Very small embryonic-like stem cells (VSELs) are a Sca-1(+)Lin(-)CD45(-) cell population that has been isolated from the bone marrow of mice. The similarities and differences between the mRNA profiles of VSELs and embryonic stem (ES) cells have not yet been defined. Here, we report the whole genome gene expression profile of VSELs and ES cells. We analyzed the global gene expression of VSELs and compared it with ES cells by microarray analysis. We observed that 9,521 genes are expressed in both VSELs and ES cells, 1,159 genes are expressed uniquely in VSELs, and 420 genes are expressed uniquely in ES cells. We found that although VSELs are similar to ES cells in their expression of genes associated with stem cell behavior and pluripotency, there are also differences in their mRNA expression. We further analyzed the expression of stem cell-associated genes in VSELs and ES cells, and found that there were differences in these genes. For instance, the Pkd2 and Yap1 gene were reduced in their expression in VSELs when compared with ES cells. But we also found Zfp54 gene expression was higher in VSELs compared with ES cells. More interestingly, we demonstrated that VSELs express c-kit, the stem cell factor (SCF) receptor. In vitro, SCF promoted VSEL differentiation into hepatic colonies in the presence of hepatocyte growth factor. In vivo, transplantation of VSELs directly into CCl4-induced injured livers significantly reduced serum ALT and AST levels. Ape1 is an important redox protein, essential for specific cytokine-induced signal transduction. Ape1 signaling is also important in regulating the growth of cancer cells, including colon cancer cells. The present study investigated whether Ape1 signaling plays a role in the regulation of colon cancer stem cell (CCSC) growth. The results showed that Ape1 was aberrantly expressed in CCSCs, as determined by quantitative (q)PCR assay. A laser confocal microscopy assay demonstrated that the Ape1 protein was mainly distributed in the nuclei, but not the cytoplasm, of the CSCs. Treatment of CCSCs with Ape1 redox inhibitor (E3330) significantly affected growth in vitro. In colon cancer xenograft mice, in vivo administration of E3330 enhanced tumor responses to the chemotherapeutic drug, 5-fluorouracil (5-FU). Furthermore, the combination of E3330 and 5-FU evidently increased the cytotoxicity of 5-FU in CSC growth. In the qPCR assay, the CCSCs were demonstrated to express the dominant ATP-binding cassette sub-family G member 2 (ABC-G2), but not the multidrug resistance 1, genes. Thus, we hypothesized that drug resistance in CCSCs is mediated by ABC-G2. Since CSCs are involved in cancer metastasis, the Ape1 inhibitor may be a potential agent in the inhibition of colon cancer growth and metastasis.

Objective The study investigated the clinical features of the elder cancer for the purpose of increasing the diagnostic rate in the early stage of lung cancer.Methods We compared the clinical materials of 86 elder lung cancer patients diagnosed by the cell pathologic examination with 22 non-elderly lung cancer patients.Results The pathogenetic age peak of the elder lung cancer was 65~75 years old(about 86.05% in the elderly lung cancer group).Smokers in the elder group account for 84.88%and 68.18% in the non-the elderly group.Those who had cough and hemoptysis accounted for 77.91%,45.35% respectively in the elderly group and 77.27%,54.55% in the non-the elderly group.Those who had chest pain accounted for 25.58% in the elderly group and 68.18% in the non-the elderly group respectively(P<0.05).The results of the cytopathologic examination showed that squamous carcinoma,positive rate was 62.79% in the elder group and 22.73% in the other group(P<0.05),and the positive rate of small cell carcinoma was 19.77% in the elder group and 59.09% in the non-the elder group(P<0.05).And the tumor distant metastasis rate was 36.05% in the elder group and 68.18% in the non-the elder group.Conclusion The old age,the smoking and the time were closely related with the lung cancer.In lung cancer the cough and hemoptysis were the main clinical manifestation.The incidence rate of the elder lung cancer was inferior to that of the non-the elder group obviously.Squamous carcinoma was the most common in the elder patients with distant metastasis.
Objective To explore clinical value of C-reactive protein(CRP) in acute aggressive period old patients with COPD. Methods 210 old patients in acute aggressive period with COPD were selected,who used antibiotic ,cortin or febrifuge and anodyne before. CRP,WBC,ESR and temperature were detected before and after controlling infection. Results The positive rate of CRP, WBC, ESR and temperature was 92. 38%, 27.62% ,48.57% and 46.67% respectively. The level of CRP is significant over WBC,ESR and temperature( P <0.01). The level of CRP were 58. 66±15.20 and 8.26±1.24 respectively at before and after controlling in-fection(P<0.01). The level of WBC were 8.69±2.60 and 5.34±1.48 respectively at before and after can-trolling infection(P<0.01). The level of ESR were 36.81±14.23 and 24.52±14.61 respectively at before and after controlling infection (P<0.01). The level of temperature were 37.29±0.78℃ and 36.26±0.37℃ respectively at before and after controlling infection (P<0.01). Conclusion CRP is a good marker in acute aggressive period old patients with COPD and it isn't disturbed by antibiotic,cortin or febrifuge and anodyne.
OBJECTIVE:To study the quality standard of Changsheng oral solution.METHODS:Radix Astragali,Radix Platycodi,Spica Prunellae and Radix et Rhizoma Glycyrrhizae in the formula were identified qualitatively by TLC.The content of Liquiritin in Changsheng oral solution was determined by HPLC.The chromatographic separation was performed on Nova-Pak C18 column(150 mm×4.6 mm,4 μm)and the mobile phase consisted of acetonitrile-2.5% acetic acid(13∶87)at a flow rate of 1.0 mL·min-1.The detective wavelength was set at 276 nm.RESULTS:Clear TLC spots were noted.The linear range of Liquiritin was 0.025 04~0.626 00 μg(r=0.999 9).The average recovery rate of Liquiritin was 98.66%(RSD=0.52%,n=6).CONCLUSION:The established standard can be used for the quality control of Changsheng oral solution.
Objective:To establish the methods for quality control of Yindong-liyan oral solution.Methods:Rhizoma Belamcandae,Radix Scrophulariae and Radix Et Rhizoma Sophorae Tonkinensis in Yindong-liyan oral solution were identified by TLC method.The content of chlorogenie acid in the solution was determined by HPLC method.Nova-Pak C_(18) column (150 min×4.6 mm,4μm) was used with the mixture of methanol-1.2% acetic acid (23:77) as the mobile phase.The flow rate was 1.0 mL/min and the detection was carried out at 327nm.Results:Rhizoma Belameandae,Radix Scrophulariae,Radix Et Rhizoma Sophorae Tonkinensis in Yindong-liyan oral solution could be detected by TLC method.The spots appeared sepa- rately in thin layer chromatograms at the corresponding positions of the control and sample solutions.The chtorogenic acid showed a good linear relationship at the range of 6.688-66.880μg/mL,and the regression equation was A=54959.21 c- 96 350.88 (r=0.9999).The average recovery was 98.66%,and the RSD was 1.66%(n=6).Conclusion:The methods are simple,sensitive and accurate,and can be used for quality control of Yindong-liyan oral solution.
Objective To research the quality analysis method of “Jiawei shenmaiyin Oral Solution”. Methods Radix et Rhizoma Glycyrrhizae and Fructus schisandrae chinensis were identified by TLC. The contents of Liquiritin in “Jiawei Shenmaiyin Oral Solution” was determined by HPLC. The chromatographic condition included Nova-Pak C18 column(4.6 mm×150 mm,4 μm) and the mobile phase consisting of acetonitrile -2.5% acetic acid(13:87). Detector wavelength was at 276 nm. A flow rate was 1.0 ml/min-1. Results Good relationship at the ranges of 0.0313 μg-0.3130 μg with correlation coefficient 0.9999. The average recovery of Liquiritin was 98.47%, and RSD was 1.10%. Conclusion It was proved that the method was reliable, simple, well-separated and precise, and thus could be used for quality control of “Jiawei Shenmaiyin Oral Solution”.
OBJECTIVE: To establish the quality standards for Yinlian oral solution. METHODS: TLC was adopted to identify Radix Scutellariae, Rhizoma Coptidis, Fructus Forsythiae in Yinlian oral solution. HPLC was applied to determine the content of Baicalin in Yinlian oral solution, using Nova- Pak C18( 150mm× 4. 6mm, 4μ m) as column, the mixture of methanol- 0. 3% phosphoric acid ( 40∶ 60) as mobile phase, with flow rate set at 1. 0mL· min- 1 and detection wavelength at 276 nm. RESULTS: Radix Scutellariae, Rhizoma Coptidis and Fructus Forsythiae were clearly detected with TLC. The content of baicalin showed a good linear relationship at a range of 1. 68~ 16. 8μ g· mL- 1( r=0. 999 8) . The average recovery was 99. 06% ( RSD=0. 79% ) . CONCLUSIONS: The standard is accurate, reliable, and can be used for quality control of Yinlian oral solution.

近年来,人工肝支持系统治疗肝病的研究发展较为迅速,血浆置换治疗是较成熟的肝脏替代疗法,它可提高肝功能衰竭病人的存活率,但对晚期重型肝炎病人的疗效仍不令人满意.
目的:以鸡爪为动物模型,研究不同分子量的透明质酸钠(Sodium hyaluronate product;以下简称SHP),对防止肌腱粘连的不同作用.方法:72只莱亨鸡分成实验组A 、B 、C和对照组D,每组又根据术后时间不同分为3周、5周、8周三组(n=6);实验手术斜坡形横断左足第Ⅲ趾屈趾深肌腱,鞘管内注射不同分子量的SHP,A组分子量为250万,B组为100万,C组为50万,对照组为生理盐水.结果:经生物力学测定和粘连范围病理组织学评分,实验组A、B与对照组D有显著性差异(P<0.01),而实验组C和对照组D无显著性差异(P≥0.01).结论:不同分子量的SHP防止肌腱粘连的作用不同;分子量大于100万的SHP具有防止粘连的作用;分子量为50万的SHP和生理盐水作用相似,不具有防止粘连的作用.
AIM:To establish a reversed-phase high pressure liquid chromatography(HPLC) method to determine the concentration of clindamycin phosphate and to investigate the stability of its injections. METHODS: Lichrosorb C 18 colum (150 mm × 4.6 mm, 5 μm )was used with the mobile phase of triethylamine chlorhydric acid solution (30mmol·L -1 ,pH=2.65)∶acetonitrile being 80∶20(V/V),flow rate of 1.0 mL·min -1 and detection wave lenth of 210 nm. RESULTS: The standard curve equation was y =597075c-62541.8( r =0.9997, n =5).The linear range was 0.4 - 1.6g·L -1 . The average recovery rate was 98.9 %.The intra day and inter day precision rate were less than 5 %.There was no difference between four products from different factories with respect to their stabilities to the light, whereas there was a few difference between the sample 1,4, and sample 2, 3, with respect to their stabilities to the heat. CONCLUSION: The RP-HPLC method was simple, rapid and sensitive. The clindamycin phosphate injection is stabilized to the light, but not stabilized to the heat.
OBJECTIVE:A HPLC method was established to determine the concentration of diclofenac sodium in transdermal absorption liquid.METHOD:A YWG-C18(10μm) colum was used with a mobile phase of CH3OH-H2O(including HAc 0.36%)(80∶20) at a flow rate of 1.0ml/min.UV detector was set at 280nm.The average recovery was 99.59% and the RSD were less than 3.28%.RESULTS:The No.1 and No.3 ointments had similar good effect on transdermal absorption(P>0.05) while the percutaneous release rate of No.1 was higher.The No.1 was absorbed more quickly than the No.2 and No.4(P<0.05).CONCLUSION:The No.1 ointment is valuable to be further developed.
