目的 探讨研究白介素-6(Interleukin-6,IL-6)对核因子NF-κB受体活化因子配体(Receptor activator of nuclear kappa B ligand,RANKL)及对破骨前体细胞的成熟分化和溶骨效应.方法 破骨前体细胞RAW264.7细胞经50ng/mL RANKL诱导1 d后将其分为:1、空白对照组(RANKL+PBS)2、低浓度IL-6组(RANKL+50ng/mL IL-6)3、中浓度IL-6组(RANKL+100ng/mL IL-6)4、高浓度IL-6组(RANKL+150ng/mL IL-6).连续培养9 d后,进行HE染色检测成熟破骨细胞生成量;通过抗酒石酸酸性磷酸酶(Tartrate resistant acid phosphatase,TRAP)染色法观察TRAP阳性多核细胞的情况;运用扫描电镜检测破骨细胞在骨片上的骨吸收陷窝形成情况.结果 HE染色中,成熟破骨细胞生成量中、高浓度IL-6组明显少于低浓度IL-6组(P<0.05),低浓度IL-6组和空白对照组间无明显差别(P>0.05).②通过TRAP染色后,经染色阳性区域面积与视野面积的百分比计算,中、高浓度IL-6组与明显少于低浓度和空白对照组(P<0.05).③扫描电镜观察发现骨吸收陷窝面积与视野面积的百分比随着IL-6浓度的增高,相比空白对照组有显著减少,且高浓度IL-6组中陷窝形成最少(P<0.05).结论 IL-6能直接作用于经RANKL诱导的RAW264.7细胞,能明显抑制破骨细胞激活分化,并降低破骨细胞所致的骨吸收效应.当IL-6浓度超过50ng/mL时,其抑制破骨细胞的骨吸收效应更加明显.
目的 观察薏苡仁对扁平疣患者皮损中T淋巴细胞的影响,初步探讨薏苡仁治疗扁平疣的可能作用机制.方法 收集扁平疣患者20例,经知情同意后予薏苡仁治疗,以薏苡仁粉30 g调糊口服,2次/d,同时外敷疣体,1次/晚,连续应用至疣体脱落或60 d止.采用免疫组织化学法对扁平疣患者治疗前后皮损中CD4+、CD8+T淋巴细胞水平进行检测,并与10例正常皮肤进行对照.结果 扁平疣患者治疗前较正常对照组CD4+/CD8+比值明显降低,差异有统计学意义(t=2.295,P<0.05);扁平疣患者治疗后较治疗前CD4+/CD8+比值明显升高,差异有统计学意义(t=2.121,P<0.05).结论 薏苡仁可能通过调节扁平疣患者皮损中T淋巴细胞的水平,从而增强机体免疫功能.
OBJECTIVETo investigate the effects of Weile Powder (WLP) on bicarbonate transporters in rats with gastric ulcers, and to probe its functional mechanisms.METHODSThe 48 SD rats were randomly divided into the normal control group, the model group, the low dose WLP group (at the daily dose of 0.075 g/mL), the middle dose WLP group (at the daily dose of 0.150 g/mL), the high dose WLP group (at the daily dose of 0.030 g/mL), and the ranitidine group (at the daily dose of 0.030 g/mL), 8 in each group. The gastric ulcer rat model was prepared by the glacial acetic acid cauterization method. Rats in each medication group were administered from the 2nd day of modeling. Rats were sacrificed after 14-day successive medication. The protein was extracted from the ulcer tissue. The protein expressions of solute carrier26A3 (SLC26A3)and solute carrier26A6 (SLC26A6) were detected using Western blot. The gastric ulcer and its peripheral tissue were sectioned. The changes of cystic fibrosis transmembrane conductance regulator (CFTR) were measured by immunofluorescence.RESULTSCompared with the model control group, the expression levels of SLC26A3 increased in the high dose WLP group and the ranitidine group with statistical difference (P < 0.05). The expression levels of SLC26A6 increased in the high and middle dose WLP groups and the ranitidine group with statistical difference (P < 0.05). The expression level of CFTR also obviously increased in the high and middle dose WLP groups (P < 0.01).CONCLUSIONWLP could elevate the expression levels of SLC26A6, SLC26A3, and CFTR, increase the secretion of bicarbonate, thus protecting the gastric mucosa.
目的 观察IGF2、H19在原发性肝癌中的表达并了解其意义.方法 建立原发性肝癌模型,模型建立后第20周处死小鼠取肝癌及癌旁组织储存于-80℃备用,另取正常小鼠肝脏作对照,用Real time RT-PCR方法检测IGF2、H19 mRNA的变化.结果 与对照组比较,IGF2、H19在癌旁及癌组织中的表达都显著增加,在癌组织中增加更为显著P<0.05.结论 IGF2、H19的异常变化与小鼠原发性肝癌的发生发展有关.
AIM To investigate the effects of pantoprazole on the migration and invasion of liver carcinoma HepG2 cell and to examine the mechanisms.METHODS Logarithmic growth phase HepG2 cells were used in cell migration experiment and invasion experiment.Cell migration experiment were divided into normal control group and pantoprazole groups(20,40,80,120,160μmol·L~(-1)).Transwell chamber assay was used to detect the migration of HepG2 cells.Cell invasion experiment were divided into normal control group,12-O-tetradecanoylphorbol13-acetate(TPA) group and pantoprazole(20,40,80,120,160μmol·L~(-1)) + TPA groups.Transwell chamber assay was used to detect the invasion of HepG2 cells.The expression of matrix metalloproteinases-9(MMP9) mRNA was determined by real-time RT-PCR.RESULTS In the migration experiment,the number of cells acrossing membrane in the pantoprazole(40,80,120,160 μmol·L~(-1))groups was lower compared with that in the normal control group(P < 0.01).In the invasion experiment,the number of cells acrossing membrane in the TPA group was higher than those in the normal control group and pantoprazole(40,80,120,160 μmol·L~(-1)) + TPA groups(P < 0.05 or P < 0.01).The expression of MMP9mRNA in the 80 μmol·L~(-1) pantoprazole group was decreased compared with normal control group(P < 0.05).CONCLUSION Pantoprazole is effective in inhibiting HepG2 cell migration and invasion and its mechanism may be involved in suppression of MMP9 mRNA.
Objective To investigate the effects of wei-le-san on thromboxane A2(TXA2),prostaglandin I2(PGI2) and cyclooxygenase-2(COX-2) in gastric ulcers of rats,and probe its functional mechanism.Methods Forty-eight rats were randomly divided into normal group,model group,low dose of wei-le-san group,middle dose of wei-le-san group,high dose of wei-le-san group and ranitidine group.The rats of gastric ulcer were induced by glacial aceticacid.After drug treatment for 14 days,the rats gastric mucosa were collected to check the contents of TXB2 and 6-Keto-PGF1α with ELISA,COX-2 by Western blot.Results Compared with control group,the levels of 6-Keto-PGF1α,COX-2 in model group were lower(P0.05),the contents of TXB2,TXB2/6-Keto-PGF1α were higher(P0.05),and wei-le-san elevated the level of 6-Keto-PGF1α in the rats gastric mucosa.Compared to the model group,the levels of 6-Keto-PGF1α,COX-2 in middle dose of wei-le-san group,high dose of wei-le-san group and ranitidine group increased(P0.05),and the contents of TXB2,TXB2/6-Keto-PGF1α decreased(P0.05).Conclusion Increasing the level of PGI2 and correcting the unbalance of TXA2/PGI2 may be one of the possible mechanisms of wei-le-san to cure gastric ulcer.
OBJECTIVE To investigate the effects of Weilesan on IL-2,IL-4 and immune globulin in gastric ulcers of rats,and discuss its functional mechanism.METHODS 48 rats were randomly divided into normal group,model group,low dose of Weilesan group,middle dose of Weilesan group,high dose of WeiLesan group and ranitidine group.The gastric ulcer model was induced by the glacial aceticacid.After drug treatment for 14 days,the blood samples were collected to measure the content of IL-2 and IL-4 in serum by ELISA.IgG,IgA,IgM were measured by Immunoturbidimetry.RESULTS Compared to the control,the levels of IL-2 and IL-4 in model group were significantly lower.Welesan elevated the level of IL-2 and IL-4 in serum(P0.05),also increased the content of IgG,IgA,IgM(P0.05).CONCLUSION One of the possible mechanism is that Weilesan improves the cellular and humoral immune function to cure the gastric ulcer.
中药夏天无[Corydalis decumbens(Thunb.)Pets.]为罂粟科植物伏生紫堇的干燥块茎,我们以往的研究显示,夏天无总碱(Coryadlis ambailis Migo tatal alkaloids,COAMTA)对脑缺血/再灌注损伤具有保护作用[4].夏天无注射液(InjectionCoryadlis Decumbens Pers,ICDP)为含夏天无生物碱的制剂,具有通络活血、止痛等功效,临床主要用于脑梗死、高血压偏瘫、小儿麻痹后遗症、坐骨神经痛、跌打损伤等病症的治疗.
