Objective:To measure serum cell-free DNA (cf-DNA) and donor-derived cell-free DNA (ddcf-DNA) in simultaneous pancreas-kidney transplantation recipients by the next generation sequencing technology and evaluate the relationship between ddcf-DNA and early acute rejection (AR).Methods:A total of 20 recipients who underwent simultaneous pancreas-kidney transplantation in the Department of Organ Transplantation, the Second Affiliated Hospital of Guangzhou Medical University from June 2019 to June 2021 were enrolled. According to whether there was AR after operation, the 20 recipients were divided into two groups: control group (10 cases) and AR group (10 cases). Three recipients in the control group were selected as the observation group by the random number table. The serum levels of ddcf-DNA were measured before transplantation, 2 h after recanalization of blood vessel of transplanted pancreas, and 1, 3, 7, 14, 30, and 60 d after transplantation. The levels of serum ddcf-DNA in the other 7 recipients in the control group were measured before and 14 and 30 d after transplantation. Meanwhile, the testing time points of serum ddcf-DNA in AR group were as follows: before operation, during rejection and after reversal of rejection. The measurement data of normal distribution between the two groups were compared by group t test. Fisher′s exact probability method was used for counting data comparison. P<0.05 means the difference was statistically significant.Results:The length of ddcf-DNA fragment was 110-350 bp. The length of cf-DNA fragment from the recipients was 100-420 bp. Three recipients in the observation group were marked as cases 1, 2 and 3, respectively. The content of ddcf-DNA of the 3 recipients before operation were 0.49%, 0.55% and 0.32%, respectively. The content of ddcf-DNA increased sharply to 12.05%, 10.64% and 11.05% 2 h after recanalization, and reached its peak after blood flow restored. At the first day after transplantation, the level of ddcf-DNA showed a significant downward trend, and reached a stable state about 30 d after transplantation. The content of ddcf-DNA was (0.71%±0.09%) in the control group and (0.66±0.10)% in the AR group. There was no significant difference between the two groups (t=1.126, P>0.05). The time of diagnosis of rejection in AR group was (17±4) d after transplantation, and the content of serum ddcf-DNA was (5.4±2.0)% when AR was confirmed; while the content of serum ddcf-DNA in the control group was (1.70±0.48)% 2 weeks after transplantation, and the difference was statistically significant (t=-5.59, P<0.05). The rejection was reversed and the recipients condition were stable at (27 ± 4) d after transplantation in the AR group, at which time the content of ddcf-DNA was (0.93 ± 0.32)%. The content of ddcf-DNA in the control group was (0.87±0.25)% on the 30th day after transplantation, and the difference was not statistically significant between the 2 groups (t=-0.497, P>0.05).Conclusions:The serum ddcf-DNA in simultaneous pancreas-kidney transplantation recipients increased significantly when AR occurred. When AR was reversed, the content of ddcf-DNA showed a significant downward trend.
目的:研究人参对大鼠创面愈合的作用及血清TNF-α、IL-1β、TGF-β1的影响。方法:30只SPF级8周龄SD大鼠随机均分为健康对照组、模型安静组、模型人参组,雌雄各半。麻醉及消毒后切除大鼠背部2 cm方形全层皮肤造模。伤后第3天开始人参灌胃,持续14 d。常规观察和测量创面愈合情况;伤后第10天、第17天断尾取血备血清,ELISA法测IL-1β、TNF-α、TGF-β1。结果:人参使皮肤创面愈合时间加快(2±0.5)d,愈合质量较好;伤后第10天、第17天人参组血清TNF-α、IL-1β水平均明显低于模型安静组(P〈0.05);TGF-β1水平在第10天明显高于模型安静组(P〈0.05),第17天与模型安静组比较无显著性差异(P〉0.05)。结论:人参可能通过改善创面炎症免疫反应及TGF-α、IL-1β、TGF-β1相关通路促进创面愈合。
Objective: To study the effect of ginseng on wound healing and serum TNF-α,IL-1β,TGF-β1 of rats.Methods: 30 SPF SD rats aged 8 weeks were randomly divided into control group,model of quiet group,model of ginseng group,half male and half female.2cm squares of rat back skin were cut off to make model after anesthesia and disinfection.On the 3rd day after injury,the rats were given gastric perfusion of ginseng for 14D.Regular observation and measurement of wound healing.On the 10th day and 17th day after injury,the tail was cut to take blood serum;ELISA method was used to test IL-1β,TNF-α,TGF-β1.Results: Ginseng made the healing time of skin wound increase 2 ± 0.5 d,the healing quality was better;On the 10th day and 17th day after injury,serum levels of IL-1β,TNF-α in ginseng group were significantly lower than those of model group(P<0.05);TGF-β1 level on the 10th day was significantly higher than that in model group(P<0.05),on the 17th day,there was no significant differences(P>0.05) compared with the model group.Conclusion: Ginseng may promote the wound healing by improving the wound inflammatory immune response and the relative pathways of TGF-α,IL-1β,TGF-β1.
Objective According to the intervention of far infrared ray physiatrics in traumatized rats to explore the immune and the cytokine mechanisms of the far infrared ray intervention.Methods By selecting 30 SPF grade healthy SD big rats of eight years old as experimental subjects,randomly divided into health+quiet group(A),injured+quiet group(B),injured+ infrared+ quiet group(C).The method created by Fu Xiao-bing adopted,after mold-building treatment,the rats were cut off tails to obtain blood,centrifugated to get blood serum,measuring IL-1β,TNF-a,TGF-β1 with ELISA method.Results After mold building,the levels of IL-1βand TNF-α were increased obviously in injured rats.In group C,the levels of IL-1βand TNF-α were significantly lower than those in group B(P0.01).TGF-β1 in group C was significantly higher than that in group B(P0.05).Conclusion The intervention of far infrared ray can improve immuno-stress ability of injured rats,weaken the persistent inflammatory reaction of wound surface,promote fibroblastic proliferation.The immune function coincides with the changed TGF-β1.

通过对14名我国女子优秀拳击运动员进行运动创伤调查分析,旨在寻找预防或减少创伤的措施,采用问卷调查及访谈形式获取资料并进行常规数理统计和分析,结果表明:总伤病率为100%,损伤多发生于眼、鼻、手等部,自我保护意识欠缺是首位损伤原因。此外,与过度疲劳与带伤训练、准备活动不够充分亦有明显关系。
