Рассмотрены свойства ретинального белка (ESR) психротрофной бактерии Exiguobacterium sibiricum, представляющего собой светозависимую протонную помпу. Уникальной структурной особенностью ESR является наличие остатка лизина в положении, соответствующем внутрибелковому донору протонов для основания Шиффа. Мы показали, что Lys96 успешно выполняет в молекуле ESR функцию донора, облегчая доставку протонов с цитоплазматической поверхности белка к основанию Шиффа. Поскольку поглощение протонов предшествует репротонированию основания Шиффа, можно предположить, что в исходном состоянии этот остаток не заряжен и приобретает протон в течение короткого промежутка времени после депротонирования основания Шиффа и образования интермедиата М. Это отличает ESR от родственных ретинальных белков - бактериородопсина (BR), протеородопсина (PR) и ксантородопсина (XR), в которых функцию донора выполняют остатки с карбоксильной группой, протонированные в исходном состоянии. Как и другие протонные помпы эубактерий (PR и XR), ESR содержит остаток гистидина, взаимодействущий с акцептором протонов Asp85. Это взаимодействие приводит к сдвигу рКа акцептора в более кислую область по сравнению с PR, обеспечивая его способность к функционированию в широком диапазоне рН. Наличие сильной водородной связи между остатками Asp85 и His57, структура вероятных протонпереносящих путей с цитоплазматической поверхности к основанию Шиффа и к наружной поверхности и другие особенности ESR продемонстрированы благодаря расшифровке его пространственной структуры, которая выявляет ряд отличий от известных структур BR и XR. Структура ESR, схема фотоцикла и реакций переноса протонов обсуждаются в сравнении с гомологичными ретинальными белками.
Small-angle X-ray scattering technique has been used to study multilayer lipid membranes of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) and the 3:1-mixture DPPC/1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) in excess water. The temperature dependences of the repetition period for lipid bilayers in the temperature range 20–55 ◦C are obtained. A comparative analysis of the scattering curves obtained for multilayer membranes showed that, below a temperature of 40 ◦C, there emerges an additional ordering with a repetition period of 66 Å in the lipid mixture, which we associate with the lipid phase separation. A disappearance of the so-called ripple (wave-like) phase of DPPC lipid in the mixture is also observed.
У роботі методом малокутового розсіяння рентгенівських променів досліджено мультишарові ліпідні мембрани ДПФХ (1,2-дипальмітоіл-sn-гліцеро-3-фосфатидилхолін) та суміші ДПФХ/ПОФХ (1-пальмітоіл-2-олеіл-sn-гліцеро-3-фосфатидилхолін) у співвідношенні 3:1 у надлишку води. Отримано температурні залежності періодів повторювання ліпідних бішарів у діапазоні температур 20–55 ºС. Порівняльний аналіз кривих розсіяння відмультишарових мембран показав, що нижче температури 40 ºС в суміші ліпідів існує додаткове впорядкування з періодом повторювання 66 Å, яке пов'язано, на нашу думку, з поділом фаз ліпідів. Також у суміші спостерігається зникнення так званої ріпл-фази (хвилеподібної фази) ліпіду ДПФХ.
The results of a study of the temperature and pressure dependence of isothermal compressibility of dimyristoylphosphatidylcholine (DMPC) are presented. The data were obtained using a small-angle X-ray scattering instrument in the Institute of Synthetic Polymer Materials (Moscow, Russia) and a device designed in the Laboratory of Neutron Physics, Joint Institute for Nuclear Research (Dubna, Russia), for studying the liquid systems by a volumetric P-V-T method. The minute baric investigation of the region of the main phase transition (transition from the gel phase into the liquid crystalline phase) was carried out. The points of the main phase transition were determined, and the phase diagram of DMPC was plotted over the pressure range of 1–800 bar at temperature from 24 to 35°C.
The software package for the YuMO spectrometer has been updated. The MS DOS programs used before the updating are compared to the similar new programs operating in the OS-9 environment. The merits and demerits of each program are discussed. The main features of the new software are described, and the results of the long-term operation of the new software package are analyzed. It is shown that upgrading the software has improved spectrometer control.
The structure of allylcarbosilane dendrimers of the fifth, sixth, and seventh generations in solutions was studied using SANS and SAXS methods. It was shown that small-angle scattering profiles of dendrimers can be accurately modeled by scattering from uniform globular particles. Using the SANS contrast variation method, it was demonstrated that dendrimer molecules in solutions do not contain closed inner cavities inaccessible to the solvent. Based on the intensity of small-angle neutron scattering measured in absolute units, the partial volume of the dendrimer in solution was determined and the volume fraction (30-40%) of open inner cavities in the efficient dendrimer volume accessible to the solvent was calculated. It was discovered that the end groups of dendrimers are localized in their surface layer.
