目的:探讨miR-375 调控RWDD3 参与脑胶质母细胞瘤U251 细胞增殖的作用机制.方法:采用RT-PCR技术检测正常脑组织及脑胶质瘤组织中miR-375 和RWDD3 的表达.以人脑胶质母细胞瘤细胞U251 为研究对象,设立对照组和相应的miR-375 组,空载体组和RWDD3 过表达组,分别过表达miR-375 及RWDD3 蛋白,利用CCK8 及细胞划痕实验检测miR-375 或RWDD3 过表达对U251 细胞增殖和迁移能力的影响.在miR-375 组中,利用RT-PCR及Western blot免疫印迹法检测脑胶质母细胞瘤U251 细胞中RWDD3 mRNA及蛋白水平的变化;在miR-375 联合RWDD3 组细胞中共转miR-375 和RWDD3 质粒,观察RWDD3 是否可以恢复miR-375 对脑胶质母细胞瘤U251 的抑制作用.结果:高级别胶质瘤组织中miR-375 的表达水平低于正常和低级别胶质瘤组织,而 RWDD3 mRNA的表达水平高于正常和低级别胶质瘤组织,差异均有统计学意义(P<0.05).CCK8 及细胞划痕实验结果显示,与对照组比较,miR-375 可抑制脑胶质瘤细胞的增殖和迁移能力;与空质粒组比较,RWDD3 可促进脑胶质瘤细胞的增殖和迁移能力.RT-PCR及Western blot结果表明,在miR-375 联合RWDD3 组中,miR-375水平升高可有效抑制 RWDD3 mRNA及蛋白水平的表达;共转染miR-375 及RWDD3 后,RWDD3 可恢复由 miR-375 抑制的脑胶质瘤细胞增殖和迁移能力.结论:miR-375 在胶质瘤组织中表达水平降低,而RWDD3 的表达水平升高.miR-375 表达下调可以促进RWDD3 的表达,进而促进对脑胶质母细胞瘤U251 细胞的增殖和迁移能力.
目的:探讨滋阴补肾序贯法对肥胖型多囊卵巢综合征大鼠血清性腺相关激素及胰岛素抵抗指数的影响.方法:56只SD大鼠随机分为空白对照组(CN=8只),多囊卵巢综合征组(PCOS=48只).将造模成功的PCOS组大鼠随机分为安慰剂组(AW=12只)和药物治疗组(滋阴组,ZY组=12只;补肾组,BS组=12只;序贯组,XG组=12只),连续20 d分别灌服生理盐水和相应药剂.用血糖仪检测空腹血糖(FBG),用ELISA法检测睾酮(T)、血清总胆固醇(TC)、血清空腹胰岛素(FINS)、黄体生成素(LH)和卵泡刺激素(FSH),并采用胰岛素抵抗指数(HOMA-IR)评估胰岛素抵抗情况.结果:与CN组相比,AW组和药物治疗组大鼠体重均显著升高(P<0.05);AW组、ZY组和XG组大鼠血清TC较CN组显著升高(P<0.05).与CN组相比,AW组血清T显著升高(P<0.01),XG组较AW组、ZY组和BS组显著降低(P<0.01).与CN组相比,AW剂组血清FSH显著降低(P<0.01);XG组血清FSH较AW组和ZY组显著升高(P<0.01);XG组血清FSH高于BS组,但差异无统计学意义(P=0.948).与CN组相比,AW组LH/FSH值显著升高(P<0.01),BS组和XG组LH/FSH值较AW组显著降低(P<0.01),且XG组比ZY组和BS组低(P<0.01).与CN组相比,AW组FINS水平显著升高(P<0.01);药物治疗组FINS较AW组显著降低(P<0.01),且XG组比ZY组和BS组低(P<0.01).AW组HOMA-IR较CN组显著升高(P<0.01);药物治疗组HOMA-IR较AW组显著降低(P<0.01);XG组HOMA-IR比ZY组和BS组有所降低,但差异无统计学意义(P=0.079,0.060).结论:滋阴补肾序贯法可通过改善胰岛素抵抗和LH/FSH比值,从而降低血清高雄激素状态.
Objective To investigate the expression and clinical significance of Galectin-3 and Cyclin D1 in Bethesda-Ⅲ nodules of thyroid cytology.Methods A total of 128 specimens of fine needle aspiration with Bethesda-Ⅲ nodules of thyroid cytology were carried out of sediment embedding slice, and the expression of Galectin-3 and Cyclin D1 in the specimens was detected by Envision method.Results There were 68 cases of papillary thyroid carcinoma(PTC), and 60 cases of benign lesions, including 45 cases of nodular goiter, 11 cases of follicular adenoma, and 4 cases of lymphocytic thyroiditis.The expression of Galectin-3 and Cyclin D1 in PTC was significantly higher than that in benign lesion.The positive rates of Galectin-3 and Cyclin D1 were 92.6% and 89.7% respectively and the difference was statistically significant(P<0.001).The sensitivity, specificity and accuracy of the diagnosis were improved by the combination of the two tests, and the difference of combined expression between PTC and benign disease was statistically significant(P<0.001).Conclusion The combined detection of Galectin-3 and Cyclin D1 is helpful for the diagnosis and differential diagnosis of Bethesda-Ⅲ nodules of thyroid cytology, and provides reliable basis for clinical treatment.
目的 探讨前列腺原发性血管内大B细胞淋巴瘤(intravascular large B-cell lymphoma,IVLBCL)临床表现、病理学特征、诊断及鉴别诊断.方法 对1例前列腺原发性IVLBCL行HE及免疫组化EnVision两步法染色,分析其临床病理学特征和免疫表型,并复习相关文献.结果 镜下,前列腺间质血管内见异型淋巴样细胞,细胞核大而深染,可见突出核仁,核分裂象易见.免疫组化:肿瘤细胞CD20、MUM1、Bcl-2阳性,CD34显示瘤细胞生长在血管内.Ki-67增殖指数80%.患者术后未经化疗,随访6个月后死亡.结论 前列腺原发性IVLBCL比较罕见,临床表现常不典型,易导致漏诊、误诊,确诊需结合病理形态学检查和免疫标记.
目的:对应用补肾活血中药组方,进行动物实验,观察种植体与牙槽骨骨性结合界面的组织切片及颌骨骨密度检测.方法:选取28只新西兰家兔,随机分实验组与对照组,造模后实验组服用补肾活血中药制剂,分批取下标本,进行组织学比对,术后及后三批动物进行骨密度结果统计学对比研究.结果:表明实验组中药促进了种植体界面的骨钙的沉积,提高骨矿化沉积率,种植体骨整合的效果优于对照组;骨密度检测结果实验组与术后组差异显著有统计学意义,P<0.05.结论:补肾活血法由甘枸杞、熟地、制黄芪、山萸肉、丹参、巴戟天、益母草、杜仲组方,质优价廉且取材方便;动物实验研究证明,能提高颌骨骨密度,促进种植体的骨整合.
目的 分析液基膜式薄层细胞学技术(thin prepliquid based cytology testing,TCT)应用于B超引导下甲状腺细针穿刺病理诊断的临床效果.方法 选取3 376例2018年1月—2019年1月在本院进行甲状腺细针穿刺病理检查的患者,对所采集的标本均进行常规细胞涂片制片(cytological smear,CS)和TCT制片,比较两种方法 的诊断效果.结果 TCT标本中不满意标本占比17.0%,显著低于CS标本的19.3%,差异有统计学意义(P<0.05).TCT与CS标本的Ⅱ级、Ⅲ级、Ⅳ级和Ⅴ级标本所占比重比较,差异均无统计学意义(P>0.05).TCT标本的阳性检出率为26.9%,CS标本的阳性检出率为25.5%,组间差异无统计学意义(P>0.05).结论 B超引导下甲状腺细针穿刺采用TCT制片,具有较高的涂片质量,且诊断准确率高,在甲状腺疾病诊断中具有重要应用价值.
目的 分析TCT(Thin-prep cel test)、HPV(Human papiloma virus)联合阴道镜技术在宫颈癌筛查中的应用价值.方法 选择本院2017年12月—2018年12月在本院进行宫颈癌筛查的80例患者作为本次研究的对象,分别对80例患者实施TCT、HPV单一检查以及TCT、HPV、阴道镜联合检查,并对不同检测方式下宫颈癌病变筛查效果进行分析.结果 80例患者病理学检查癌前病变确诊53例,TCT检查诊断准确率为66.04%,HPV检查诊断准确率为77.36%,TCT、HPV联合阴道镜检查诊断准确率为100.00%;TCT、HPV联合阴道镜方法下的诊断准确率高于单一诊断的准确率,差异具有统计学意义(P<0.05).结论 针对早期宫颈癌筛查采取HPV、TCT联合阴道镜检查方式,在病变诊断中具有较高的准确性,利于早期治疗.
目的:探讨细胞角蛋白19(CK19)、甲状腺过氧化物酶(TPO)在甲状腺细针穿刺(FNA) BethesdaⅢ类结节组织中的表达情况及临床意义.方法:对156例细胞学为BethesdaⅢ类且有组织病理学诊断的甲状腺FNA标本进行沉渣包埋切片,采用EnVision法检测CK19及TPO在沉渣包埋切片中的表达情况.结果:术后组织病理学确诊为甲状腺乳头状癌(papillary thyroid carcinoma,PTC)95例,良性病变61例,包括结节性甲状腺肿(nodular goiter,NG)33例,滤泡性腺瘤(follicular adenoma,FA)24例,淋巴细胞甲状腺炎(lymphocytic thyroiditis,LT)4例.免疫细胞化学检测CK19在PTC中明显高表达,阳性表达率为98.95%,敏感度为98.95%,特异度为85.25%;TPO在PTC中明显阴性表达,阴性表达率为93.68%,敏感度为93.68%,特异度为96.72%;两种抗体在PTC中联合表达的阳性率为98.95%,敏感度为98.95%,特异度为100.00%.PTC中CK19阳性表达率及TPO阴性表达率显著高于良性病变(P<0.01),两种抗体联合表达率在PTC与良性病变间的差异具有统计学意义(P<0.01).结论:CK19和TPO联合检测有助于甲状腺FNA BethesdaⅢ类结节组织中PTC与良性病变的鉴别诊断.
目的 探讨应用免疫细胞化学(immunocytochemistry,ICC)技术检测甲状腺细针穿刺细胞学(fine needle aspiration cytology,FNAC)标本中CK19、MC及TPO表达情况,并将结果与免疫组织化学(immunochemistry,IHC)进行比较.方法 收集85例甲状腺乳头状癌(papillary thyroid carcinoma,PTC)患者的FNAC标本和相应组织学标本,采用EnVision法检测CK19、MC及TPO在细胞学和组织学中的表达情况.结果 在ICC检测结果中,CK19、MC及TPO分别有81例、76例、7例阳性;在IHC检测结果中,CK19、MC及TPO分别有83例、79例、8例阳性.对于CK19,ICC与IHC一致性为97.65%,敏感性为97.59%,特异性为100%,阳性预测值(PPV)和阴性预测值(NPV)为100%和50%.对于MC,ICC与IHC一致性为94.12%,敏感性为94.94%,特异性为83.33%;PPV和NPV为98.68%和55.56%.对于TPO,ICC与IHC一致性为96.47%,敏感性为98.70%,特异性为75%;PPV和NPV为97.44%和85.71%.结论 ICC技术在PTC的诊断结果与IHC技术相当,可以作为诊断PTC重要辅助手段.
目的:研究兔直肠粘膜内脱垂(Internal Rectal Prolapse,IRP)发展的阶段性特征,旨在推断IRP的发生机制.方法:将156只兔(雄兔、 雌兔各78只),按随机数字表随机分为4组,分别为正常组、 造模早期组、 造模中期组、造模晚期组,分别于不同阶段取兔解剖,进行力学与病理学分析.结果:造模不同时期兔直肠组织的力学指标均存在统计学差异;造模不同阶段兔直肠组织的镜下征象存在差异;在造模不同时期雌兔与雄兔的力学指标与病理特征差异无统计学意义.结论:不同病理学阶段的兔肛门直肠力学特性存在差异,可能是IRP的发生机制之一.
目的:观察消痔灵注射治疗兔直肠黏膜内脱垂(IRP)的疗效.方法:将16只兔按直肠黏膜内脱垂兔造模方案造模,造模后随机分为两组各8只.第1组为IRP组,肛门镜下观察后解剖取兔直肠进行病理分析;第2组为消痔灵注射组,在注射后第14天肛门镜下观察后解剖取兔直肠进行病理分析.结果:IRP组兔肛门镜下观察见直肠黏膜遮挡肛镜视野范围均在3/4以上,直肠显微镜下见腺体组织增生,血管扩张充血,间质炎细胞浸润聚集,间质水肿、疏松;消痔灵注射组在注射后第14天肛门镜下观察见直肠黏膜遮挡肛镜视野范围均在1/6以下,直肠显微镜下见黏膜下层呈慢性炎细胞浸润、纤维组织增生,黏膜层与肌层排列较IRP兔直肠紧密.结论:消痔灵注射治疗兔IRP效果肯定.
目的 探讨术中冰冻切片对甲状腺病变的诊断价值及误诊原因.方法 回顾性分析670例甲状腺病变术中冰冻切片诊断结果,并与石蜡切片比较,分析甲状腺冰冻切片病理形态特征及误诊和延迟诊断情况.结果 670例甲状腺冰冻切片病例中,确诊647例(准确率96.58%),误诊15例(误诊率2.24%),延迟诊断8例(延迟率1.19%);误诊病例中假阳性1例(0.15%),假阴性14例(2.09%).结论 直径小于0.2 cm的微小癌和不确定包膜侵犯的滤泡性肿瘤是甲状腺冰冻切片的主要诊断陷阱,准确取材、精心制片及熟练掌握冰冻切片甲状腺病变形态特征可提高冰冻切片诊断准确率并降低误诊率.
目的 探讨p53在慢性胃炎腺上皮增殖中的作用.方法 选择正常体检者20例和符合纳入标准的慢性胃炎患者124例,取胃黏膜活体组织检查,免疫组化检测黏膜组织中ki67、p53蛋白表达.结果 慢性胃炎组胃黏膜上皮细胞ki67、p53表达水平明显高于正常对照组,差异有统计学意义(P<0.05).结论 p53表达在慢性胃炎腺上皮增殖过程中发挥一定作用.
Objective To explore the application value of Liquid based cytology combine with sediment embedding biopsy and immunohistochemical detection in the diagnosis of hydrothorax.MethodsTo detecte liquid based cytology of 368 cases of hydrothorax and we found that 65 cases were malignant or suspicious malignant,and we further diagnosis and trace their organization sources by the means of sediment embedding biopsy combined with immunohistochemical staining.Results By the method of liquid based cytology ,we found that 49 cases (75.38%) were positive and 16 cases were suspicious positive . On the other hand ,by the method of sediment embedding biopsy conbined with immunohistochemical detection , 61 cases were positive,with the positive rate of 93.85%,and the difference was statistically significant (p < 0.05). Immunohistochemical markers results Confirmed that 41 cases of patients were lung adenocarcinoma, 2 cases were lung squamous carcinoma,1case was pulmonary neuroendocrine carcinoma, 2 cases were malignant mesothelioma, 9 cases were gastrointestinal carcinoma, 2 cases were breast cancer ,1 case was bile duct carcinoma , 3 cases were ovarian cancer and 4 cases were reactive mesothelium hyperplasia.Conclusion By using Liquid based cytology combined with sediment embedding biopsy and immunohistochemical detection can improve the accuracy of hydrothorax cytology diagnosis and positive detection rate,and be of great clinical application value in estimate the source of tumor cell tissue.
Objective:To develop IRP rabbit model .Methods: Thirty New Zealand rabbits were randomly divided into normal group and model group .Three methods were conducted to make the rabbit models .The three methods contain rhubarb and senna gavage , peri-anal injection of anhydrous alcohol and stand .The prolapse of rectal mucosa under the anus mirror and the microscopic structure of rec-tum between the normal group and the model group were compared .Results:Compared with the normal group , internal rectal prolapse were significantly different in the model group .Pathological findings were in accordance with IRP standards .Conclusion: Composite method of IRP rabbit model is reliable .

Objective:To investigate the influence of Total Panax Notoginseng Saponins(tPNS) on hemodynamics of rats with endotoxic shock and its mechanism.Methods:tPNS was injected to rats with endotoxic shock and their blood pressure,heart rates,left ventricular stroke volume were observed.Results:The hemodynamics indicaters of endotoxi shock rats which injected tPNS were obviously improved than rats which not injected tPNS.Conclusion:It is showed that tPNS has a beneficial effects on the hemodynamics in rats with endotoxic shock.

Objective:To know about the Expression and implication of c-kit gene in malignancy epithelial tumor.Methods:634 cases of malignant tumour specimens that had been stained with immunohistochemistry method in routin were collected from the second people hosipital of Fujian province and Fuzhou general hospital of Nanjing command from Aug,2005 to Aug,2006.The positive expression of CD117 were observed and analyzed on malignancy epithelial tumor.Results:CD117 were positive on 16 cases of malignancy epithelial tumor,of which 8 cases were gastrointestinal tract cancer,6 cases of breast cancer and 2 cases of lung cancer.Conclusion:The expression of c-kit gene is present on a few malignancy epithelial tumor and may suggest an indication of Imatinibmesylate(STI571).
