5.3 software. Quality of the included studies will be estimated by using the Cochrane Collaboration risk of bias tool, and publication bias will be assessed by using Egger test and Begg test. In addition, quality of evidence will be evaluated by using Grading of Recommendations Assessment, Development, and Evaluation. Results: We will analyze the effect of acupuncture on time to first flatus and time to bowel sound recovery as the primary outcomes of this review. Meanwhile, frequency of bowel sounds, time to defecation, time of hospital stay, biochemical indicators related to gastrointestinal motility, inflammation factors, responder rate, and adverse events for patients receiving gynecological surgery. Conclusion: Our findings will benefit researchers and provide reference for the treatment and prevention of POI for the patients undergoing gynecological surgery.
目的 探讨表没食子儿茶素没食子酸酯(EGCG)对人子宫肌瘤细胞增殖的影响及其机制.方法 获取人子宫肌瘤组织后提取人子宫肌瘤细胞.将细胞分为对照组、50μmol/L EGCG组、100μmol/L EGCG组、200μmol/L EGCG组,各EGCG剂量组分别加入相应浓度的EGCG,对照组加入等体积的磷酸缓冲盐溶液.干预24 h、48 h、72 h后,检测细胞存活率;干预48 h后观察细胞形态,并检测Wnt5 b、β-连环蛋白(β-Catenin)、上皮生长因子受体(EGFR)蛋白的表达情况.结果 干预48 h、72 h后,对照组、50μmol/L EGCG组、100μmol/L EGCG组、200μmol/L EGCG组人子宫肌瘤细胞存活率依次降低;随着干预时间的延长,各干预组细胞存活率有下降趋势(均P<0.05);免疫组化检测结果显示,各浓度EGCG干预组Wnt5 b、β-Catenin蛋白表达水平均低于对照组;200μmol/L EGCG组Wnt5 b蛋白表达水平低于其他两个浓度干预组,50μmol/L EGCG组、100μmol/L EGCG组、200μmol/L EGCG组β-Catenin蛋白表达水平依次降低(均P<0.05).免疫印迹试验结果显示,各浓度EGCG干预组EGFR、β-Catenin蛋白表达水平均低于对照组,100、200μmol/L EGCG组Wnt5 b蛋白表达水平低于对照组;200μmol/L EGCG组的EGFR、Wnt5 b蛋白表达水平均低于其他两个干预组(均P<0.05).结论 EGCG可抑制人子宫肌瘤细胞增殖,其机制可能与抑制Wnt/β-Catenin信号通路和EGFR的表达有关.
目的:该研究通过网络药理学的方法探讨归肾丸治疗薄型子宫内膜的关键核心靶点及相关信号通路等作用机制,进一步运用动物实验加以验证所得靶点,验证归肾丸对薄型子宫内膜的影响.方法:利用中药系统药理学数据库与分析平台(TCMSP)数据库检索归肾丸中8味中药组成的有效化学成分、活性成分靶点和靶点简称;运用GeneCards数据库和在线人类孟德尔遗传数据库(OMIM)检索薄型子宫内膜相关的靶点基因;运用韦恩软件对归肾丸的药物靶点和薄型子宫内膜的疾病靶点进行取交集操作,将交集靶点导入STRING数据库及Cytoscape 3.7.2软件进行可视化分析获得"药物-疾病"的蛋白质-蛋白质相互作用(PPI)网络;再将交集靶点输入Enrichr数据库和David数据库分别进行基因本体(GO)富集分析及京都基因与基因组百科全书(KEGG)富集分析.以得到的可能核心关键靶点作为理论基础,建立大鼠薄型子宫内膜模型,归肾丸及雌激素干预21d后,通过苏木素·伊红(HE)染色法观察大鼠的内膜厚度;通过蛋白免疫印迹法(Western blot)和实时荧光定量聚合酶链式反应(Real-time PCR)检测表皮生长因子受体(EGFR),基质金属蛋白酶9(MMP9),白细胞介素1β(IL-1β),丝裂原激活蛋白激酶14(MAPK14)这4个核心关键靶点的蛋白和mRNA的表达水平.结果:通过韦恩软件共获得130个交集靶点;将130个交集靶点导入STRING数据库和Cytoscape软件中,得到包括33个节点,107条边的蛋PPI网络;DAVID 6.8数据库进行GO分析中的功能注释分析,共涉及167个生物过程(BP),22个细胞组分(CC),39个分子功能(MF);DAVID 6.8数据库进行KEGG富集分析,与薄型子宫内膜相关的通路共34条;Western blot和Real-time PCR分别对6组大鼠子宫内膜组织中的EGFR,MMP9,IL-1β,MAPK14蛋白和mRNA的表达结果进行分析,归肾丸能够增强EGFR,MMP9,IL-1β,MAPK14蛋白和mRNA在薄型子宫内膜上的表达.结论:通过网络药理学的理论分析及动物实验的研究结果发现,归肾丸能够有效改善薄型子宫内膜的相关指标,并通过增加EGFR,MMP9,IL-1β,MAPK14蛋白和mRNA表达水平,促进内膜组织增殖,改善内膜偏薄的症状.

Quality of the included studies will be estimated by using the Cochrane Collaboration risk of bias tool, and publication bias will be assessed by using Egger test and Begg test. In addition, quality of evidence will be evaluated by using Grading of Recommendations Assessment, Development, and Evaluation. Results: We will analyze the effect of acupuncture on time to first flatus and time to bowel sound recovery as the primary outcomes of this review. Meanwhile, frequency of bowel sounds, time to defecation, time of hospital stay, biochemical indicators related to gastrointestinal motility, inflammation factors, responder rate, and adverse events for patients receiving gynecological surgery. Conclusion: Our findings will benefit researchers and provide reference for the treatment and prevention of POI for the patients undergoing gynecological surgery.
目的 验证同源基因A10(hoxA10)对10号染色体缺失的磷酸酶、张力蛋白同源基因(pten)、磷脂酰肌醇-3-激酶(pi3k)和蛋白激酶B(Akt)信号通路的影响.方法 设计SD大鼠骨髓间充质干细胞hoxA10基因的短发夹RNA(shRNA)靶序列,并构建pLVSHG01-Rat HoxA10重组慢病毒.用敲低型慢病毒感染SD大鼠的骨髓间充质干细胞(BMSCs),将BMSCs分为感染组(感染hoxA10敲低型慢病毒)和空白对照组(未感染).用嘌呤霉素筛选出感染成功的细胞株后,以蛋白质印迹法检测感染后BMSCs中HoxA10蛋白表达,用实时荧光定量反转录-PCR法检测各组骨髓间充质干细胞中hoxA10、pten、pi3k和akt基因的表达水平.用双染流式细胞凋亡试剂盒检测各组细胞的凋亡率和周期.结果 成功构建了hoxA10低表达的SD大鼠骨髓间充质干细胞,空白对照组和感染组的hoxA10基因表达量分别为6.22±0.20和3.09±0.06;这2组的pten基因表达量分别为1.97±0.18和5.09±0.07;这2组的pi3k基因表达量分别为6.06±0.05和2.72±0.10;这2组的akt基因表达量分别为5.98±0.09和2.48±0.35.上述指标:空白对照组与感染组比较,差异均有统计学意义(均P<0.001).空白对照组的细胞凋亡率(18.88±1.71)%明显低于感染组(36.01±0.45)%,组间差异有统计学意义(P<0.05).结论 HoxA10基因能够负向调控pten/pi3 k/akt信号通路.
背景:补肾类方剂对骨髓间充质干细胞的促增殖作用已经被证实,但其作用机制尚不明确,该研究以归肾丸为例,探讨其可能的作用机制.目的:通过观察归肾丸水提物对骨髓间充质干细胞增殖的影响,探究归肾丸滋补肾精与骨髓间充质干细胞增殖之间的关系.方法:从3周龄雄性SD大鼠股骨骨髓中分离提取骨髓间充质干细胞,采用全骨髓贴壁法将其纯化,选用纯度较高的第3代细胞进行药物干预,将其分为空白对照组和10,20,40,80,100 mg/L归肾丸水提物组,在体外连续培养5 d以观察量效关系和时效关系.倒置显微镜下观察细胞形态,CCK-8法评估细胞增殖活性,Western blot检测细胞中PI3K、AKT蛋白表达.结果与结论:①归肾丸水提物组的细胞增殖能力明显高于空白对照组,存在一定的量效和时效关系,尤以80 mg/L归肾丸水提物干预3 d时作用效果最佳;②归肾丸水提物组骨髓间充质干细胞中PI3K、AKT蛋白的表达明显高于空白对照组;③结果提示,归肾丸水提物能有效促进骨髓间充质干细胞的增殖,可能与其参与调控PI3K/AKT信号通路有关.
OBJECTIVE To evaluate the effectiveness of electroacupuncture (EA) for female stress urinary incontinence (SUI). METHODS We searched 12 databases electronically from inception to November 2018 without language restrictions. We included randomized controlled trials (RCTs) involving women with SUI, but excludd other types of urinary incontinence or studies that were not RCTs. Two independent reviewers extracted study characteristics, with disagreements resolved by consensus. Data were pooled and expressed as mean difference (MD) for continuous outcomes and relative risk (RR) for dichotomous outcomes, with 95% confidence intervals (CI). This study was registered with the International Prospective Register of Systematic Reviews (number CRD42018089734). RESULTS We found very low to high level evidence that EA improved the effective rate (RR = 2.03, 95%CI: 1.40, 2.95; P = 0.0002) and reduced urine leakage as measured by the 1-hour pad test (MD = 3.33, 95%CI: 0.89, 5.77; P = 0.008), International Consultation on Incontinence Questionnaire Short Form score (MD = 3.14, 95%CI: 2.42, 3.85; P < 0.00001), and 72-hour incontinence episodes (MD = 1.17, 95%CI: 0.56, 1.78; P = 0.0002) compared with sham electroacupuncture (SA), pelvic floor muscle training, and medication. CONCLUSION The effectiveness and safety of EA for key outcomes for women with SUI are statistically significantly better than those of SA, but most available evidence is very low or low quality. More well-designed RCTs are needed to confirm these findings.
Background: Decreased ovarian reserve (DOR) is a common reproductive barrier in female. Bushen Huoxue (BSHX) method of TCM is widely applied to treat DOR clinically. The purpose of this study is to provide a systemic and comprehensive evaluation of BSHX in the treatment of DOR. Methods: We have registered this protocol with OSF registry and the DOI is 10.17605/OSF.IO/QNUE2. We will search 4 English databases (PubMed, EMBASE, MEDLINE, Cochrane Library) and four Chinese databases (China national knowledge infrastructure database, Wanfang database, VIP and Superstar database) from their inception to August 10, 2020. Two authors will search and extract independently all related studies. RevMan 5.3 software will be applied to synthesize data. Results: The results of this study will be published in a scientific journal after peer-review. Conclusion: This systematic review will provide reliable evidences for clinicians, and help them make decisions in DOR management.
目的 探讨差速贴壁法获取人子宫腺肌病病灶细胞的平滑肌细胞的纯化时间.方法 采取组织块酶消化法进行子宫腺肌病病灶细胞原代培养,待生长融合80%~90%时根据细胞贴壁时间分为未处理组及15 min、30 min、60 min、90 min四个时间组,各时间贴壁组与未贴壁组进行细胞筛选与培养,以免疫组化方法鉴定平滑肌细胞及其纯度.结果 400倍镜视野下每组16个视野计数算滑肌细胞数百分比,未处理组最低,为12.60%;未贴壁15 min组最高,为73.38%,与未处理组比较有统计学差异(P<0.01);贴壁90 min组与未处理组比较无统计学差异(P>0.05).结论 用差速贴壁法分离人子宫腺肌病病灶的平滑肌细胞可通过贴壁15 min剔除大部分杂质细胞,选择15 min内未贴壁细胞继续培养传代可进一步获取纯度更高的子宫平滑肌细胞.
