Aim To investigate the protection of oxymatrine on focal crerebral ischemic injury in rats and its mechanism of inhibition of opoptosis.Methods The method of permanent middle cerebral artery occlusion in rats(permanent middle cerebral artery occlusion,pMCAO) was adopted to establish cerebral ischemia model,Through the abdominal cavity pMCAO postoperative rats were given OMT(30,60,120 mg·kg-1).Evaluation index like cerebral infarction volume,water content and behavioral symptoms of brain were taken account in terms of OMT in cerebral ischemia neural protection.Through HE dyeing method the influence of OMT on form change and number of ischemic cortex of nerve cells was observed.Western blot was applied to detect OMT of ischemic cerebral cortex Caspase-3,Bcl-2,Bax protein level of expression.Results In rat focal cerebral ischemia body model,OMT(30,60,120 mg·kg-1) could significantly reduce the cerebellum infarction volume,water content and improve brain behavior signs(P0.01).HE dyeing results indicated: OMT could significantly increase the survival rate of nerve cells and improve the form of nerve cells.Western blot results showed: compared with false surgery group,PMCAO rats after 3,6,12,24 h,ischemic cortex Caspase-3,Bax protein expression level in 3 h began to rise and peaked at 24 h;Bcl-2 in the expression level 3h began to drop,24 h to a minimum.After given the OMT,the levels of Caspase-3,Bax protein in PMCAO rat cortex ischemia could down-regulated and the levels of Bcl-2 protein up-regulated.Conclusions The current finding provide the first evidence that OMT has a direct neuroprotective activity in cerebral ischemic neuronal injury,which is probably related to the inhibition of opoptosis by up-regulation of the levels of Bcl-2 and down-regulation of the levels of Bax and Caspase-3 singnal way.
AIM:To construct a 3D model of the chimeric antibodies (AntiCD28: ch-2F5) with corresponding antigen molecule docked to theoretically verify the rationality of the binding of antibody with its antigen and to provide a method of 3D identification between antigen and antibody and spatial structure analysis.METHODS:We analyzed the sequence by submitting it to http://www.ncbi.nlm.nih.gov/ and made a comparison using integratly the 3 databases of GenBank, Protein data bank and GENO-3D. The 3D model was constructed by Swiss-model homology modeling server and molecular docking online was performed by GRAMM-X Protein Docking Web Server. Chimeric heavy chain, light chain, heavy-light chain complex, heavy-light chain and antigen complex were displayed and photographed by the Chimera Software. Meanwhile, the spatial structures of heavy, light chains, variable region, constant region, CDR and frame area were marked by different colours respectively to exhibit the 3D structure on every side.RESULTS:The 3D structure of the heavy-light chain and antigen complex we constructed was consistent well with the theory of antigen binding to antibody molecules.CONCLUSION:The structure of the chimeric antibody we constructed with the bioinformatic method was in accordance with the general structure of antibody, and its antigen binding site was also consistent with the molecular theory. Thus, the model helps to analyze the 3D structure of antibody and antigen-antibody interaction.
采用粒子群优化算法(PSO,Particle Swarm Optimization)调整径向基神经网络(RBFNN,Radial Basis Function Neural Network)参数,有效地改进了传统的K-均值聚类算法易使网络受初始参数选取的影响而收敛于局部极小值的缺点,并将其应用于环境中硝基苯类化合物毒性的测定,结果表明基于PSO的RBFNN算法显著提高了数据处理的准确性和稳定性。
Objective To research the effects of a chiral aminoalcohol in the asymmetric allylation of acylhydrazones.Methods The asymmetric allylation of acylhydrazones with allyltrichlorosilane under the stoichiometric amounts of(1S,2S)-1-(p-nitrophenyl)-2-(3′,4′-methylenedioxabenzylamino) propane-1,3-diol(compoundⅠ) was carried out and the enantioselective excess(e.e.) value of productions were detected by high performance liquid chromatography.Results The chiral aminoalcohol ligand compound Ⅰcan promote the asymmetric allylation of acylhydrazones and the yield was 57%;The highest values of e.e.obtained for this reaction was 22%.Temperature can affect the yield,reaction in 0 ~ 15 ℃conditions are relatively appropriate.Conclusion Chiral compounds Ⅰ for the first time was applied to allylation of acylhydrazones with allyltrichlorosilane and the results showed that the compound Ⅰ can contribute to the occurrence of the reaction.
A neural network based on Particle Swarm Optimization(PSO) is adopted to solve the problem of simultaneously analyzing As(Ⅲ) and As(Ⅴ) using Spectrophotometer Analysis.The weights of multi-layer forward neural network are trained by the PSO algorithm,which overcomes the disadvantages of error converging slowly and dropping into the local minimum inhering in traditional Back Propagation(BP) neural network.And the results show that the method improves the accuracy and stability of data processing greatly.
Objective To investigate the correlation between the polymorphisms of CYP11B2(C-344T) and essential hypertensive risk stratification.Methods According to Chinese guideline for prevention and treatment of hypertension,244 cases with essential hypertension were divided into medium risk group,high risk group and very high risk group.Polymorphisms of CYP11B2(C-344T)gene were detected by PCR-restriction fragment length polymorphisms.The possible association was tested by χ2 test.Results The frequency of genotype and allele of CYP11B2(C-344T)in all groups were consistent with Hardy-Weinberg equilibrium.The frequency of CC and TT genotype in essential hypertension(EH)group was higher than that in control group,the frequency of TC genotype was lower than that in control group,but the differences were no statistically significant (P0.05).T and C allele frequency in EH group was similar with that in control group(P0.05).There was no statistically significant in the frequency of TT,TC,CC,T allele and C allele among medium risk group,high risk group,very high risk group and control group(P0.05).Conclusion The polymorphisms of CYP11B2(C-344T)gene are unrelated with essential hypertensive risk stratification.
Objective To determine the content of polysaccharide in retinervus luffac fructus produced in Henan province and to explore the medical value and developing perspective.Methods The content of polysaccharide was determined with the method of phenol-sulfuric acid,and the glucose(drying at 105 ℃ to keep constant weight) was used as reference substance.Results The sample concentration was good linear relationship with absorbance within the range of 6~91 μg·mL-1,the equation of linear regression is A=8.995 8c-0.007 8(r=0.999 7),the average recovery was 101.8%(n=5,RSD=1.59%) and the average concentration of polysaccharide was 7.66%.Conclusion The content of polysaccharide in Retinervus luffac fructus produced in Henan is higher than in Guangdong,Shanxi,Zhejiang and other provinces,and it has higher medical value and development foreground.
To study the stability of glucosamine-α-aminoacids-copper(Ⅱ) ternary complexes,the formation constant of α-aminoacids-glucosamine-copper(Ⅱ) ternary complexes were determined at 298 K,I=0.1 mol/L KNO_3 by pH method.It was found that the two biologically active ligand,α-aminoacid and glucosamine accommodate each other when coordinated to copper(Ⅱ) ion.The stability of the ternary complexes is higher than that of the corresponding binary complex.The formation constant of the ternary complexes linearly increases with increase of the protonation constant of aminoacid and the stability of its binary complexes.Linear free energy relationship(LFER) does exist in the ternary system.
[Objective] The aim of the paper was to determine the content of polysaccharide in tree peony bark of Henan province. [Method] The content of polysaccharide was determined by phenol-sulfuric acid method, and was compared with glucose(drying at 105 ℃ to keep constant weight).[Result] The sample concentration was good linear relationship with absorbance within 40~102 μg/ml. And the equation of linear regression was A=9.368 3 C-0.017 9(r=0.999 65), the average recovery was 96. 47 %(n=4, RSD=1.82 %), and the average concentration of polysaccharide was 7.56 %. [Conclusion] The content of polysaccharide in rree peony bark of Henan province was high, and it had higher medical value and development foreground.
The total content of flavone in Acanthopanax senticosus injection is determined by spectrophotometry compared with Rutin.The result reads that the sample concentration is in linear relationship with absorbance in the range of 4~36μg·mL-1.The average recovery is 102.3%(RSD=0.59%,n=5) and the average concentration of total flavone is 5.251mg·mL-1.This method is simple,quick,accurate,sensitive and suitable for the content determination of total flavone in Acanthopanax senticosus injection.
