We examined postoperative material from 28 patients aged 39-61 years with gliomas of different degrees of anaplasia (the diagnosis was histologically verified according to the WHO classification of CNS tumors) who had not previously received antitumor treatment. In glioma tissue, the glucose concentration was significantly higher than in the brain tissue of subjects dead from traumas (control), while lactate concentration did not differ from that in the control group or was lower. Hexokinase activity demonstrated a tendency to an increase in grade I and significant elevation in grades II and III, while in grade IV gliomas, this parameter did not differ from the control. Activities of the pentose-phosphate pathway enzymes glucose-6-phosphate dehydrogenase and transketolase increased with increasing of tumor anaplasia. Activity of glycogen synthase 3β kinase was significantly higher than in the control group. IDH1 mutation was discovered in 40% cases, the MGMT promoter methylation was detected in more than 50%, the Ki-67 level increased with increasing tumor anaplasia. The most significant correlations with glioma markers were detected for glucose-6-phosphate dehydrogenase and glycogen synthase 3β kinase. Activities of the studied enzymes of carbohydrate metabolism significantly correlated with Ki-67 marker.
Blood plasma of 39 cancer patients who had not previously undergone anti-tumor treatment, and 21 patients with recurrent epithelial tissues cancer before treatment and after the first course of chemotherapy were investigated. The treatment effectiveness of patients with relapse was evaluated throughout the year. The control group was the blood plasma of 14 healthy people. Free radical activity and oxidative modification of proteins were assessed in blood plasma. In malignant neoplasms of epithelial tissues, free radical activity and oxidative modification of plasma proteins are significantly higher. At the initial stages of carcinogenesis, the oxidative modification level of plasma proteins increases both due to aldehyde and ketone carbonyl derivatives. In the terminal stage of malignant tumors, an increase in the oxidative modification of proteins occurs due to aliphatic ketone dinitrophenyl hydrazones, which are the markers of protein aggregation characteristic of the late stages of oxidative stress. In case of an objective response to polychemotherapy, there is a significant decrease in free radical plasma activity after the first course of therapy and a significant increase in carbonyl derivatives recorded at a wavelength of 230 nm, which can serve as a marker of its effectiveness.
Aim. Analysis of the electrophoretic mobility of red blood cells in patients with the diseases caused by increased cell proliferation activity, and its relationship with the lipid spectrum of erythrocyte membranes. Methods. The blood of 42 patients with malignant tumors of epithelial tissues (colon, kidney, prostate, urine bladder and cervical cancer) and 16 patients with skin diseases was studied. The control group consisted of 20 healthy individuals. Electrophoretic mobility of red blood cells was assessed by microelectrophoresis. Lipid fractions levels in erythrocytes was determined by one-dimensional thin layer chromatography. Results. Over than two-fold decrease of electrophoretic mobility of erythrocytes was observed in patients with malignant tumors of epithelial tissues and skin diseases associated with cell proliferation processes (psoriasis) - by 59 and 60%, respectively In inflammatory skin diseases (microbial eczema), the decrease was less pronounced (30%). The contents of charged phospholipid in erythrocyte membranes in case of cancer and psoriasis has changed in a similar way: a significant reduction in sphingomyelin level in the absence of significant differences in phosphatidylethanolamine and phosphatidylcholine levels. Multidirectional statistically significant changes in the contents of lysophosphatidylcholine were revealed depending on the stage of cancer, their level was decreased two-fold in case of psoriasis. The changes of cholesterol, diacylglycerols, free fatty acids levels in the membranes of red blood cells in case of cancer and psoriasis were divergent. Conclusion. In diseases caused by cell proliferation activation, electrophoretic mobility of erythrocytes is reduced by more than 50%. Kinetic characteristic of erythrocytes may be used for detecting the conditions of pathologic cell proliferation, including malignant tumors, since they alter in cases of phospholipid membranes lipid spectrum disorders.
Abstract. The article presents data about blood serum cytokine profile in patients with chronic widespread dermatoses, associated with increase in blood markers of endogenous intoxication (EI) syndrome, i.e., lowand medium-molecular mass substances (LMWS). A close interrelation has been revealed between EI levels, and cytokine profiles. In the patients with high amounts of blood LMWS, increased levels of anti-inflammatory IL have been shown. Correlation analysis of clinical and laboratory parameters has demonstrated a leading role of EI in pathogenetic mechanisms of chronic extended dermatoses, and brings about altered regulation of metabolic processes. A severe clinical course of dermatoses (i.e., therapy resistance) was associated with a considerable decrease in blood IL-8 and IL-10 levels, along with high IgE and LMWS amounts. (Med. Целью работы явилось изучение влияния препаратов изотретиноина (акнекутан и роаккутан) с различной биодоступностью на белковый гомеостаз больных с тяжелыми формами акне. Под наблюдением находились 35 больных c тяжелыми формами акне (основная группа), в группу сравнения были включены 23 человека. Исследовались общая окислительная модификация белков, в том числе альбумина, а также характер собственной и инициированной кристаллизации различных биосубстратов (сыворотка крови, слюна) у данной группы больных с использованием методов классической кристаллоскопии и сравнительной тезиграфии. Установлено нарушение белкового гомеостаза у больных тяжелыми формами акне. По данным биохимических и биокристалломных методов выявлены различия во влиянии на белковую составляющую гомеостаза разных форм изотретиноина при их сопоставимом клиническом эффекте.
32 patients with acne were surveyed to study changes in total protein oxidative modification of blood. Along with the generally accepted clinical and laboratory findings in patients electrophoresis of serum proteins by the severity of oxidative stress on the content of carbonyl derivatives (in spontaneous oxidative modification of proteins) was determined.It is proved that the treatment of acne patients with systemic retinoids is accompanied by oxidative modification of serum proteins. The consequence of these processes can be aggravating oxidative stress and increased body harmful effects caused by free radicals in tissues.Considering not sufficient study of the effects of systemic retinoid therapy, the treatment of acne should be strictly controlled, and in the selection of the dosage form should be dominated by the principles of more bioavailability and maximum effective dosage.
32 patients with inflammatory skin diseases have been examined to study the degree of oxidative modification of serum albumin. The quantitative content of modified and effective serum albumin and its carbonyl derivatives were evaluated. We have found out a statistically significant decrease in the effective albumin concentration and an increase in the level of ketone carbonyl groups (370 nm) during spontaneous and induced oxidative modifications of protein in patients with inflammatory skin processes relative to the control group. It has been revealed that by the end of treatment with systemic retinoids the level of spontaneous oxidative modification of albumin decreases and the content of effective albumin increases.
The aim of the investigation was to assess the interaction of tumor marker level, the indices of protein and mineral metabolism, free radical oxidation in blood plasma and the tumor-forming stage in malignant processes in epithelial tissues.Materials and Methods. Blood plasma of 73 patients with epithelial tissue malignant neoplasms and 31 apparently healthy people were studied. Blood plasma biochemical parameters were assessed using the analyzers: free-radical activity - by induced biochemiluminescence; oxidative protein modification - by the level of carbonyl derivatives; elemental analysis - using atomic emission spectrometry.Results. Low diagnostic value of the determination of oncomarkers at early stage of the studied oncological diseases was shown. We observed the change of blood plasma biochemical parameters at early carcinogenesis stage: the albumin level reduction and the concentration increase of urea, alpha 1-and gamma-globulin fractions. Cancer patients were found to have impaired element homeostasis: Na, Fe, Cu, Li level decrease, K, P, Sr increase. We revealed the activation of free-radical oxidation and oxidative protein modification, the correlation of the intensity of these processes with the content of some elements in blood plasma. Integral analysis of blood plasma biochemical parameters increases diagnostic value of the determination of tumor markers in the detection of malignant tumors of epithelial tissues.

It is demonstrated that in comparison with healthy patients in examined group of patients this indicator is increased mainly in patients with atopic dermatitis (up to 62%) and least of all with psoriasis (up to 25%) and bullous dermatitis (up to 18%). The statistically reliable differences of this indicator from control values under endogenic intoxication and different degree of severity of disease are determined. The detection of level of 8-OH-2-desoxiguanosin in blood serum is an informative technique of analysis to increase quality of laboratory monitoring of oxidizing stress.
Aim. To investigate the relationship between blood plasma biochemical indicators in patients with solid tumors before a treatment and after the first course of chemotherapy with objective treatment response. Methods. Blood plasma samples taken from 14 patients with cancer relapse were studied before the treatment and after the first course of specific chemotherapy (carboplatin, methotrexate, vinblastine in patients with urine bladder cancer and irinotecan, leukovorin and fluorouracil in patients with colorectal cancer). The first group included patients with colorectal cancer relapse (males - 3, females - 4) aged 57-62 years. The second group included patients with urine bladder cancer relapse (males - 5, females - 2) aged 48-64 years. Free radical activity and protein oxidative modification, as well as endogenous intoxication and major mineral and trace elements levels were studied. Results. When achieving the objective chemotherapy effect in patients after the first treatment course, an increase of alpha-1 globulin and gamma globulin level, as well as an increase of phosphorus, zinc, lithium and iron blood plasma level was observed. In case of following disease progression, the opposite dynamics of above-mentioned indicators was revealed. In patients with good objective effect of polychemotherapy after the first treatment course, a significant increase of endogenous intoxication and blood plasma free radical activity decrease were observed. If the following tumor progression was observed, the tendency of blood plasma free radical activity increase, accompanied by the total protein oxidative modification activation, was revealed after the first treatment course. Conclusion. The increase of alpha-1 and gamma globulin levels, as well as an increase of phosphorus level in blood plasma after the first course of polychemotherapy can be used as prognostic factors of anticancer therapy efficiency. High level of endogenous intoxication as well as an increase of copper, iron, zinc and lithium blood plasma concentrations can be applied as the additional markers.

Проведен анализ степени окислительной модификации липопротеинов высокой (ЛПВП) и низкой плотности (ЛПНП), выделенных из сыворотки крови 54 больных хроническими распространенными дерматозами (псориаз, атопический дерматит, пузырные дерматозы). Сравнивались группы больных с обычным течением заболевания и хорошим эффектом после проведенной комплексной терапии и очень тяжелые больные с часто рецидивирующими проявлениями дерматозов, резистентными к применяемой терапии и торпидным течением. Показано, что наибольшему влиянию активных форм кислорода подвержены молекулы ЛПНП. При этом нарушаются процессы пероксидации как белковой, так и липидной ее части. Зарегистрирован высокий статистически значимый по сравнению со здоровыми лицами уровень альдегидных и кетонных карбонильных производных апо-белков и сниженнное количество малонового диальдегида – продукта перекисного окисления липидов, но не выявлено статистически значимых различий показателей в зависимости от вида дерматоза. Однако степень выраженности окислительных процессов как белковой, так и липидной составляющей ЛПНП имеет зависимость от тяжести течения заболевания.
