Предложен новый способ консервирования ядросодержащих клеток в электрическом холодильнике с использованием ксенона. После суточного криоанабиоза (80°С) при медленном охлаждении биообъекта более 60% лейкоцитов обладает устойчивой к витальному красителю клеточной мембраной, у 85% гранулоцитов сохраняется исходный уровень лизосомально-катионных белков, снижается интенсивность перекисного окисления липидов и активность ферментативных систем, регулирующих содержание перекисей. Криоконсервирование биообъектов в среде инертных газов является перспективным направлением в практической медицине и может стать альтернативным способом традиционному способу с использованием жидкого азота.
10-, 50-, 100-, 500-, and 1000-fold dilutions of venous serum of healthy women, healthy men and men with coronary heart disease were studied in the experiments with 195 ring segments of the porcine right coronary artery. Human serum provides M-cholineblocking activity that can be explained by the presence of an endogenous non-selective M-cholinoreceptor blocker whose content depends on the gender and the presence of coronary heart disease.
The aim of this work is the estimation of dehydrational structurization the blood serum in mouses takin into account the conditions of formation the lynphoid leucosis (AKR line). The authors studied crystallogenous properties of the blood plasma of mouses AKR (n=20) and Black (n=15) lines. For biological fluids own and initiated crystallogenesous the authors used special semiquantitive criterias. It was established, that experimental lymphoid leucosis allows to change the blood composition and transformation of its crystallogenous potential.
Acetylcholine (ACh, 10-10–10-6 g/ml) dose-dependently increases the rate of erythrocyte agglutination in men. This effect is reduced by selective blockers of M1and M3-cholinergic receptors (ChR): gastrotsepin and 4-DAMP, inhibitor of cyclooxygenase and phospholipase A2 indomethacin, inhibitor of calmodulin trifluoperazine, blocker of Ca2+-dependent K+-channels BaCl2, and blocker of Ca2+-channels verapamil. This means that the effect of ACh is caused by the activation of M1and M3-ChR and involves cyclooxygenase, phospholipase A2, calmodulin and Ca2+and К+-channels.
On strips of the isolated myocardium of right hearts auriculum of the 43 patients with ischemic illness of heart and 9 patients with heart diseases of various ethyology at statement venous canule during aorto-coronary shunting, estimated influence of adrenaline (10(-9)-10(-4) g/ml) on amplitude caused by electrostimulus (1H, 5ms, 25-30 V) contractions, and also inotropic and adrenomodulation activity of serum blood (in dilution 1 : 10000, 1: 1000, 1 : 500, 1: 100, 1 : 50, 1: 10 and 1 : 5) nonpregnant women. Direct dependence of amplitude of contraction on size of fraction of of blood emission on Teyholts is revealed. It means, that strips of right auriculum myocardium reflect contractility of a left ventriculum myocardium. Adrenaline in concentration 10(-7)-10(-6) g/ml dependent of dose raised amplitude of the caused contraction not influencing it in concentration of 10(-9) and 10(-8) g/ml (the constant of dissotiation has 2 x 10(-7) g/ml), that as a whole, speaks about decrease in efficiency of activation beta-AP. Blood Serum in dissolutions 1 : 10000-1 : 50 did not influence on amplitude of contraction, and in dissolutions 1 : 10 and 1 : 5 strengthened it, that speaks presence in blood the endogenous activator of myocyte contractility (EAMC). Serum showed beta-adrenomodulation activity that speaks presence in it endogenous sensitizer of beta-adrenoreceptors (ESBAR) and endogenous blocker of beta-adrenoreceptors (EBBAR). In particular, in experiences with adrenaline in subthreshold concentration (10(-8) g/ml) serum showed ESBAR-activity (in dissolutions 1 : 1000, 1 : 500, 1 : 100 and 1 : 50), and in experiences with adrenaline in as much as possible effective concentration (10(-6) g/ml) serum showed ESBAR-activity (in dissolutions 1 : 50 and 1 : 10) and EBBAR-activity (in dissolutions 1:500) Hence, containing in blood serum endogenous modulators of beta-adrenoreactivity - ESBAR and EBBAR can modulate efficiency of beta-adrenoreceptors activation of human cardiomyocytes. It speaks about perspectivity of application of ES BAR analogues in cardiology.
In experiments with longitudinal stripes of cow renal artery, serum taken from healthy people and patients with Stage II arterial hypertension (AH), in titrations 1:100, 1:500, 1:1000, and 1:10000, did not influence tonotropic, alpha-drenoreceptor (AR) activating effect of adrenaline (10–6 g/ml). In titration 1:50, this effect was enhanced, due to serum alpha-adrenosensitizing activity. In Stage III AH, female serum did not affect tonotropic effect of adrenaline, and male serum reduced it in titrations 1:50, 1:100, and 1:1000, due to alpha-adrenoblocking activity. Therefore, 1) serum of cormotensive people and patients with Stage II AH could contain endogenous sensitizer of alpha-AR (ESAAR), and in Stage III AH its level might be reduced; 2) serum of males with Stage III AH could contain endogenous blocker of alpha?AR (EBAAR). The nature of ASAAR and EBAAR and their role in AH pathogenesis is discussed.
