Objective: To investigate the effects of the diet rich in different proportions of methionine on endothelial injury in sprague-dawley (SD) rats.Methods: Male SD rats were randomly divided into the normal diet group (CR), 1% methionine loading diet group (M1 R), 3% methionine loading diet group (M3 R) and 5%methionine loading diet group (M5 R), and blood in tail vein were drawn at 0, 2nd, 4th, 6th and 8th week, respec-tively. After 8 weeks, serum concentrations of homocysteine (Hcy) were measured, and the endothelial ultrastruc-ture changes in aorta wall were observed using scanning electron microscope.Results:A signiifcant interaction between high methionine diet and feeding duration on endothelial injury was found (P<0.05), and serum levels of Hcy were signiifcantly increased in rats fed with high methionine diet (P<0.05), with feeding duration increasing. Compared with CR, serum level of Hcy in M3 and M5 R was signiifcantly higher at 2nd week (P<0.01), whereas serum Hcy in M1 R was signiifcantly higher at 4th week (P<0.05). Endothelial cells in M1 R were imperfectly unanimous in size, and the cell surface shrinked. Endothelium in M3 R and M5 R were found to be damaged by worms kinds of harming typical, with enclose wall thrombus and the fat deposition.Conclusion:High dietary intake of methionine is signiifcantly associated with endothelial injury, which suggests that a method setting up endothelial injury animal models would be established.
目的 探讨游泳运动对大鼠血管抗衰老的影响.方法 30只健康雄性SD大鼠(180~220g)随机分为3组:溶剂对照组(n=10),D-半乳糖组(n=10)和有氧运动+D-半乳糖组(运动组,n=10).D-半乳糖组和运动组大鼠每日颈部皮下注射D-半乳糖(125mg/kg),溶剂对照组大鼠每日颈部皮下注射同体积生理盐水,且运动组大鼠每日进行60 min的无负重游泳运动训练,持续8周.以血浆总一氧化氮合成酶(T-NOS)活力,一氧化氮(NO),内皮素(ET)和NO/ET平衡建立血管衰老指标评价体系,并测量主动脉组织超氧化物歧化酶、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)和丙二醛(MDA)含量.结果 与溶剂对照组相比,D-半乳糖组大鼠主动脉MDA和血浆ET含量显著升高,而GSH-Px、SOD活力、血浆NO含量和NO/ET比值显著下降.与D-半乳糖组相比,运动组大鼠主动脉MDA含量和T-NOS活力显著下降,GSH-Px、SOD、T-NOS活力、NO含量和NO/ET比值显著升高(P<0.05),且与对照组相比上述各项指标无显著差异.结论 D-半乳糖皮下注射可诱导大鼠主动脉脂质过氧化反应,血浆NO/ET失衡;适宜的有氧运动能显著增加大鼠血管的抗氧化能力,减少脂质过氧化反应,改善NO/ET失衡,预防血管衰老.
目的 观察蜂胶对幽门螺杆菌(Hp)感染长爪沙鼠胃黏膜的影响.方法 5周龄长爪沙鼠15只,随机分为对照组、感染组和蜂胶组.所有沙鼠禁食24h后,感染组和蜂胶组灌胃Hp菌液109cfu/ml,0.5 ml/只,连续灌胃3次,感染后4周每隔5d灌胃一次,感染后第16周连续5d每天灌胃一次加强感染;对照组灌胃无菌肉汤;蜂胶组在第22周和第24周灌胃蜂胶溶液,感染组和对照组灌胃生理盐水;27周处死所有试验动物,测量肝、肾、脾重量并计算脏器系数,测定血清IL-8,取胃黏膜进行Hp定量培养和病理学观察.结果 与对照组相比,感染组血清IL-8显著降低(P<0.05),与感染组相比,蜂胶组血清IL-8显著升高(P<0.01);各组肝脏、肾脏和脾脏脏器系数差异均不显著(P>0.05);蜂胶组的Hp数量(246±172 cfu/mg)显著低于感染组(1075±688 cfu/mg)(P<0.05);病理检查结果显示感染组胃黏膜炎症较严重.结论 蜂胶可抑制沙鼠胃内Hp的生长,具有一定的黏膜修复和器官保护作用.
[Purpose] To study the long-term and repeated infection of Helicobacter pylori (H. pylori) on Mongolia gerbil’s gastric mucosa cells. [Methods] Twenty 4-week-old Mongolia gerbils were randomly divided into experimental group and control group. In the experimental group,the gerbils were reinfected at 4 weeks after H. pylori colonization infection,and repeated for 6 times,while the gerbils of control group were treated with sterile broth. All gerbils were sacrificed at the 100th week after the first time of H. pylori colonization,then the gastric mucosa were aseptically obtained immediately and processed for transmission electron microscopy observation. [Results] The gastric mucosa in the experimental group showed the ultrastructural changes of chronic gastritis,appeared different degree of atrophy,the number of gastric glands reduced,glandular cells arranged irregularly,connections between cells loosed,vacuolar degeneration and even necrosis in some cells. The majority of gastric parietal cell secretion of tubular dilatation,microvilli sparse,the quantity of mitochondria decreased, some vacuolated; chief cell’s endoplasmic reticulum dilation,nuclear pyknosis,secretory granules are pale or vacuolization,mitochondrial swelling. Four of the experimental gerbil’s gastric mucosal cells became atypically,with larger and irregularly nucleus,derangement arranged of cells,offset nucleoid secretory granules,loosed cells connections,similar to adenocarcinoma cells. Most of gastric interstitial were infiltrated with eosinophils. [Conclusions] The long-term and repeated H. pylori infection may cause the ultrastructure pathological changes abnormal changes,and even tumorigenesis of Mongolia gerbil gastric epithelial cells.
Objective To investigate the effects of Quercetin(QC) on oxidative stress induced by exhausted swimming in rats,so as to provide scientific basis for developing anti-fatigue food.Methods Male SD rats were randomly divided into resting control group,exhausted swimming group,and exhausted swimming with administration of Quercetin group.The content of malondialdehyde(MDA),the activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px) in blood serum were measured.At the same time the survival time after exhausted swimming was determined.Results The survival time in exhausted swimming+Quercetin group was longer than exhausted swimming group((348.5±52.4)min vs(171.7±37.1)min,P0.01).The content of MDA in exhausted swimming+QC group was lower((4.4±1.0) nmol/ml vs(6.2±1.9) nmol/ml,P0.01),whereas the activity of SOD,and GSH-Px was higher((105.0±3.4)U/ml vs(80.5±4.2)U/ml,(873.6±72.3)U/ml vs(538.6±63.6)U/ml respectively,P0.01)than in exhausted swimming group.Conclusion Quercetin could increase the antioxidative enzyme activity,decrease the lipid peroxidation,thus enhance the ability of exercise in exhausted fatigue rats.
OBJECTIVE:To observe the effects of training on plasma content of NO, ET and NO/ET system in rats fed with high methionine diets. METHODS:Male Wistar rats were randomly divided into normal diet group (CR), high methionine diet group (MR) and high methionine diet plus 90 min swimming group (T+ MR). Rats in MR and T+ MR group were fed with the diet which contained 3%of methionine for eight weeks, but rats in control group were fed with ordinary diet. After breeding for eight weeks homocysteine (Hcy) level, T-NOS, NO and ET were measured. RESULTS:Plasma homocysteine of MR increased twofold as compared with that of CR,whereas plasma NO/ET of MR descended significantly (P < 0.05), suggesting that hyperhomocysteinemia was induced by the high methionine diet, and plasma NO/ET system was out of balance. Plasma homocysteine content of T+ MR obviously descended, plasma T-NOS, NO and NO/ET ascended obviously (P < 0.05) as compared with that of MR, and the above indicators of group T + MR were not different as compared with that of CR (P > 0.05). CONCLUSION:Proper training can descend plasma homocysteine level of rats fed with high methionine diet and better balance between NO and ET to prevent developing of hyperhomocysteinemia.
Objective:To investigate the ultrastructure changes in mitochondrion of pancreatic cells from streptozotocin-induced type-2 diabetic rat remodeled intestinal flora.Methods:64 male SD rats were randomly divided into four groups,including control group,type-2-diabetic-modeling group,probiotic group and depollutional group.All groups were fed with high-fat diet and inoculated Streptozotocin(30mg/ml) by intraperitoneal injection one time except control group. Results:Fasting blood glucose increased significantly comparing with control group,while there were no significant differences among those three groups(P>0.05).Mitochondria were significantly impaired in probiotic group revealed by TEM,while the impairment in depollutional group was relatively light.Conclusion:Remodeling intestinal flora resulted in more severe ultrastructural pathology of mitochondria in pancreatic cells from streptozotocin-induced type-2 diabetic rats.
Objective To observe the effects of aerobic training on plasma tissue plasminogen activator/plasminogen activator inhibitor-1 and postaglandin I2/thromboxanes A2 in rats fed a high methionine diet. Methods Ninetysix male Wistar rats were randomly divided into a normal diet control group (CR), a high methionine diet group (MR)and a high methionine diet plus 90 min aerobic training intervention group (T + MR). After 8 weeks of training, the plasma level of homocysteine (Hcy) 6-keto-PGF1α, TXB2, t-PA and PAI-1 were measured. Results Plasma Hcy in the MR group increased two-fold as compared with the CR group, whereas t-PA/PAI-1 and 6-keto-PGF1α/TXB2 of the MR group decreased significantly. Plasma Hcy in the T + MR group significantly decreased, while plasma t-PA/PAI-1 and 6-ketoPGF1α/TXB2 increased significantly compared with the MR group. The above parameters were not different in the T + MR group from those of the CR group. Conclusions Hyperhomocysteinemia could be induced by a high methionine diet.Proper training can decrease the plasma Hcy level of rats fed with a high methionine diet, and correcting the imbalance of t-PA/PAI-1 and 6-Keto-PGF1α/TXB2 could prevent the development of hyperhomocysteinemia.
目的 研究拘束冷应激对大鼠血清中促肾上腺皮质激素(ACTH)、皮质醇(CORT)、肌酸激酶(CK)和丙氨酸转氨酶(ALT)浓度的动态变化及可能的机制.方法 50日龄清洁级大鼠56只,随机分为7组,即0、0.5、1.0、1.5、2.0、2.5和3.0h组.采用拘束冷应激法,在应激相应的时间后,检测大鼠血清中ACTH、CORT、CK、ALT浓度和胃溃疡指数的测定.结果 ①大鼠血清中的ACTH浓度随着应激时间的增加呈现迅速升高的趋势,0.5 h达到最高值,之后又迅速下降,逐渐趋于平缓;②血清中的CORT浓度呈现逐渐升高的趋势.1.5 h达到最高值,之后又迅速下降,逐渐趋于平缓;③血清中的CK和ALT活力显著升高,且随着冷应激时间的增加而递增;④大鼠胃溃疡指数在应激初期呈现迅速升高的趋势,2.5 h时达到最高值.结论 拘束冷应激可使大鼠血清中的ACTH和CORT的浓度上升,至最高值后迅速下降;而血清中CK和ALT的浓度则随着拘束冷应激时间的增加而递增.拘束冷应激导致大鼠发生急性胃溃疡,应激2.5 h时最严重,提示拘束冷应激大鼠模型的造模时间以2.5 h为最佳.
To observe the effects of endothelial secretion function in rats fed with high methionine diet,male Wistar rats were randomly divided into the normal diet group(CR) and high methionine diet group(MR).After 8 weeks,plasma homocysteine level,NO,ET,t-PA and PAI-1were measured.Plasma homocysteine of MR increased twofold as compared with that of CR,whereas NO /ET and t-PA/PAI-1 of MR decreased significantly(P<0.05).Rats fed with high methionine diet could be led to hyperhomocysteinemia,and a serious disequilibrium of plasma t-PA /PAI-1 and NO/ET.
目的 探讨二巯基丁二酸(DMSA)与维生素C、蛋氨酸联合应用对染铅ICR小鼠血铅及肝脏的影响. 方法 实验组小鼠饮用0.1%的醋酸铅溶液6周后, 随机分为模型对照组、低剂量DMSA组、高剂量DMSA组及低剂量DMSA与维生素C、蛋氨酸联合应用组(联合组).给药4周后,测小鼠的血铅.并取肝脏组织进行苏木素-伊红(HE)染色,观察肝脏的形态学改变. 结果 低剂量DMSA组、高剂量DMSA组与联合组小鼠血铅浓度和模型对照组差异有显著性( P<0.01); 正常对照组血铅浓度和模型对照组差异有显著性(P <0.01).肝病理学表现为肝细胞可见变性水肿,其中以联合组水肿程度最轻.结论 DMSA与维生素C、蛋氨酸联合应用对小鼠体内铅的排泄具有更好的效果.
目的探讨提高含少量骨髓细胞恶性血液病标本染色体核型检出率的方法。方法用常规浓度秋水仙酰胺1h、2h、4h、6h、8h处理10例骨髓标本,寻找最佳阻滞时间,以该时间阻滞含少量骨髓细胞恶性血液病标本,用R带技术分析染色体核型分析情况。结果(1)用常规浓度秋水仙酰胺6h阻滞细胞中期得到的分裂相数最多,而4h可分析分裂相数最多,与常规1h相比有统计学意义(P<0.05)。(2)以秋水仙酰胺4h处理15例含少量骨髓细胞恶性血液病标本,有9例核型分析成功,成功率为60%;而常规1h处理7例标本,只有1例成功,成功率只有14.3%。结论以常规浓度秋水仙酰胺4h延长细胞中期分裂可能提高含少量骨髓细胞恶性血液病标本染色体核型检查成功率。
