从生物化学的角度来看,中西医在认识上存在一致性:①维生素、必需氨基酸、必需脂肪酸等营养素必须要通过食物摄取,这与中医的食疗养生理论是一致的;②糖、脂肪、蛋白质必须被消化变成小分子才能吸收,这与中医的“脾主运化”理论是一致的;③物质代谢作为相互联系的整体,同时又要和机体内外环境的变化协调一致,这和中医的整体观是一致的;④高等生物体内存在三个层次的代谢调节,即细胞水平的代谢调节、激素水平的代谢调节和整体水平的代谢调节,这和“天人合一”思想是一致的,同时整体水平的代谢调节,有中枢神经系统的参与,这和中医的“情志养生”理论是一致的;⑤基因和遗传病与中医的“先天禀赋”理论是一致的.
To investigate the potential antitumor effects of ginsenoside Rg3 in prostate cancer cells, the androgen-insensitive prostate cancer cell line PC3 was cultured and incubated with ginsenoside Rg3 in vitro. Cell number counts, cell proliferation assays and senescence-associated β-galactosidase (SA-β-gal) staining were performed to evaluate cell proliferation. The results demonstrated that ginsenoside Rg3 led to cell proliferation arrest; ginsenoside Rg3 decreased the number of cells and increased the positive SA-β-gal staining rate in PC3 cells. Cell cycle analysis by flow cytometry revealed that ginsenoside Rg3 interfered with the G1/S transition in PC3 cells. The mechanism involved in ginsenoside Rg3-induced cell proliferation arrest was then further investigated. This indicated that the level of reactive oxygen species (ROS) in PC3 cells was upregulated by ginsenoside Rg3 treatment. Furthermore, pretreatment with N-acetyl-L-cysteine, a scavenger of ROS, was able to reverse the effects on cell number and cell cycle arrest induced by ginsenoside Rg3 in PC3 cells. These results indicate that ginsenoside Rg3 exhibits anticancer effects on prostate cancer cells through ROS-mediated arrest of the cell cycle.

<正>健身气功八段锦是国家体育总局向全国乃至世界推广的身体锻炼方法,是中医养生与治疗学的一部分〔1〕。八段锦是一套由八节动作编成的功法,以人自身形体活动、呼吸吐纳、心理调节相结合为要素的民族传统运动方法。作为以肢体运动为主的导引术,其运动强度和动作的编排次序符合运动学和生理学规律,是适合中老年人练习的中小强度有氧运动。本实验从八段锦对肺活量的调节作用入手,研究练习八段锦对不同血脂水平中老年人肺活量的调节作用。
通过总结生物化学PBL教学法的实践经验,对目前基础医学PBL教学法应用中存在的问题进行探讨,认为PBL教学模式的实施,应与课程内容、学生层次、教师素质、教学资源等因素契合,灵活应用,才能充分发挥其优势。
Objective:To study the effects of sallvianolic acid B on the expression of Bcl-2 and Bax proteins in ApoE-/-mice induced by high fat feed combined with STZ.Methods:The eight-week old mice were given intraperitoneal injection with stz and fed in high fat diet for 12-weeks to make diebatic atherosderosis model,then assigned them randomly into four groups:model group,atorvastatin group,high Salvianolic acid B group and middle Salvianolic acid B group.After treating 8 weeks,the immunohistochemistry were used to monitor the expression of Bcl-2 and Bax proteins in aortas and ratios of both were compared.Results:In modle group,the expression of Bcl-2 or Bax proteins was higher than those in control group,the latter was increased more obviously;and ratio of both expressions was in imbalance.In high sallvianolic acid B group,compared with model group,the expression of Bcl-2 protein was increased,while the expression of bax protein was reduced;and ratio of both was recovered.Conclusion:Sallvianolic acid B could up-regulate the expression of bcl-2 protein,down-regulates the expression of bax protein simultaneously and recovers the ratio of both,which might be one of mechanisms of sallvianolic acid B preventing and treating diabetic atherosclerosis disease.
[Objective]To study the mechanism of apoptosis of human umbilical vein endothelial cell(HUVEC) induced by AGEs and ox-LDL.[Methods]HUVECs were incubated at some concentration of AGEs and ox-LDL for 24 h.The apoptosis was observed by Hochest 33258 staining.The mitochondria membrane potential(MMP) was analyzed by flowcytometry after Rhodamine-123.[Results]AGEs and ox-LDL could increase apoptosis significantly and decrease mitochondria membrane potential of the cultured HUVECs significantly also(P0.05).[Conclusion]The results indicated that AGEs and ox-LDL could promote endothelial cell apoptosis,the mechanism in which may be related to decreasing of mitochondria membrane potential.
Objective: To investigate the protective effect of salvianolic acid B(Sal B) on hyperglycemia-induced apoptosis in cultured human umbilical vein endothelial cells (ECV-304). Methods: The dosage of the high glucose and oxidized low density lipoprotein and the concentration of the drugs were confirmed by MTT. ECV-304 was identified by CD31 immunofluorescent staining. The cell models of endothelial cell oxidative damage were established by oxidized low density lipoprotein (OX-LDL). The model cells were divided into 5 groups. The cell apoptosis was detected by flow cytometry. Results: The optimum concentration of the glucose was 44 mmol/L, OX-LDL was 50 mg/L, the effective concentration of Sal B was 10-4 mol/L and lovastatin was 50 μmol/L. The number of apoptotic cells was significantly increased in ECV-304 after exposure to hyperglycemia compared with that of normal glucose level (P 0.05). The hyperglycemia-induced apoptosis was significantly attenuated after pretreatment with Sal B (P 0.01). The apoptosis rates were significantly higher in the middle and high concentrations of Sal B group than those of lovastatin group (P 0.05). Conclusion: Results of our study indicate that Sal B could inhibit high glucose-induced apoptosis in human umbilical vein endothelial cells, which provide a basis for future therapeutic interventions in diabetic vascular complications.
目的 观察丹酚酸B抑制高糖高脂诱导的内皮细胞凋亡的作用.方法 以糖基化终末产物(AGEs)和氧化低密度脂蛋白(ox-LDL)联合作用诱导人脐静脉内皮细胞凋亡,通过Hochest33258染色,观察并比较丹酚酸B及洛伐他汀作用不同时间(12、24、36、48、72 h),抑制高糖高脂诱导的内皮细胞凋亡的效果.结果 研究表明,洛伐他汀作用12、24 h时,抑制内皮细胞凋亡效果明显,而丹酚酸B作用36、48 h及72 h抑制内皮细胞凋亡效果显著.结论 与洛伐他汀相比,丹酚酸B抑制高糖高脂诱导的内皮细胞凋亡的作用发挥得较迟缓,但效果持久.
[Objective] To observe the effect of Radix Dipsaci containing serum on the proliferation of osteoblasts in rats withdiscontinued administration and different sex.[Methods] The drug containing serum in rats was prepared.Twenty-four Wistar rats withequal male and female were randomly divided into four groups:male control group(I), male drug-added group(II), female control group(Ⅲ), female drug-added group(IV), Intragastric perfusion with Radix Dipsaci(4.5g crude drug/kg body weight) and saline respectivelyaccording to the animal body weight was given for one week and then the blood was gained.After centrifugation the supernatant fluid wasobtained.Then the osteoblasts were isolated and cultured.The proliferation of osteoblasts was analyzed quantitatively.The cells weredisposed and added with different concentration of rat serum, and divided into 2.5%, 5% and 10% Radix Dipsaci group.The proliferationof cells was tested with MTT.[Results] When the medium concentration was 2.5% of Radix Dipsaci significant difference was foundbetween male and female drug-added group(P0.05).There was also significant difference between female drug-added and femaleantitheses group when the medium was 5% concentration of Radix Dipsaci rat serum(P0.01).While the medium was 10% concentrationof Radix Dipsaci significant difference could be seen between female drug-added group and female antitheses group(P0.05).However,no matter the concentration of Radix Dipsaci was, no significant differences could be seen between male drug-added group and malecontrol group.[Conclusion] Radix Dipsaci can stimulate the proliferation of osteoblasts.This function in female drug-added group wasmore significant than that of male drug added group.
[Objective] To observe the effect of Salvianolic acid B on diabetic atherosclerosis in Apolipoprotein E (ApoE)-gene knocked mice. [Methods] eight-week old mice were given intraperitoneal injection with stz and fed with high fat diet for 12-weeks to establish a diebatic atherosderosis model,then assigned them randomly into three groups:model group (fed with saline) Salvianolic acid B group (fed with Salvianolic acid B )and atorvastatin calcium positive control group (2.3 mg/kg everyday). After treating for 8 weeks,all the mice were killed and the aortas were stripped for morphological research and photograph analysis. [Results] In model group,some plaques were bigger,merged into flake,aortic wall thickened diffusively. Compared with that,In Salvianolic acid B treatment group and Lovastatin positive control group,the aortic sinus had a more limited plaque lesion. Morphological research showed that As lesions were found in aortas of model group and Salvianolic acid B group. The lesions of model group were more serious than those of Salvianolic acid B group. [Conclusion] Salvianolic acid B can relieve the diabetic As lesions of the ApoE gene knockout mice.
Purpose:To investigate the changes of the lengths of tibia and femur,serum IGF-I level and bone IGF-I level after sports training.Fur-thermore to explain the reasons of the changes.Methods:The4weeks old female Wistar were randomized into four groups:control group,running group,jumping group and swimming group.Experiment duration was nine weeks.Their blood and bone were detected:1.the length of tibia and fe-mur;2.the serum IGF-I level;3.the bone IGF-I level.Results:The lengths of tibia and femur in running group and jumping group were significant longer than those in swimming group and control group.There was no significant difference in the serum IGF-I level of four groups.In running group and jumping group the tibia IGF-I level were significantly higher than those in swimming group and control group.Conclusion:The effect of exercise promoting longitudinal growth of bone was significant.Serum IGF-I level was not significantly influenced by exercise.Bone IGF-I level was affected significantly by exercise.We speculated that force be key to the elevation of bone IGF-I level.And IGF-I was an important factor to bone longitudinal growth.
目的 以洛伐他汀为阳性对照,选取不同作用时间,观察丹酚酸B抑制高糖高脂诱导的内皮细胞凋亡的作用。方法 以糖基化终末产物(AGEs)和氧化低密度脂蛋白(ox-LDL)联合作用诱导人脐静脉内皮细胞凋亡,通过hochest33258染色,观察并比较丹酚酸B及洛伐他汀作用不同时间(12、24、36、48、72h),抑制高糖高脂诱导的内皮细胞凋亡的效果。 结果 研究表明,洛伐他汀作用12、24h时,抑制内皮细胞凋亡效果明显,而丹酚酸B作用36、48及72h抑制内皮细胞凋亡效果显著。结论 与洛伐他汀相比,丹酚酸B抑制高糖高脂诱导的内皮细胞凋亡的作用发挥得较迟缓,但效果持久。
目的 探讨丹酚酸B对小鼠MC3T3-E1细胞核结合因子a1(Cbfa1)基因mRNA表达和Cbfa1启动子活性的影响。方法 分别采用不同浓度丹酚酸B干预小鼠MC3T3-E1前成骨细胞系24和48小时,real-time PCR检测丹酚酸B对细胞中Cbfa1基因mRNA表达的影响;构建小鼠Cbfa1基因启动子荧光素酶报告基因载体并转染MC3T3-E1细胞,通过检测荧光素酶活性观察丹酚酸B对Cbfa1启动子活性的调节。结果 1μM和10μM的丹酚酸B分别处理MC3T3-E1细胞24小时和48小时,可上调Cbfa1 mRNA的表达(F=23.24,P<0.01),其中以1μM丹酚酸B处理48小时上调效应最显著,约为对照组的1.71倍(t=10.36,P<0.01)。1μM和10μM的丹酚酸B干预MC3T3-E1细胞后,Cbfa1启动子活性升高,分别为对照组的2.63、1.89倍(F=13.27,t分别为5.83、4.53,P<0.01)。结论 丹酚酸B可促进MC3T3-E1细胞中Cbfa1基因表达并上调Cbfa1启动子活性。
