Background Periodontitis represents a chronic inflammatory disease resulting from the interaction between plaque-associated microorganisms and the host immune response. PANoptosis, a newly recognized programmed cell death, is linked to multiple inflammatory disorders. However, its role in periodontitis pathogenesis and diagnostic potential remains unclear. This study aimed to identify key PANoptosis-related biomarkers in periodontitis using bioinformatics analysis followed by experimental verification. Methods Three machine-learning algorithms were applied to identify key differentially expressed PANoptosis-related genes (DE-PRGs), followed by ROC analyses in public datasets and sequencing data to assess their diagnostic performance for periodontitis. Functional enrichment, immune infiltration analysis, and drug prediction analyses were conducted on these DE-PRGs. The mRNA expression of these genes in human gingival tissue was confirmed by qPCR, and protein expression was validated in a rat periodontitis model through immunohistochemistry. Results ALOX5, CDK5R1, MMP14, and JAK3 were identified as key candidate genes, primarily enriched in inflammation, oxidative stress and immune pathways. These genes showed good diagnostic value in both public periodontitis databases and sequencing data. qPCR and IHC findings showed higher expression levels of ALOX5, MMP14, and JAK3 in human and rat periodontitis tissues compared to healthy controls, while CDK5R1 expression was reduced (P< 0.05). Drug prediction identified tofacitinib, zileuton, and melatonin as potential periodontitis therapeutics. Conclusion This study provides new evidence linking PANoptosis to periodontitis. ALOX5, CDK5R1, MMP14 and JAK3 were identified as PANoptosis-related biomarkers and potential therapeutic targets. These findings support the precision diagnosis and treatment of periodontitis.
This study investigated the protective role and molecular mechanism of WNT4 in preserving the viability and neurogenic potential of dental pulp stem cells (DPSCs) within an inflammatory microenvironment following facial nerve injury (FNI). Human DPSCs (hDPSCs; authenticated by surface marker profiling and multi-lineage differentiation) and a rat model of facial nerve transection (n = 15 per group) were utilized. In vitro, hDPSCs were treated with TNF-α(10 ng/mL) to model inflammation. In vivo, WNT4-overexpressing or control DPSCs (1 × 106 cells) were injected into the tail vein immediately after nerve transection. PANoptosis and neurogenic differentiation were assessed by Western blot, flow cytometry, and immunofluorescence staining. Transcriptional regulation was analyzed by dual-luciferase reporter assay and ChIP-qPCR. Protein interaction was examined by LC–MS/MS and docking. In vivo recovery was monitored by functional assessments and histology. WNT4 overexpression significantly attenuated TNF-α-induced PANoptosis in hDPSCs and restored their neurogenic potential. Mechanistically, KLF7 transcriptionally activated WNT4. WNT4 interacted with NOTCH1, activating Wnt/β-catenin signaling and upregulating c-Jun, CYCD1, and VEGFA. In vivo, engineered hDPSCs reduced local inflammation and accelerated functional recovery and histological repair. WNT4 protects DPSCs against inflammatory PANoptosis and enhances neuroreparative potential, providing a basis for optimizing stem cell therapy in FNI.
PURPOSE:Refractive surgery may exacerbate the existing dry eye symptoms in patients with dry eye syndrome. Therefore, we explore the therapeutic and protective effects of using optimal intense pulsed light (IPL) therapy combined with meibomian gland expression (MGX) before and after surgery on post-laser corneal refractive surgery dry eyes. METHODS:This was a prospective, self-controlled, single-center clinical trial. Using optimal IPL therapy and MGX, a total of 68 patients with mild-to-moderate evaporative dry eye or meibomian gland dysfunction who underwent refractive surgery received 2 sessions of treatment. The treatment was administered once a week preoperatively and once a week postoperatively. Baseline measurements were taken before optimal IPL therapy and MGX, and the parameters were assessed at 1 week and 1 month after surgery. RESULTS:Out of 68 patients, 54 patients received small-incision lenticule extraction (SMILE), while 14 patients received femtosecond laser-assisted in situ keratomileusis (FS-LASIK) (results primarily reflect SMILE outcomes due to sample imbalance). In the SMILE group, significant improvement was already observed in uncorrected/corrected distance visual acuity (P = 1.42 × 10-3), meibomian gland expressibility score (MGYSS) (P = 0.01), and meibum yielding secretion score (MGS) (P = 5.39 × 10-4) between 1 week before and after the surgery. First noninvasive breakup time (NIBUTf) (P = 8.98 × 10-3), average noninvasive breakup time (NIBUTav) (P = 2.30 × 10-4), and meibomian gland dropout score (bottom) (P = 9.05 × 10-3) were observed to have significant improvement till 1 month after the surgery. Patients in the FS-LASIK group exhibited a comparable trend of change. CONCLUSION:IPL treatment combined with MGX may improve ocular discomfort and dry eye symptoms in MGYSS, MGS, NIBUTf, NIBUTav, and meibomian gland dropout score (bottom) for patients with post-laser corneal refractive surgery dry eyes in short term.
This study first investigated how FNDC5 affected the development of oral cancer and revealed the role of FNDC5 in the migration and invasion of oral cancer. The present work evaluated differential FNDC5 expression within oral cancer samples versus matched non-carcinoma samples based on GEO database analysis and immunohistochemistry. We then generated oral cancer cell lines with FNDC5 overexpression and knockdown to determine the role of altered FNDC5 expression in the migration and invasion of oral cancer. PI3K inhibitor was used for investigating the possible mechanism underlying FNDC5 during EMT of oral cancer. Finally, these in-vitro results were validated using the lung metastatic nude mouse model. According to our results, FNDC5 level markedly decreased within oral cancer compared with adjacent samples and FNDC5 overexpression inhibited migration, invasion as well as EMT of oral cancer, while FNDC5 knockdown promoted oral cancer cell EMT. In addition, PI3K inhibitors blocked the induction of oral cancer cells EMT by FNDC5 knockdown. In vivo experiments further demonstrated the above results. This work is the first to illustrate the impact of FNDC5 on inhibiting migration and invasion of oral cancer, and our results suggest that FNDC5 affects EMT of oral cancer via the inhibition of PI3K/Akt/Snail pathway.
BackgroundThe aberrant expression of AEG-1 is significantly correlated with tumorigenesis, development, neurodegeneration and inflammation. However, the relationship between AEG-1 expression and immune infiltration in OSCC, as well as other tumor types, has yet to be comprehensively analyzed.MethodsThe expression levels, prognostic and clinicopathological characteristics, mutation patterns and methylation landscapes of AEG-1 in various tumors were obtained from multiple databases, including TIMER, GEPIA, HPA, TCGA, UALCAN, cBioPortal, SMART and TISIDB, in addition to single-cell RNA-seq data. The integration of these datasets facilitated the elucidation of the relationships among pan-cancer cellular heterogeneity, immune infiltration and AEG-1 expression levels. In vitro experiments created AEG-1 overexpressing cell lines, and mRNA-seq analyzed AEG-1-related differential genes in OSCC. RT-PCR validated these findings in vivo using xenograft tumors. Tumor cell lines were developed to study AEG-1’s effects through H&E, Masson, and PAS staining. Immunohistochemistry examined AEG-1-related gene expression patterns.ResultsOur analysis demonstrated that AEG-1 is highly expressed across various cancer types and is associated with tumor grade and patient prognosis. Additionally, AEG-1 amplification was observed in multiple cancers. Notably, we identified a significant elevation of AEG-1 expression in OSCC, which strongly correlated with patient prognosis and immune infiltration. Through mRNA-seq analysis of differentially expressed genes and immune-related gene sets, we identified a strong correlation between AEG-1 and immune infiltration markers such as LCP2, CD247, HLA-DPA1, HLA-DRA, HLA-DRB1, CIITA and CD74 in OSCC. Additionally, AEG-1 was found to regulate Th1/Th2 immune homeostasis, promote glycogen accumulation, and contribute to tumor fibrosis.ConclusionIn conclusion, AEG-1 significantly correlates with prognosis and immune infiltration across various cancer types and holds potential as a novel prognostic immune biomarker for OSCC. This finding may facilitate the identification of patients who are most likely to benefit from adjuvant immunotherapy.
Background Facial nerve injury often results in poor prognosis due to the challenging process of nerve regeneration. Neuregulin-1, a human calmodulin, is under investigation in this study for its impact on the reparative capabilities of Dental Pulp Stem Cells (DPSCs) in facial nerve injury. Methods Lentivirus was used to transfect and construct Neuregulin-1 overexpressed DPSCs. Various techniques assessed the effects of Neuregulin-1: osteogenic induction, lipid induction, Reverse Transcription Polymerase Chain Reaction, Western Blot, Cell Counting Kit-8 assay, wound healing, immunofluorescence, Phalloidin staining, nerve stem action potential, Hematoxylin-eosin staining, transmission electron microscopy, and immunohistochemistry. Results Neuregulin-1 effectively enhanced the proliferation, migration, and cytoskeletal rearrangement of DPSCs, while simultaneously suppressing the expression of Ras homolog gene family member A (RhoA) and Microfilament actin (F-actin). These changes facilitated the neural differentiation of DPSCs. Additionally, in vivo experiments showed that Neuregulin-1 expedited the restoration of action potential in the facial nerve trunk, increased the thickness of the myelin sheath, and stimulated axon regeneration. Conclusion Neuregulin-1 has the capability to facilitate the repair of facial nerve injuries by promoting the regenerative capacity of DPSCs. Thus, Neuregulin-1 is a significant potential gene in the reparative processes of nerve damage.
背景:牙髓干细胞由于其来源广泛、免疫原性低并具有优秀的向神经系统细胞分化的能力,因此对神经系统疾病有潜在的治疗价值.目的:总结牙髓干细胞神经源性标记物的表达特点,回顾以往牙髓干细胞向神经系统细胞分化的研究进展,为其应用于临床神经系统疾病的治疗提供研究基础.方法:应用计算机以"牙髓干细胞、神经分化、神经细胞标记物、神经"为中文检索词,以"dental pulp stem cells,neuro-differentiation,nerve biomarker,nerve"为英文检索词,检索中国知网、万方数据和PubMed数据库中的相关文献,根据纳入标准并筛选,最终选择85篇文献进行综述.结果 与结论:①牙髓干细胞向神经细胞分化过程中所表达的几种常用的神经标记物主要包括:巢蛋白、性别决定基因相关转录因子2、βⅢ微管蛋白、胶质纤维酸性蛋白、微管相关蛋白2、S100钙结合蛋白B、神经元特异性烯醇化酶、神经元特异性核蛋白,文章总结归纳了上述标记物的特点、表达的时期以及在牙髓干细胞神经向分化研究中的应用情况.②目前研究表明,判断牙髓干细胞向神经细胞分化是否成功,需要结合形态学特征以及分化后细胞的功能进行综合判断,同时使用单一标记物很难专门识别单个细胞谱系,因此,未来有必要将多个标记物应用于一种细胞类型进行分析.③文章还从生长因子、信号通路及生物支架等方面介绍了牙髓干细胞向神经样细胞分化的方法,以期为诱导牙髓干细胞神经向分化研究提供理论参考.
ObjectiveThe aim of this research was to investigate the effect of vascular endothelial growth factor A (VEGFA)-overexpressing rat dental pulp stem cells (rDPSCs) combined with laminin-coated and yarn-encapsulated poly(l-lactide-co-glycolide) (PLGA) nerve guidance conduit (LC-YE-PLGA NGC) in repairing 10 mm facial nerve injury in rats.Study DesignrDPSCs isolated from rat mandibular central incisor were cultured and identified in vitro and further transfected with the lentiviral vectors (Lv-VEGFA). To investigate the role and mechanisms of VEGFA in neurogenic differentiation in vitro, semaxanib (SU5416), Cell Counting Kit-8 (CCK-8), real-time quantitative polymerase chain reaction (qPCR) and Western blotting were performed. Ten-millimeter facial nerve defect models in rats were established and bridged by LC-YE-PLGA NGCs. The repair effects were detected by transmission electron microscopy (TEM), compound muscle action potential (CMAP), immunohistochemistry and immunofluorescence.ResultsExtracted cells exhibited spindle-shaped morphology, presented typical markers (CD44+CD90+CD34−CD45−), and presented multidirectional differentiation potential. The DPSCs with VEGFA overexpression were constructed successfully. VEGFA enhanced the proliferation and neural differentiation ability of rDPSCs, and the expression of neuron-specific enolase (NSE) and βIII-tubulin was increased. However, these trends were reversed with the addition of SU5416. This suggests that VEGFA mediates the above effects mainly through vascular endothelial growth factor receptor 2 (VEGFR2) binding. The LC-YE-NGC basically meet the requirements of facial nerve repair. For the in vivo experiment, the CMAP latency period was shorter in DPSCS-VEGFA-NGC group in comparison with other experimental groups, while the amplitude was increased. Such functional recovery correlated well with an increase in histological improvement. Further study suggested that VEGFA-modified DPSCs could increase the myelin number, thickness and axon diameter of facial nerve. NSE, βIII-tubulin and S100 fluorescence intensity and immunohistochemical staining intensity were significantly enhanced.ConclusionVEGFA-modified rDPSCs combined with LC-YE-PLGA NGCs have certain advantages in the growth and functional recovery of facial nerves in rats.
Mitochondria play a critical role in nerve regeneration, yet the impact of gene expression changes related to mitochondria in facial nerve regeneration remains unknown. To address this knowledge gap, we analyzed the expression profile of the facial motor nucleus (FMN) using data obtained from the Gene Expression Omnibus (GEO) database (GSE162977). By comparing different time points in the data, we identified differentially expressed genes (DEGs). Additionally, we collected mitochondria-related genes from the Gene Ontology (GO) database and intersected them with the DEGs, resulting in the identification of mitochondria-related DEGs (MIT-DEGs). To gain further insights, we performed functional enrichment and pathway analysis of the MIT-DEGs. To explore the interactions among these MIT-DEGs, we constructed a protein-protein interaction (PPI) network using the STRING database and identified hub genes using the Degree algorithm of Cytoscape software. To validate the relevance of these genes to nerve regeneration, we established a rat facial nerve injury (FNI) model and conducted a series of experiments. Through these experiments, we confirmed three MIT-DEGs (Myc, Lyn, and Cdk1) associated with facial nerve regeneration. Our findings provide valuable insights into the transcriptional changes of mitochondria-related genes in the FMN following FNI, which can contribute to the development of new treatment strategies for FNI.
神经损伤是临床中的常见及多发疾病,可分为周围神经损伤(peripheral nervous injury,PNI)和中枢神经损伤(central nervous injury,CNI),目前采取的治疗方法主要有自体神经移植、异体神经移植、细胞促进疗法、组织工程法等.相比于其他三种方法,组织工程法的发展将从根本上解决组织和器官缺损所致的功能障碍或丧失治疗等问题,以达到完美的形态修复.神经调节素-1(neuregulin-1,NRG-1)作为一种经典的生长和分化因子,应用到组织工程上可以抑制细胞衰老、促进细胞增殖和分化,并且能够通过相关信号通路促进PNI和CNI后的神经修复.本文主要说明NRG-1的结构、功能及其在PNI与CNI修复中发挥的作用及作用机制.
Background: Periodontitis is associated with periodontal tissue damage and teeth loss. Nonalcoholic fatty liver disease (NAFLD) has an intimate relationship with periodontitis. Nevertheless, interacted mechanisms between them have not been clear. This study was intended for the exploration of shared gene signatures and latent therapeutic targets in periodontitis and NAFLD. Methods: Microarray datasets of periodontitis and NAFLD were obtained from the Gene Expression Omnibus (GEO) database. The weighted gene co-expression network analysis (WGCNA) was utilized for the acquisition of modules bound up with NAFLD and periodontitis. We used ClueGO to carry out biological analysis on shared genes to search their latent effects in NAFLD and periodontitis. Another cohort composed of differential gene analysis verified the results. The common microRNAs (miRNAs) in NAFLD and periodontitis were acquired in the light of the Human microRNA Disease Database (HMDD). According to miRTarbase, miRDB, and Targetscan databases, latent target genes of miRNAs were forecasted. Finally, the miRNAs–mRNAs network was designed. Results: Significant modules with periodontitis and NAFLD were obtained via WGCNA. GO enrichment analysis with GlueGo indicated that damaged migration of dendritic cells (DCs) might be a common pathophysiologic feature of NAFLD and periodontitis. In addition, we revealed common genes in NAFLD and periodontitis, including IGK, IGLJ3, IGHM, MME, SELL, ENPP2, VCAN, LCP1, IGHD, FCGR2C, ALOX5AP, IGJ, MMP9, FABP4, IL32, HBB, FMO1, ALPK2, PLA2G7, MNDA, HLA-DRA, and SLC16A7. The results of differential analysis in another cohort were highly accordant with the findings of WGCNA. We established a comorbidity model to explain the underlying mechanism of NAFLD secondary to periodontitis. Finally, the analysis of miRNA pointed out that hsa-mir-125b-5p, hsa-mir-17-5p, and hsa-mir-21-5p might provide potential therapeutic targets. Conclusion: Our study initially established a comorbidity model to explain the underlying mechanism of NAFLD secondary to periodontitis, found that damaged migration of DCs might be a common pathophysiological feature of NAFLD and periodontitis, and provided potential therapeutic targets.
牙髓干细胞(dental pulp stem cells,DPSCs)是一类具有高度增殖、多向分化潜能的成体干细胞,其来源丰富,具有提取较为便捷、伦理争议较小的优势.由于DPSCs具有多向分化的潜能,可以在一定条件下诱导成神经向分化,在神经损伤的修复和神经遗传性疾病的治疗等方面应用前景广阔.神经营养素(neurotrophin,NT)是一类能够促进轴突生长、利于神经细胞分化与存活的由神经所支配的组织或细胞产生的蛋白质分子,可以分为神经营养因子(neurotrophic factor,NTFs)家族、胶质细胞源性神经营养因子(glial cell-derived neurotrophic factor,GDNF)家族、白细胞介素-6(interleukin-6,IL-6)细胞因子家族、成纤维细胞生长因子(fibroblast growth factor,FGF)家族等,NT作为促进神经细胞分化的营养因子,也能够促进干细胞神经向分化,其对DPSCs神经向分化有一定的引导作用.该文简要介绍了NT在促进细胞神经向分化方面的作用,就NT对DPSCs神经向分化的影响和应用前景进行概述.
目的:探讨银离子藻酸盐敷料对不同类型手术切口的影响.方法:共选取患者100例,对照组50例,给予普通无菌敷料治疗.其中Ⅰ类切口(清洁切口)5例,Ⅱ类切口(清洁-污染切口)7例,Ⅲ类切口(污染切口)38例.实验组50例患者采用银离子藻酸盐敷料治疗,其中Ⅰ类切口(清洁切口)5例,Ⅱ类切口(清洁-污染切口)7例,Ⅲ类切口(污染切口)38例.比较两组患者的感染情况、愈合时间及视觉模拟评分(VAS),观察换药过程中的疼痛情况.结果:(1)Ⅰ类切口两组感染率均为0,组间无差异(P>0.05);Ⅱ类、Ⅲ类切口对照组感染率明显高于观察组(P<0.05);(2)Ⅰ类切口两组愈合时间差异不显著(P>0.05);Ⅱ类、Ⅲ类切口愈合时间对照组明显长于观察组(P<0.05);(3)Ⅰ类、Ⅱ类、Ⅲ类切口 VAS评分观察组均低于对照组(P<0.05).结论:银离子藻酸盐敷料对伤口感染有显著效果,特别是在Ⅱ类、Ⅲ类切口中,不仅可以缓解疼痛,还可以降低伤口感染率,缩短伤口愈合时间.
Peripheral nerve injury (PNI) is a common disease in the oral cavity that can easily lead to loss of function and abnormal appearance. The application of dental pulp stem cells (DPSCs) combined with tissue engineering in the repair of PNI is a research hotspot. DPSCs have the advantages of abundant sources, simple extraction, low immunogenicity and a high proliferation rate in vitro. They can differentiate into Schwann cells (SCs). SCs can induce autophagy and secrete key neurotrophic factors, such as nerve growth factor, brain-derived neurotrophic factor, ciliary neurotrophic factor and glial cell-derived neurotrophic factor. SCs are beneficial for the repair of nerve injury. DPSCs in different periods have differences in immune regulation, anti-inflammatory effects, expression of neural markers, angiogenesis and so on, which provide more diversified choices for nerve repair. At present, the introduction of tissue engineering provides a more controllable and improved microenvironment for DPSCs, which is conducive to the application and development of DPSCs in regenerative medicine and tissue engineering. However, there are still many problems to be solved, such as the selection of stem cells, functional link recovery, uncontrollable direction of axon regeneration, regulation of the peripheral nervous system and mechanism of repair.
Oral squamous cell carcinoma (OSCC) is a common cancer that develops from oral epithelial cells, it has a high incidence, mortality and teratogenic rate, which poses a serious threat to people's life and health.The Hippo signaling pathway plays a key role in tumorigenesis, regulation of stem cell homeostasis, tissue regeneration, and organ size control. In OSCC, activation of Hippo signaling pathway can inhibit malignant biological behavior, epithelial mesenchymal transformation and distant metastasis of tumors, and improve the survival rate of patients. Considering the importance of the Hippo signaling pathway in the development of cancer, this paper summarized the composition and regulatory mechanism of Hippo pathway, elaborated the role of Hippo signaling pathway in the occurrence and development of OSCC.At the same time, make a simple generalization about the potential therapeutic approaches and strategies to reduce the risk of drug resistance for OSCC patients targeting this pathway.
目的:研究二甲双胍(metformin,MF)对成牙骨质细胞(cementoblasts,CB)增殖、分化、矿化能力的影响,为临床牙骨质缺损、牙根修复提供新的用药思路.方法:对成牙骨质细胞系OCCM-30分别给予不同浓度的MF培养,MTS法检测CB的增殖;实时荧光定量PCR法(quantitative real-time polymerase chain reaction,qPCR)及蛋白免疫印迹法检测骨涎蛋白(bone sialoprotein,BSP)和Runt相关转录因子2(runt-related transcription factor 2,Runx2)在基因及蛋白层面的表达;碱性磷酸酶(alkaline phosphatase,ALP)活性测定及茜素红染色法检测MF对CB矿化的影响.结果:不同浓度的MF作用于OCCM-30细胞24、48、72小时后,200μmol/L和400μmol/L MF组对细胞的增殖能力具有促进作用(P<0.05),高浓度1600μmol/L MF组在作用细胞72小时后对细胞增殖产生抑制作用(P<0.05).与对照组相比,用200μmol/L MF处理OCCM-30细胞12、24、48小时后,Runx2和BSP mRNA及蛋白的表达量明显增高(P<0.05).200μmol/L MF作用OCCM-30细胞3天时ALP活性高于对照组(P<0.05),作用7天时则无统计学差异(P>0.05).200μmol/L MF作用OCCM-30细胞21天后,矿化结节形成百分率明显高于对照组(P<0.01).结论:一定浓度范围内的MF能促进CB的增殖、分化及矿化,提高CB的成骨能力.
The Bmp2 and Bmp4 expressed in root mesenchyme were essential for the patterning and cellular differentiation of tooth root. The role of the epithelium-derived Bmps in tooth root development, however, had not been reported. In this study, we found that the double abrogation of Bmp2 and Bmp4 from mouse epithelium caused short root anomaly (SRA). The K14-cre ; Bmp2 f/f ; Bmp4 f/f mice exhibited a persistent Hertwig’s Epithelial Root Sheath (HERS) with the reduced cell death, and the down-regulated BMP-Smad4 and Erk signaling pathways. Moreover, the Shh expression in the HERS, the Shh-Gli1 signaling, and Nfic expression in the root mesenchyme of the K14-cre ; Bmp2 f/f ; Bmp4 f/f mice were also decreased, indicating a disrupted epithelium- mesenchyme interaction between HERS and root mesenchyme. Such disruption suppressed the Osx and Dspp expression in the root mesenchyme, indicating an impairment on the differentiation and maturation of root odontoblasts. The impaired differentiation and maturation of root odontoblasts could be rescued partially by transgenic Dspp . Therefore, although required in a low dosage and with a functional redundancy, the epithelial Bmp2 and Bmp4 were indispensable for the HERS degeneration, as well as the differentiation and maturation of root mesenchyme.
目的:探讨星形胶质细胞升高基因-1 (astrocyte elevated gene-1 ,AEG-1)对5 -氟尿嘧啶(5 -FU)诱导的口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)细胞系耐药性的影响.方法:验证高(SCC15)、中(转染空质粒的对照组SCC15、CAL27)、低(CAL27)四组不同AEG-1 表达量细胞模型AEG-1 基因转染效果.MTT实验观察四组细胞对5 -FU耐受性的变化;蛋白质印迹法和细胞划痕实验观察四组细胞在5 -FU作用下caspase-3、MMP-2、MMP-9蛋白表达水平变化及细胞迁移能力的变化.结果:AEG-1 基因在细胞中转染成功.MTT实验显示不同浓度5 -FU作用四组细胞后,过表达组细胞的半数抑制浓度(IC50 )为423.7 μg/ml,其对照组为213.5 μg/ml(P<0.05);沉默细胞组IC50值为291.6 μg/ml,其对照组为463.1 μg/ml(P<0.05).Western blot显示过表达组及其对照组中5 -FU的浓度分别为400 μg/ml和200 μg/ml时,MMP-2、MMP-9及caspase-3的表达水平开始升高(P<0.05 );沉默组及其对照组中5 -FU的浓度分别为240 μg/ml和480 μg/ml时,MMP-2、MMP-9及caspase-3的表达水平开始升高(P<0.05 ).划痕实验显示加入相同浓度5 -FU后过表达组细胞迁移率明显高于其对照组(P<0.05);而沉默组细胞迁移率明显低于其对照组 (P<0.05).结论:上调/下调AEG-1 表达能增加/降低OSCC细胞对5 -FU的耐药性,提示OSCC细胞的耐药性可能与AEG-1 表达量相关,OSCC细胞系中AEG-1 的表达情况可能能间接干扰临床药物化疗的效果.
目的:观察在上皮组织中条件性敲除Bmp2和Bmp4基因对小鼠牙根发育的影响,为深入研究牙根发育的分子机制提供前期基础.方法:利用Jackson Laboratory推荐的引物,对鼠尾组织进行聚合酶链式反应,鉴定小鼠基因型.取出小鼠下颌骨组织,经过相应处理后,进行石蜡包埋,用于HE染色.在牙根发育早期过程中比较Bmp2和Bmp4基因敲除组(double gene knockout,DcKO)与同源对照组(control,Con)小鼠的上皮根鞘(hertwig's epithelial root sheath,HERS)和根部成牙本质细胞的形态差异,在牙根发育完成后经小动物高清X线观察两组小鼠牙根状况.结果:出生后0 d,两组小鼠牙根尚未发育;出生后7 d,与对照组相比,实验组小鼠根部成牙本质细胞极化更差;出生后14 d,DcKO小鼠的HERS组织比Con组断裂程度低.牙根发育完成后,21 d和28 d的X线结果显示实验组小鼠的牙根比对照组短,根部髓腔比对照组小鼠大.结论:Bmp2和Bmp4基因参与小鼠牙根发育,Bmp2和Bmp4基因缺失可导致DcKO组小鼠的牙根变短.
目的:探讨银离子藻酸盐敷料对术后切口感染的应用效果.方法:将80例患者随机分为观察组和对照组,观察组在常规清创后应用银离子藻酸盐敷料,而对照组在常规清创后采用利凡诺敷料,观察两组患者的换药次数和切口完全愈合时间.结果:与对照组相比观察组的换药次数为(27±8.49)次,切口完全愈合时间为(16.58±6.57)d,明显缩短,二者相比有统计学意义(P<0.05).结论:银离子藻酸盐敷料具有良好的控制伤口感染及促进伤口愈合的作用,使用银离子藻酸盐敷料处理感染伤口的换药能够缩短住院天数,减少换药次数及费用.
