BACKGROUND: Small nucleolar RNA host gene 12 (SNHG12) has been shown to be a long noncoding RNA (lncRNA) that facilitates the progression of a number of malignancies. However, the expression pattern and biological function of SNHG12 in nasopharyngeal carcinoma (NPC) have not been investigated. OBJECTIVE: The aim of our study is to investigate the expression, clinical significance and function of SNHG12 in NPC. METHODS: RT-PCR was used to detect the expression of SNHG12 in NPC cell lines and primary tumor tissues. The correlation of SNHG12 with clinicopathological features and patient prognosis was analyzed. The biologic functions of SNHG12 in NPC were explored by MTT assay, colony formation assay, wound healing assays, transwell assay and flow cytometric analysis in vitro. The expression of EMT markers and Notch signal pathway markers were determined by western blotting. RESULTS: The expression levels of SNHG12 were up-regulated in both NPC tissues and cell lines. High SNHG12 expression was significantly associated with clinical stage, grade and poor prognosis. Multivariate analysis demonstrated that high lncRNA SNHG12 expression was an independent poor prognostic factor for NPC patients. Functionally, knockdown of SNHG12 suppressed NPC cells proliferation, migration and invasion. Mechanistic investigations showed that knockdown of SNHG12 suppressed the activation of EMT and Notch-1 signal pathway. CONCLUSIONS: Our data suggest that SNHG12 promotes the progression of NPC and is a potential therapeutic target for NPC intervention.
Nasopharyngeal carcinoma (NPC) is an epithelial malignancy with high prevalence in Southeast Asia. Concurrent chemoradiotherapy is the most popular therapeutic method. NPC is mainly characterized by insidious onset and deep lesion site, and thus is difficult to treat. In clinical, the lack of targeted drugs could further affects the therapeutic efficacy for NPC. Folic acid receptor alpha (FR alpha) is an abnormally expressed molecule in various tumor tissues, and is involved in the occurrence and progression of these tumors. However, the potential role and mechanism of FR alpha in NPC remains unclarified. In this study, we detected overexpression of FR alpha in human NPC cell line CNE2 compared with normal nasopharyngeal epithelial cell line NP69 (P<0.05). Further, CNE2 cells were randomly divided into control, scrambled siRNA (negative control) and FR alpha siRNA groups. FR alpha expression was determined by Western blot and real-time PCR analysis. Cell proliferation, invasion and apoptosis were detected by MTT assay, transwell assay, and flow cytometry, respectively. It was found that down-regulation of FR alpha by siRNA significantly inhibited cell proliferation and invasion, but markedly induced cell apoptosis of CNE2 cells compared with control group (all P<0.05). Further mechanism study showed that pERK1/2 and NF-kappa B expression was significantly decreased when compared to control group (P<0.05). In conclusion, our study suggests that FR alpha might facilitate NPC via the ERK1/2/NF-kappa B signaling pathway. FR alpha might be an effective biomarker of NPC and might be useful for the diagnosis and targeted treatment for the malignant disease.
Folic acid (FA), which is necessary for normal cell division of mammals, has been implicated to be involved in many tumors. Dietary FA intake has been reported to be associated with a lower risk of nasopharyngeal cancer (NPC). However, the molecular mechanisms of FA in NPC cells remain unclear. In the present study, we found that FA treatment dose dependently inhibited the proliferation, invasion and migration of NPC cells, via folate receptor α (FRα). We further found that FA, bound to FRα, induced the activation of MEK/ERK1/2, and increased the expressions of TSLC1 and E-cadherin. Moreover, blocking of ERK1/2 activation attenuated FA-mediated increase in TSLC1 expression. In addition, knockdown of TSLC1 abolished the FA-mediated inhibition of cell proliferation, invasion and migration, and suppressed the FA-mediated increase oinE-cadherin expression in NPC cells. Taken together, our data suggest that FA treatment inhibits NPC cell proliferation and invasion via activation of FRα/ERK1/2/ TSLC1 signaling pathway. Therefore, FA could be explored as a therapeutic drug for the treatment of NPC, and TSLC1 may act as a tumor suppressor in NPC.
Objective:To explore the curative effect of operation,radiotherapy and photodynamic therapy in combi-nation with small dose of chemotherapy in the treatment of early glottic cancer.Methods:A retrospective analysis was performed on clinical data of 122 patients with early glottic cancer,and were divided into operation group(83 cases), radiotherapy group(20 cases),photodynamic therapy group(19 cases)according to treatment methods,were given joint of small dose chemotherapy.3,5 years survival rate and 2 years tumor free survival rate and local control rate at 2 years of the three groups were observed and compared.Results:Eight patients were lost to follow up,destined for death or recurrence.In 3 groups total of 6 patients died of laryngeal carcinoma,1 patient died in second primary malig-nant tumor,1 patient died of other diseases.Three patients of the 3 groups,the 5 years survival rate and 2 years tumor free survival rate and local control rate after 2 years were not significantly different(P >0.05).Complications oc-curred in 13 cases in operation group,2 cases occurred complications of photodynamic therapy group,radiotherapy group 7 cases with complications.A total of 27 cases developed recurrence and metastasis,including 4 cases of cervi-cal lymph node metastasis,23 cases of recurrence in primary lesion.Photodynamic therapy group and radiotherapy group,operation group were 5 cases,3 cases,15 cases relapsed.Conclusion:It have similar clinical efficacy of surger-y,radiotherapy and photodynamic therapy combined with low -dose chemotherapy in the treatment of early glottic car-cinoma.
1.1 临床资料。随机选取我科2013~2014年行手术治疗的嗜酸性粒细胞型鼻息肉(eosinophilic chronic rhinosinusitis with nasal polyps,ECRSwNP)和非嗜酸性粒细胞型鼻息肉(non-eosinophilic chronic rhinosinusitis with nasal polyps,nECRSwNP)标本各15例,取同期因行鼻中隔偏曲手术切除的下鼻甲黏膜标本15例作为对照组。所有标本均经病理学证实,其中鼻息肉分型标准为[1]:平均每高倍视野嗜酸性粒细胞计数>10为ECRSwNP,≤10为nECRSwNP。ECRSwNP组中,男8例,女7例,年龄18~62岁。nECRSwNP组中,男9例,女6例,年龄18~62岁。对照组中,男10例,女5例,年龄20~58岁。3组在性别、年龄等方面对比,差异无统计学意义。
OBJECTIVE:To study the expression of hyaluronan synthasel-3 (HAS1-3) in nasal polyps, and discuss their clinical significance. METHOD:The expression of HA, HAS1, HAS2 and HAS3 in nasal polyps group (25 cases) and inferior turbinate group (15 cases) was detected by immunohistochemistry and semi-quantitative RT-PCR. RESULT:Compared with the control, the expression of HA, HAS1 and HAS3 was higher in nasal polyps (P<0. 05), while there was no significant difference in HAS2 expression (P>0. 05). CONCLUSION:The increase of HASI and HAS3 expression may be the main cause of the excessive deposition of HA in nasal polyps, which may play an important role in the pathogenic process of nasal polyps, and become a potential target for therapy of nasal polyps.
Neoplatin plus 5-fluorouracil (5-FU) is the routine method to treat nasopharyngeal carcinoma. The present study aims to investigate the potential drug-drug interaction for this therapy regimen, and the therapeutic mechanism towards nasopharyngeal carcinoma. The inhibition of neoplatin + 5-FU towards human liver microsomes (HLMs)-catalyzed SN-38 (the active metabolite of irinotecan) glucuronidation was investigated. Metabolomics-based serum analysis was used to elucidate the therapeutic mechanism of neoplatin + 5-FU towards nasopharyngeal carcinoma. The results showed that neoplatin + 5-FU can inhibit HLMs-catalyzed SN-38 glucuronidation, indicating the potential drug-drug interaction between this treatment regimen with irinotecan. The treatment of neoplatin + 5-FU towards nasopharyngeal carcinoma was attributed to the prevention of neoplatin + 5-FU towards alteration of amino acids glutamine and glutamic acid. In conclusion, potential drug-drug interaction existed for the clinical application of neoplatin + 5-FU to treat nasopharyngeal carcinoma in which prevention of nasopharyngeal carcinoma-induced alteration of glutamine and glutamic acid is the potential mechanism.
目的 探讨紫杉醇加顺铂(TP)方案与顺铂加氟尿嘧啶(PF)方案化疗同步放疗对局部晚期喉癌的效果和毒性.方法 随机选取2011年3月-2014年10月南京医科大学附属淮安第一医院耳鼻喉科收治的局部晚期喉癌患者80例,依据治疗方法将这些患者分为TP组(n=40)和PF组(n=40),给予二组患者化疗同步放疗,然后对二组患者的近期疗效、毒性反应发生情况及生存情况进行统计分析.结果 TP组患者治疗的总有效率92.5%(37/40)显著高于PF组60.0%(24/40)(P<0.05),Ⅲ~Ⅳ度放射性咽喉炎、胃肠道反应、骨髓抑制发生率均显著高于PF组(P<0.05).结论 在局部晚期喉癌的治疗中,TP方案较PF方案化疗同步放疗具有较好的效果和较小的毒性.
BACKGROUND Unclear pathogenesis existed for nasopharyngeal carcinoma. AIMS to analyze the role of bile acids in the pathogenesis of nasopharyngeal carcinoma. METHODS 20 healthy volunteers and 20 patients with nasopharyngeal carcinoma were enrolled between January 1(st), 2013 and December 31(st), 2014. ESI-QTOF-MS analysis of serum was performed to find altered bile acids components. The biological function of changed bile acids was investigated using in vitro experiment. RESULTS Compared with healthy volunteers, the level of DCA and GDCA exhibited higher abundance in patients with nasopharyngeal carcinoma (p<0.01). Furthermore, the biological function was investigated for the inhibition of DCA and GDCA towards the secretion of IL-10 by CD4+CD25- T cells. Both DCA and GDCA significantly inhibited the secretion of IL-10 by CD4+CD25- T cells. Furthermore, DCA+GDCA can show stronger inhibition towards the secretion of IL-10 than DCA and GDCA. CONCLUSION The inhibition of IL-10 secretion by elevated DCA and GDCA components in nasopharyngeal carcinoma patients is the inducer for nasopharyngeal carcinoma.
Lasiodin-related potential drug-drug interaction and clinical therapy of lasiodin towards nasopharyngeal carcinoma were investigated in this study. In vitro inhibition of lasiodin towards the metabolism of irinotecan was investigated. Metabolomics-based analysis of serum was carried out towards healthy volunteers, treated and untreated nasopharyngeal carcinoma with lasiodin-containing Rabdosia serra tea. The results showed that lasiodin inhibited 30% activity of SN-38 glucuronidation. Metabolomics analysis showed that treatment with lasiodin-containing R. serra tea treatment presented the elevation of glutamic acid and reduction of glutamine in patients with nasopharyngeal carcinoma. In conclusion, lasiodin-related potential drug-drug interaction and clinical therapy of lasiodin towards nasopharyngeal carcinoma were demonstrated in the present study.
Objective To explore the treatment mechanism of safflower injection on sudden deafness through the changes of the hearing threshold, serum superoxide dismntase(SOD) and malonyldialdehyde(MDA) before and after treatment. Methods 40 cases of sudden deafness were divided into two groups : group Ⅰ(group with safflower) and group Ⅱ(group without safflower). The concentrations of serum MDA and SOD and hearing threshold were detected before and after treatment. 25 cases of normal hearing as the control group. Results(1) The total efficiency in the group Ⅰ was higher than the group Ⅱ(P 0.05).(2)The concentration of SOD was lower and the concentration of MDA was higher than that of the control group before treatment in both two groups(P 0.05). The concentration of SOD was higher and the concentration of MDA was lower after treatment in group I(P 0.05). The concentrations of SOD and the MDA had no significant change in group Ⅱ(P0.05). Conclusion Safflower injection could scavenge oxygen free radical and improve the quality of hearing recovery.
目的 分析不同开口切割头对腺样体切除术疗效及术中、术后出血的影响.方法 回顾性分析2009年7月至2010年7月因儿童阻塞性睡眠呼吸暂停低通气综合征行腺样体切除术的患儿129例,其中采用正向开口切割头85例,反向开口切割头44例,对比两种不同开口切割头对手术疗效和术中、术后出血的影响.结果 不同开口切割头对手术疗效及术后出血的影响差异无统计学意义(P>0.05),但采用正向开口切割刀头的病例(7.1%)术中出血较反向切割刀头(22.7%)少,差异有统计学意义(P<0.05).结论 采用正向切割刀头能很好的把握切割的深度,减少术中出血,值得临床推广.
目的:探讨局麻下和全麻下扁桃体切除手术疗效对比。方法选取2014年3月至2014年09月在我院行扁桃体切除术的62例患者住院病例资料进行回顾性分析,对局麻下和全麻下扁桃体手术疗效进行分析。结果31例全麻手术患者术后反应相对较小,术后恢复较好,术后出血的病例比率低,具有统计学意。结论在目前全民医保的大环境下,随着麻醉技术的不断的发展进步,及患者对医疗服务要求不断提高下,全麻下扁桃体切除术已成必然趋势。
目的:研究低温等离子消融(LTPA)方案应用在下鼻甲骨折外移术式(TFSO)时,对慢性肥厚性鼻炎(CHR)患者的效果影响。方法收集2010年3月到2013年3月,该院被确诊为CHR的患者92例。以数字表法将患者随机分成观察组(46例)及对照组(46例)。观察组行LTPA联合TFSO术式,对照组则行下鼻甲成形术(ITP)联合TFSO术式。对比两组疗效、治疗前后视觉模拟评分法(VAS)评分情况以及不良预后情况。结果观察组疗效显著优于对照组,差异有统计学意义(P<0.05)。观察组在治疗后VAS评分显著低于治疗前及对照组治疗后的水平,差异均有统计学意义(P<0.05)。观察组治疗无效患者1例,对照组有8例,经相应处理后好转。结论以LTPA联合TFSO方案治疗CHR患者,疗效显著,可明显改善患者VAS评分水平,效果较好,值得临床推荐。
Objective To explore the effects of intranasal glucocorticoid on the expression of epidermal growth factor (EGF) and epidermal growth factor receptor (EGFR) in nasal polyps. Methods Forty-two patients with nasal polyps were equally randomized divided into two groups:group A (n=21, treated with budesonide 128μg, BID, 12 weeks) and group B (n=21, normal saline, BID, 12 weeks). The expression of EGF and EGFR in nasal polyps were detected with immunohistochemistry and the therapeutic effect of nasal polyps were evaluated with the nasal en-doscopic score system. Results The expression of EGF in the epithelial cells [(0.15±0.04) vs (0.24±0.06)] and endo-scopic staging of nasal polyps were significantly lower in group A than that in group B (P<0.05). The expression of EGFR in group B did not differ significantly from group A (P>0.05). Conclusion Inhibiting the expression of EGF and not EGFR in the epithelial cells of nasal polyps may be one of mechanisms for the therapeutic effect of intranasal glucocorticoid on the nasal polyps.
目的研究鼻内窥镜治疗难治性鼻出血的疗效。方法回顾性分析2010年6月至2012年6月34例难治性鼻出血患者内窥镜下诊治过程。结果 30例有明确出血点,其中嗅裂20例(67%)、下鼻道后穹隆5例(17%)、中鼻道后下部4例(13%)、后鼻孔上方1例(3%),经手术治疗后治愈。4例患者未发现明确出血点,其中2例经鼻内镜下凡士林纱条填塞后未再出血,2例因术后再次出血经第二次手术治愈。结论鼻内窥镜手术是治疗鼻出血的有效方法,治疗的关键是寻找到出血点。
目的 探讨鼻内窥镜下微创治疗鼻窦真菌球性鼻窦病变的疗效.方法 对98例鼻窦真菌球患者采用鼻内镜下经中鼻道扩大的上颌窦自然口取出上颌窦真菌球,累及筛窦、蝶窦亦经内窥镜开放去除病灶,观察治疗效果.结果 所有患者术后症状消失,鼻内镜下创面愈合、窦口开放,窦腔黏膜上皮化完整;79例术后3~6个月复查CT,有8例上颌窦局部黏膜稍增厚,余均未见异常;随访1年未见复发.结论 经内窥镜微创治疗真菌球性鼻窦病变操作方便,术后出血少、反应轻、并发症少,无复发.
目的 分析手术切除儿童肥大腺样体后对分泌性中耳炎致听力下降疗效的影响.方法 对本院住院的腺样体肥大并发分泌性中耳炎的30例(49耳)患儿经鼻内镜行腺样体切除术,回顾性分析对患儿听力的治疗效果.结果 在0.25~8kHz的气导阈值,术后与手术前有显著性差异(P<0.05),术后各频率骨导阈值与手术前无显著性差异(t=1.022,P>0.05).结论 对于伴有腺样体肥大的分泌性中耳炎患儿,手术切除其肥大的腺样体可明显改善患儿的听力.
目的 探讨糖皮质激素对鼻息肉组织中CD44s表达的影响.方法 鼻息肉患者30例,随机分为激素组(口服强的松10mg·kg-1·d-1,×10 d)和对照组,每组15例.采用免疫组织化学染色方法检测两组鼻息肉组织中CD44s的表达情况.结果 与对照组比较,激素组鼻息肉组织中血管内皮细胞(0.15±0.03 vs.0.30±0.05)和炎症细胞(0.20±0.05 vs.0.36±0.04)CD44s的表达明显减少(P<0.05).结论 糖皮质激素抑制鼻息肉组织中血管内皮细胞、炎症细胞CD44s的表达,这可能是其治疗鼻息肉的作用机制之一.
