Objective To analysis the pulmonary tissue proteomic a-mong the normal , pulmonary hypertensive induced by monocrotaline and Puerarin rats , How the Puerarin inhibits the form of pulmonary hyperten-tion and pneumoangiogram rebuilding cleared.Methods SD ras random-ly divided into 5 groups, the normal group, the solvent group, the model group in two times, monocrotaline and puerarin group ( experimental group ) , 10 in each group .The solvent group was intraperitoneal injected of ethanol mixed with 0.9% sodium chloride solution ( 50 mg · kg -1 weight ) every morning and bred three weeks .The model group was intra-peritoneal injected monocrotaline (50 mg· kg -1 weight)once and respec-tively bred 2 or 3 weeks.The experimental group, after the injection of monocrotaline solution once , was intraperitoneal injected puerarin liquid (40 mg· kg -1 weight ) every morning in three weeks .Set up the pulmo-nary hypertensive rat model sucessfully , two dimensional gel electropho-resis of the total proteins from these five group , gel enzymolysis.Matrix-assisted laser desorption ionization -time of flight mass spectrometry ( MALDI-TOF-MS) and retrieval databank in NCBInr and discuss the results.Results The pulmonary hypertention , pneumoangiogram rebuil-ding , the right ventricle hypertrophy was inhibited by puerarin.The MALDI-TOF-TOF-MS discovered that the elongation factor 2 express very little in normal group and express a great quantity in 3-week-monocrotaline group , but in puerarin group ex-press down quickly.Conclusion The puerarin could inhibit the form of pulmonary hypertention by interfering in elon-gation factor 2 , it worth to study more about it.
Objective:To investigate the rhubarb combined Shenfu Injection for respiratory function improvement in patients with ARDS.Methods:78 cases of ARDS patients were randomly divided into the rhubarb combined Shenfu Injection group(traditional Chinese medicine group) and the conventional treatment group(control group).Both groups were given conventional treatment,TCM group on the basis of conventional treatment.Conduct Murray score,Marshll rate,oxygenation index(PAO2/FiO2),duration of mechanical ventilation and respiratory mechanics difference were evaluated after treatment.Results:In improving the differences in ARDS patients with acute lung injury,organ function,PAO2/FiO2,duration of mechanical ventilation and respiratory mechanics,the traditional Chinese medicine group was better than the conventional treatment group,with a significant difference,P0.05.Conclusion:The rhubarb combined with Shenfu Injection has a significant effect on the improvement of respiratory function in patients with ARDS.
Objective To observe the change of CX3CL1/fractalkine (FKN) in the pulmonary hypertension rats induced by monocrotaline.Methods Thirty male Sprague-Dawley rats (270-310 g) were randomly divided into three groups:control group (C group),monocrotaline two-weeks group (M2 group),monocrotaline three-weeks group (M3 group).The rats were weighed and anesthetized,then a catheter was placed through the right external jugular vein to the pulmonary artery to test the mean pulmonary arterial pressure (mPAP) and the mean right ventricle pressure (mRVP).The mean carotid arterial pressure (mCAP) and the weight ratio of right ventricle (RV) to left ventricle plus septum (LV+S)were also measured.The pathological model was assessed with mPAP,mCAP and RV/(LV+ S).The structure change of pulmonary arterioles was observed under optical microscope.Remodeling of lung blood vessel was detected by measurement and calculation of the ratio of vessel wall area to total area (WA/TA) and the thickness of pulmonary artery media (PAMT).The concentration of sFKN in plasma was measured by ELISA.Expression of FKN in the pulmonary artery wall was measured by the technique of immunohistochemistry.The level of FKN mRNA in lung tissue was detected by RT-PCR.Results ①The mPAP,mRVP and RV/(LV+S) in model groups (M2,M3) were significantly higher than those in C group.The data of WA/TA and PAMT in model groups (M2,M3) were higher than those in C group.② sFKN and FKN mRNA in M2 [(677.76± 101.75) ng/L、0.49±0.09] were higher than those in C group [(412.09±57.38) ng/L,0.11± 0.06] (p <0.01),M3 group [(1 078.02±254.05) ng/L,0.64±0.04]was higher than M2 group and C group ( P <0.01).③FKN protein expression in the vessel wall of pulmonary arterioles in each group rats:FKN in model groups (M2 group:0.192±0.006,M3 group:0.198±0.019) were more than that in group C (0.171±0.010,P <0.01).There were no difference between group M3 and M2.The sFKN had a positive correlation with PAMT and RV/(LV+S) ( r =0.796,0.710 respectively,P < 0.01 ),FKN mRNA had a positive correlation with PAMT and RV/(LV+S) ( r =0.934,0.757 respectively,P <0.01).Conclusions FKN and inflammatory reaction play a key role in the formation and development of pulmonary hypertension induced by monocrotaline,and they probably participate in the progress of the pulmonary artery remodeling.
肺动脉高压是以肺动脉血管阻力进行性升高为主要特征的一组疾病,发生机制复杂,肺源性心脏病是其终末阶段.趋化因子CX3CL1(fractalkine,FKN)是趋化因子CX3C亚族的惟一成员,具有可溶形式及膜结合形式,既有趋化性蛋白的功能也有细胞黏附分子的功能,还具有促进平滑肌细胞增殖的生长因子作用[1].本研究通过野百合碱诱导的肺动脉高压大鼠模型观察肺动脉高压发展过程中FKN的表达变化,探讨FKN在肺动脉高压形成过程中的作用和可能的机制.
Objective To explore the effect of cold restraint stress on heart and liver of rats,the expression of HSP70 and the levels of CK and ALT in serum was studied. Methods 77 SD rats of 50 days old were randomly divided into 7 groups,namely 0 h,0. 5 h,1. 0 h,1. 5 h,2. 0 h,2. 5 h,3. 0 h group. The cold-restrained rats lasted the corresponding time,then the distributions of HSP70 in heart and liver tissues were examined by means of immunohistochemistry method and the level of CK and ALT in serum was determined. Results (1)The HSP70 expression in heart increased with stress time 1. 5 h group had an obvious increase compared with the 0 h group ( P 0. 05),and after 2. 0 h group,the increase was more significant. (2)The HSP70 expression in liver increased slowly with stress time. Only 3. 0 h group had an obvious increase compared with the 0 h,0. 5h and 1. 0h group (P 0. 05). (3)The level of CK rose quickly at the beginning stage of the stress,then changed smoothly. The level of ALT in serum rose with stress time,during the first 2. 5 h changed smoothly but significantly increased at 2. 5 h group. Conclusion The cold restraint stress could induce the expression of HSP70 in heart and the level of CK in serum markedly increasing in a short time,and increased with stress time,while the expression of HSP70 in liver and the level of ALT in serum had no markedly change in a short time,only after 3 hours the increase became obvious,it suggests that heart is more sensitive to the cold restraint stress,while liver has more powerful resistance.
Objective:To study the dynamic effects of cold restraint stress on HSP70 expression in kidney and serum corticosteroid content in rats,and explore its possible mechanism.Methods:Seventy-seven 50-day-old SD rats were randomly divided into 7 groups,namely 0 h,0.5 h,1.0 h,1.5 h,2.0 h,2.5 h and 3.0 h group.At corresponding restraint stress time,the expression of HSP70 in kidney tissues were tested by means of immunohistochemistry and the level of CORT in serum was also examined.Results:The level of CORT reached its peak in 1.5 h group,but there was no significant difference in other groups.Under the light microscope,the expressions of HSP70 in the nuclei of renal glomerular capillary endothelial cells,in the nuclei and cytoplasm of renal tubular epithelial cells could be observed.The HSP70 expression in kidney increased continuously first,then it was kept in a high level at stress time of 1.5 h,2.0 h and 2.5 h(P0.01),and since then it declined gradually(P0.05).Conclusion:Suitable cold restraint stress can induce the synthetic of HSP70,and it increased with stress time.While excessive cold restraint stress may decreased or block the expressions of HSP70 in kidney.
目的 研究拘束冷应激对大鼠血清中促肾上腺皮质激素(ACTH)、皮质醇(CORT)、肌酸激酶(CK)和丙氨酸转氨酶(ALT)浓度的动态变化及可能的机制.方法 50日龄清洁级大鼠56只,随机分为7组,即0、0.5、1.0、1.5、2.0、2.5和3.0h组.采用拘束冷应激法,在应激相应的时间后,检测大鼠血清中ACTH、CORT、CK、ALT浓度和胃溃疡指数的测定.结果 ①大鼠血清中的ACTH浓度随着应激时间的增加呈现迅速升高的趋势,0.5 h达到最高值,之后又迅速下降,逐渐趋于平缓;②血清中的CORT浓度呈现逐渐升高的趋势.1.5 h达到最高值,之后又迅速下降,逐渐趋于平缓;③血清中的CK和ALT活力显著升高,且随着冷应激时间的增加而递增;④大鼠胃溃疡指数在应激初期呈现迅速升高的趋势,2.5 h时达到最高值.结论 拘束冷应激可使大鼠血清中的ACTH和CORT的浓度上升,至最高值后迅速下降;而血清中CK和ALT的浓度则随着拘束冷应激时间的增加而递增.拘束冷应激导致大鼠发生急性胃溃疡,应激2.5 h时最严重,提示拘束冷应激大鼠模型的造模时间以2.5 h为最佳.
肺栓塞(pulmonary embolism,PE)在住院及长期卧床患者中常常发生.当前,溶解血栓治疗被认为是一种主要的治疗方法,但常常引起严重的出血并发症以至威胁生命.研究发现,一些特殊的新型肺血管内皮细胞抗体与溶栓药物相结合,能在溶栓治疗中提高溶栓的效率并减少出血的发生,以下即对肺栓塞溶栓治疗中内皮细胞的靶向作用的研究进展作一综述。
目的探讨年龄、肥胖、颈围等临床相关易患因素与不同性别阻塞性睡眠呼吸暂停低通气综合征(OSAHS)患者病情和程度的关系。方法对因睡眠打鼾就诊的422例患者行身体参数测量,检测整夜多导睡眠图,分析不同性别患者年龄、颈围、主客观评分、最低血氧饱和度、氧减指数与睡眠呼吸暂停低通气指数(AHI)的关系。结果不论男女性别,OSAHS组与鼾症组各指标比较差异均有显著性,与AHI呈一定相关关系(P<0.05)。两组主观评估(ESS)、最低氧饱和度、氧减指数比较差异有显著性,与AHI有较强的相关性(P<0.05),男性OSAHS者与鼾症者比较年龄差异不明显,颈围明显偏大,而女性OSAHS者年龄较鼾症者大,颈围与AHI无关。结论超重和腹部脂肪蓄积的打鼾者有更大的呼吸暂停的危险性,年龄和颈围在对病情评估和预测时受性别影响较大,ESS、最低氧饱和度或氧减指数(ODI)可作为病情预测的简易指标。
Objective To study the clinical characteristics of patients with obstructive sleep apnea-hypopnea syndrome (OSAHS), and look for a simple way to assess the patient’s condition. Methods 419 patients snoring during sleep were examined by all-night polysomnography and nocturnal oxygen saturation monitor. Then we analyzed the clinical characteristics of the patients divided into 4 groups according to apnea-hypopnea index (AHI). Results The morbidity rate is 84.0% in the snorers. There were significant difference among Epworth Sleep Scale (ESS), Clinical Scale (CS), AHI, blood pressure, obesity parameters, min-SpO2 and oxygen desaturation index (ODI) in patients of each group of OSAHS and simple snoring disease. We used stepwise multiple linear regression to analyze the commonly used clinical parameters, and find that ODI, min-SpO2, and ESS were statistical significant correlation with AHI (R=0.862). Conclusions OSAHS has a high morbidity rate in outpatients with snoring. Age and obesity are liability factors of OSAHS. With the development of OSAHS, blood pressure and blood oxygen will become worse and worse. ODI, min-SpO2 and ESS can be used to assess the pathogenetic condition, even make a primary diagnosis.
OBJECTIVE:To study the diagnostic value of DNA, RNA and proliferating cell nuclear antigen (PCNA) examination in malignant effusion by multiparametric flow cytometry, and therefore to provide proof for clinical application.METHODS:Forty seven patients with pleural effusions in our hospital from August 2003 to February 2004 were divided into two groups: 19 suffering from benign pleural effusions and 28 from malignant effusions confirmed by pathologic examination. The cells for diagnosis were divided into four groups stained by PI (Propidium-iodide), PY (Pyronin), PCNA-FITC and PCNA-mouse-alpha-2a. The specimens were analyzed by a flow cytometer (FacS Caliber, Becton Dickinson). The sensitivity and the specificity of each examination and combined examination were calculated by statistic software SPSS 13.0.RESULTS:(1) The expression of DI, RI, and PI in benign pleural effusion was 1.03 +/- 0.06, 10.03 +/- 0.54, and (4.86 +/- 0.72)%, respectively, and those in malignant ones was 1.26 +/- 0.17, 11.65 +/- 1.45, and (11.97 +/- 1.50)%, respectively, the difference being statistically significant. The cutoff value of DI, RI and PI was 1.10%, 10.75% and 4.56%, and the sensitivity of DI examination was 89.3%, 78.6%, 75.0%, and the specificity was 89.5%, 98.5%, 84.2%, respectively. (2) In 6 cases suffering from malignant pleural effusions, RI was positive but DI was negative, indicating that DI combined with RI examination was better than DI examination alone. (3) In 5 cases suffering from malignant pleural effusions confirmed by tissue examination, the cytology was negative, but the result of DI and RI was abnormal, indicating that flow cytometry was complementary to pathologic examination. (4) The sensitivity of DI + RI, DI + PI, RI + PI and DI + RI + PI combined examination was 98.2%, 89.3%, 89.3%, 92.9%; the specificity was 84.2%, 89.5%, 84.2%, 94.2% respectively. The results demonstrated that DI + RI + PI combined examination was the best, which showed the least false negative and false positive results. The sensitivity of DI + RI combined examination was 98.2%, but the specificity was 84.2%, the false positive rate being higher than DI + RI + PI combined examination. In none of the benign pleural effusions was the DI + RI + PI higher than the cutoff value, suggesting that combined examination can exclude benign pleural effusions.CONCLUSIONS:DNA, RNA and PCNA examinations by flow cytometry are of value in the diagnosis of malignant effusion, especially for cases which can not be diagnosed by cytological examination. DI + RI + PI combined examination showed better results with the lowest false negative and false positive rates.
The cellular responses to hypoxia are characterized by alterations in the expression of a number of genes.This article reviews previous and recent studies that have examined the changes in the expression of proteins in response to chronic sustained and intermittent hypoxia.Results showed that hypoxia affects the level and the state of proteins related to many domain about energy metabolism.This research results provide the possibility for the further ananlysis of protein function and definition of the mechanism about the reaction of cell to hypoxia,which then can play an effective role in control the consequence resulting from hypoxia.
目的:观察肺血栓栓塞症的各种临床资料并对其进行统计,为更好地诊断、治疗提供依据。方法:对两院确诊的肺血栓栓塞症病例详细的临床表现、实验室检查情况、危险因素、治疗方案及结果、误诊情况进行统计分析。结果:70例病例符合肺血栓栓塞症诊断与治疗指南的临床表现、辅检、治疗方案等。结论:肺血栓栓塞症具有高发病率、易误诊、病死率高的特点,临床医师对缺乏典型临床表现的病人应进行全面地诊断及鉴别诊断,寻找合适有效的检查来明确病因。
