A large-scale and comprehensive survey of public environmental awareness was carried out in Liaoning Province, China, where one of the largest heavy industrial bases is located. This survey was organized by the EU-CHINA Liaoning Integrated Environmental Program. The aim of the survey was to collect public information and awareness of environmental issues and therefore provide a valuable reference source for the government to formulate environmental policy. Statistical survey specialists and environmental protection experts worked together to establish survey items and questionnaires, and the design and survey were performed in strict accordance to the requirements of the project. The survey involved five large cities and 5996 respondents, representing residents, policy-makers, teachers, students, workers in enterprises, and farmers in suburban regions of the five cities. The evaluation of public opinion on current environmental issues and diversity among different survey groups were summarized statistically. The findings of the survey research are: the public is very concerned about air, water, and food pollution, improvement of the environment for community life and further global environmental changes. The survey also emphasized the need for better education to raise environmental awareness and the importance of public initiatives in environmental protection and management.
Public environmental awareness is of significance in formulating and implementing the policies of national environmental protection.In this study,an environmental awareness survey was made in five heavy chemo-industrial cities of Liaoning Province.The target groups included residents,policy-decision makers,teachers,students,enterprise staffs and farmers,and the main survey contents were public evaluation on the changes of environmental quality,satisfaction degree on the environment of living communities,environmental issues concerned about,types of environmental pollution,origins of environmental pollution and ecological degradation,causes of the differences in regional environmental quality,and cognition levels of environmental protection and its national development objectives.The results showed that 78.7% of the publics considered that the environmental quality was getting better during last 5 years,but 65.3% of them didn't satisfy their living environment.The publics held that the fund use for environmental protection should be in order of garbage disposal tree plantingwaste water treatmentair quality control.The survey also demonstrated that most of the publics lacked of needful knowledge about environmental protection,and sufficient understanding on water and food contamination.The weak public environmental awareness had an adverse effect on the improvement of environmental quality,and the publics strongly called for the propaganda of environmental awareness.This study would provide bases to the administrations of Liaoning Province for regional environmental protection,ecological construction,sustainable economic development,and formulation of the policies and regulations on environmental protection.
Accurate interpretation of microscopic changes in tissues is critical in hazard identification and risk assessment. To address a possible confounder, the effects of postmortem interval on hepatocyte vacuolation and liver weight were studied in fasted and nonfasted Sprague-Dawley rats. Male and female rats (5/sex/interval) were euthanized with CO2, weighed, and necropsied either immediately or after remaining in the closed CO2 chamber for 5, 10, or 25 minutes after respirations ceased. The liver was removed, weighed, and fixed for light microscopy, immunohistochemistry, and electron microscopy. The liver weight and liver to body weight ratio increased significantly in both male and female rats. Postmortem hepatocellular vacuolation was more prominent in males than in females. Both fasted and nonfasted males were similarly affected, however, fasted females were affected more than nonfasted females at the 25-minute interval. Ultrastructurally, intracytoplasmic vacuoles in hepatocytes and/or endothelial cells contained electron-lucent material that was morphologically similar to plasma in sinusoidal spaces. Results of our study suggest that hepatocyte vacuoles were formed in a postmortem time-dependent manner as a result of plasma influx into the cytoplasm. This change was associated with hepatic sinusoidal congestion and increases in liver weight. Males were more sensitive than females to postmortem hepatocyte vacuolation.
TRAF4 is one of six identified members of the family of TNFR-associated factors. While the other family members have been found to play important roles in the development and maintenance of a normal immune system, the importance of TRAF4 has remained unclear. To address this issue, we have generated TRAF4-deficient mice. Despite widespread expression of TRAF4 in the developing embryo, as well as in the adult, lack of TRAF4 expression results in a localized, developmental defect of the upper respiratory tract. TRAF4-deficient mice are born with a constricted upper trachea at the site of the tracheal junction with the larynx. This narrowing of the proximal end of the trachea results in respiratory air flow abnormalities and increases rates of pulmonary inflammation. These data demonstrate that TRAF4 is required to regulate the anastomosis of the upper and lower respiratory systems during development.
Previous studies have shown that the pan CD28/cytotoxic T lymphocyte antigen (CTL)A-4 antagonist CTLA4 immunoglobulin (Ig) inhibits eosinophilic airway inflammation in Schistosoma mansoni-sensitized and airway-challenged mice. In the present study, the importance of CD28 as well as the individual roles of CD80 and CD86 were examined in this system using wild-type and CD28 knockout (KO) mice. Unlike wild-type controls, CD28KO mice did not produce systemic IgE or eosinophilic airway inflammation after antigen challenge. However, a lymphocytic infiltrate and continued production of interferon-gamma was observed in these animals. Thus, CD28 is not essential for the initial recruitment of lymphocytes into antigen-challenged airways but critically regulates the allergic T-helper 2 phenotype. We next determined by polymerase chain reaction and flow cytometry that CD80 and CD86 molecules are constitutively expressed in the naive murine lung and on eosinophils in the allergic lung, suggesting a potential important role for both ligands in the development of asthma. Combined anti-CD80/anti-CD86 treatment throughout the antigen challenge period fully blocked the development of allergic airways, whereas a partial reduction was observed in mice treated with either anti-CD80 or anti-CD86 antibody alone. However, only anti-CD86 blocked systemic IgE production. Therefore, signaling through either CD80 or CD86 is sufficient to generate a partial local allergic response, whereas CD86 costimulation is essential to induce systemic allergic (IgE) reactions. Finally, combined anti-B7 monoclonal antibody treatment after sensitization reduced airway eosinophilia and interleukin (IL)-4/IL-5 cytokine secretion consistent with an ongoing role for CD28/B7 interactions in the effector phase of the disease. These results emphasize the importance of differential B7 expression on different cells and in different organs on subsequent CD28/B7-mediated immune events, including the potential for CD28/B7 blockade in the treatment of atopic airway disease in people.
We studied the effects of an anti-interleukin (IL)-5 monoclonal antibody (TRFK-5) or dexamethasone (DEX) to reverse already established airway hyperresponsiveness (AHR) and tissue eosinophilia in a Schistosoma mansoni antigen-sensitized and airway-challenged mouse model of chronic asthma. In this model at 4 d after antigen challenge there is dramatic bronchoalveolar lavage fluid (BAL) eosinophilia, AHR to intravenous methacholine (MCh), and histologic evidence of peribronchial eosinophilic infiltration and mucoid cell hyperplasia. These changes persist for up to 2 wk after antigen challenge. Treatment with DEX from Days 4 through 10 significantly reduced established airway eosinophilia compared with animals sham-treated with saline from Days 4 -10 (120 +/- 29 eosinophils/microl BAL for DEX-treated mice versus 382 +/- 60 eosinophils/microl BAL for sham-treated animals, p < 0.01). DEX-treated mice also had dramatically reduced mucoid cell hyperplasia, and airway responsiveness returned to normal. In contrast, TRFK-5 given during the same time period reduced airway eosinophilia (86 +/- 32 eosinophils/microl BAL versus 382 +/- 60 eosinophils/microl BAL, p < 0.01) but did not reduce goblet cell hyperplasia or reverse already established AHR. Treatment with DEX but not TRFK-5 also inhibited interferon gamma (IFN-gamma) content of BAL fluid (0.49 +/- 0.09 ng/ml BAL fluid for DEX versus 1.50 +/- 0.24 ng/ml BAL fluid and 1.36 +/- 0.13 ng/ml BAL fluid for TRFK-5 and sham-treated mice, respectively, both p < 0.001 versus DEX). Thus, treatment with DEX reduces established eosinophilic airway inflammation and AHR in S. mansoni-sensitized and airway-challenged mice but treatment with TRFK-5 reversed established eosinophilia without ameliorating established AHR. Together, these data suggest that once airway inflammation develops, neutralizing the effects of IL-5 or reducing eosinophilia alone may not result in inhibiting established AHR in atopic asthma.
Lemierre's syndrome, oropharyngeal infections induced by anaerobic bacteria, leading to fatal septic thrombophlebitis of the internal jugular vein and pulmonary embolic abscesses in humans, was diagnosed in a 6-month-old, male, New Zealand White rabbit. After acute onset of anorexia, lethargy, and depression, the rabbit died suddenly despite emergency clinical care. Necropsy revealed swelling, necrosis, and abscess in the soft tissues around the left caudal mandibular ramus, oral mucosa, and molar teeth, with systemic embolic abscesses and necrosis, especially in the jugular vein, lungs, and brain. Histologic examination revealed necrosis and embolic abscesses with filamentous bacteria in the mandibular soft tissues, salivary gland, jugular veins, alveolar bone and marrow, periodontal tissues and dental pulp, oral and pharyngeal mucosa, lungs, brain, liver, myocardium, meninges, and small intestine. Bacterial culture of the mandibular abscess and heart blood yielded Fusobacterium necrophorum.
A case of epidermotropic lymphoma with systemic spread into lymph nodes and visceral organs was observed in a 7- to 8-month-old, female ICR mouse. The mouse developed progressive and generalized alopecia and lymphadenopathy of several weeks' duration. The affected skin was markedly and diffusely thickened, with multiple serous to hemorrhagic crusts, ulcerated plaques, and raised nodules. Microscopically, random and/or perivascular infiltration of pleomorphic lymphoid cells was present in the skin, lymph nodes, thymus, spleen, lungs, and liver. On cytologic examination, the lymphoid cells were similar in all affected tissues, and had hyperchromatic and irregularly oval, cleaved, and occasionally convoluted nuclei, approximately 6 to 9 mm in diameter. On immunohistochemical examination, most infiltrating cells were labeled with anti-CD3 (panT cell) antibody. A smaller proportion of the cells (, 5%) were labeled with anti-CD79a (panB cell) antibody.
Background. Insulin-dependent diabetes mellitus (IDDM) is the second most prevalent chronic illness of children. Investigation of the treatment of IDDM is hindered by the lack of a reproducible and easily maintained non-human primate model of this disorder. Methods. We induced IDDM in 11 juvenile cynomolgus monkeys after a single (150 mg/kg) intravenous injection of streptozotocin (STZ). All diabetic monkeys were treated with insulin twice daily, based on a sliding scale. Subcutaneous vascular access ports were surgically placed in each monkey to facilitate serial blood sampling and drug administration. Allogeneic pancreatic islet cells from unrelated donors were subsequently transplanted into the mesenteric circulation of all STZ-treated monkeys. Results. Mild, transient nausea and vomiting occurred in all animals after STZ injection; however, no additional signs of toxicity occurred. Within 36 hr, all monkeys required twice daily administration of exogenous insulin to maintain a non-ketotic state. Serum C-peptide levels decreased from >1.2 ng/ml before STZ, to between 0.0 and 0.9 ng/ml after STZ, confirming islet cell destruction. Animals were maintained in an insulin-dependent state for up to 147 days without any observable clinical complications. Subcutaneous vascular access port patency was maintained up to 136 days with a single incidence of local infection. Islet cell transplantation resulted in normoglycemia within 24 hr. Serum C-peptide levels increased (range: 2-8 ng/ml) for 6-8 days in immune competent animals, and for 39-98 days after transplant in immunosuppressed monkeys. Conclusions. IDDM can be consistently induced and safely treated in juvenile cynomolgus monkeys. Chronic vascular access can be maintained with minimal supervision and complications. This model is appropriate for studies investigating potential treatments for IDDM including islet cell transplantation.
Diseases of bulbourethral glands have rarely been reported in mice. The present report describes bulbourethral gland cysts in three male mice: a >1-year-old DBA/2J mouse (case 1), a 10-month-old nude mouse (case 2), and a 4.5-month-old chimeric mouse (case 3). Round to oval fluctuant nodules were observed on their dorsal perineum. The nodules in cases 1 and 2 were pale, pink to tan, soft, and glistening; in case 3, the mass was purple-red and soft. All nodules were located subcutaneously between the bulbospongiosus and ischiocavernous muscles, with a deep connection to the urethra. The nodules were 13 mm (case 1), 6 mm (case 2), and 8 mm (case 3) in diameter. The cut surface of the nodules was cystic; the lumina were filled with clear gelatinous or viscous (cases 1 and 2) or thin blood-tinged fluid (case 3), The nodules comprised irregular cysts and acini that were lined by a single layer of cuboidal, pyramidal to tall columnar epithelial cells with little stroma. Bulbourethral gland cysts should be included as differential diagnoses for perineal nodules or swellings in mice.
Renal cell carcinoma was diagnosed in 2 Siberian hamsters (Phodopus sungorus). One hamster had a unilateral solitary papillary-tubular carcinoma. The other hamster had a clear-cell, a papillary-tubular, and mixed types of carcinomas in the left kidney and a cystic-papillary-tubular type of carcinoma in the right kidney. In the papillary-tubular carcinoma. there were cuboidal to columnar cells arranged in tubular or papillary patterns supported by delicate fibrous stroma. Nuclei were located basally, often perpendicular to the subjacent basement membrane. Cytoplasm was pale to eosinophilic and variably abundant. Pleomorphic oval to polygonal cells found in the clear-cell carcinoma were arranged in broad trabeculae, lobules, or nests, with congested capillary stroma and multifocal necrosis. Cytoplasm varied from eosinophilic and granular to pale and clear or vacuolated. In the cystic-papillary-tubular carcinoma, cuboidal to columnar cells were arranged in cystic, papillary, or tubular patterns. Cysts varied in size and shape and contained secretory material and erythrocytes.
Complete T-cell activation requires two distinct signals, one delivered via the T-cell receptor, and the second "co-stimulatory" signal through CD28/B7 ligation. Previous studies showed that the blockade of CD28/B7 ligation alters differentiation of Th1/Th2 lymphocyte subsets in vitro and in vivo. The present study was designed to determine the effect of a CD28/B7 antagonist (CTLA4Ig) on Th1/Th2 development in Schistosoma mansoni-sensitized and airway-challenged mice. Treatment of mice with CTLA4Ig beginning 1 wk after sensitization abolished airway responsiveness to intravenous methacholine determined 96 h following antigen challenge. We also found a significant reduction in bronchoalveolar lavage (BAL) eosinophilia, and reduced peribronchial eosinophilic infiltration and mucoid-cell hyperplasia. Furthermore, CTLA4Ig treatment significantly decreased interleukin (IL)-4 and IL-5 content in BAL fluid in vivo, and the production of IL-5 by lung lymphocytes stimulated with soluble egg antigen (SEA) in vitro. In contrast, the content of interferon-gamma in BAL fluid and supernatant from SEA-stimulated lung lymphocytes from CTLA4Ig-treated mice was increased significantly compared with untreated animals. Thus, CTLA4Ig inhibits eosinophilic airway inflammation and airway hyperresponsiveness in S. mansoni-sensitized and airway-challenged mice, most likely due to attenuated secretion of Th2-type cytokines and increased secretion of Th1-type cytokines.
ABSTRACT Hepatitis, proliferative typhlitis, and colitis were characterized in young adult and older SCID/NCr mice naturally infected withHelicobacter hepaticus. Liver lesions consisted of Kupffer, Ito, and oval cell hyperplasia along with multifocal to coalescing coagulative hepatocyte necrosis. Numerous Warthin-Starry-positive bacteria were observed in the parenchyma, and there were minimal to mild accumulations of monocytic cells and neutrophils. Proliferative typhlitis was characterized by moderate to marked mucosal epithelial cell hyperplasia with mild monocytic and neutrophilic infiltration. Minimal to mild colitis with mucosal epithelial cell hyperplasia of the colon was most marked in older mice. Comparable gastrointestinal lesions were not observed in uninfected control SCID/NCr mice. H. hepaticus was cultured from fetal viscera of 2 of 11 pups sampled late in gestation from infected SCID/NCr females, suggesting transplacental infection of H. hepaticus. As expected, most of the naturally infected SCID/NCr mice had no serum immunoglobulin G response against H. hepaticus. These findings contrast with those in infected immunocompetent A/JCr mice, which develop a significant immune response to H. hepaticus associated with prominent multifocal mononuclear cell infiltrates in the liver, with only rare bacteria observable at the periphery of inflammatory foci or in the biliary canaliculi. The results demonstrate that chronic inflammatory and proliferative lesions simultaneously affecting the liver, cecum, and colon are associated with natural infection of SCID/NCr mice with H. hepaticus and that lesions are progressive with age. Concurrent infection with H. hepaticus may confound studies that have been attributed to similar lesions due to other experimental manipulations of SCID/NCr mice.
Objective-To determine incidence of neoplastic disease in ferrets.Design-Retrospective study.Animals-574 ferrets with neoplastic disease.Procedure-Medical records from the Veterinary Medical Data Base at Purdue University from 1968 to May 1997 were reviewed to identify ferrets with neoplastic disease. Data on tumor type, organ or system affected, sex, age, geographic location of affected ferrets, participating institution, and year of diagnosis were retrieved.Results-639 tumors of various types were diagnosed in 574 of 4,774 (12%) ferrets in the database. Sixty-one ferrets had multiple tumor types. Primary tumors were found in every system; endocrine (254; 39.7%), hemolymphatic (97; 15.2%), integumentary (83; 12.9%), and digestive (54; 8.4%) systems were most commonly affected. The most common tumor types were pancreatic islet cell (139; 21.7%) and adrenocortical cell (107; 16.7%) tumors and lymphoma (76; 11.9%). Most (94.2%) pancreatic islet cell tumors were functional. Age of affected ferrets ranged from less than 1 month to more than 15 years old. Tumor incidence was highest in ferrets between 4 and 7 years old. A sex predilection was not found, although tumors were found more commonly in spayed females and castrated males than in sexually intact females and males, respectively. Number of tumors diagnosed increased as the number of ferrets examined increased. Neoplastic disease accounted for an increasingly greater percentage of diseases diagnosed in ferrets during the study period.Clinical Implications-ferrets have an incidence and spectrum of neoplastic disease similar to other mammalian species.
The objective of this study was to determine whether 4-ipomeanol toxicosis in calves impairs alveolar macrophage functions important in pulmonary defense against infectious agents. Male Holstein calves were given either 4-ipomeanol (3 mg kg−1, i.v.) or vehicle (polyethylene glycol 400). Alveolar macrophages were recovered by pulmonary lavage 3 days later, and their capacities to phagocytose and kill E. coli, migrate toward zymosan-activated immune bovine serum, and produce interferon and interleukin-1 activity were evaluated in vitro. Alveolar macrophages recovered from 4-ipomeanol-treated calves had over a 70% decrease (p < 0.01) in chemotactic activity and over a 37% decrease (p < 0.005) in their capacity to phagocytose E. coli as compared to macrophages from control calves. Interleukin-1 activity in macrophages from 4-ipomeanol-treated calves tended to the higher than that from control calves, but the differences were not statistically significant (p = 0.06). 4-ipomeanol did not affect macrophage bactericidal activity or production of interferon. These results indicate that 4-ipomeanol suppresses select functions of alveolar macrophages in cattle that may be important in pulmonary defense against bacterial pathogens.
