Aim:To identify the key candidate genes and proteins related with esophageal and gastric cardia carcinogenesis by differential analysis on the patients with single esophageal or gastric cardia cancers or both from the same patient and proteomics.Methods:The surface-enhanced laser description/ionization time-of-flight mass spectrometry (SELDI-TOF-MS) and comparative genomic hybridization (CGH) were applied to the 4 patients with concurrent esophageal and gastric cardia cancers (CC),107 patsents with single esophageal cancer(EC) and 86 patients with gastric cardia cancer.Results: Based on the consistent analysis by CGH and SELDI-TOF-MS, 14 cross proteins were identified both by CGH and SELDI-TOF-MS on EC from CC and single EC patients (52%, 11/21); 2 cross proteins (Beta-defens in 123, Gamma-aminobutyric acid receptor-associated protein) on gastric cardia adenocarcinoma (GCA) from CC and single GCA patients, and 1 cross protein (Pho-spholipase A2, membrane associated) from the patients with CC, single EC and single GCA were identified.Conclusion: The seventeen cross proteins identified in this study may be a group of key candidates related with esophageal and gastric cardia carcinogenesis.
Objective To characterize the profile of chromosomal imbalances in gastric cardia adenocarcinoma (GCA) with or without family history (GCA FH+ or GCA FH-) in Linzhou, Henan province, and to provide molecular basis to identify the related genes to GCA hereditary susceptibility. MethodsComparative genomic hybridization (CGH) was used to examine 14 cases with GCA FH+ and 28 cases with GCA FH-. ResultsMore frequent DNA copy number gains were observed in GCA FH+ than in GCA FH-. Those with the frequency over 20% were as follows: 20p (FH+ 50% vs FH-21%), 5p (FH+ 50% vs FH-18%), 6q (FH+ 43% vs FH-18%), 16q (FH+ 36% vs FH-14%, P0.05). More frequent DNA copy number losses were seen in GCA FH+ than in GCA FH-. Those 20% were as follows: 4q (FH+ 43% vs FH-14%, P0.05). Frequent DNA copy number gains in 1q, 3q, 6q/p, 7p, 8q, 13q/p, 20q/p and frequent DNA copy number losses in 1p, 17q/p, 19p were both frequent in GCA FH+ and GCA FH-(20%, P0.05). ConclusionFrequent DNA copy number gains on chromosome 20p, 5p, 6q, and 16q and losses on chromosome 15q in GCA FH+ indicate that these chromosome sites may harbor the genes related with GCA high susceptibility. The similar frequent DNA copy number gains or losses on chromosome 1q/p, 3q, 6q/p, 7p, 8q, 13q/p, 20q/p, 17q/p and 19p in GCA FH+ and GCA FH-suggest that genes located in these chromosome sites may be related with environmental risk factors in gastric cardia carcinogenesis.
Objective To characterise the alterations of serum autoantibodies for cyclinB1,p62,Koc-IMP1 and survivin in the subjects with esophgeal and gastric cardia carcinoma and precancerous lesion and their expres-sions in the esophageal and gastric cardia cancer tissue.Methods Enzyme-linked immunoassay and tumor-associated antigen mini-array (consisting of five full-length recombinant proteins,including eyefinB1-p62-Koc,IMP1 and Survivin)were applied to determine the serum level of the autoantibodies of these antigens on 376 subjects with e-sephageal and gastric cardia carcinoma and precancerous lesions.At the same time,the expression of these antigens was detected by immunohistochemical method(ABC)on 13 patients with esophageal cancer and 16 with gastric car-dia cancer.Results All of the 5 antigens determined,the linear correlation Was observed for the detection frequency of cyclinB1,IMPI and p62 in esophageal carcinogenesis,and for p62 in gastric cardia multi-stage progression from normal to precancerous and cancerous lesions(P<0.05).The detection rale with single positive antoantibody im-munoreactivity for both esophageal and gastric cardia cancers were low.However.the positive detection mte for both esophageal and gastric cardia cancer increased apparently when the multiple positive markers were combined together for analysis,which increased tO 3~5 and 3~4 folds respectively.Furthermore,the difference in autoantibody immu-noreactive rate was significant with the lesion progressed from mild tO severe precancerous lesions and to cancer both in esophageal and gastric cardia cancers(P<0.05).The positive immunoreactions of the 5 antigens were detected in cancer tissues.The positive immunostaining rates for cyclinB1,Koc,IMP1 and Survivin both in esophageal and gastric cardia cancers were higher compared to their serllin positive rate of autoantibodies I P<0.05).Of the pa-tients with positive immunostaining in the two cancer tissues,the autoantibodies in the serum for the corresponding antigens could be detected in the same patient.Conclusion The production of the tumor-associated autoantibodies is related tO antigens.The screening rate through serum tumor-associated antigen mini-array for the patients with e-sophageal and gastric cardia carcinoma and precancerous lesions has been increased apparently with combined analy-sis of multiple autoantibodies than with single one.
Aim: To detect the promoter methylation of RASSF1A gene in esophageal cancer and adjacent normal tissue.Methods:Methylation special PCR was applied to detect RASSF1A methylation on 79 cases of esophageal squamous cell carcinoma (SCC) and 20 cases of adjacent normal tissue.Results: The detection rate of RASSF1A methylation in SCC (67%, 53/79) was much higher than that in adjacent normal tissue (15%, 3/20). The methylation of RASSF1A gene had significant difference among different age groups and differentiation degree, P0.05. However, the RASSF1A methylation was not associated with lymph node metastasis or tumor staging. The methylation rate in SCC was apparently higher than that in adjacent normal tissue from the same patient (50%(10/20) vs 15%(3/20)), P0.05. Conclusion: RASSF1A methylation is a common event in SCC; methylation in SCC is much higher than that in adjacent normal tissue and correlated with the age and differentiation of the patients.
动态观察河南科技大学第一附属医院1996~2005年收治的4683例食管癌和贲门癌患者的临床特征,分析住院患者的例数、年龄和性别分布特征和规律。结果4683例患者中,食管癌3648例,贲门癌1035例,二者均为男性明显多于女性。提示食管癌和贲门癌仍然是洛阳地区最常见的恶性肿瘤之一,应加强对其流行病学特征及发病因素的研究。
Aim:To detect the expression of RASSF1A protein in young patients with esophageal squamous cell carcinoma (SCC) and gastric cardia adenocarcinoma (GCA).Methods:Immunohistochemistry (ABC) method was applied to detect the expression of RASSF1A protein in SCC, GCA and the corresponding normal tissue in young patients from Linzhou, Henan.Results: The positive immunostaining rate for RASSF1A in SCC and GCA tissue was 83% (10/12), 89% (8/9), respectively, which was similar to that in corresponding normal tissue (P0.05). However, the immunostaining patterns were different. In SCC, there were scattered (4 cases) , focal (4 cases) and diffused (2 cases) immunostaining patterns; in normal esophageal tissue the papillary (3 cases) and focal (3 cases) immunostaining patterns were common; in GCA the focal (6 cases) pattern was predominant, and scattered (2 cases) pattern was present; there were diffused (5 cases) and focal (4 cases) patterns in the normal gastric cardia tissue. No significant relationship was observed among the rate of RASSF1A protein positive expression and differentiation, metastasis, or invasion depth both in SCC and GCA. Conclusion: The results demonstrate that RASSF1A protein expression is a frequent molecular event in SCC and GCA, suggesting that RASSF1A may play an important role in esophageal and gastric cardia carcinogenesis.
Objective To determine the prevalence of reflux esophagitis(RE) from the subjects in the high incidence area for esophageal cancer in Linzhou,Henan province and to further understanding the mechanism of esophageal carcinogenesis.Methods 508 subjects in high risk area for esophageal cancer were intervened with RE questionnaire.RE diagnosis was made on the subjects with ≥12 score based on the RE symptom scoring.Results 28 subjects were observed with ≥12 score for RE symptom(6%).The detection rate for the female(4%,20/508) was apparently higher than in male(2%,8/508)(P<0.05).Conclusions The prevalence of RE in females is higher than in males,which is contrast with the prevalence of esophageal cancer(higher in males than in females),and is lower than in western country,suggesting that RE may be correlated well with esophageal adenocarcinoma rather than in esophageal squalors cell carcinoma.

Aim: To determine the impact of genetic and environment factors on esophageal and gastric cardia carcinogenesis by comparing the alternations of esophageal and gastric cardia precancerous lesions from the symptomGAAB2free subjects at Linzhou, Henan, the high incidence area for esophageal cancer and the subjects migrated to Changzhi, Shanxi from Linzhou since 1905. Methods: Two natural villages were selected for the study fields, one was migrated villages in Changzhi, Shanxi, the other in Linzhou, Henan. All the subjects with 30 years old in these two villages were invited to attend the mass survey with epidemiological questionnaire, endoscopic, biopsy and histopathological examinations. Results: In esophagus, the detection rate of basal cell hyperplasia (BCH) at the middle segment was apparently higher in the migrated subjects than in Linzhou subjects (38% vs 28%, P0.05). In gastric cardia: the prevalence of chronic superficial carditis (CSG), DYS, and intestinal metaplasia (IM) in the migrated subjects were higher than that in Linzhou subjects (P0.05), however, the prevalence of chronic atrophic carditis (CAG) and gastric cardia adenocarcinoma (GCA) was similar between the migrated subjects and Linzhou subjects (P0.05). In gastric antrum, the prevalence of DYS and IM in migrated subjects was higher than that in Linzhou subjects (P0.05).but, the prevalence of CSG, CAG, and GCA was very similar between the migrated and Linzhou subjects (P0.05).Conclusion: After 100 years, the prevalence of SCC and GCA in the migrated subjects is similar with Linzhou subjects, the prevalence of esophageal precancerous lesions in the migrated and Linzhou subjects is also similar, but the prevalence of severe precancerous lesions at gastric cardia and antrum in the migrated subjects is higher than in Linzhou subjects. The present results demonstrate that the environment and time do not show apparent effect on prevalence of esophageal and gastric cardia precancerous and cancerous lesions, suggesting that genetic factor may play an important role in esophageal and gastric cardia carcinogenesis.
Aim: To further understand the prevalence of familial esophageal cancer (EC) and to establish familial esophageal cancer model through the epidemiological investigation on the subjects at the highGAAB2incidence area in Henan. Methods: Standard questionnaires were distributed to 1 107 subjects from 448 households. Results: The investigation on 264 households demonstrated that, within continuous three generations, there were 5 households (2%) with more than 10 EC patients; 8 households with 7 EC patients (3%); and the number of households with 6, 5, 4 and 3 EC patients was 13(5%), 21(8%), 28(11%) and 46(17%), respectively. Of the 264 households interviewed, there were 153 households (58%) with familial esophageal cancer history.Conclusion: The prevalence for familial EC in the highGAAB2incidence area for EC in Henan is high, which suggests that genetic changes may play an important role in EC.
Objective: To characterize the patterns of ABO blood type and alteration of whole blood analysis in esophageal squamous cell carcinoma (SCC) and gastric cardia adenocarcinoma (GCA) from the patients in high incidence area for SCC and GCA in Huixian, HenanMethods: All the SCC and GCA patients were enrolled from Huixian People's Hospital from 1987 to 2003 Blood type and blood cell alteration were recorded and correlated with SCC and GCAResults: In SCC,B blood type was the main pattern for both male and female (39% and 35%)In GCA,however, O blood type was more frequently observed in male (40%), B blood type was the main type in female (47%) The decreasing with different degrees of blood cells was observed both in SCC and GCA, especially in GCA with a decreasing rate of red blood cell with 50% in male and 56% in female The similar pattern was observed in hemoglobin The decreasing rate of lymph cells was more apparent in SCC than that in GCAConclusion: The present results indicate that SCC occurs more frequently in patients with B blood type, GCA seems to occur more frequently in male with O blood type and B blood type in femaleAnemia and lymph cell decreasing are very common in GCA, and its clinical significance needs to be further characterized
