Ideological and political curriculum is a new teaching concept that has been widely promoted in colleges and universities in China in recent years. The core idea is to integrate ideological and political education into the teaching of various professional courses under the premise of "combining knowledge teaching, ability training and value shaping" as the curriculum goal to realize the concept of educating people in the whole process and curriculum. Taking the course of Veterinary Parasitology as an example, this paper explores and practices the ideological and political elements of the course. The echelon structure of the teaching team is reasonable with rich teaching experience and mature teaching methods. By deeply digging into the contents of professional courses, which contain many natural science spirits, humanities and social sciences connotation and socialist core values, ideological and political elements are integrated into the course contents and teaching practice in various ways. We explore the ideological and political excavation path and integration method of the curriculum,study and investigate the teaching effect of the ideological and political implementation of the curriculum, evaluate and reflect on the practical teaching integration effect of professional courses and ideological and political education, hoping to provide the theoretical support,empirical data and teaching experience for the ideological and political education reform of courses related to animal parasitology and veterinary disciplines, and provide a reference and a reference for the ideological and political education reform of other natural science courses.
为了解吉林市周边猪场弓形虫的感染情况及其基因型分型,试验于2019年10月份—2020年9月份在吉林市周边9个养猪场采集326份血清样本,利用ELISA方法进行血清中弓形虫抗体检测,计算猪场弓形虫阳性率.利用动物试验进行阳性虫株的分离并进行细胞培养,利用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法在SAG3、GRA6和L358 3个位点进行分离虫株的基因型初步鉴定.结果表明:吉林市周边猪场血清学检测弓形虫平均阳性率为28.31%;分离得到1株虫株,命名为JLpig1株.临床分离株JLpig1在位点GRA6处表现为基因Ⅰ型和基因Ⅱ型;在位点SAG3处表现为基因Ⅰ型和基因Ⅱ型;在位点L358处表现为基因Ⅱ型和China1型(Toxo DB#9株).初步判断吉林分离株JLpig1是强毒株或中毒株可能性较大,以基因Ⅱ型为主.说明吉林市周边猪场弓形虫病感染较为普遍,且感染率较高,应提高猪场人员对猪弓形虫病的认识,加强对猪场猪弓形虫病的防控.
小规模限制性在线课程(Small private online course,SPOC)教学模式可以将课程碎片化,让学生随时随地学习课程知识点,能有效地提高教师与学生的讨论与交流,解答线上学习遇到的疑点、难点,增加学生学习的主动性和课程参与度,提升学习效率和教学效果[1].SPOC是目前比较流行且实用的一种"线上+线下"混合式教学模式,可以解决大型开放式网络课程(MOOC)在高校实施过程中遇到的瓶颈,起到扬长避短的作用[2].
教学督导是教学质量的专业评价者,是提升人才培养质量的根本保障.督导不应流于形式,而要通过督导真正调动广大教师的教学积极性,深挖学校的内生动力,提高人才培养质量,实现高等教育内涵式发展.因此,加速督导体制机制的实质性改革势在必行且迫在眉睫.本文以吉林农业大学为例,针对目前高校教学督导普遍存在的问题进行剖析,并提出相应的教学督导改革措施,以期为其他相关高校提供参考和借鉴.
本试验于2019年7~10月对吉林市江南公园圈养的棕熊进行了寄生虫感染情况调查.采用直接涂片法、饱和蔗糖溶液漂浮法、离心沉淀法和锦纶筛兜淘洗法对棕熊的粪便进行检查,对检出的虫卵采用麦克马斯特氏法检测出每克粪便中的虫卵数(EPG);采用煤油浸泡法、皮屑溶解法对棕熊的脱落皮屑进行检查.通过试验,共检出4种寄生虫,分别为棕熊圆线虫、棕熊蛔虫、棕熊蛲虫和棕熊球虫,其中以棕熊圆线虫感染较为严重,感染率为50%;其次为棕熊蛲虫,感染率为31.25%;棕熊蛔虫的感染率为25%;棕熊球虫的感染率为18.75%.建议公园管理人员根据棕熊体内的寄生虫种类进行用药,加大驱虫力度,做好预防措施.
弓形虫是一种专性细胞内寄生的人兽共患寄生原虫,能够感染包括人、家畜、家禽及野生动物在内的几乎所有的温血动物,可引起严重的人兽共患弓形虫病.该病主要引起孕妇或孕畜流产、早产、死胎、畸胎甚至胎儿死亡.免疫功能低下或免疫缺陷者感染弓形虫是导致死亡的一个主要原因.弓形虫的基因分型和遗传多态性的发现是近些年弓形虫研究的重要进展,具有重要的生物学意义和临床意义.本文主要针对弓形虫基因分型进行归纳和总结,以期为今后弓形虫的基因型研究提供理论参考.
Metorchis orientalis belongs to the genus Metorchis of Opisthorchiidae, which mainly parasitizes in liver and bile ducts of waterfowl, causing liver dysfunction of the host. It has been reported that M. orientalis also infects humans. As a natural species in Australia and a popular ornamental animal, Black Swan (Cygnus atratus) has been imported into many countries. At present there has been no report of M. orientalis infection in Black Swan. In the present study M. orientalis infection in Black Swan was identified by a combination of different techniques, including morphological observation and molecular analysis. M. orientalis adults were found in the gallbladder and bile duct of a three-year-old female Black Swan, which was further confirmed by internal transcribed spacer (ITS) sequence analysis. In addition, the intermediate and definitive hosts of M. orientalis from the 'Qing' lake (a man-made lake in Changchun, China) that Black Swan lived were investigated and the infection route was preliminarily determined. Parafossarulus striatulus functioned as the first intermediate host which contained M. orientalis DNA, and fishes such as Pseudorasbora parva and Rhodeinae served as the second intermediate hosts with M. orientalis metacercariae in the fish flesh. M. orientalis eggs were found in the feces of three other Swans and six ducks that lived in the 'Qing' lake. This was the first reported case about M. orientalis infection of Black Swan. Our study described the course of the infection and provided new information about potential carriers and disseminators of M. orientalis.
To study the localization and characteristics of the recombinant antigens of Clonorchis sinensis,two amino acids highly repetitive sequences were obtained by construction and screening of a cDNAexpression libraries of C.sinensis etacercaria and adult worms.Then we synthesized peptides with highly repetitive gene sequences and coupling with the carrier protein.Animals were immunized and sera was collected.Then antibodies were purified by ammonium sulfate precipitation and affinity chromatography.At last antigenic genes were located by immunofluorescence technique.In results,recombinant antigens CsSQ1 and CsSQ2 of C.sinensis mainly distributed in oral sucker and ventral suckers,respectively.The localization of the recombinant antigens of C.sinensis was confirmed.We speculate that the recombinant antigens may be related to the immune adhesion of C.sinensis.It provided the reference for the applied research in diagnosis and immunological characteristics of antigens of C.sinensis in the future.
本文针对现行《动物寄生虫病学》课程教学中存在的不足,结合应用型人才培养目标,改变传统的授课模式,对该课程进行教学手段、 教学内容及考核方式等方面的改革探索与实践,以进一步激发学生学习该门课程的积极性和主动性,培养学生自学能力、总结分析问题能力.
笔者于2018年6月接诊吉林市某养猪场发生小规模母猪流产、死胎症状的病例,经过临床观察、病理剖检及实验室检验等检测方法进行诊断,确诊该猪群感染病为弓形虫急性病例,并对其进行治疗控制了病情,该病的诊治过程介绍如下.
华支睾吸虫病是由华支睾吸虫引起的危害严重的鱼源性人兽共患寄生虫病,主要引起人和多种动物的肝胆病变,严重者可引起胆管上皮癌,因此华支睾吸虫被国际癌症署(IARC)归为第Ⅰ类生物性致癌因子.我国华支睾吸虫病主要流行于淡水资源丰富的地区,在我国27个省、市、自治区均有该病人体感染的报道.近年来,我国华支睾吸虫感染率呈上升趋势,且受威胁地区和人数不断增加.对该病的诊断和防控已刻不容缓,传统的病原学诊断方法存在着一定的局限性和弊端.免疫学方法和分子生物学技术的不断发展为该病的诊断提供了新的途径,今后在华支睾吸虫病的基础研究与防控中起到更加重要的作用.论文就华支睾吸虫病的流行特点及诊断方法研究进展进行综述,以期为华支睾吸虫病的研究提供参考.
对吉林农业科技学院动物科技学院转型发展阶段中的教学实践和经验进行总结,并对动物寄生虫病学课程的教学模式和方法进行探索,以期为转型背景下该课程的建设、兽医学科的发展及应用型人才的培养提供建议和参考.
为了对吉林某土鸡养殖场发生大规模的肿瘤性疾病进行诊断,经过流行病学调查、临床症状检查、病理剖检及组织学观察,初步诊断为禽白血病和鸡马立克病;进行实验室鉴别诊断,设计特异性引物,利用PCR方法,经过BLAST序列比对,实验结果确定该病为鸡的马立克病,而且同时表现出4种混合型的临床症状.
为了研究三氯生能否引起细胞自噬并探讨其可能的诱导通路,揭示三氯生作为抗菌药物的新作用机制,利用免疫荧光、菌落计数、免疫印迹等技术检测经三氯生处理过细胞的自噬水平、相关蛋白表达量的变化及杀菌能力的改变.结果表明:三氯生能引起Hela细胞和Raw264.7细胞的自噬,并且该自噬为完全自噬.三氯生诱导的小鼠巨噬细胞系Raw264.7细胞自噬依赖的是胞外信号调节激酶(Extracellular signal-regulatedkinase,ERK)途径,而非经典的雷帕霉素靶蛋白(Mammalian target of rapamycin,mTOR)途径.此外,三氯生通过该作用机制加强了巨噬细胞对胞内菌的杀伤作用.研究表明,三氯生诱导自噬的现象在细胞中普遍存在,自噬在对抗病原微生物中起到重要作用,诱导自噬是三氯生作为抗菌药物的新作用机制.
采用结晶紫染色检测结核分枝杆菌生物被膜的形成能力,实时荧光定量PCR(qRT-PCR)检测生物被膜态的不同结核分枝杆菌Rv3519表达水平,分析Rv3519基因表达与结核分枝杆菌生物被膜形成能力的关系.结果表明:临床分离株产膜能力较H37Rv明显偏低,差异显著(P<0.01);临床分离株之间生物被膜产量差异不显著(P>0.05);生物被膜态的分离株较标准株Rv3519基因表达量偏低,差异显著(P<0.01).说明结核分枝杆菌生物被膜形成能力与Rv3519基因表达水平相关,Rv3519基因可以作为结核分枝杆菌生物被膜形成能力的潜在分子诊断标志.
To investigate the differences of small intestine villi histology in different varieties of beef cattles,The histological observations of small intestine villi in four varieties of beef calves were used by paraffin sections and H.E.routine staining methods,the images were processed and analysed with the Image-Pro-Plus software and Statistics in Medicine SPASS 16.0.The results showed the villus length of small intestine of Charolais cattle was the longest,and there was no significant difference compared with Simmental cattle(P0.05),there were highly significant differences compared with Red Steppe and Limousine cattle(P0.01).The villus width of small intestine of Red Steppe was the widest,there were no significant differences compared with Charolais cattle and Simmental cattle(P0.05),and there was a highly significant difference compared with Limousine cattle(P0.01).The crypt depth of small intestine of Red Steppe was the deepest,there were highly significant differences compared with others(P0.01).The ratio of villous length to crypt depth(V/C) for Charolais cattle was the biggest,there were no significant differences compared with Limousine cattle and Simmental cattle(P0.05),but there was a highly significant difference compared with Red Steppe(P0.01).It displayed that there were different degrees of differences in villi structure of small intestine mucosa during the sexual maturation for different varieties of beef cattle,indicating existence of differences on absorption capacity,growth rate,feed efficiency and other performance indicators.
To investigate the differences of small intestine histology in different varieties of beef cattle,samples of small intestine jejunum in adult Grassland Red,Limousine and Charolais beef cattle were taken out and embedded in paraffin sections.Sections were stained by HE conventional methods and the images were processed and analysed with the Image-Pro-Plus software and Statistics in Medicine SPASS16.0.The results showed the difference of the Villus length of small intestine between Red Steppe and Limousine cattle was significant(P0.01),between Red Steppe and Charolais cattle was not significant(P0.05),between Limousine cattle and Charolais cattle was significant(P0.01);The difference of the Crypt depth and the Mucous depth of small intestine among the 3 varieties was significant(P0.01);The difference of V/C value between Red Steppe and Limousine cattle was significant(P0.01),between Red Steppe and Charolais cattle was significant(P0.01),between Limousine cattle and Charolais cattle was not significant(P0.05);The difference of Submucous muscle layer depth between Red Steppe and Limousine cattle was significant(P0.01),between Red Steppe and Charolais cattle was not significant(P0.05),between Limousine cattle and Charolais cattle was significant(P0.01);It indicated that there were significant differences of small intestine histological among different varieties of beef cattle which have an effect on Growth rate and Feed efficiency of performance indicator.
T and B lymphocytes from blood of different species of animals were stimulated by purified soybean agglutinin(SBA),and detected through MTT method,while compared with ConA and LPS groups,so the effect of SBA on immune function of different species of animals could be compared.The results showed that the SI of T and B lymphocytes in dog,chicken and rabbit were significantly lower than corresponding ConA and LPS groups(P 0.05).It was lower in pig and higher in sheep than in control group,but both had no significant difference(P 0.05).The SI of T and B lymphocytes stimulated by SBA in different species of animals were different,and the dominant feature was that chicken were the highest and sheep were the lowest.In the same species of animals,the effects of proliferation function of T and B lymphocytes were different,the SI of T lymphocytes were higher than that of B lymphocytes in rabbit,dog,and pig,but it was just against in sheep and chicken.These results indicated that the effects of proliferation function of T and B lymphocytes stimulated by SBA in different species of animals exist,especially in dog,rabbit and chicken.Except sheep,the proliferation function of T and B lymphocytes from other species of animals were more or less inhibited.It showed that the immunocytes from different species of animals have different glycosylation mode,cytodifferentiation process and signal transmission mechanism.
Arbor Acres breeders,at 75 days old,fed the diet containing 20 % raw soybean to investigate the binding rule of soybean agglutinin to the small intestinal epithelia. After two weeks,samples of small intestinal tissue were taken out containing duodenum,front-jejunum,middle-jejunum,behind-jejunumand ileum,and embedded in paraffin. Sections were stained by the SP immunohistochemistry to analyze the distribution of soybean agglutinin with the Image-ProPlus software. The results showed that the average optical values of soybean agglutinin binding with duodenum and front-jejunum were significantly higher than that of middle jejunum,behind-jejunum and ileum in the different intestine segments (P0.05). Binding with intestinal vills was significantly higher than that of crypts and intestinal wall in the different sites of epthelia (P0.05). It indicated that glycosylation pattern is different among epithelial of different intestine in broiler.
