Growing lines of evidence supported the importance of CD8+ lung tissue resident memory T (T RM ) cells in protection against respiratory viruses, exemplified by influenza A virus. However, the underlying in vivo mechanism remains largely undetermined. Here, we used mouse infection models to dissect in vivo cross-protective activity of lung CD8+ T RM cells. By simultaneously interrogating transcriptional dynamics in lung CD8+ T RM cells and surrounding tissues during the early course of infection, we demonstrated that lung CD8+ T RM cells react to antigen re-exposure within hours, manifested by IFN-γ upregulation, and a tissue-wide interferon-stimulated gene (ISG) program is subsequently elicited. Using antibody-mediated IFN-γ neutralization and IFN-γ receptor knockout mice, we could show that the induction of several important antiviral ISGs required IFN-γ signaling, so did the suppression of key inflammatory cytokines. Interestingly, there were also examples of ISGs unaffected in the absence of IFN-γ activity. Collectively, focusing on in situ characterization of lung CD8+ T RM cells during very early stage of infection, a critical period of host antiviral defense that has been poorly investigated, our studies highlight that these cells, once triggered by antigen re-exposure, are programmed to produce IFN-γ expeditiously to promote a lung-wide antiviral response for effective virus control.
BACKGROUND:Hypoxia is a striking feature of most solid tumors and could be used to discriminate tumors from normoxic tissues. Therefore, the design of hypoxia-conditioned Chimeric Antigen Receptor (CAR) T cells is a promising strategy to reduce on-target off-tumor toxicity in adoptive cell therapy. However, existing hypoxia-conditioned CAR-T designs have been only partially successful in enhancing safety profile but accompanied with reduced cytotoxic efficacy. Our goal is to further improve safety profile with retained excellent antitumor efficacy. METHODS:In this study, we designed and constructed a hypoxia-inducible transcription amplification system (HiTA-system) to control the expression of CAR in T (HiTA-CAR-T) cells. CAR expression was determined by Flow cytometry, and the activation and cytotoxicity of HiTA-CAR-T cells in vitro were evaluated in response to antigenic stimulations under hypoxic or normoxic conditions. The safety of HiTA-CAR-T cells was profiled in a mouse model for its on-target toxicity to normal liver and other tissues, and antitumor efficacy in vivo was monitored in murine xenograft models. RESULTS:Our results showed that HiTA-CAR-T cells are highly restricted to hypoxia for their CAR expression, activation and cytotoxicity to tumor cells in vitro. In a mouse model in vivo, HiTA-CAR-T cells targeting Her2 antigen showed undetectable CAR expression in all different normoxic tissues including human Her2-expresing liver, accordingly, no liver and systemic toxicity were observed; In contrast, regular CAR-T cells targeting Her2 displayed significant toxicity on human Her2-expression liver. Importantly, HiTA-CAR-T cells were able to achieve significant tumor suppression in murine xenograft models. CONCLUSION:Our HiTA system showed a remarkable improvement in hypoxia-restricted transgene expression in comparison with currently available systems. HiTA-CAR-T cells presented significant antitumor activities in absence of any significant liver or systemic toxicity in vivo. This approach could be also applied to design CAR-T cell targeting other tumor antigens.
We discuss a deep network for temporal compressed imaging. The data collected by an experimental platform are used for training and testing. Spatial variant point spread functions are calibrated for reconstruction.
The early application of compressive sensing in optical imaging focuses on spatial compressive imaging. In recent years, increasing compressive imaging systems have employed detector array instead of a single detector for collecting measured values. Moreover, the scope of compressive imaging expands from two-dimensional space to three-dimensional ranging, high-speed imaging, multispectral imaging, ghost imaging, and holography imaging. Herein, we analyzed recent works on high-resolution compressive imaging, compressive sensing ranging, and temporal high-speed compressive imaging with details, summarized the research progresses of measured matrix design by combining spatial compressive imaging, works on sensing matrix design in spatial compressive imaging, discussed their challenges and future development opportunities, and reviewed the applications of compressive sensing in multispectral imaging, ghost imaging, and holography imaging. Furthermore, we summarized the improvement of reconstruction performance of system targets by applying deep learning to compressive imaging.
In compressive imaging, the data acquisition speed of sensors is one of the restrictions limiting system imaging speed. To relax the speed requirement of sensors, we combine temporal and spatial compressive sensing idea for block-wise compressive imaging(BCI).

大学生实验室安全教育是高校实验室安全管理工作的重要内容之一.为强化学生的安全意识,学院将“实验室安全及试剂基础知识”作为一个教育专题列入学院全体新生的专业导论课教学中.针对不同专业,有的放矢设置教学内容,寻找合适案例,精心设计课件.多种教学法相结合,以激发学生的学习兴趣.为使学生重视培训,在课程结束前安排10 min的随堂测试,并要求学生做出承诺:时刻保持高度警惕,严防实验室安全事故发生.通过培训,学生的实验室安全意识明显提高,为安全管理奠定了良好基础,充分体现了学院“以人为本,预防为先”的工作原则.
藏药藏茵陈川西獐牙菜是传统高寒藏药,在治疗肝胆疾病方面具有独特疗效,已成为目前研究的热点.文章系统查阅近20年国内外有关川西獐牙菜的研究进展,对川西獐牙菜生物学特性及资源分布、组织培养、分子生物学和引种栽培研究的进展进行分析、概括和总结,为藏茵陈资源保护和可持续发展提供科学的参考依据,对地区生物、生态多样性保护起到积极作用.
探索建立了一种新型“大型仪器自主操作资格”获得机制.仪器使用申请者在分析测试中心仪器预约平台上提出申请,运用虚拟实验训练系统进行针对性自主培训.通过无人监督考核后,预约上机操作考核,2项考核均合格即可获得申请使用仪器的自主操作资格.该获得机制的核心是运用虚拟实验训练系统进行自主培训,具有很强的适用性、针对性和灵活性,尤其适用于申请者专业背景和操作水平有较大差异的状况.该机制可有效促进开放式共享平台管理模式的科学优化,切实提高大型仪器的使用效率,为实验学科的教学改革提供新的思路.
Large-scale instruments & equipment are tools and supports for scientific experiments,the achievements of undergraduate teaching and scientific research largely depend on conclusions from a large number of scientific experiments for university faculties of science and engineering.In recent years,large-scale instruments & equipment in teaching,research and innovation education play an increasingly important role, and the number of large-scale instruments & equipment is increasing.For the implementation of standardized management and improvement of the utilization of instruments,a large-scale instruments & equipment sharing platform is established by the enforcement of online booking and the establishment of a training system,which has achieved good results,making it truly boost power of teaching and scientific research.
MISA (MicroSAtelite) software was employed to screen SSRs in 68 787 contigs of Swertia mussotii transcriptome sequences. 5 610 SSRs were distributed in 5 099 contigs which accounted for 7.41% of 68 787 contigs. There are 220 kinds of SSR motifs existing in S. mussotii transcriptome. On average, SSRs occurred every 12.60 kb in length. In the SSRs, the tri-nucleotide repeat motif was the most abundant (45.99%), followed by the di-nucleotide (41.62%). AT/TA and AAT/TTA were the main types of motif in di-, tri-nucleotide repeats. The repeat numbers of SSRs which from S. mussotii transcriptome SSRs were mainly from 5 to 10 and motif length of them mostly ranged from 12 bp to 30 bp. A total of 30 651 contigs were annotated, and only 1 447 SSRs were occurred in protein-coding regions. In the six repeat motifs, tri-nucleotide repeats were the most abundant in coding regions (928). There are abundant SSRs in S. mussotii transcriptome with high frequency and various types, indicating their usefulness in theory. This research may lay the foundation for designing the targeted SSR primers and developing SSR molecular markers by mining the information of SSRs loci in S. mussotii transcriptome sequences data.
Wulingshan region is located on the border between Hunan,Hubei,Chongqing,Guizhou. In the area,the geographical composition is complicated,the climate is warm and humid,ecological environment is complicated,gestates rich biological resources,but there are also many endangered plants in urgent need of research and conservation. This paper reviewed researches on the survey of the endangered plants resource,the genetic diversity research,conservation and use of it in Wulingshan region. It can provide scientific reference for the protection and sustainable development of these endangered plants,and play an active role in protection of biological diversity in Wulingshan region.
(口山)酮是一种特殊的黄酮类化合物,也是藏药中含量最高、生物活性较强、药用价值和经济价值丰富的一类有效成分.目前认为,3-羟基苯甲酸辅酶A连接酶、二苯甲酮合成酶和(口山)酮合成酶是山酮生物合成路径中的关键酶.文章对(口山)酮的结构类型、功能及其在植物中的分布,以及(口山)酮的生物合成路径与关键酶进行综述,并对相关研究进行了展望.
Cauliflower-like N doped nano titania film,of which the surface was stacked by nano-particles with diameter of 10~60 nm,was prepared on titanium substrate by dipulse microarc oxidation in the electrolyte of Na2B4O7·10H2O,NaOH,and C6H12N4.The frequency was a main factor on constituents,porosity and roughness of the film,while the processing time would impact the surface roughness through accumulation of particles.Under higher frequency conditions,TiO2 films have more anatase phase than rutile phase,and it could be attributed to the control of outgrowth temperature during single pulse.While under lower frequency conditions,the band gap narrowing of N doped TiO2 films became more obvious.
【Objective】To investigate the genotype frequency of aldehyde dehydrogenase-2(ALDH2),and evaluate the relationship between ALDH2 polymorphism and drinking behavior in Hezhen race population.【Methods】 Epidemiological data were collected from 101 Hezhen race in Hezhen settlements,and ALDH2 genotypes were detected by PCR-RFLP.【Results】The frequency of ALDH2 mutant allele(GA/AA) was 24.8% in Hezhen race.No difference was found between GG and GA/AA in drinking frequency and alcohol consumption per occasion.Drinking behavior was not influenced by the individual genotypes of ALDH2 in Hezhen population.【Conclusion】1/4 of Hezhen race have GA/AA allele.Drinking behavior was not influenced by the individual genotyoe of ALDH2.

The HLA system is a very important genetic marker for human beings with the race and region differences,which par- ticipates in and adjusts immunity function.HLA-DRB1 allele genes have the highest polymorphism in HLA classⅡ.The accurate classi- fication of HLA-DRB1 directly affects the donor selection in organ transplant,individual identification in forensic medicine,disease-cor- relation,and anthropology research.HLA-DRB1 polymorphism in different people,its value in the human origin and national fusion,the typing and detecting methods of HLA-DRB1,the correlation between HLA-DRB1 and HBV or SLE were reviewed in this article.

Thrombin as a hemostatic drug can be extracted from blood. Because there are more than 100 plasma proteins in blood,it is natural for thrombin to contain some plasma proteins. α-2 macroglobulin is one of the important plasma proteins and it can pavilion and protect trypsin. Soybean trypsin inhibitors are used to inactivate the trypsin that can not be pavilioned,and then the activity of α-2 macroglobulin included in the thrombin mixture is detected by using the special substance-BAPNA. It shows that the thrombin extracted from blood does contain the α-2 macroglobulin. This work provides some guidance for thrombin purification.
