目的:观察扶土疏木解毒汤对同时行化疗及靶向治疗的晚期大肠癌患者免疫功能及生存质量的影响.方法:将60例晚期大肠癌患者随机分为治疗组与对照组,每组30例.对照组予化疗+靶向治疗(贝伐珠单抗联合FOLFIRI),治疗组在对照组治疗的基础上加用中药汤剂扶土疏木解毒汤口服,均连续治疗8周后观察疗效.比较2组患者实体瘤疗效评价结果及治疗前后中医证候积分、行为状态评分(Karnofsky评分)、免疫功能(CD4+、CD8+、CD4+/CD8+)、生存质量评分变化情况.结果:治疗后治疗组实体瘤疗效评价总有效率(ORR)为60.00%、疾病控制率(DCR)为83.33%,对照组ORR为46.67%、DCR为80.00%,组间比较差异均无统计学意义(P>0.05).治疗后治疗组患者乏力、纳差等单项中医证候评分及总分均较治疗前明显降低(P<0.05),也明显低于对照组治疗后(P<0.05),而对照组上述评分治疗前后比较差异均无统计学意义(P>0.05).治疗组治疗后患者Karnofsky评分较治疗前明显升高(P<0.05),也明显高于对照组治疗后(P<0.05),而对照组治疗前后Karnofsky评分比较差异无统计学意义(P>0.05).治疗后,对照组患者CD4+、CD4+/CD8+均较治疗前明显降低(P<0.05),也明显低于治疗组(P<0.05),而治疗组CD4+、CD8+、CD4+/CD8+指标治疗前后比较差异均无统计学意义(P>0.05).治疗后,治疗组的生理状况、情感状况、功能状况、附加关注评分及生存质量总分均较治疗前显著升高(P<0.05),也明显高于对照组治疗后(P<0.05).结论:扶土疏木解毒汤辅助化疗、靶向治疗可显著改善晚期大肠癌患者临床症状,稳定免疫功能,提高患者的生存质量.
目的 观察健脾养正消癥方加减联合安罗替尼二线治疗晚期食管癌的临床疗效.方法 选取经病理组织学检查确诊的60例经一线化疗后进展的晚期食管癌患者为研究对象,将其随机分为对照组(30例)和试验组(30例).对照组给予安罗替尼12 mg,1次/d,连服2周,停药1周,2个周期后评价疗效.试验组在安罗替尼治疗基础上,以健脾养正法为理论指导采用健脾养正消癥方加减.观察2组疗效、不良反应及临床症状的改善情况.结果 试验组卡氏评分(KPS)评定的生活质量好转率为46.67%(14/30),高于对照组的20.00%(6/30),差异有统计学意义(P<0.05).试验组疲乏、高血压及腹泻的发生率分别为46.67%、30.00%和23.33%,分别低于对照组的56.67%、40.00%和40.00%,差异有统计学意义(P<0.05).试验组客观缓解率(ORR)为26.67%,高于对照组的20.00%;试验组疾病控制率(DCR)为70.00%,高于对照组的63.33%,但差异无统计学意义(P>0.05).试验组恶心、困倦、食欲改善情况优于对照组,差异有统计学意义(P<0.05).结论 健脾养正法联合安罗替尼治疗晚期食管癌能明显提高患者生活质量且不良反应可耐受,可以作为晚期食管癌的有效姑息治疗方法.
目的:观察健脾养正法联合促红素治疗晚期胃癌肿瘤相关性贫血的临床疗效.方法:选取经病理组织学确诊的44例晚期胃癌患者,随机分为对照组(22例)和实验组(22例).对照组给予促红素150 IU/kg,皮下注射,每周3次,8周后评价疗效.实验组在促红素治疗基础上,以健脾养正法为理论指导,服用健脾养正消癥方加减.观察两组血液学指标变化及临床症状的改善情况.结果:实验组患者治疗前后红细胞计数、血红蛋白及血细胞比容均明显升高,差异有统计学意义(P<0.05).反映T细胞免疫功能的指标CD4+/CD8+比值明显升高,差异有统计学意义(P<0.05).实验组KPS评分好转率、疲乏症状评分改善率、恶心、抑郁、焦虑、困倦、食欲症状的改善情况均优于对照组,差异有统计学意义(P<0.05).结论:健脾养正法联合促红素对于晚期胃癌肿瘤相关性贫血治疗效果较好,能明显提高患者生活质量,可以作为晚期胃癌肿瘤相关性贫血的有效姑息治疗方法.
目的:基于网络药理学探索川楝素对胃癌MKN-28细胞增殖的影响及其分子机制.方法:通过网络药理学筛选出川楝素治疗胃癌的分子通路;MTT法观察川楝素对人胃癌MKN-28细胞增殖抑制的量效关系;Hoechst 33258染色法观察川楝素对细胞凋亡的影响;Western Blot法检测凋亡及丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)信号通路相关蛋白表达.结果:网络药理学方法提示川楝素通过MAPK信号通路作用于胃癌,川楝素能显著抑制人胃癌MKN-28细胞增殖,并能诱导人胃癌MKN-28细胞凋亡.Western Blot实验提示,随着给药剂量增加,Cleaved Caspase-3、Cleaved-PARP、p38和p-p38表达量递增.结论:川楝素能高效抑制人胃癌MKN-28细胞增殖;川楝素是通过激活MAPK通路,诱导细胞凋亡,发挥其抗肿瘤药理作用.

Objective: To discuss the inhibitory effects of replenishing-Qi invigorating-spleen detoxifying prescription on gastric cancer cells and its mechanism. Methods: The inhibitory effects of different concentrations of replenishing-Qi invigorating-spleen detoxifying prescription on the proliferation of gastric cancer cell line after the prescription acting on them in vitro for different times were observed by MTT method; the inducing effects of different concentrations of replenishing-Qi invigorating-spleen detoxifying prescription on the apoptosis of human gastric cancer cell lines were studied by Annexin-V/PI double-staining method; cycle arresting effects of the prescription on human gastric cancer cell line were detected by flow cytometry; the changes of Bax, Bcl-2 and Caspase-3 were observed by Western blot method. Results:The prescription showed better inhibitory and proliferative effects to MGC-803 cells of human gastric cancer cell line (P <0.05), and it was in a dose-dependent manner; after the prescription acting on human gastric cancer cells for 24 hours, early apoptotic rate of the cells presented the rising trend as the medication concentrations increased, and it was in a dose-dependent manner; it demonstrated that the prescription could arrest MGC-803 cells at S stage, which was in a dose-dependent manner; the expressions of Bcl-2 and Caspase-3 in the groups of 4, 2 and 1 mg/mL decreased; while the expressions of Bax and Cleaved-caspase-3 in the groups of 4, 2 and 1 mg/mL increased. Conclusion: Replenishing-Qi invigorating-spleen detoxifying prescription could inhibit the proliferation of human gastric cancer cell line and induce its apoptosis, kill tumor cells, which might be realized by triggering cascade reaction of caspase, lowering Bcl-2 and increasing Bax.
Objective: To investigate the relationship between the expression of RNA-binding protein 38 (RBM38) and distant metastasis and prognosis of breast cancer. Methods: A total of 161 cases of breast cancer tissues were selected, and the expression of RBM38 was detected by immunohistochemical SP method. The relationship between the expression of RBM38 and the distant metastasis and prognosis of breast cancer patients were analyzed. Cox regression model was applied to analyze the influence factors of distant metastasis of breast cancer. Results: Among the 161 breast cancer patients, high expression of RBM38 was observed in 85 cases and low expression was observed in 76 cases. Log rank test showed that the distant metastasis rate of RBM38 highly expressed group was significantly lower than that of RBM38 lowly expressed group (P=0.010), while the survival rate was significantly higher (P=0.009). Univariate Cox regression analysis showed that the pathological stage (P=0.001), axillary lymph node positive (P=0.001), and the expression of RBM38 (P=0.015) were dramatically related to the distant metastasis of breast cancer patients. Multivariate Cox regression analysis showed that positive axillary lymph node was an independent risk factor (RR=4.222, 95% CI: 1.235-14.432), and high expression of RBM38 was an independent protective factor (RR=0.328, 95% CI: 0.128-0.839) of distant metastasis of breast cancer patients. Conclusion: High expression of RBM38 is positively related with low distant metastasis rate and good prognosis of breast cancer patients. Hence, high expression of RBM38 is an independent protective factor of distant metastasis.
Objective:To investigate the effect of Jianpi Shugan Kangdu decoction (JSKD) on the growth and metastasis of MDA-MB-231-pAcGFP xenograft tumors in nude mice,and explore its mechanism.Method:Human triple-negative breast cancer cells (MDA-MB-231) were transfected with pAcGFP-C1 by fugene HD transfection reagent,and then the breast cancer cells with stable expression of green fluorescent protein (MDA-MB-231-pAcGFP) were obtained by geneticin (G418) screening.The percentage of MDA-MB-231-pAcGFP was detected by flow cytometry (FCM).The mice bearing MDA-MB-231-pAcGFP cells were obtained with subcutaneous inoculation,and then the volume and the weight of the xenograft tumors were observed by using fluorescence imaging technology.Result:The transfection rate of the MDA-MB-231-pAcGFP cells was 96.5% and the rate of tumor formation of these cells was 100%.The average weight of the xenograft tumors was (1.30 ± 0.11) g (group A,model group),(0.75-±0.12) g (group B,JSKD group),(0.48 ±0.09) g (group C,CXCR4 inhibitor group) and (0.44 ± 0.07) g (group D,JSKD + CXCR4 inhibitor group),respectively,and their tumor volumes were (6.635 ± 0.500),(4.017 ± 0.466),(2.664 ± 0.700),(2.323 ± 0.492) cm3,respectively.The tumor weight and volume were changed in three treatment groups as compared with the model group (P <0.05).The tumor inhibition rates were 42.18%,62.95%,66.28% in three treatment groups,respectively.Fluorescent metastatic nodules were found in liver of group A and group C,however,fluorescent metastatic nodules were not found in liver of group B and group D.Conclusion:Jianpi Shugan Kangdu decoction could significantly inhibit the growth of the MDA-MB-231-pAcGFP xenograft tumors and prevent tumor metastasis in liver,providing experimental basis for the prevention and treatment of triple-negative breast cancer by Jianpi Shugan Kangdu decoction.
Objective To explore the effect of Jianpi Yangzheng Xiaozheng recipe on tumor abnormal protein (TAP) and T cell subset in patients with gastric cancer. Method 82 cases of gastric cancer patients were enrolled in the research as study group or control group, as per a random number table, with 41 cases in each. The study group was given com-bined treatment of Jianpi Yangzheng Xiaozheng recipe and systemic chemotherapy, while the control group was treated with chemotherapy alone. The TAP and T cell subset before treatment, every 2 cycles during the 4-cycle treatment were compared between two groups, respectively. Result The TAP of study group in every 2 two cycles during the whole course declined significantly compared to that before treatment, the difference was statistically significant (P<0.05);in T subset, after 2 cycles and 4 cycles, the ratio of CD4+/CD8+of study group showed no significant decrease as compared with that before treatment (P>0.05), but the ratio in control group after 4 cycles of treatment reduced significantly (P<0.05). Conclusion The combined therapy of Jianpi Yangzheng Xiaozheng recipe and systemic chemotherapy could low-er the level of TAP, which is associated with metastasis, meanwhile, it improves the positive immune function by elevat-ing CD4+/CD8+and CD4+levels, minimizing the risk of metastasis and recurrence as compared with simple chemotherapy, and may improve the cellular immune function of gastric cancer patients.
Objective:To explore the clinical effects of acupuncture and moxibustion combined with modefied Siwu decoction in the treatment of adenomyosis. Methods:Total 60 cases of patients with adenomyosis were randomly divided into the acupuncture and moxibustion group and the acupuncture and moxibustion com-bined with modefied Siwu decoction group,both groups received the treatment of acupuncture and moxibus-tion,at the meanwhile the latter group taking modefied Siwu decoction. Dynamic differences of visual ana logue scale(VAS),menstrual flow,uterine volume and uterus length between the two groups were observed. Results:After treatment of 3 and 6 months,the VAS,menstrual flow,uterine length and uterine volume in the two groups were reduced ( P<0 . 05 ) . There were significantly differences in VAS , menstrual flow , uter ine length and uterine volume between the two groups after treatment of 6 months (P<0.05). Conclusion:The treatment of acupuncture and moxibustion combined with modefied Siwu decoction could effectively re-lieve the dysmenorrhea and menorrhagia in patients with adenomyosis ,with significantly analgesic effect.

Objective:To compare the efficacy and safety between raltitrexed and irinotecan with FOLFIRI for pa-tients with advanced colorectal cancer. Methods:A total of 44 patients with advanced colorectal cancer were divided randomly into two groups. There were 22 patients for FOLFIRI regimen group( control group:irinotecan 180mg/m2 iv d1 ,leucovorin 400mg/m2 iv d1 ,fluorouracil 400mg/m2 iv d1 ,fluorouracil 2000mg/m2 iv 46h d1 )and 22 patients for raltitrexed plus irinotecan regimen group(treatment group:irinotecan 180mg/m2 iv d1,raltitrexed 3mg/m2 iv d1). Both two regimens were 21 days as a cycle,and a total of 2 cycles was applied. The efficacy and side effects were evaluated after 2 cycles. Results:Patients in control group received 3 PR,5 SD,14 PD to contrast treatment group 4 PR,6 SD, 12 PD,no CR case. No statistical difference was found between the two groups in regard to response rate(13. 6% vs. 18. 2%)and disease control rate(36. 4% vs. 45. 5%). FOLFIRI regimen had higher frequency hypodynamia,periph-eral nerve venenosity,mucositis and angiitis(p﹤0. 05),while raltitrexed plus irinotecan regimen had higher frequen-cy of hemophthisis(p﹤0. 05). Conclusion:Raltitrexed plus irinotican regimen is effective for metastatic colorectal cancer.
目的:观察疏肝健脾解毒汤配合T ACE术治疗原发性肝癌患者的疗效及安全性。方法:用疏肝健脾解毒汤联合T ACE术(治疗组)与单纯采用 T ACE术(对照组)治疗原发性肝癌患者46例,比较治疗后两组患者肿瘤大小的改变、临床症状的缓解及不良反应的程度。结果:疏肝健脾解毒汤配合T ACE术治疗原发性肝癌治疗组近期客观疗效的有效率(CR+ PR)与对照组相比差异无统计学意义。中医证候疗效的有效率治疗组与对照组比较差异有统计学意义。治疗组副反应的发生率均较对照组低,其中血红蛋白减少、白细胞减少和肝功能的损害治疗组与对照组相比,差异有统计学意义。治疗组的严重不良反应(III度,IV度)发生率与对照组相比,差异有统计学意义。结论:疏肝健脾解毒汤配合 T ACE术治疗原发性肝癌,在改善患者的临床症状,提高生活质量,减轻不良反应的方面有一定作用。
Objective To evaluate the clinical effi cacy and adverse reactions of perfusion with mannatide plus cytotoxic drugs (cisplatin and fluorouracil) in treatment of malignant effusion.Methods 44 patients with malignant effusion were randomly divided into two groups. All patients underwent serous cavity puncturation, continuously draining the effusion via central venous catheter. Then 23 cases in treatment group were treated with mannatide (40~50 mg) plus cisplatin (40~60 mg) orfl uorouracil (0.5~1.0 g) through the central venous catheter; 21 cases in control group only received cisplatin (40~60 mg) orfl uorouracil (0.5~1.0 g). The treatment for two groups was repeated once per week and continued for 2 weeks.Results The response rate and the disease control rate of the treatment group were 47.8% (11/23) and 73.9% (17/23), better than 38.1% (8/21) and 66.7% (14/21) of the control group, but there was no signifi cant statistic difference between the two groups (P>0.05). Life quality improvement rate of the treatment group was still better than the control group (P<0.05). The main side effect of two groups were myelosuppression, gastrointestinal reaction and regional pain.Conclusion Perfusion of mannatide plus cytotoxic drugs to treat malignant effusion is a certain effective and hypotoxic therapy, and can also improve the life quality of patients.
Objective:To study the effect of every Chinese herb in Jianpi Huayu Recipe on the CEA of the colon cancer cell.Methods: Sandwich ELISA and Immunohistochemical detection were used to detect the CEA of the cancer cell culture supernatant.Results: Chinese herbs of Jianpi Huayu Recipe all could inhabit the growth of colon cancer cell,and with the decreasing concentration of herbs,effect correspondingly also reduced.Used the method of Sandwich ELISA and immunohistochemical detection,the expression of CEA can be decreased by all herbs.ZhiJun and ZaoXiu were the most significant in reduc the expression of CEA.Conclusion: All Chinese herbs of Jianpi Huayu Recipe can inhibit the expression of CEA of the colon cancer cell.

Objective:This study was designed to explore:①The effect of Jianpi Huayu Fang on the biological behaviors including proliferation,adhesion and invasion capabilities of human hepatoma cell line HepG2 in vitro.②The effect of Jianpi Huayu Fang in vitro on the apoptosis of human hepatoma cell line HepG2.Methods:①After human hepatoma cell line HepG2 was treated with Jianpi Huayu Fang at different concentrations,cell proliferation and adhesion capabilities was assessed using MTT assay.The invasion capabilities of HepG2 cells were determined using Transwell method(Boyden chamber),respectively.②Flow cytometry,AnnexinV/PI double staining method was used to observe the early apoptosis induced by different concentrations of Jianpi Huayu Fang in human hepatoma cell line HepG2.Results:①Jianpi Huayu Fang significantly inhibited the proliferation of HepG2 cells in a concentration-dependent manner.The inhibition ratio of cell proliferation in different concentration of Jianpi Huayu Fang had significantly different at the same time(P0.01).②Jianpi Huayu Fang(2mg/mL,1mg/mL)significantly decreased the adhesion capability of HepG2 cells at 40,60,and 120min,respectively(P0.01).Jianpi Huayu Fang significantly decreased the adhesion capability of HepG2 cells(P0.01).③Jianpi Huayu Fang applied on human hepatoma cell line HepG2 for 48h,induced the apoptosis of human hepatoma cell line HepG2 with a degree of concentration-dependence(P0.01).Conclusion:①Jianpi Huayu Fang inhibited the proliferation of human hepatoma cell line HepG2.②Jianpi Huayu Fang supressed the adhesion and invasion capability of HepG2 cells.③Jianpi Huayu Fang induced the apoptosis of human hepatoma cell line HepG2.
目的:探讨参芪重楼合剂体内对鸡胚绒毛尿囊膜(CAM)血管网生长的影响.方法:利用CAM模型观察参芪重楼合剂对CAM血管网的形成的作用.结果:参芪重楼合剂50 mg/mL对(CAM)血管形成的抑制作用与环磷酰胺0.4 mg/mL的抑制作用相当,均为100%,参芪重楼合剂25、12.5、6.25 mg/mL对于(CAM)血管形成的抑制作用分别为70%、30%、10%.结论:参芪重楼合剂对CAM血管网的形成具有抑制作用.
OBJECTIVE To explore into the in vitro effect of drugs for invigorating the spleen and removing blood stasis on tumor cells in the alimentary tract and the coordinating effect of drugs for invigorating the spleen and drugs for removing blood stasis and toxins.METHOD MTT method was used to observe the effect of the drugs in different concentrations,and WEB coefficient method was used to evaluate the coordinating effect of the two kinds of drugs.RESULTWith the exception of Radix Astragali seu Hedysari and Rhizoma Curcumae,all the other drugs in the prescription had tumor-inhibiting effect,and the coordinating effect occurred 48 hours after drug administration.CONCLUSION Most of the drugs in the prescription for invigorating the spleen and removing blood stasis have inhibiting effect on tumor cells and drugs for invigorating the spleen and drugs for removing blood stasis and toxins assume in vitro coordinating effect.
OBJECTIVE To investigate into the compound Chinese prescription Keliu Pill on the growth and metastasis of Lewis lung tumor and VEGF and MVD expression,in order to explore into the mechanism of Keliu Pill in inhibiting the growth and metastasis of tumors.METHOD Lewis lung cancer mice models were established for observation.RESULT In the large,medium and small dosage groups and the group with additional medications,the tumor-inhibiting effect was statistically different from that in the NS group(P0.01),the metastasis-inhibiting effect was also statistically different from that in the NS group(P0.01),the MVD expression markedly decreased as compared with the NS group(P0.05),and the VEGF expression markedly decreased as compared with the NS group(P0.01).CONCLUSION Keliu Pill can inhibit the growth and metastasis of Lewis lung tumor and reduce MVD and VEGF expression in Lewis lung cancer mice.
