目的:通过观察针刺、艾灸对环磷酰胺(CTX)化疗后,健康小鼠骨髓造血细胞中Notch信号通路相关差异基因numb1、numb2表达的影响,从分子生物学方面探讨针灸改善化疗后骨髓抑制、提升外周血白细胞的机制.方法:取雄性昆明种SPF级小鼠40只,运用随机分层分组的方法分为空白组(A组)、模型组(B组)、针刺组(C组)、艾灸组(D组).B组、C组、D组的小鼠注射CTX建立骨髓抑制模型,A组给予同等剂量生理盐水.针刺和艾灸治疗后,运用免疫组化法、RT-PCR法、Western-Bolt法检测Notch信号通路上相关差异基因numb1、numb2的蛋白表达水平及m RNA表达,进行数据统计分析.结果:numb1、numb2属于小鼠骨髓造血细胞Notch信号通路相关性基因,且具有显著差异性,针刺、艾灸可以上调numb1、numb2 mRNA的表达,促进numb1、numb2蛋白含量的增加.结论:上调Notch信号通路相关差异基因numb1、numb2的表达,抑制过度激活的Notch信号通路,增强骨髓造血功能,可能是针灸减轻CTX化疗引起的骨髓抑制、改善造血功能的微观机制之一.
目的:通过观察黄连素对2型糖尿病大鼠血脂调节作用及对IL-6、IL-10及CRP水平影响,探讨其对2型糖尿病的作用机制.方法:80只成年雄性健康SD大鼠,随机取20只作为对照组,60只以高糖高脂饲料喂养建立2型糖尿病(T2DM)模型,造模成功后随机分为模型组及黄连素治疗组.黄连素组给予黄连素125mg/(kg·d)灌胃,模型组和正常对照组给予同等量生理盐水,共4周.4周后处死大鼠,取血测定FBG、TG、TC、CRP、IL-6、IL-10水平.给药期间记录大鼠的体重、饮食、饮水及尿量.结果:黄连素可以改善2型糖尿病大鼠的临床症状及精神状态;黄连素组FBG、TG、TC水平低于模型组,差异有统计学意义(P<0.05);模型组和黄连素组的CRP和IL-6水平高于正常组,差异有统计学意义(P<0.05);黄连素组CRP和IL-6水平低于模型组,差异有统计学意义(P<0.05);模型组和黄连素组IL-10的水平低于正常组,差异有统计学意义(P<0.05);黄连素组IL-10水平高于模型组,差异有统计学意义(P<0.05).结论:黄连素对2型糖尿病大鼠有治疗作用,降低血糖血脂,通过调控血清炎症因子水平发挥其降糖的作用机制.
目的:观察透穴埋线治疗单纯性肥胖症的临床疗效.方法:选择河南中医药大学第三附属医院针灸门诊收治的单纯性肥胖症患者1206例,给予透穴埋线治疗(中脘、梁门、天枢、大横、阴交、章门、京门、丰隆、局部阿是穴加减)治疗1次为1个疗程,30 d后判断疗效.结果:男性及女性使用透穴埋线减肥的疗效对比无统计学意义(P>0.05);透穴埋线治疗肥胖症的疗效与其肥胖程度基本成正比(P<0.01);青少年患者的疗效优于中、老年患者;春季和夏季治疗的患者疗效优于秋季患者.结论:透穴埋线法减肥的疗效和性别无明显关联,和肥胖度基本成正比;青少年比中、老年患者疗效显著;春、夏季比秋冬季疗效显著.
秋季,很多人会出现口干、咽干、鼻干、皮肤干燥、便秘等上火症状.将一头蒜捣碎后分成两份,敷在脚心处,然后用塑料薄膜将脚缠绕起来,固定住,对于去虚火非常有效.成人最好是敷到脚心发辣为止(以不超过10分钟为宜).
目的 通过观察针灸对CTX荷瘤小鼠骨髓细胞周期的影响,探讨针灸对抗化疗毒副反应,改善骨髓抑制、促进细胞修复效应的机制.方法 将雄性清洁级昆明种小鼠192只种植S180肉瘤,随机分为荷瘤空白组、荷瘤模型组、荷瘤针刺组、荷瘤艾灸组,每组48只,其中荷瘤模型组、荷瘤针刺组、荷瘤艾灸组腹腔注射CTX,制备CTX荷瘤小鼠模型,荷瘤空白组注入相同体积的生理盐水.荷瘤针刺组、荷瘤艾灸组分别采用针刺、艾灸治疗,荷瘤空白组、荷瘤模型组每日陪同抓取、固定,不做任何治疗.各组分别于治疗后第2~6天采用流式细胞仪测定骨髓细胞各细胞周期的百分率.结果 烷化剂CTX损伤骨髓细胞的同时,干扰了正常的细胞周期规律.针灸可以调节这种不正常的细胞周期状态,加速G1期细胞向S期细胞、G2期细胞向M期细胞的转化,从而提高机体细胞抗损伤能力和修复能力.结论 促进骨髓细胞周期转化,是针灸改善化疗后骨髓抑制,保护造血功能的关键机制之一.
Objective: To evaluate the protein expression and distribution of Notch signaling-related, differentially expressed genes (DEGs) in the bone marrow cells of mice. Additionally, verification of acupuncture regulated protein expression of these DEGs to protect bone marrow stem cells and alleviate cyclophosphamide (CTX) chemotherapy-induced myelosuppression was examined as well. Methods: Thirty two Healthy male Kunming mice of clean grade were randomized into blank group (A), model group (B), acupuncture group (C) and moxibustion group (D). Mice in groups B, C, and D were intraperitoneally injected with 100mg/kg/d CTX for three successive days to generate the myelosuppressive model; while mice in group A were injected with an equivalent volume of sterile saline. After immobilizing the animals, mice in groups C and D received daily acupuncture and moxibustion in points “DU14”, “BL17” “ BL23” and “ ST36”respectively,while those in group A and B were received no treatment. After five days of treatment,the mice were euthanized by cervical dislocation and their femurs collected for marrow harvest , Small-throughput cDNA microarray was used for initial DEG screening, followed by immunohistochemistry, realtime quantitative PCR (RT-qPCR), and Western Blot analysis to detect protein expression and quantity of the Notch signaling-related DEGs in the bone marrow cells. Results: Expression of numb proteins (i.e., numb1 and numb2) was found to be upregulated while notch2 and jag1 were down regulated in bone marrow cells of acupuncture-treated, tumor-bearing mice. Conclusion: Notch signaling pathway serves as an important signal transduction pathway for acupuncture and moxibustion to relieve CTX chemotherapy-induced myelosuppression. Regulation of associated DEGs in notch signaling pathway in marrow cells is the key mechanism by which acupuncture and moxibustion improve marrow hematopoiesis after chemotherapy.
目的:通过观察针灸对环磷酰胺(CTX)化疗小鼠骨髓细胞DNA含量的影响,探讨针灸改善化疗后骨髓抑制、升高外周血白细胞的分子生物学机制.方法:将昆明种雄性小鼠160只按体质量随机分为正常组、模型组、针刺组、艾灸组,每组40只.用CTX建立骨髓抑制模型.模型成功后,每天定时对针刺组、艾灸组小鼠“大椎”“膈俞”“肾俞”“足三里”给予针刺、艾灸治疗,连续6d.正常组、模型组每日陪同抓取固定,不治疗.镜检外周血白细胞数、流式细胞仪检测各组小鼠骨髓细胞G1期、G2期DNA含量,进行数据统计分析.结果:针刺和艾灸可使CTX小鼠外周血白细胞提前1天回升并提前1天超过基础水平;针刺和艾灸可以上调骨髓细胞G1期、G2期DNA含量,在治疗第5天针刺组、艾灸组骨髓细胞G1期、G2期DNA含量高于模型组,差异具有统计学意义(P<0.05).结论:针刺和艾灸可以通过上调CTX小鼠骨髓细胞G1期、G2期DNA的含量,加速骨髓细胞DNA的修复,改善化疗所致骨髓细胞增殖、分裂受抑制的状态,发挥提升外周血白细胞,维护造血功能的作用.
目的:观察针灸对环磷酰胺(CTX)化疗后的荷瘤小鼠白细胞及骨髓细胞DNA修复蛋白O6-甲基鸟嘌呤-DNA甲基转移酶.(MGMT)、DNA聚合酶β(POLβ)、切除修复交叉互补基因(XPD)表达的影响.方法:160只雄性清洁级昆明种小鼠植种S180肉瘤,按体质量随机分为荷瘤空白(A)组、荷瘤模型(B)组、荷瘤针刺(C)组、荷瘤艾灸(D)组.B、C、D组腹腔注射CTX 150mg/kg制备CTX荷瘤模型小鼠,A组注射同等剂量的0.9%氯化钠溶液.C、D组分别选取“大椎”“膈俞”“肾俞”“足三里”进行治疗,A、B组每日陪同抓取固定,不治疗.镜检各组小鼠外周血白细胞,免疫组化法检测骨髓细胞DNA修复蛋白MGMT、POLβ、XPD的表达情况,统计分析数据.结果:针灸可提升化疗后荷瘤小鼠白细胞数量(P<0.05),亦能上调CTX化疗后荷瘤小鼠骨髓细胞DNA修复相关蛋白MGMT、POL β、XPD的表达.结论:针灸升高CTX化疗后荷瘤小鼠外周血白细胞的疗效确切,其升白机制与上调CTX化疗后荷瘤小鼠骨髓细胞DNA修复相关蛋白MGMT、POL β、XPD的表达及改善化疗后骨髓抑制关系密切.
目的 通过测定针刺和艾灸前后,骨髓抑制小鼠DNA修复基因XPD蛋白表达量的改变,探讨针灸减轻骨髓抑制、增加外周血白细胞的分子生物学机制.方法 取清洁级、雄性昆明种小鼠224只,分成正常组、模型组、针刺组、艾灸组.模型组、针刺组、艾灸组的小鼠腹腔注射环磷酰胺(CTX),造成CTX骨髓抑制小鼠模型,正常组小鼠腹腔注入等量的0.9%氯化钠溶液.针刺组、艾灸组分别进行针刺、艾灸,正常组、模型组陪同固定,不治疗.各组均在2~6d用免疫组化染色法(Elivison二步法)、Western Blot法检测和分析骨髓细胞XPD蛋白表达量的变化.结果 针灸可以通过上调CTX模型小鼠骨髓细胞中XPD的蛋白表达,促进骨髓细胞受损DNA的核苷酸切除修复,减轻因CTX化疗引起的骨髓抑制.结论 上调受损骨髓细胞DNA修复基因XPD的表达,促进细胞受损DNA核苷酸切除修复,是针灸减轻骨髓抑制、保护造血功能、增加外周血白细胞的重要机制之一.
OBJECTIVE:To observe the effect of acupuncture and moxibustion intervention on the contents of serum IL-7 and IL-18 and white blood cell counts in cyclophosphamide (CTX)-induced hypofunction of immunity in tumor-bearing mice, so as to explore its mechanism underlying improvement of immune suppression caused by chemotherapy. METHODS:Sixty-four male Kunming mice were implanted with S 180 sarcoma at the left armpit (for making tumor-bearing model) , and then, randomized into control group, model group, acupuncture group and moxibustion group( 16 mice in each group). The mice of the latter 3 groups were injected with CTX (i. p. , 150 mg · kg(-1) · d(-1)), and those of the control group were injected (i. p.) with normal saline. Filiform acupuncture needle or moxibustion was applied to "Dazhui" (GV 14) , unilateral "Geshu" (BL 17) , "Shenshu"(BL 23) and "Zusanli"(ST 36) once daily for 3 or 5 days. Contents of serum IL-7 and IL-18 were detected by ELISA. RESULTS:In comparison with the control group, the plasma leukocyte counts from day 2 to day 5, the spleen index and serum IL-18 contents on day 3 and 5 after administration of CTX were significantly decreased, and serum IL-7 contents on day 3 and 5 after CTX remarkably increased in the model group (P < 0.05). Following the treatment, the spleen index on day 3 and serum IL-18 content on day 5 in the moxibustion group, serum IL-7 and IL-18 contents on day 3 and 5 in the acupuncture group were significantly up-regulated (P < 0.05). The effect of acupuncture was markedly superior to that of moxibustion in up-regulating serum IL-18 contents on day 3 and 5 after the treatment (P < 0.05). No significant differences were found between the acupuncture and moxibustion groups in the leukocyte counts and spleen index after the treatment, and in serum IL-7 contents on day 3 (P > 0.05). CONCLUSION:Acupuncture and moxibustion can up-regulate the serum IL-7 and IL-18 and white blood cell levels in immuno-suppression mice, wnicn may contribute to their effects in relieving immune hypofunction caused by alkylating agent CTX.
目的:观察埋线与针刺治疗颈型颈椎病临床疗效及优势.方法:符合诊断标准病例120例随机分为两组,埋线组60例,针刺组60例,依据循证医学临床研究的原则观察治疗效果,两组用相同的穴位,埋线组埋线1次,针刺组每天针刺1次,共10次,两组10 d后判断疗效,30 d后随访,按等级资料判断疗效结果.结果:埋线组有效率为96.7%、针刺组有效率为90.0%;30d远期疗效埋线组有效率为100.0%、针刺组有效率为86.7%.结论:针刺和埋线均能对颈型颈椎病产生很好的疗效,埋线疗效优于针刺,特别是远期疗效,埋线省时省事,能够满足现代快节奏生活,是治疗颈型颈椎病的有效方法.
Objective To study the content of serum granulocyte macrophage colony stimulating factor(GM-CSF)and granulocyte colony stimulating factor(G-CSF)in cyclophosphamide(CTX)-induced tumor-bearing mice before and after acupuncture and moxibustion treatment,so as to find out the mechanism of acupuncture and moxibustion treatment on recovery from myelosuppression and increasing of leukocyte counts.Methods 64 Kunming male mice of clean grade were transplanted subcutaneously S180 sarcoma cells to induce the trans-planting tumor model,then were randomly divided into control group,model group,acupuncture group and moxibustion group (each n =16)according to the body weight.Mice in model group,acupuncture group and moxibustion group were intraperitoneally injected with CTX to establish CTX-induced tumor-bearing mice mod-el,and the control group were injected intraperitoneally with normal saline.Acupoints Dazhui(GV14),Geshu (BL17),Shenshu (BL23)and Zusanli (ST36)were needled in acupuncture group and were moxa moxibusted in moxibustion group once daily at regular time for consecutive 5 d.Peripheral leukocyte counts were meas-ured by detecting the blood collected from caudal vein before modeling and at every day of treatment proce-dure.The serum concentrations of GM-CSF and G-CSF were determined by using ELISA method at the third day of treatment and one day after five-day-treatment.Results Compared with control group,peripheral leu-kocyte counts decreased,even to the lowest level at the fourth day in the three groups of CTX-induced tumor-bearing mice,and the serum concentrations of GM-CSF and G-CSF also decreased in the three groups signifi-cantly(P <0.05).At the fifth day of treatment,leukocyte counts started to increase,and the serum concen-trations of GM-CSF and G-CSF increased significantly compared with model group(P <0.05).At the day after treatment,leukocyte counts increased to nearly normal level and the serum concentrations of GM-CSF and G-CSF increased dramatically compared with model group.Conclusion CTX damaged the myeloid element to reduce peripheral leukocyte counts and the serum concentrations of GM-CSF and G-CSF.Acupuncture and moxibustion can obviously increase the concentration of GM-CSF and G-CSF,that is to say,both the Chinese medical treatment can induce colony growth of the granulocyte precursor and macrophage precursor cells and stimulate differentiation of the hematopoietic cells,leading to proliferation,differentiation and maturation of the myeloid hematopoietic cells,resulting in recovery from myelosuppression and increasing of leukocyte counts induced by chemotherapy.
Objective: To observe the clinical effect of acupoint-catgut-embedding on insomnia. Methods: Catgut embedding therapy was applied at insomnia for 35 cases, and the results were compared with the acupuncture group that included 32 cases. Results:The total efficacy was 91.4%in the treatment group higher than 78.1%in the control group. Conclusion:The therapeutic effect of catgut embedding on insomnia is superior to that of acupuncture group.

目的:从针灸对环磷酰胺(CTX)化疗小鼠骨髓造血微环境的影响,探讨针灸改善化疗后骨髓抑制,提升外周血白细胞的作用机制。方法:选用昆明种清洁级小鼠64只,称质量后依据体质量随机分为空白组、模型组、针刺组、艾灸组4组,每组8只。空白组小鼠腹腔注射生理盐水,剩余3组小鼠腹腔注射CTX溶液造成骨髓抑制模型。造模完成4h后,针刺组、艾灸组小鼠分别选取大椎、膈俞、肾俞、足三里施以针刺和艾灸,空白组、模型组陪同固定。同时分别在造模前、造模后及治疗3d、5d后割尾取静脉血,检测外周血白细胞,并分批处死小鼠取股骨内骨髓组织,用免疫组织化学法检测骨髓组织中细胞间黏附分子(ICAM-1)、血管细胞黏附分子(VCAM-1)蛋白表达。结果:针刺、艾灸均可以提高CTX化疗小鼠骨髓组织中黏附分子ICAM-1、VCAM-1蛋白表达,升高外周血白细胞。结论:针灸提高CTX化疗小鼠骨髓组织中黏附分子ICAM-1、VCAM-1蛋白表达,利于造血细胞与组织细胞之间的黏附和信息交换,增强造血细胞对细胞因子反应的敏感性,促进造血细胞的分裂、增殖、释放,应当是针灸参与机体造血调控,修复骨髓造血微环境的损伤,改善化疗后骨髓抑制,提升白细胞的重要机制之一。

目的:探讨透穴埋线法对肥胖大鼠总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)的影响,为临床运用透穴埋线法治疗肥胖及其相关疾病提供客观的实验依据。方法:将60只健康雄性SD大鼠随机分成两组,一组12只,喂以普通饲料;另外一组48只,喂以高脂饲料。6周后,选出造模成功的20只符合要求的高脂大鼠随机分成模型组10只、透穴埋线组(简称埋线组)10只;空白对照组选取普食组的10只大鼠,共3组大鼠。实验期间,各组实验大鼠均普通饲料喂养,空白对照组不做任何处理,模型对照组陪同固定,透穴埋线治疗组予透穴埋线治疗。治疗后观察并分别比较大鼠TC、TG、LDL-C、HDL-C水平的变化。结果:造模6周后,高脂组大鼠的TC、TG、LDL-C水平均明显高于空白对照组,HDL-C水平低于正常组(P<0.05);实验结束后,埋线组大鼠的TC、TG、LDL-C水平较模型组明显下降,HDL-C水平明显上升(P<0.05)。结论:经高脂饲料喂养可有效、快速的建立营养性肥胖大鼠模型,成功率达65%。透穴埋线法能降低肥胖大鼠的血清TC、TG、LDL-C水平及其他肥胖相关指标,促进脂肪代谢,提高LEP受体、INS受体的敏感性和表达,有效调控肥胖大鼠的摄食行为和脂肪代谢。
Objective:To make basic research on cupping therapy and observe the coloration law of cupping plaque on healthy persons in order to provide basis for establishing the cupping plaque map.Methods:40 healthy persons equally divided into 4 groups were given cupping therapy at 12 acupoints on the back and waist for 10 min and the pressure in the cups were-0.07MPa,-0.06 MPa,-0.05 MPa,and-0.04 MPa respectively.After 24 hours,the colors of cupping plaque were quantized and assessed and the changing law of it was analyzed.Results:With the same cupping parameter,the cupping plaque color on the lumbar was darker,which had statistical significance;under the different pressure,the shade of the cupping plaque showed-0.06 MPa>-0.07 MPa>-0.05 MPa>-0.04 MPa,while there was no statistical significance between groups of-0.06 MPa and-0.07 MPa.Conclusion:With the same cupping parameter,the coloration of cupping plaque is supposed to be the same at the same acupoints on both sides of the back,while the shade of the cupping plaque is different in different regions of the back.The cupping plaque color gets darker along with the increase of the subatmospheric pressure but the speed will slow down as the subatmospheric pressure increases to-0.06MPa.To study the coloration law of cupping plaque on healthy persons,the subjects should be selected seriously.
