Aim or purpose: This study aimed to develop a keratin-based microneedle patch loaded with hop extract (KMNs@HE) to overcome limitations of current oral ulcer therapies, such as short retention time and poor drug penetration (C. Vitamia, G.N. Iftinan, I.R. Latarissa, et al. Drug Des Devel Ther, 2024,18:1297 - 1312.). Materials and methods: Eight-week-old male Sprague-Dawley rats (approved by Ethics Committee, No. QYFY WZLL 27832) with induced oral ulcers were divided into control (CON), keratin-only microneedle (KMNs), KMNs@HE, and commercial chitosan film (CTS) groups. Methods included antimicrobial/antioxidant assays, cytotoxicity testing (CCK-8 on L929 fibroblasts), microneedle characterization (SEM, mechanical tests), histopathology (HE, Masson, Sirius Red), and immunohistochemistry (CK5, CK13, TNF-α, IL-6). Results: The 1000 μg/mL hop extract exhibited potent antimicrobial activity (100% inhibition at 12 h), high antioxidant capacity (DPPH: 90%, ABTS: 96%), and biocompatibility (cell viability >70%). KMNs@HE demonstrated sustained drug release (50% in 2 h, full release in 12 h), robust mechanical strength (120 N), and effective mucosal penetration (500 μm depth). In vivo, KMNs@HE achieved 85% ulcer closure by day 5 (P = 0.0184 vs. CTS), complete healing by day 7 (P < 0.001 vs. CON/KMNs), enhanced collagen regeneration, and reduced inflammatory markers (IL-6, TNF-α). Conclusions: KMNs@HE offers a pain-free, sustained-release system with superior healing efficacy and biocompatibility, positioning it as a promising alternative for oral ulcer therapy.
Oral ulcers are a prevalent manifestation of various oral diseases, often resulting in notable pain and discomfort, potentially leading to infection or even malignancy without proper intervention. However, the effectiveness of local therapeutic drugs is limited owing to their short residence time in the oral cavity, with the unique environment such as saliva flow and mechanical actions of lips and tongue, leading to difficulty in penetrating the mucosal and pseudomembrane barriers. Herein, keratin and hyaluronic acid-based microneedle patches loaded with hop extract (KMNs@HE) have been developed for the treatment of oral ulcers. The prepared microneedle patches exhibit a well-arranged microarray and good mechanical properties, which successfully penetrate the pseudomembrane barrier of buccal mucosa ulcer of SD rats. It also shows good antimicrobial activity, acceptable biocompatibility, with a sustained release profile of hop extract for 12 h. Histopathological analysis reveals that the KMNs@HE promotes collagen formation and ameliorates inflammation by downregulating the protein expression of TNF-α and IL-6, thereby accelerating the healing process in the SD rat ulcer model. The developed KMNs@HE has the potential to enhance the comfort and effectiveness of oral ulcer treatment, offering a promising avenue for future research in the treatment of oral mucosal diseases.
The healing of tooth extraction wounds is significantly influenced by various factors, including interference from the oral microenvironment, invasion of gingival tissue, and inflammation of the alveolar socket, all of which contribute to the substantial loss of bone tissue in the alveolar socket. This study employed electrospinning technology to fabricate polyvinyl alcohol (PVA)/sodium alginate (SA) nanofiber scaffolds infused with freeze-dried concentrated growth factor (CGF) and nanohydroxyapatite (nHA). The objective was to investigate the repair mechanism of the PVA/SA/CGF/nHA nanofibers for oral alveolar bone defects, thereby offering novel treatment strategies for bone defect repair. In vitro experimental results demonstrate that the addition of nHA significantly enhances both the degradation rate and swelling ratio of the PVA/SA/CGF/nHA fiber membrane. Additionally, the scaffold exhibits favorable microstructural properties and biocompatibility. The sustained release of fibrin in CGF suggests that the fibrous membrane maintains a stable three-dimensional structure, facilitating the slow and sustained release of CGF. Alkaline phosphatase (ALP) and alizarin red staining indicate a significant promotion of osteogenic differentiation of human bone marrow stem cells (hBMSCs). Furthermore, qRT-PCR results reveal increased expression levels of the ALP, collagen type-1 (Col1), runt-related transcription factor 2 (Runx2), osteocalcin (OCN), and osteopontin (OPN) genes. To further investigate the impact of the local application of the nanofiber membrane on bone regeneration, a rat tooth extraction wound model was established. These results confirm that the electrospun PVA/SA/CGF/nHA nanofiber membrane significantly promotes the proliferation, migration, and osteogenic differentiation of hBMSCs. This finding offers a novel treatment for oral alveolar bone defects and has potential clinical applications in the oral cavity.
Objective This study investigated the clinical effects and applicability of minimally invasive impacted teeth extraction using digital robots. Methods A marker was bonded to the non-surgical area before surgery. A Cone-Beam Computed Tomography (CBCT) scan was obtained and uploaded to the robot software to determine the drilling position of the ring drill. During the surgery, the robot arm automatically navigated to a predetermined position, and the ring drill removed part of the bone tissue and exposed and extracted the impacted teeth. Finally, the surgeon tightly sutured the wounds to the surgical area. Results Three minimally invasive extractions of impacted teeth with robotic assistance were performed without complications. The surgical area showed good healing during the one-month follow-up examination. Conclusions Digital robot-assisted minimally invasive extraction of impacted teeth is a highly feasible clinical procedure as it minimises trauma to the surgical area and protects the surrounding blood vessels and nerve bundles, making it a safe and valuable technique with significant potential for clinical application.
The extract of Hops (Humulus lupulus L.), a herb medicine with a rich history spanning over 2000 years, has recently received extensive attention in food, health, and biomedicine. However, its reports in oral application are rare. To provide fundamental information for further clinical study in oral treatment, in this work, pleiotropic activity of hop extract including the stability, cytotoxicity, antioxidant activity, and antimicrobial activity of hop extract are investigated. The UV and thermal stability of hop extract were assessed using FT-IR analysis, while antioxidant activity was determined through experiments measuring DPPH, ABTS, and hydroxyl radical scavenging activities. Cytotoxicity was evaluated using the CCK-8 assay. Furthermore, we examined the antimicrobial activities of hop extract against four pathogenic microorganisms commonly associated with oral diseases—namely, E. faecalis, S. aureus, E. coli, and C. albicans—through inhibition zone and colony-forming unit assays. Experimental results show that hop extract has good UV and thermal stability, as well as good biological biocompatibility and antioxidant activity. Importantly, antimicrobial tests indicate that hop extract exhibits not only strong antibacterial activity against both Gram-positive and Gram-negative bacteria, but also effective antifungal ability. This study highlights the potential of hop extract as a promising antimicrobial agent for oral treatment. It serves as a crucial first step towards further applications and paves the way for the development of new treatments in oral therapies.
Objective: The aim of this study was to investigate the presence of phosphorylated SMAD2/3 (P-SMAD2/3) in periapical lesions in humans and its possible correlation with matrix metalloproteinase 9 (MMP9) during the development of apical periodontitis.Design: In this study, a total of 38 samples were collected, consisting of 16 healthy controls and 22 periapical lesions. These samples underwent fixation, dehydration, and embedding for further histologic and immuno-chemical analysis. The expression of phosphorylated SMAD2/3 and MMP9 was quantified using the average integrated optical density. Additionally, immunofluorescence analysis was conducted to investigate the colocalization of phosphorylated SMAD2/3 and MMP9. Results: The study found that periapical lesions exhibited a stronger expression of MMP9 compared to healthy controls. Additionally, the expression of phosphorylated SMAD2/3 was observed to increase in the periapical granulomas and radicular cysts group, as compared to the normal group (P < 0.01). The results of the immunofluorescence test showed that phosphorylated SMAD2/3 was colocalized with MMP9.Conclusions: The study found that SMAD2/3 phosphorylation is correlated with matrix metalloproteinase 9 expression in human periapical lesions, suggesting its potential involvement in tissue destruction and immune cell infiltration in periapical lesions.
ABSTRACT The precise movement of the maxilla is particularly important for orthognathic surgery, especially for patients with maxillary segmentation. In this preliminary study, the authors present a new tooth bone combined with a supporting osteotomy guide and positioning guide to guide the osteotomy and reduction of the maxilla. Through our preoperative simulation and postoperative image fusion, the authors found that the overlapping area is more than 90%. According to compare of the virtual plans and the postoperative results based on distances from the maxillary land- marks to the horizontal plane, sagittal plane, and coronal plane, the surgical error was about 2mm. Our T-shaped guide provides a reliable method for patients with maxillary segmental osteotomy, which may be a useful alternative to the intermediate.
临床上早期至现今,教科书、参考书乃至各种相关文献杂志均有"阻塞性睡眠呼吸暂停综合征"这一医学术语,又名"阻塞性睡眠呼吸暂停低通气综合征",其英文缩写为OSAHS。鉴于OSAHS的命名体现不出该病的病理生理特点及其因果关系,本文提出"睡眠性上呼吸道通气不畅或梗阻-呼吸暂停综合征"(SURFOAS)这一命名,并通过与前两者的全方位比较,认为SURFOAS更能体现该病的主要病理生理特征,尤其从命名中还可明确其通气不畅与梗阻的部位。本文从SURFOAS患者上呼吸道解剖中列举出诸多异常结构的改变(论据),进而阐明睡眠与上呼吸道通气不畅或梗阻乃至呼吸暂停之间的因果关系。因此,SURFOAS的提出与命名,相比OSAHS而言,无论横向比较,还是纵向比对,均具有合理性,合乎逻辑,且符合临床疾病命名原则,对于医学基础理论指导临床实践而言,相对更加规范和标准。

MicroRNAs are noncoding RNAs of 21 to 23 nucleotides in length that play important roles in almost all biological pathways. The roles of microRNA-299-3p in the development and progression of oral squamous cell carcinoma remain unclear. Expression level of microRNA-299-3p in oral squamous cell carcinoma cell lines was analyzed. Then, the effects of microRNA-299-3p on oral squamous cell carcinoma cell proliferation and migration were investigated. Moreover, bioinformation algorithm and Western blot were conducted to explore whether forkhead box P4 was a direct target of miR-299-3p. We showed that microRNA-299-3p expression was significantly reduced in oral squamous cell carcinoma cell lines. Next, overexpression of microRNA-299-3p was found to inhibit oral squamous cell carcinoma cell proliferation and migration but promote apoptosis. In addition, forkhead box P4 was identified as a functional target of microRNA-299-3p. Our results provide a new perspective for the mechanisms underlying the progression of oral squamous cell carcinoma and a novel target for the treatment of oral squamous cell carcinoma.
Production process of cotton fabric involves numerous water and chemical consumptions, causing serious energy waste and environmental problem. In this study, a waterless atmospheric plasma treatment (APT) was applied on cotton fabric to desize the starch coated in the weaving process. Plasma parameters, i.e., plasma voltage, plasma duty cycle and oxygen flow rate, are varied in order to systematically evaluate their influence on fabric physical properties such as the impurity removal, whiteness, capillary effect, tensile strength and breaking elongation of fabric. Optimal plasma parameters for oxygen APT were determined, with plasma voltage of 320 V, plasma duty cycle of 60 % and oxygen flow rate of 1000 l/h. The APT desizing method attained comparable fabric performances with the traditional amylase desizing method. The results were further supported by the scanning electron microscope (SEM) and Fourier transfer infrared spectroscopy (FTIR) investigations.

PURPOSE:To study the clinical effect of "gelatamp" colloidal silver gelatin sponge on preventing dry socket after extracting the mandibular impacted teeth.METHODS:1350 teeth extracted from 976 patients who needed to extract the mandibular impacted teeth were divided into group A, group B and group C randomly. "Gelatamp" colloidal silver gelatin sponge was implanted into alveolar socket after teeth extraction in group A, with absorbable gelatin sponge was implanted into alveolar socket in group B and nothing was implanted into alveolar socket in group C. The incidence of dry socket was observed, the data was analyzed using SPSS10.0 software package.RESULTS:The incidence of dry socket was 0.44% in group A, 2% in group B and 4.44% in group C. There was significant difference in the incidence of dry socket between group A and group C(P<0.01). There was also significant difference between group B and group C(P<0.05) and between group A and group B(P<0.05).CONCLUSIONS:The results demonstrate that "gelatamp" colloidal silver gelatin sponge can prevent the occurrence of dry socket after teeth extraction, which is of wide clinical use.
Objective To investigate the morphology of the first permanent premolars from some youth and junior in Shandong region and provide anatomic data and therapy basis for clinical treatment,education and scientific research in department of stomatology.Methods One hundred and forty six freshly extracted first permanent premolars of youth and junior in Shandong region were collected and grouped by position and gender for 8 groups.The teeth were measured according to the established method,including the length of tooth,height of dental crown,length of dental root,the ratio of height of dental crown and length of dental root,width of dental crown,width of dental neck,the ratio of width of dental crown and width of dental neck,deep of dental crown,depth of dental neck,the ratio of deep of dental crown and depth of dental neck.Then the data were analyzed using the SPSS 13.0 software package.Results Some anatomic data of the first permanent premolars of hobbledehoy in Shandong region were obtained.Conclusion The anatomic data between the right and left position of the same gender had no difference and between different gender of the same position had some differences.
Objective To Investigate the root canal configuration and canal curvature of permanent mandibular anterior teeth in labiolingual direction.Methods A total of 105 anterior teeth were collected and examined by indirect digital X-ray radiography from labiolingual direction.The canal configuration and curvature was analyzed by Weine and Schneider methods.Results Most root canals of mandibular anterior teeth were type I,and more than half were mesial-distally curved.The most curved place was located in the apical third of the canal.Conclusion Root canal configuration of mandibular anterior teeth are relatively complex and have a higher bending rates.The direction of pulp explosure should be close to the incisal edge.
Mandibular incisors are the smallest human permanent teeth with complex root canals. Double root canal, lateral branch of root canal, apical ramification or apical furcation usually happen, which make the clinical therapy more difficult. In this paper, a case of mandibular incisors with double root canals was reported, the root canal type and its incidence were discussed.
