目的 采用Minitab软件优选内部沸腾法提取冬虫夏草多糖工艺参数的设计空间.方法 以冬虫夏草多糖提取率为考察指标,首先采用部分因子试验,对5个影响提取效率的影响因素进行筛选,然后利用中心复合实验设计原理开展响应面实验,对冬虫夏草多糖提取工艺参数空间进行优化.结果 确定冬虫夏草多糖提取率≥2.4%的关键工艺参数设计空间,解析剂浓度范围80% ~ 86%,提取剂温度范围87~92℃.结论 所得提取工艺设计空间稳定可行,为冬虫夏草多糖提取物规模化生产提供数据支持.
目的 优化鲜冬虫夏草中肿瘤细胞生长抑制活性提取物的低温超声波提取条件.方法 采用3因素3水平中心组合试验,以鲜冬虫夏草提取物对黑色素瘤B16细胞的抑制率为响应值,进行响应曲面分析.结果 鲜冬虫夏草的最佳低温超声波提取工艺参数为:超声机功率320 W,超声提取时间43 min,超声提取温度30℃.在此条件下提取,提取物实际黑色素瘤B16细胞的抑制率达72.80%.结论 该研究所得优选提取工艺参数是稳定可行的,为鲜冬虫夏苹资源的合理利用及产品开发提供支持.
通过全面系统的调研文献,以2010年以来74篇辅助肿瘤治疗的中成药研究文献为依据,总结了国内已上市的中成药在肿瘤辅助治疗领域的实验研究和临床应用现状,为今后开发肿瘤辅助治疗中成药提供参考.
OBJECTIVE To compare the contents of sterols compounds in silkworm C. militaris, oak silkworm C. militaris and C. militaris fruiting body. METHODS A HPLC-ELSD method for the analysis of sterols compounds was developed. Agilent SB C18 column ( 4. 6 mm × 150 mm, 5 μm) was used. Gradient elution buffer consisted of water and methanol ( 0. 005% formic acid) .RESULTS The contents of ergosterol were C. militaris fruiting body ﹥ silkworm C. militaris>oak silkworm C. militaris. Cholesterol and sitosterol were detected in the silkworm C. militaris and oak silkworm C. militaris, but not in the C. militaris fruiting body.CONCLUSION The contents of sterols in three different type of C. militaris are varied. The result of this research is helpful for the quality control, rational use and comprehensive development of C. militaris.
An online SPE-HPLC method for simultaneous determination of cordycepin (3'-deoxyadenosine) and 2'-deoxyadenosine in Cordyceps genus (C. sinensis,C. militaris,Hirsutella sinensis and C. sobolifera) was developed. The samples were enriched on a ZORBAX SB-AQ (4.6 mm×12.5 mm,5 μm) column with isocratic elution by 9% methanol solution. The separation of analytes was performed on a ZORBAX SB-AQ (4.6 mm×150 mm,5 μm) column with gradient elution by 0.1% formic acid solution and methanol (91∶9). The flow rate was 1.0 mL•min⁻¹. Column temperature was 40 ℃ and detection wavelength was 260 nm. This method has been applied for analysis of different Cordyceps genus. The 2'-deoxyadenosine was detected in C. sinensis,Hirsutella sinensis and C. sobolifera. The cordycepin was detected in C. militaris. In summary,the cordycepin chromatographic peak from C. sinensis in some past reports may be the 2'-deoxyadenosine chromatographic peak or the mixture peak of 2'-deoxyadenosine and cordycepin in which 2'-deoxyadenosine content was higher than cordycepin. The developed method is suitable for analysis of cordycepin and 2'-deoxyadenosine in Cordyceps genus.
