Inflammatory response is a complex process of the body as it delivers the appropriate defense against harmful stimuli. lambda-carrageenan induced paw edema mice model is also known to be used in study for anti-inflammatory activity. Therefore, we established lambda-carrageenan induced acute paw edema in C57BL/6J mice and detected a variety of indexes for more roundly evaluating the effect of anti-inflammatory drug. Mice were stimulated with lambda-carrageenan for the induction of inflammatory model and orally treated with indomethacin (10 mg/kg), which as a positive drug, should be injected 1 h before lambda-carrageenan injection. Mice were divided into three groups: control group, lambda-carrageenan (CAR) group and lambda-carrageenan + indomethacin (CAR + INDO) group. Evaluation indexes of the inflammatory response in mice model were detected by paw thickness, histological analysis, serum levels of cytokines in vivo. Results showed that paw thickness was seriously swelling in CAR group, moreover it was significantly improved in CAR + INDO group compared with that in CAR group. Histological analysis also indicated that the obvious inflammatory cell infiltration and synovial hyperplasia were found in the CAR group, whereas the CAR + INDO group revealed mild clinical symptom expression. The concentration of tumornecrosis fatoralpha (TNF-alpha), interleukin-1 beta (IL-1 beta) and interleukin-17 (IL-17) in CAR + INDO group were significantly reduced compared with those in CAR group. We established acute inflammatory model and roundly evaluated a variety of indexes in C57BL/6J mice, which exhibited excellent stability and repeatability. Our research has provided a systematic research method for the study of screening and evaluation of activity in anti-inflammatory drug.
目的:评价葡萄糖-6-磷酸异构酶(GPI)、抗环瓜氨酸肽(CCP)抗体、抗突变型瓜氨酸波形蛋白(MCV)抗体、抗聚角蛋白微丝蛋白抗体(AFA)在类风湿关节炎(RA)诊断中的意义,并比较多种指标联合检测在RA诊断中的意义.方法:对178例确诊为RA的患者进行血清学指标检测,并进行回顾性分析.血清GPI、抗CCP抗体、抗MCV抗体、AFA检测均采用酶联免疫吸附法,采用 χ2检验和Pearson相关分析对多种指标在RA诊断中的价值及相关性进行比较,并对不同指标间的联合检测进行讨论分析.结果:GPI、抗CCP抗体、抗MCV抗体和AFA在RA组的特异性和灵敏度分别为65.00%和70.22%、95.00% 和84.83%、78.33% 和72.47%、75.71% 和63.48%.RA组中GPI、抗CCP抗体、抗MCV抗体和AFA阳性率分别为70.22%、84.83%、72.47%、63.48%,显著高于对照组的11.67%、5.00%、21.67%、13.33%,差异有统计学意义(P<0.001);任意3种或4种指标联合检测特异性较高,但敏感度降低,明显低于单项检测抗CCP抗体,差异有统计学意义(P<0.001);任意3种或4种指标联合检测较单一指标检测均具有较高的阳性率,差异有统计学意义(P<0.001),尤其是GPI+抗CCP抗体+抗MCV抗体+AFA联合检测,RA组中阳性率可达96.07%.结论:GPI、抗CCP抗体、抗MCV抗体和AFA均可作为RA诊断的血清学指标,多种血清学指标联合检测有助于提升RA诊断的阳性率.
目的:了解本院尿路感染常见病原菌的分布及耐药情况,为临床合理选用抗菌药物提供依据。方法:对本院2010年1月至2011年12月住院及门诊患者尿培养阳性非重复分离的268株病原菌进行回顾性分析。结果:尿路感染常见病原菌以革兰阴性杆菌为主有177株(66.0%),其中尤以大肠埃希菌分离率最高有117株占革兰阴性杆菌的66.1%,产超广谱β-内酰胺酶(ESBLS)的大肠埃希菌有72株占大肠埃希菌的61.5%,产ESBLS大肠埃希菌的耐药率明显高于非产ESBLS株。革兰阳性球菌81株(30.2%),革兰阳性球菌耐药情况也较严重,真菌10株(3.7%)。结论:尿路感染常见病原菌以革兰阴性杆菌为主,临床应重视病原学检查,根据耐药特点合理使用抗菌药物,以得到有效的治疗效果。
目的:了解铜绿假单胞菌各病区分布情况及耐药性变迁,预防和控制院内感染的发生,减缓耐药菌株的出现,延长现有抗生素的使用期限。结果:临床分离铜绿假单胞菌103株,38株来自呼吸科,81株来自痰标本,产金属酶的有7株,占分离菌株的6.8%,其中4株分离自神经外科昏迷伴器官切开的患者痰液中,3株分离自ICU病房。结论:分离的铜绿假单胞菌科室以呼吸科居多,标本类型以痰居多;耐亚胺培南的铜绿假单胞菌对许多抗生素的耐药率均高于敏感株。
目的:观察2型糖尿病患者早期肾病生化指标的变化。方法:将2型糖尿病339例按照尿微量清蛋白排泄是否≥20μg/min分为糖尿病早期肾病组(观察组)164例,无糖尿病早期肾病组(对照组)175例。比较两组血清胱抑素C(Cys C)、糖化血红蛋白、血清肌酐、血清β2微球蛋白、尿β2微球蛋白和尿α微球蛋白水平的差异。结果:观察组血清Cys C、尿β2微球蛋白和尿α微球蛋白水平均非常显著高于对照组(P<0.01);糖化血红蛋白、血清肌酐及血清β2微球蛋白水平均有不同程度升高,但两组差异不显著(P>0.05)。Logistic回归分析结果显示,血清Cys C、尿β2微球蛋白和尿α微球蛋白与糖尿病早期肾病显著相关(P<0.05)。结论:血清CysC、尿β2微球蛋白及尿α微球蛋白水平升高可作为糖尿病早期肾损害的指标,有利于糖尿病早期肾病的诊断和治疗。
