目的 从网络视角出发,探讨枸骨叶抗肿瘤的的可能作用机制,为其临床使用提供依据.方法 通过中药系统药理学数据库与分析平台(TCMSP)、中医百科全书数据库(ETCM)及查阅相关文献,获取枸骨叶的有效成分及潜在靶点.利用人类基因组注释数据库(GeneCards)结果对比,获得枸骨叶与肿瘤重合的潜在靶点基因,并通过Cytoscape 3.6.0软件构建枸骨叶的"候选成分-作用靶点"网络.利用String数据库和Cytoscape 3.6.0软件构建药物抗肿瘤的蛋白质相互作用(PPI)网络;并通过分子对接验证和筛选潜在的核心化合物,利用DAVID生物信息资源数据库对核心靶点进行基因本体论(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析.结果 共筛选出枸骨叶抗肿瘤的作用的靶点137个,对应13种有效成分.关键靶点涉及TP53、MAPK1、STAT3等.主要通过调控肿瘤坏死因子(TNF)信号通路、缺氧诱导因子-1(HIF-1)信号通路、磷脂酰肌醇3-激酶(PI3K/AKT)信号通路、肿瘤抑制因子P53信号通路、T细胞受体信号通路等发挥抗癌作用.涉及的癌种包括前列腺癌、胰腺癌、非小细胞肺癌等.分子对接结果显示,长梗冬青苷与MAPK1能稳定地结合并通过蛋白氨基酸Gly32、Glu33、Gly34、Tyr36等发生相互作用.结论 枸骨叶抗肿瘤作用的主要机制涉及槲皮素、熊果酸、长梗冬青苷、芒柄花黄素为代表的多成分,以TP53、MAPK1及STAT3为代表的多靶点,以及细胞增殖、细胞凋亡、信号传导为代表的多通路.
目的:观察中药通络壮骨散外敷治疗乳腺癌内分泌治疗后骨质疏松的疗效.方法:220例乳腺癌内分泌治疗后骨质疏松患者,随机分为治疗组和对照组,每组110例,对照组口服碳酸钙D3片,治疗组在对照组基础上外敷通络壮骨散.疗程均为25周,对比治疗前后腰椎骨密度的变化.结果:两组骨密度均增加(P<0.05),治疗组骨密度高于对照组(P<0.05);并两组的I型原骨胶原蛋白N-端肽(s-PINP)均降低(P<0.05),治疗组s-PINP低于对照组(P<0.05);两组碱性磷酸酶(ALP)降低、骨钙素(BGP)增加(P<0.05),治疗组ALP低于对照组,BGP高于对照组(P<0.05).结论:钙剂联合中药通络壮骨散外敷治疗乳腺癌内分泌治疗后骨质疏松效果更好.
目的:观察四神丸治疗脾肾阳虚型肠易激综合征的疗效.方法:48例脾肾阳虚型肠易激综合征患者,随机分为A组和B组,各24例.A组口服四神丸汤药治疗,B组常规口服培菲康治疗.比较两组疗效.结果:A组总有效率为83%,B组总有效率为67%,两组总有效率比较差异有统计学意义(P<0.05).结论:四神丸对脾肾阳虚型肠易激综合征患者脾胃功能恢复有良好效果.
Objective To determine the effect of water extracts of Zuojinfang(ZJF) and Fanzuojinfang(FZJF) on SHSY5 Y cells apoptosis induced by MPP+. Methods SH-SY5 Y cells were cultured in vitro and MPP+ induced SHSY5 Y cells apoptotic model was established and treated with water extracts of ZJF and FZJF. The cell activity was measured by MTT test, the cell apoptosis rate was detected by flow cytometry, and the morphology was observed by Hoechst 33258 staining. Results The apoptosis of SH-SY5 Y cells was induced by 0.5 mmol·L- 1 MPP+ for 48 h, MTT decreased remarkably to(50.87±2.31)%. the apoptosis rate increased to(43.38±1.49)%, and Hochest staining showed cell nuclear condensation, granular fluorescence and other typical features of apoptosis. After the cells were incubated with ZJF/FZJF and 0.5 mmol·L- 1 MPP+, the apoptosis was increased obviously. When the carnitine concentration increased to 2.0 mg·m L- 1, the cell activity increased to(63.16±1.89)% and(75.19±1.97)%, and the percentages of apoptosis decreased to(20.17±1.43)% and(9.41±1.27)% respectively. The difference in comparison with the model group(43.38±1.49)% was statistically significant(P 0. 01). Conclusion Both ZJF(cold) and FZJF(hot) can withstand the apoptosis of SH-SY5 Y cells induced by 0.5 mmol·L- 1 MPP+, where the effect of FZJF is stronger than that of ZJF under the same conditions. With different natures of transitional Chinese medicine, the anti-apoptotic effect is different.
目的研究左金方和反左金方水提物对四氢吡啶离子(MPP+)诱导SH-SY5Y细胞凋亡的影响。方法体外培养SH-SY5Y细胞,建立MPP+致SH-SY5Y细胞凋亡的模型,同时给予左金方和反左金方水提物干预。应用MTT比色法检测细胞活力,流式细胞仪检测细胞凋亡率,Hochest染色观察凋亡细胞形态学的改变。结果0.5 mmol·L^-1 MPP+作用于SH-SY5Y细胞60 h可诱导细胞凋亡,细胞存活率降低至(50.87±2.31)%,凋亡率增加至(43.38±1.49)%,Hochest 33258染色可见明显的颗粒状和固缩状荧光。左金方和反左金方水提物与0.5 mmol·L^-1 MPP+同时处理SH-SY5Y细胞后,能够明显改变细胞的凋亡学特性,当药物浓度增加到2.0 mg·m L^-1时,细胞存活率分别升高至(63.16±1.89)%和(75.19±1.97)%,与模型组比较,差异均有统计学意义(P〈0.01);其凋亡率由模型组的(43.38±1.49)%分别降低到(20.17±1.43)%和(9.41±1.27)%(P〈0.01),反左金方(热性)抗凋亡的作用更强。结论左金方(寒性)和反左金方(热性)均能对抗由MPP+诱导的SH-SY5Y细胞的凋亡,而反左金方(热性)抗凋亡作用更强,中药药性不同,其抗凋亡作用亦不同。
目的 研究左金方诱导人胃癌细胞SGC-7901凋亡的作用机制.方法 采用MTT比色法观察左金方对SGC-7901的生长抑制情况;流式细胞仪PI单染亚二倍体峰分析细胞凋亡率;Hoechst 33258染色观察细胞形态学变化;Western blot方法检测Bcl-2、Bax蛋白表达以及JC-1试剂盒测定细胞膜电位.结果 左金方能抑制细胞活性,与时间-剂量呈依赖关系.0.1、0.5和1.0 mg·mL-1左金方作用于SGC-7901细胞48 h后,细胞凋亡率分别为(11.60±1.24)%、(21.80±0.28)%、(36.40±1.59)%,与对照组(2.66±1.33)%比较差异均有统计学意义(P<0.01);Hochest染色细胞出现核固缩状和颗粒状荧光等典型的凋亡学特征;左金方可降低SGC-7901细胞Bcl-2基因表达和增加Bax基因表达;左金方作用SGC-7901 4 h后,可以明显引起细胞MMP的下降;1.0 mg·mL-1左金方组的作用尤其明显.结论 左金方通过上调Bax/Bcl-2比率,下调MMP诱导了人胃癌细胞SGC-7901的凋亡.
Objective: To investigate the protective effects of water extracts of Zingiber officinale,Rheum palmatum and Salvia miltiorrhiza on SH-SY5Y cells apoptosis induced by Aβ25-35.Methods: SH-SY5Y cells were cultured in vitro and Aβ25-35 induced SH-SY5Y cells apoptotic model was established and treated with Water extracts of Z.officinale,R.palmatum and S.miltiorrhiza.The cell Viability was measured by MTT test,the morphology of SH-SY5Y cells was observed by Hoechst33258 staining and the percentage of apoptosis by FCM.Results:The cell activity decreased remarkably to(65.69±3.26)%(P0.01 vs.control) induced by 0.5 mmol/L Aβ25-35 for 72 h and were positively stained with fluorochrome Hoechst 33258,presenting the percentage of apoptosis as(45.17±3.84) %(P0.01 vs.control),However,water extracts of Z.officinale obviously improved MTT value,reduced the percentage of apoptosis,and water extracts of R.palmatum and S.miltiorrhiza.almost had no effect on the apoptosis of SH-SY5Y cells.Conclusion: Z.officinale has markedly protective effects on SH-SY5Y cells apoptosis induced by Aβ25-35,but R.palmatum and S.miltiorrhiza.show no protective effects,as the different natures of TCM,the anti-apoptotic effects of TCM are different neither.
