目的 探讨低氧环境下间充质干细胞在SDF-1复合PLGA/胶原三维支架上的增殖和迁移的影响,为治疗肌腱损伤提供理论依据.方法 于2021年6月—2022年6月齐齐哈尔医学院分子生物学实验室实施,购买大鼠骨髓间充质干细胞,随机分为4组,分别为低氧对照组,低氧P/C支架组,低氧S-P/C支架组,正常对照组,每组各进行5次CCK-8细胞增殖实验检测细胞的增殖情况,每组各进行3次Transwell细胞迁移实验检测细胞的迁移情况.结果 在第7天各组细胞培养活性显示,低氧对照组吸光度值(1.26±0.10)高于正常对照组的(1.07±0.12),差异有统计学意义(t=2.724,P=0.027).低于低氧P/C支架组吸光度值(1.50±0.15)高于低氧对照组的(1.26±0.10),差异有统计学意义(t=2.909,P=0.020).低氧S-P/C支架组吸光度值(1.71±0.13)高于低氧P/C支架组的(1.50±0.15),差异有统计学意义(t=2.486,P=0.038).各组细胞迁移数量,低氧对照组细胞迁移数量为(35.2±5.4)×103 个相对正常对照组的(21.6±3.5)×103 个,差异有统计学意义(t=3.661,P=0.022).低氧P/C支架组细胞迁移数量为(47.8±4.9)×103 个高于低氧对照组的(35.2±5.4)×103 个,差异有统计学意义(t=2.993,P=0.040).低氧S-P/C支架组细胞迁移数量为(56.6±2.3)×103 个高于低氧P/C支架组的(47.8±4.9)×103个,差异有统计学意义(t=2.816,P=0.048).结论 在低氧环境下,SDF-1复合PLGA/胶原三维支架可促进间充质干细胞的增殖和迁移.
目的:探讨大黄?虫丸对下肢动脉硬化闭塞症模型大鼠的作用及其机制.方法:取90只SD大鼠依据随机数字表随机分为6组,每组各15只,分别为空白对照组、模型组、阳性对照组、高剂量组、中剂量组和低剂量组.除空白对照组外,其余各组大鼠采用复合方法制作动脉(髂-股动脉)粥样硬化模型大鼠.模型复制成功后,空白对照组和模型组灌胃生理盐水,阳性对照组每日灌胃通塞脉片生理盐水溶液(380 mg/kg),低、中、高剂量组每日分别灌胃大黄?虫丸生理盐水溶液(2.16、1.08、0.54 g/kg),共灌胃4周.应用双抗体夹心ABC-ELISA法检测大鼠血清中血脂(TC、TG、LDL、HDL)及ET-1、IL-6的含量;Western blot法检测血小板活化因子(PAF)、脂蛋白相关磷脂酶A2(LP-PLA2)表达水平.结果:与空白对照组比较,模型组大鼠ET-1、IL-6、TC、TG、LDL水平明显升高(P<0.01),HDL水平明显降低(P<0.01),PAT和LP-PLA2蛋白表达水平明显升高(P<0.01).与模型组比较,高剂量组可以明显降低ET-1、IL-6、TC、TG、LDL的含量,升高HDL的含量(P<0.01),降低PAF、LP-PLA2蛋白表达水平(P<0.05);中剂量组与阳性对照组比较,差异无统计学意义(P>0.05).结论:大黄?虫丸可通过调控PAF/LP/PLA2的信号通路,从而抑制炎症因子的激活,减轻动脉粥样硬化和血栓的形成,从而有效治疗下肢动脉硬化闭塞症.
目的:探讨大黄蛰虫丸联合血府逐瘀汤加减预防下肢动脉硬化闭塞症(ASO)球囊扩张及支架术后支架内再狭窄(ISR)的临床疗效及作用机制.方法:将拟行支架植入术的符合纳入标准的ASO患者60例按照随机数字表分为对照组和治疗组,对照组采用常规抗血小板及降脂治疗,治疗组在对照组基础上口服大黄蛰虫丸和血府逐瘀汤.比较两组支架植入后3个月和6个月的狭窄程度、踝肱指数,并在术前、术后6个月检测两组患者血液纤维蛋白原(Fib)、血脂(TC、TG、LDL-C、HDL-C)水平.结果:与对照组比较,治疗组术后6个月踝肱指数明显提升,差异有统计学意义(P<0.05),术后6个月重度狭窄患者的比例降低(20.0%vs 13.3%),但差异无统计学意义(P>0.05);与本组术前比较,两组术后6个月TC、TG、LDL-C均下降,HDL-C升高(P<0.05),且治疗组优于对照组(P<0.05);与本组术前比较,两组术后6个月Fib均下降(P<0.05),且治疗组优于对照组(P<0.05).结论:大黄蛰虫丸联合血府逐瘀汤加减可降低血脂、血浆纤维蛋白原,改善血液成分,具有防治ISR的作用.
根据下肢动脉硬化闭塞症的可能发病机制,为解除患者病痛,提升患者生存质量,降低下肢动脉硬化闭塞症病老年人的威胁,充分挖掘八段锦在医学领域的宝贵功能,本文分析了八段锦的调养功能、作用和特点.发现通过八段锦锻炼的练习可以有效防治下肢动脉硬化闭塞症的发生发展,可滑利关节、发达肌肉、增长气力、强壮筋骨,气血通畅、脏腑协调,可调节血脂的异常,改善内皮细胞功能,保护血管内皮,不但能够抑制血栓形成和血小板聚集,并且能够降低血液的粘稠度,从而发挥抗动脉粥样硬化的作用,对下肢动脉硬化闭塞症有很好地治疗效果.
目的 探讨三维培养条件下,骨形态发生蛋白-12(BMP-12)对人骨髓间充质干细胞(hBMSC)增殖和成肌腱分化的影响.方法 静电纺丝技术制备PLGA/明胶材料.hBMSCs分为6组:对照组(Con),细胞正常培养;BMP-12组,细胞中添加10 ng/mL BMP-12培养;Triciribine组,BMP-12组细胞添加5μmol/L Triciribine;材料组(Material),hBMSCs种植在PLGA/明胶材料;3D-BMP-12组,材料组中添加10 ng/mL BMP-12;3I-Triciribine组,3D-BMP-12组添加5μmol/L Triciribine.各组hBMSCs均进行成肌腱诱导分化.扫描电子显微镜检测材料结构;CCK-8法检测各组细胞增殖;Western blot检测各组Scleraxis(Scx)、Tenomodulin(TNMD)和Tenascin-R、Akt、p-Akt蛋白表达.结果 与对照组相比,材料组、BMP-12组细胞生长数目均明显升高.与材料组相比,3D-BMP-12组hBMSCs细胞数量、Scx、TNMD和Tenascin-R蛋白表达均明显升高(P<0.01),3D-Triciribine组hBMSCs上述检测指标均分别比3D-BMP-12组显著降低(P<0.01).3D-BMP-12组hBMSCs的Akt、p-Akt蛋白表达均显著升高,3D-Triciribine组Akt、p-Akt蛋白表达均明显降低(P<0.01).结论 BMP-12能激活Akt信号通路,促进生物材料上hBMSCs增殖和成肌腱分化.
Circular RNAs (circRNAs) act as a key role in mediating carcinogenesis. Nevertheless, the functions and mechanisms of circRNAs in osteosarcoma (OS) are still not fully understood. In the present study, we aim to investigate the functions of circ-XPO1 in OS and its potential mechanism underlying OS progression. CircRNA microarray indicated elevation of circ-XPO1 in OS specimens relative to normal samples. Elevation of circ-XPO1 and XPO1 mRNA was identified in OS tissue specimens and cells by qRT-PCR. In addition, enhanced expression of circ-XPO1 and XPO1 mRNA both correlated with poor prognosis for the patients with OS, as estimated by Kaplan-Meier analysis. Functionally, circ-XPO1 and XPO1 both facilitated the growth and invasion and decreased the apoptosis of OS cells. Moreover, we constructed the circ-XPO1-miRNAs-XPO1 3'-UTR interaction network and verified that circ-XPO1 could sponge miR-23a-3p, miR-23b-3p, miR-23c, and miR-130a-5p to regulate XPO1 expression. Furthermore, rescue assay indicated that the effect of circ-XPO1 on cell progression was partly relying on these miRNAs. Taken together, we found that circ-XPO1 regulated the expression of XPO1 through sponging miRNAs as a competing endogenous (ceRNA), providing the possibility that circ-XPO1 might play as a new therapeutic target for OS.
目的 探讨低氧环境下PLGA/胶原纤维支架对小鼠骨髓间充质干细胞(mBMSCs)增殖及成肌腱分化的影响.方法 利用静电纺丝技术制作出30组PLGA/胶原纤维支架,正常条件下培养的mBMSCs为正常对照组;以3%O2,5%CO2,92%N2建立细胞低氧模型,在低氧环境下培养mBMSCs为低氧对照组,mBMSCs种植在PLGA/胶原纤维支架为低氧支架实验组,各组均进行成肌腱分化诱导实验14 d.CCK-8实验检测3 d、5 d、7 d各组mBMSCs增殖的吸光度值,ELISA实验检测各组mBMSCs上清液肌腱细胞标志性蛋白Tenomodulin和Scleraxis含量.结果 在3 d、5 d、7 d观察点内,正常对照组、低氧对照组、低氧支架实验组mBMSCs吸光度值差异有统计学意义(P<0.05),第3 d、5 d、7 d观察点内,低氧对照组mBMSCs吸光度值均高于正常对照组,差异有统计学意义(P<0.01);与低氧对照组相比,低氧支架实验组3 d、5 d、7 d吸光度A值均分别增高,差异有统计学意义(P<0.01).在细胞低氧环境下,低氧支架实验组Tenomodulin蛋白(4.526±0.002)μmol/L和Scleraxis蛋白(3.752±0.004)μmol/L含量高于低氧对照组Tenomodulin蛋白(2.831±0.023)μmol/L和Scleraxis蛋白(2.457±0.032)μmol/L,低氧对照组Tenomodulin蛋白(2.831±0.023)μmol/L和Scleraxis蛋白(2.457±0.032)μmol/L含量高于正常对照组Tenomodulin蛋白(1.542±0.071)μmol/L和Scleraxis蛋白(1.136±0.056)μmol/L,3组对比差异有统计学意义(P<0.01).结论 低氧环境下PLGA/胶原纤维支架可有效促进mBMSCs增殖和成肌腱分化.
目的 观察CT引导下微波消融治疗特殊部位小肝癌的临床疗效.方法 方便选取2015年5月—2016年5月确诊为原发性肝癌(特殊部位小肝癌)的63例患者,按照随即数字表的原则,分为对照组31例和治疗组32例.对照组采用经肝动脉化疗栓塞术(TACE)治疗,治疗组在TACE基础上联合CT引导下微波消融(MWA)治疗.术后共随访2年,检测比较2组患者手术前后(术后1年、2年)的甲胎蛋白(AFP)水平情况;比较2组患者术后病灶控制以及并发症发生情况;比较2组患者术后生存率.结果 与对照组治疗后1年AFP降低有效率74.2%,2年AFP降低有效率64.5%相比,治疗组术后1、2年AFP降低有效率均更高,分别为93.8%,87.5%,两组比较差异有统计学意义(x2=4.510、4.585,P<0.05);与对照组总体控制率71.0%,并发症发生率25.8%相比,治疗组术后病灶总体控制率更高且并发症更少,分别为93.8%,6.3%,两组比较差异有统计学意义(x2=5.671、4.510,P<0.05);与对照组术后1年、2年生存率(分别为74.2%、64.5%)相比,治疗组术后1、2年生存率更高,分别为93.8%、90.6%,两组比较差异有统计学意义(x2=4.510、6.210,P<0.05).结论 与单用TACE相比,联合TACE与CT引导下MWA共同治疗特殊部位小肝癌的患者,可更加有效降低AFP,保护肝脏功能,疗效确切,减少术后并发症的发生,且能增加术后总体生存率,表明TACE与CT引导下MWA联合治疗特殊部位小肝癌更加安全有效,值得临床推广.
目的 观察前列腺动脉介入栓塞术治疗前列腺增生(BPH)的临床疗效以及血流动力学的影响.方法 随机选取2017年9月—2018年9月确诊为BPH经内科治疗无效且不愿接受传统电切术的43例患者作为治疗组,行前列腺动脉介入栓塞术(PAE);随机选取在该科行前列腺电切术(TURP)的44例患者作为对照组.术后随访3个月,记录比较2组患者国际症状评分(IPSS)、生活质量评分(QOL)、最大尿流率(Qmax)、残余尿(RU)以及前列腺体积的变化;观察比较2组患者治疗前后前列腺动脉的内径(R)、最大血流速度(Vmax)、脉动指数(PI)、阻力指数(RI)等血流动力学指标;观察比较2组患者总体疗效以及并发症发生情况.结果 治疗前,2组各项指标均差异无统计学意义(P>0.05).治疗后,2组患者IPSS、QOL、RU以及前列腺体积均下降,Qmax上升,但与对照组相比,治疗组指标改善更多,差异有统计学差异(P<0.05);治疗后,2组患者血流动力学各项指标(R、Vmax、PI、RI)均下降,但与对照组相比,治疗组指标下降更多,差异有统计学意义(P<0.05);治疗组总体治疗疗效为90.7%,对照组为70.5%(x2=5.668,P<0.05);且对照组并发症为18.2%,治疗组为4.7%(x2=3.914,P<0.05).结论 与传统TURP相比,使用PAE治疗BPH,可更加有效改善前列腺功能,减少前列腺体积,缓解下尿路梗阻症状,从而增加最大尿流率,另外可明显减少前列腺动脉内径与血供,增加治疗总有效率,且能减少术后并发症的发生,表明PAE治疗BPH更加安全有效,减少术后风险,尤其适用于不愿接受传统电切术的BPH患者,值得临床推广.
目的 观察TIPS治疗肝硬化门静脉高压(PHT)合并上消化道出血(UGH)的临床效果以及对血流学的影响.方法 将2017年4月—2018年5月确诊为PHT合并UGH的61例患者,随机分为对照组30例和治疗组31例.对照组采用胃冠状静脉栓塞术(GCVE)方式,治疗组在此基础上联合经颈静脉肝内门体分流术(TIPS).术后随访6个月,记录比较两组患者手术前后直接门静脉压力(PVP)以及门静脉压力梯度(PPG);观察比较两组患者手术前后门静脉内径、门静脉主干压、门静脉流速等血流学指标;观察比较两组患者术后并发症发生情况.结果 术前,两组各项指标均差异无统计学意义(P>0.05).术后两组患者PVP、PPG均较术前下降,但与对照组相比,治疗组PVP、PPG降低更多,差异均有统计学意义(P<0.05);治疗后,两组患者血流学各项指标均改善,但与对照组相比,治疗组改善更优,差异均有统计学意义(P<0.05);且治疗组术后并发症更少,差异有统计学意义(P<0.05).结论 与单用GCVE术相比,联合TIPS与GCVE共同治疗PHT合并UGH的患者,可更加有效降低门静脉压力,减少门静脉内径,且有效增加门静脉流速,疗效确切,且能减少术后并发症的发生,表明TIPS与GCVE联合治疗PHT合并UGH更加安全有效,减少术后风险.
随着现代信息网络化的飞速发展,数字化课程也在全国各大医学高校教学普遍开展起来.断层解剖学是研究正常人体不同方位层面上器官结构的位置、形态、毗邻的一门医学学科,是医学影像学专业学生的专业基础课程,是学好医学影像诊断学、超声学、介入放射学的桥梁学科,但本课程知识内容繁多、复杂,学生学习记忆本课程知识点困难,教学效果不够理想.本研究在本课程实验课教学上结合虚拟数字人教学,丰富教学手段,让学习者学习情景更逼真、立体、生动,自主学习主动性更强,取得较好教学效果,教学质量明显提高.
目的 探讨"电子影像胶片库"在医学影像解剖学实验教学中的应用效果.方法 随机抽取2015级医学影像学专业50人平均分成两组,每组25人,一组运用实体影像胶片进行实验授课为对照组,一组运用电子影像胶片结合实体影像胶片进行实验授课为实验组.最后进行实验胶片考试和学生调查问卷对比两种教学法.结果 实验组平均实验成绩分数为(18.46±1.41)分,对照组平均实验成绩分数为(16.22±1.64)分,实验组实验成绩分数高于对照组,差异具有统计学意义(t=10.016,P<0.01);在调查问卷中通过学习知识自主性、记忆知识准确性、分析问题能力、理论联系实践能力4个方面调查两组教学法满意度,得出实验组学生调查问卷各项内容优于对照组学生,差异均具有统计学意义(P<0.05).结论 在医学影像解剖学实验教学中运用电子影像胶片结合实体影像胶片可以明显提高学生学习效率,提高本课程教学质量,为以后进入临床学习阅片有更好的铺垫.
随着现代电子科技的飞速发展,网络化全面覆盖,各大医学高校为适应当代大学生的要求与时俱进改善教学环境,本学科2015年建立断层解剖学虚拟实验室,模拟真实场景,在断层解剖学实验教学中全方位激发学生学习兴趣,提高教学质量,使学生从基础学科走向临床实践有更好的过渡.
OBJECTIVE To explore the postoperative infection‐related complications in liver cancer patients under‐going radiofrequency ablation and observe the clinical characteristics .METHODS A total of 781 patients who un‐derwent the radiofrequency ablation of liver tumor in invasive technology department from Mar 2010 to Mar 2016 were retrospectively analyzed ,the clinical data ,involving the size ,number ,and location of tumor and use of anti‐biotics as well as incidence of postoperative infections ,were collected from the patients ,the patients who presen‐ted with severe infection were treated with CT enhancement scanning ,the indwelling tube drainage was carried out after the local infection foci were confirmed ,the bacteriological test was performed for the drainage fluid ,and the drug susceptibility testing was conducted so as to adjust the usage of antibiotics .RESULTS The 781 patients under‐went 957 times of radiofrequency ablation in total ,and 35 patients had the postoperative infection ,with the infec‐tion rate 3 .66% .The major clinical manifestations of the patients with severe infection included the fever ,chills , liver pain and abdominal pain .After the foci were confirmed by the enhanced CT and the drainage fluid specimens were cultured ,totally 39 strains of bacteria were isolated .The high risk factors included the history of choledo‐chojejunostomy ,multiple liver tumors ,and tumor adjacent intestinal tract and open radiofrequency ablation .The infections in all of the patients were controlled after the antibiotics dressing change or the anti‐infection therapy was strengthened .CONCLUSION The radiofrequency ablation is effective in the treatment of the liver cancer pa‐tients but may induce the infection‐related complications such as liver abscess ,incision infection ,and cholesteato‐ma .The CT enhancement scanning and drug susceptibility testing may facilitate the identification of the infection‐relat‐ed complications and the treatment .
Objective To investigate the expression and significance of β-catenin and p28GANK in residual hepatocellular carcinoma (HCC)cells after transcatheter arterial chemoembolization (TACE).Methods We collected forty-five cases of surgical specimens of hepatocellular carcinoma after TACE ( TACE group ) and thirty cases of surgery without any treatment (pure surgery group).The expression of β-catenin and p28GANK were detected by using immunohistochemical SP method and compared between the two groups .Results The positive expression of β-catenin and p28GANK in TACE group were 77.78%and 75.56%respectively,which were significantly higher than those in pure surgery group (46.67%and 53.33%respectively,P<0.05).In the residual hepatocellular carcinoma cells of TACE group ,the positive expression of β-catenin showed correlation with the positive expression of p28GANK(Φ=0.318,P =0.033).The high expression of β-catenin and p28GANK were closely related to portal vein thrombosis and distant metastasis (P<0.05).Conclusion The ex-pression of β-catenin and p28GANK in the residual hepatocellular carcinoma cells were increased significantly after TACE.The high expression of β-catenin and p28GANK were closely related to portal vein thrombosis and distant metastasis.The high expression of β-catenin and p28GANK may be one of the reasons of hepatocellular carcinoma invasiveness and metastasis .
Objective To discuss the value of radiofrequency ablation in the special sites of the liver cancer by ultrasound guided with CT monitoring.Methods 45 patients with a total of 75 special sites of malignant hepatic lesions were enrolled in this study.Intraoperative and postoperative complications were recorded.The therapeutic results were observed by analyzing the imaging data of the first month after Radiofrequency ablation Results No serious intraoperative or postoperative complications were occurred,15 patients(33.3%) had transient fever;16 cases(35.6%),had abdominal pain;3 cases with intractable hiccups;6 patients(13.3%) with nausea.Follow up were carried out one month after RF treatment.Of 13 lesions near the diaphragm,complete ablation was demonstrated in 12(92.31%);8 lesions adjacent gallbladder,complete ablation in 6 lesions(75%);5 lesions near adjacent bowel,complete ablation in 5 lesions(100%);9 lesions adjacent large blood vessels,complete ablation in 8 lesions(88.89%);14 lesions adjacent bile duct,complete ablation in 13 lesions(92.86%),only 1 case with the lesion near the gallbladder occurred tumorigenesis,no metastasis was found.Conclusion Clinical efficacy of radiofrequency ablation in the special sites of liver cancer by combining ultrasound guided with CT monitoring is satisfied with low complication and worthy of promotion.
影像解剖学与断层解剖学是医学院校影像专业的一门专业基础课,在其教学中我们使用了临床胶片库(X线、CT及MRI胶片)辅助教学,提高了学生的学习兴趣和考试成绩,取得了良好的教学效果,丰富了此门课程的教学工具.
Objective To discuss the variation of intraepithelial lymphocyte and lamina propria lymphocyte of small intestine(duodenum,jejunum,ileum) in rat under the condition in the patients with rheumatoid arthritis.Methods By immunohistochemical staining method,we observed the normal group and disease-causing group intestine mucosa variation of Tlymphocyte,calculated the quantity of IEL and LPL,and analyzed the result statistically.Results The amount of IEL and LPL in various part(duodenum,jejunum,ileum) of rat increased under the pathogenic group.Conclusion By immunohistochemical method that under the conditions of patients with rheumatoid arthritis,intestinal mucosa epithelium and lamina propria could be seen an increase in the distribution of T lymphocytes.IEL and LPL in different part of the intestine tract may play important biological roles in immune response of mucosa.
断层解剖学是目前新兴的一门解剖学,这门学科的实验教学模式尚处于探索阶段,如何适应社会发展开展断层解剖学实验教学,既提高学生解剖学实验技能,又加强医学生对后继临床课的学习和应用,是我们在断层解剖学实验体系中需要面对的一个重要问题.经过实践.我教研室采用了一些方法改进断层解剖学实验教学,并取得良好的效果,供国内外同行借鉴参考.
