Objectives Threatened abortion (TA) is a common condition in pregnant women. The role of CARMN in the onset and progression of TA is increasingly recognized as significant. However, the precise mechanisms of its influence are not yet fully understood. This study was to investigate the clinical value of CARMN in TA patients and the mechanism of action that may influence TA.Methods This study included 105 patients aged 22-38 years old, 6-8 weeks pregnant and with early TA of single pregnancy. In addition, 63 healthy women of the same age group who were 6-8 weeks pregnant were included as controls. qRT-PCR was employed to assess the expression levels of CARMN and miR-515-5p. The clinical diagnostic utility of CARMN in patients with TA and the predictive value of CARMN in pregnancy outcome of TA patients were analyzed using ROC curve methodology. Cell viability in HTR-8/SVneo cells was measured utilizing a cell counting kit-8 (CCK-8) kit. Apoptosis rates were quantified through flow cytometry. Cell migration was investigated using a transwell assay.Results The level of serum CARMN decreased and miR-515-5p increased in patients with TA. CARMN could predict pregnancy outcomes in patients with TA. Functionally, CARMN enhanced the viability and migration of HTR-8/SVneo cells by regulating the expression of miR-515-5p, while inhibiting apoptosis.Conclusions LncRNA CARMN may serve as a diagnostic marker for TA and participate in the progression of TA by regulating the expression of miR-515-5p.
目的:观察疏调平衡针刺法对慢性失眠伴焦虑、抑郁患者的临床疗效.方法:选择符合纳入标准的249例慢性失眠伴焦虑、抑郁患者依据随机数字表随机分为疏调平衡针刺组、传统针刺组和药物对照组,在治疗及随访过程中共脱失患者16例,最终疏调平衡针刺组纳入80例、传统针刺组纳入78例、药物对照组纳入75例.分别予以疏调平衡辨证取穴针刺、传统辨证取穴针刺、口服艾司唑仑片治疗,治疗时间均为4周.并于治疗前、治疗完成时及治疗结束后1个月比较3组患者匹兹堡睡眠质量指数(PSQI)、焦虑自评指数(SAS)及抑郁自评指数(SDS)得分的变化情况,并对比治疗完时和治疗结束后1个月的疗效.结果:①与治疗前比较,治疗完成时3组的PSQI总分均低于治疗前(P<0.01),疏调平衡针刺组改善最明显;②治疗完成时比较,疏调平衡针刺组与传统针刺组疗效优于药物对照组(P<0.05);③与治疗前比较,疏调平衡针刺组与传统针刺组SAS、SDS指数改善明显(P<0.01),药物对照组治疗前后无显著变化(P>0.05).结论:疏调平衡针刺法治疗慢性失眠伴焦虑、抑郁疗效显著,伴随的焦虑、抑郁程度也随之改善,不良反应少,具有较好的临床疗效.
Recently, Toll-like receptors (TLRs) have been extensively studied in radiation damage, but the inherent defects of high toxicity and low efficacy of most TLR ligands limit their further clinical transformation. CRX-527, as a TLR4 ligand, has rarely been reported to protect against radiation. We demonstrated that CRX-527 was safer than LPS at the same dose in vivo and had almost no toxic effect in vitro. Administration of CRX-527 improved the survival rate of total body irradiation (TBI) to 100% in wild-type mice but not in TLR4-/- mice. After TBI, hematopoietic system damage was significantly alleviated, and the recovery period was accelerated in CRX-527-treated mice. Moreover, CRX-527 induced differentiation of HSCs and the stimulation of CRX-527 significantly increased the proportion and number of LSK cells and promoted their differentiation into macrophages, activating immune defense. Furthermore, we proposed an immune defense role for hematopoietic differentiation in the protection against intestinal radiation damage, and confirmed that macrophages invaded the intestines through peripheral blood to protect them from radiation damage. Meanwhile, CRX-527 maintained intestinal function and homeostasis, promoted the regeneration of intestinal stem cells, and protected intestinal injury from lethal dose irradiation. Furthermore, After the use of mice, we found that CRX-527 had no significant protective effect on the hematopoietic and intestinal systems of irradiated TLR4-/- mice. in conclusion, CRX-527 induced differentiation of HSCs protecting the intestinal epithelium from radiation damage.
目的:探讨补肾解毒方对放射致感染性肺损伤的防护作用.方法:选取72只C57BL/1OJ小鼠,随机分为空白对照组、单纯照射组、氨磷汀组、中药组,每组18只,建立各组小鼠模型,在实验开始后第11天,空白对照组小鼠不接受照射,余各组小鼠均接受一次性6 Gy 60Coγ射线全身照射,其中氨磷汀组在照射前30 min内注射氨磷汀.并于照射后第1天、第7天、第14天每组随机抽取6只小鼠进行取材,计算肺脏系数,采用酶联免疫吸附实验(ELISA)检测降钙素原(PCT)、C-反应蛋白(CRP)、白细胞介素-6(IL-6),并在光学显微镜下观察小鼠肺组织病理形态学变化.结果:照射后第1天,空白对照组肺系数水平低于余3组(P<0.05),同时单纯照射组高于氨磷汀组和中药组(P<0.05);照射后第7天,空白对照组肺系数水平低于单纯照射组和氨磷汀组(P<0.05),同时单纯照射组高于氨磷汀组和中药组(P<0.05);照射后第14天,单纯照射组肺系数水平高于余3组(P<0.05).单纯照射组病理形态学变化最明显,氨磷汀组和中药组可明显减轻肺组织病理变化,其中中药组优于氨磷汀组.照射后第1天,空白对照组PCT、CRP、IL-6水平显著低于余3组(P<0.05),同时单纯照射组CRP、IL-6水平高于氨磷汀组与中药组(P<0.05);照射后第7天,PCT、CRP、IL-6水平中,空白对照组低于余3组(P<0.05),单纯照射组高于氨磷汀组和中药组(P<0.05),同时中药组低于氨磷汀组(P<0.05);照射后第14天,空白对照组PCT、IL-6水平低于单纯照射组和氨磷汀组(P<0.05),单纯照射组高于氨磷汀组和中药组(P<0.05),中药组低于氨磷汀组(P<0.05),单纯照射组CRP水平高于余3组(P<0.05).结论:补肾解毒方能够减轻放射致感染性肺损伤的感染程度,降低PCT、CRP水平,发挥抗炎抗损伤作用.
Objective To explore and analyze the application effect of fine nursing for neonates with jaundice. Methods 65 cases of children with jaundice who were admitted to our hospital from March 2020 to May 2020 and From January 2021 to April 2021 were selected as the subjects and were divided into routine group and observation group by simple random grouping method, with 32 cases in each group. Routine nursing was used in routine group. Observation group carried out fine nursing. The nursing quality, yellowing time, serum bilirubin, adverse reactions and nursing satisfaction of the two groups were analyzed and evaluated. Results After nursing, life nursing quality score, psychological nursing quality score, health education score and treatment nursing quality score of the observation group were significantly higher than those of the conventional group (P<0.05). Serum bilirubin, jaundice yellowing time and adverse reaction rate in the observation group were significantly better than those in the conventional group (P<0.05). Nursing satisfaction in the observation group was significantly higher than that in the conventional group (P<0.05). Conclusion Fine nursing can effectively improve nursing quality, shorten the time of jaundice yellowing, reduce the level of serum bilirubin and adverse reactions, improve nursing satisfaction, with clinical application value is significant.
Objective:To explore the effect of a Chinese medicinal composition ( Xiadanqi) on the prevention of radon exposure induced injuries of lung in vitro and in vivo. Methods:Mice were randomly divided into three groups of blank control group, radon-exposed group alone and radon-exposed group intervened with Chinese medicinal composition. The pathological changes of lung tissues in each group after 120 WLM were observed by HE and Masson staining, and the expressions of α-SMA protein and Vimentin protein in lung tissues were detected by immunohistochemistry staining. The levels of oxidative stress in lung tissue of each group were detected with SOD and MDA kits. At the same time, a radon exposed cell model and a radon exposure + Xiadanqi intervention cell model were constructed using an ecological radon chamber. The cell adhesion abilities of different groups were detected by an adhesion kit. The cell migration ability of each group was determined by the transwell migration experiment. The expression of E-cadherin and Vimentin protein was detected by Western blot. Results:Compared with the radon exposure group, the concentration of MDA was decreased ( t=4.43, P<0.05), the activity of SOD was increased ( t=3.22, P<0.05), and α-SMA and Vimentin protein expressions were decreased ( t=3.08, 7.57, P<0.05) in lung tissue of mice intervened with 2 mg/g Xiadanqi. In vitro, compared with radon exposure group, the migration ability was reduced ( t=4.78, 13.01, P<0.05), the cell adhesion property was enhanced ( t=3.41, 12.55, P<0.05), the expression of E-cadherin protein was increased ( t=2.96, 19.57, P<0.05), and the expression of Vimentin protein was obviously reduced ( t=21.00, 33.32, P<0.05) in radon-exposed cells with the treatment of Chinese medicine (150 μg/ml and 200 μg/ml). Conclusions:The Chinese medicinal composition ( Xiadanqi) has a certain radioprotective effect on radon exposure induced injury by reducing oxidative stress, attenuating EMT and fibrosis, and thus it may be applied as a protective agent for radon induced injury.
目的:探讨补肾解毒方辐射防护作用的靶点及其作用机制,为补肾解毒方作为防治辐射损伤药物使用提供实验基础.方法:将138只TLR4+/+小鼠随机分为TLR4+/+空白对照组18只、TLR4+/+辐射模型组60只、TLR4+/+补肾解毒方组60只;将138只TLR4-/-小鼠随机分为TLR4-/-空白对照组18只、TLR4-/-辐射模型组60只、TLR4-/-补肾解毒方组60只.空白对照组、辐射模型组给予生理盐水灌胃,补肾解毒方组予中药灌胃,并于第11天对辐射模型组与补肾解毒方组进行6Gy全身照射.检测并比较各组CD14、TLR4受体、MD2和CD4+CD25+Treg细胞水平.结果:CD14检测结果显示辐射后第14天,TLR4+/+辐射模型组与补肾解毒方组均高于空白对照组,且补肾解毒方组高于辐射模型组(P<0.01).辐射后第30天,两组CD14水平下降,TLR4+/+补肾解毒方组高于辐射模型组和空白对照组(P<0.01).CD4+CD25+Treg检测显示辐射后第14天,TLR4+/+补肾解毒方组高于辐射模型组和空白对照组(P<0.01);辐射后第30天,TLR4+/+补肾解毒方组高于空白对照组(P<0.01).上述指标同时间点的TLR4-/-补肾解毒方组与辐射模型组比较,差异无统计学意义(P>0.05).结论:补肾解毒方可通过TLR4信号通路发挥辐射损伤防护作用.
目的 观察补肾解毒方对核辐射孕鼠胎盘抗氧化能力的影响.方法 将60只C57BL/6J孕鼠随机分为空白对照组、辐射模型组、补肾解毒方组,每组20只,补肾解毒方组给予补肾解毒方13.5 g/(kg·d)灌胃,其他两组给予0.2 mL生理盐水灌胃,各组均灌胃21 d.辐射模型组、补肾解毒方组两组小鼠于灌胃第11天接受一次性4Gy 60Coγ-射线的全身照射.于辐射后第1天、第10天各取10只小鼠剖腹取胎盘组织检测胎盘组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)含量.结果 MDA含量:辐射后第10天,辐射组与空白组比较,孕鼠胎盘MDA含量明显升高(P<0.01),中药组与辐射组及辐射后1天比较孕鼠胎盘MDA含量下降有统计学意义(P<0.01);SOD活性(U/L):辐射后第10天,辐射组、中药组与空白组比较,孕鼠胎盘SOD活性差异有统计学意义(P<0.05),中药组与辐射组比较孕鼠胎盘SOD活性差异显著(P<0.01);中药组辐射后10天较辐射后1天孕鼠胎盘SOD活性差异无统计学意义(P>0.05);GSH-Px活性:辐射后第10天,辐射组、中药组与空白组比较,孕鼠胎盘GSH-Px活性差异有显著统计学意义(P<0.01),中药组与辐射组比较孕鼠胎盘GSH-Px活性差异有统计学意义(P<0.05);中药组辐射后10天较辐射后1天孕鼠胎盘GSH-Px活性升高有统计学意义(P<0.05).结论 补肾解毒方能改善胎盘组织电离辐射所致氧化损伤.
目的 基于代谢组学初步探究补肾解毒方对辐射损伤大鼠的防护作用机制.方法 将60只雄性Wistar大鼠随机分为假照射组、2 Gy辐射模型组、4 Gy辐射模型组、2 Gy补肾解毒方干预组和4 Gy补肾解毒方干预组,每组12只,假照射组和辐射模型组给予生理盐水灌胃,补肾解毒方干预组给予补肾解毒方灌胃[组方生药等效剂量9.45 g/(kg·d)],连续灌胃10 d.第11天,除假照射组外,其余组均进行相应剂量的γ射线全身辐射,辐射后24 h和72 h分别收集各组大鼠血清,高效液相色谱串联质谱靶向检测大鼠血清中氨基酸和酰基肉碱的含量.结果 与假照射组相比,辐射大鼠血清代谢轮廓明显改变,血清中赖氨酸含量明显升高(P<0.01),瓜氨酸、谷氨酸、组氨酸、脯氨酸含量明显降低(P<0.01),十四碳烯酰肉碱和苏氨酸水平先降低后升高(P<0.01).与辐射模型组相比,补肾解毒方可改善辐射大鼠血清代谢轮廓,明显回调辐射大鼠血清中瓜氨酸、谷氨酸、组氨酸、赖氨酸、脯氨酸、苏氨酸、十四碳烯酰肉碱含量(P<0.01或P<0.05),改善精氨酸生物合成、精氨酸和脯氨酸代谢、组氨酸代谢、D-谷氨酰胺和D-谷氨酸代谢等途径.结论 补肾解毒方可能通过回调辐射大鼠血清中部分氨基酸和酰基肉碱含量,改善相关代谢通路,发挥辐射防护作用.
目的 运用数据挖掘的方法分析研究清代京城名医孔伯华治疗眩晕的用药与治则.方法 收集整理孔伯华相关著作中治疗眩晕的医案.运用Microsoft Excel 2016进行中药频数统计,利用SPSS 21.0软件进行因子分析.结果 共筛选出139首方剂,含355味中药.使用频数排名前五味的中药为,眩晕病:旋覆花、石决明、知母、代赭石、黄柏,体现出眩晕多为肝火上逆,通过聚类分析得,眩晕病分为4类,通过因子分析提取出6个公因子.结论 孔老在治疗眩晕时:采用降逆,清肝,升清之法.
目的 探究中、低剂量辐射对小鼠胎盘结构的影响及补肾解毒方对胎盘损伤的防护作用.方法 选取受孕后孕鼠25只,随机分为5组,分别为空白对照组,1 Gy辐射组,4 Gy辐射组,1Gy辐射+中药组,4 Gy辐射+中药组,自受孕第1天开始每日分别予1 Gy辐射组,4 Gy辐射组组生理盐水灌胃,1 Gy辐射+中药组,4 Gy辐射+中药组补肾解毒方溶液灌胃,10 d后分别予1 Gy、4 Gy60Co-γ射线照射,照射后仍以以上方法灌胃10 d,剖腹取出仔鼠及胎盘,称重胎盘,电镜观察胎盘切片.结果 与空白对照组相比,1 Gy辐射组和1 Gy辐射+中药组仔鼠胎盘组织损伤轻微,胎盘重量及胎盘结构改变均不明显;4 Gy辐射组胎盘重量较空白对照组明显降低(P<0.05),胎盘结构疏松紊乱,主要可见血管管腔扩大,内有血凝块,血管内红细胞明显减少,组织间有淤血现象,纤维素样坏死等表现;4 Gy辐射+中药组胎盘重量较空白对照组亦明显降低(P<0.05),但较单纯4 Gy辐射组有所增加(JP<0.05),胎盘血管组织结构尚清晰,血管扩张不明显.结论 中高剂量辐射会导致胎盘重量减低和组织损伤,尤其血管损伤明显,补肾解毒方对中高剂量辐射具有较好的防护作用.
[目的]探讨补肾解毒方对小鼠放射损伤的防护作用.[方法]将72只小鼠随机分为空白对照组、单纯放射组、放射+中药组,每组24只.饲养3 d后开始给药,放射+中药组给予补肾解毒方汤剂灌胃,空白对照组和单纯放射组给予等体积生理盐水灌胃.在实验开始后第11天,除空白对照组外所有小鼠均接受一次性4 Gy 60Coγ射线全身照射,照射后各组每日继续灌胃至实验结束,方法同前.共灌胃25 d.并于照射后第1、7、14天,应用酶联免疫吸附分析(ELISA)检测血清白细胞介素6(IL-6)、白细胞介素10(IL-10)水平,应用全自动生化仪检测外周血白细胞数.[结果]照射后第1天,单纯放射组血清IL-6水平高于空白对照组(P<0.01),IL-10水平组间无差异(P>0.05),单纯放射组和放射+中药组白细胞水平低于空白对照组(P<0.01),而放射+中药组白细胞水平高于单纯放射组(P<0.01);照射后第7天,单纯放射组血清IL-6水平高于空白对照组和放射+中药组(均P<0.01),放射+中药组血清IL-10水平高于空白对照组(P<0.01),单纯放射组和放射+中药组白细胞水平低于空白对照组(P<0.01),而放射+中药组白细胞水平高于单纯放射组(P<0.05);照射后第14天,单纯放射组IL-6水平高于空白对照组和放射+中药组(均P<0.01),IL-10水平组间无差异(P>0.05),单纯放射组白细胞水平低于空白对照组(P<0.01).[结论]补肾解毒方可通过下调血清IL-6水平,提升血清IL-10水平,并减轻放射所致的白细胞数减少,发挥抗炎抗损伤作用,提高放射后小鼠的免疫力.
目的 探讨自拟中药补肾解毒方对小鼠胸腺、脾脏、白细胞及淋巴细胞的影响.方法 将90只清洁级小鼠随机分为空白对照组、辐射模型组、氨磷汀阳性对照组、补肾解毒方预防组、补肾解毒方防治结合组.造模前各组分别进行相应处理.除空白对照组外,其余4组小鼠均接受剂量为6 Gy的60Co γ射线的辐射;并于辐射后第1天、第7天、第14天分批处死小鼠,记录小鼠体质量及胸腺、脾脏质量,计算脾脏指数和胸腺指数;眼球采血并采用全自动生化仪测定外周血白细胞、淋巴细胞总数.结果 (1)辐射后第1天,与空白对照组相比,各组小鼠胸腺指数、脾脏指数均明显下降(P<0.01);辐射后第7天,补肾解毒方预防组和补肾解毒方防治结合组胸腺指数明显高于辐射模型组(P<0.05或P<0.01);辐射后第14天,补肾解毒方预防组、补肾解毒方防治结合组胸腺指数、脾脏指数高于氨磷汀阳性对照组和辐射模型组(P<0.01或P<0.05).(2)辐射后第1天,与空白对照组相比,各组小鼠白细胞、淋巴细胞均明显下降(P<0.01);辐射后第7天,补肾解毒方预防组和补肾解毒方防治结合组淋巴细胞数量明显高于氨磷汀阳性对照组(P<0.05);辐射后第14天,补肾解毒方预防组、补肾解毒方防治结合组小鼠白细胞、淋巴细胞明显高于辐射模型组和氨磷汀阳性对照组(P<0.01或P<0.05).结论 补肾解毒方能够促进辐射后胸腺和脾脏的恢复,提高外周血白细胞和淋巴细胞总数,具有良好的辐射防护作用.
目的 研究氨磷汀对急性放射性肠炎小鼠小肠组织中血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)和磷脂酰肌醇-3-激酶(PI3K)/蛋白激酶B(PKB)通路mRNA表达的影响及生物学意义.方法 将小鼠分成对照组、辐射组及干预组.采用6 Gy 60Coγ射线全身辐射,在第1、7及14天分批处死小鼠观察病理改变.qRT-PCR检测不同时间点对照组、辐射组及干预组VEGF、bFGF和PI3K/PKB通路mRNA相对表达量.结果 辐射后小肠黏膜下层微血管损伤明显.辐射组辐射后第1天小鼠小肠组织中VEGF、bFGF和PI3K/PKB通路mRNA相对表达量升高(P<0.05),第7天各mRNA均呈下降趋势但仍高于对照组(P<0.05),第14天VEGF、bFGF mRNA相对表达量仍高于对照组(P<0.05);而PI3K、PKB mRNA相对表达量降至对照组水平(P>0.05).干预组辐射后第1天各mRNA均升高(P<0.05),第7和14天各mRNA相对表达量与对照组比较,差异无统计学意义(P>0.05).干预组与辐射组各基因不同时间点的mRNA相对表达量有差异,干预组mRNA相对表达量较低,变化趋势有差异(P<0.05).结论 氨磷汀能下调急性放射性肠炎早期VEGF、bFGF和PI3K/PKB通路mRNA的相对表达量,对辐射后微血管损伤具有一定的保护作用.
目的:探讨补肾解毒方对低剂量辐射后孕鼠妊娠结局的影响,并评价其对低剂量辐射后孕鼠胎盘氧化损伤的防护作用.方法:将60只合笼交配受孕后的C57BL/6J小鼠随机分为空白对照组、辐射组、中药治疗组.予中药治疗组补肾解毒方汤灌胃,予空白对照组、辐射组等容积生理盐水灌胃,连续10 d,于第11天对辐射组及中药治疗组采用2 Gy 60 Coγ-射线照射,后继续灌胃至临产前.观察孕鼠孕产结局(窝仔数量、早产率、流产率、死胎率及仔鼠的一般情况),计算胎盘指数,检测胎盘组织中超氧化物歧化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-Px)活力及丙二醇(MDA)含量.结果:辐射后第1天,辐射组及中药治疗组孕鼠体质量与空白对照组相比下降明显(P<0.01),辐射后第10天,中药治疗组孕鼠体质量明显上升(P<0.01),但仍低于空白对照组(P<0.05),辐射组体质量明显低于空白对照组和中药治疗组(P<0.01,P<0.05);辐射组和中药治疗组死胎率、吸收胎率均高于空白对照组(P<0.05),活胎率、胎盘指数、仔鼠体质量、仔鼠身长均低于空白对照组(P<0.05);中药治疗组死胎率、吸收胎率均低于辐射组(P<0.05),活胎率、胎盘指数、仔鼠体质量、仔鼠身长均高于辐射组(P<0.05);相比空白对照组,辐射组和中药治疗组胎盘组织的SOD活力、GSH-Px活力都明显降低(P<0.05),中药治疗组SOD、GSH-Px活性高于辐射组(P<0.05),MDA含量低于辐射组(P<0.05).结论:补肾解毒方对低剂量辐射后孕鼠的胎盘组织氧化损伤具有防护作用并影响其妊娠的结局.
目的:探讨补肾解毒方对辐射后小鼠血清白细胞介素-12(IL-12)、干扰素-γ(IFN-γ)分泌水平的影响.方法:将90只小鼠随机分为空白组、辐射组、氨磷汀组、补肾解毒方防治结合组和补肾解毒方预防组,每组18只,并建立各组小鼠模型.在实验开始后第11天,除空白组外所有小鼠均接受一次性4Gy 60 Co γ射线全身辐照,并于辐射后第1天、第7天、第14天每组随机抽取6只进行眼球取血,分离血清,应用ELISA法检测血清中IL-12、IFN-γ水平.结果:辐射后第1天,氨磷汀组IL-12水平高于辐射组(P<0.05),各组间IFN-γ水平无差异;辐射后第7天,防治组IL-12水平高于辐射组(P<0.05),防治组IFN-γ水平高于辐射组与氨磷汀组(P<0.01,P<0.05),预防组IFN-γ水平高于辐射组(P<0.05),空白组IFN-γ水平高于辐射组和氨磷汀组(P<0.05);辐射后第14天,各组IL-12、IFN-γ水平均无差异.结论:辐射后补肾解毒方可通过提高分泌血清中IL-12和IFN-γ的水平,起到增强辐射后小鼠免疫系统防护作用.
背景胸腺是最重要的中枢免疫器官,在接受射线后易受损造成免疫损伤,目前国内外研制的胸腺防护剂仍存在缺陷,中药因其高效低毒的特性,越来越受到重视.目的 研究补肾解毒方对放射致小鼠胸腺损伤的防护作用.方法 2019年1月,将90只C57BL/10J小鼠采用随机数字表法分为5组:空白对照组、单纯放射组、放射+氨磷汀组、放射+补肾解毒方防治结合组和放射+补肾解毒方预防组,每组18只.适应性饲养3 d后,放射+补肾解毒方防治结合组、放射+补肾解毒方预防组每日予以补肾解毒方汤剂灌胃(0.25 ml/只),同时单纯放射组和放射+氨磷汀组予以等剂量0.9%氯化钠溶液连续灌胃10 d,第11天除空白对照组外,其余4组小鼠均接受与照射源同等距离的一次性4 Gy 60Coγ-射线全身照射,其中放射+氨磷汀组小鼠在照射前30 min于左侧腹腔内注射氨磷汀200 mg/kg.照射后,放射+补肾解毒方防治结合组继续予以补肾解毒方灌胃,余4组给予等剂量0.9%氯化钠溶液灌胃.观察小鼠一般情况,在照射后第1、7、14天进行取材,计算胸腺指数,在光镜下观察照射后第7天各组胸腺组织结构.结果 五组小鼠在照射后一般情况均较照射前差,具体表现为活跃度下降、精神状态不佳、皮毛光泽度下降,尤其单纯放射组程度更甚,其中5只小鼠出现毛发块状脱落症状.照射后第1、7、14天,空白对照组胸腺指数均高于其余4组(P<0.01);照射后第7天,放射+氨磷汀组、放射+补肾解毒方防治结合组胸腺指数高于单纯放射组,放射+补肾解毒方预防组低于放射+补肾解毒方防治结合组(P<0.01);照射后第14天,放射+补肾解毒方防治结合组胸腺指数高于单纯放射组、放射+氨磷汀组和放射+补肾解毒方预防组(P<0.05).空白对照组胸腺结构完整,皮质内可见髓质,中央可见圆形或椭圆形胸腺小体;单纯放射组小梁结构破坏,界限不清,胸腺小体模糊;其余3组病理表现基本一致,部分小梁小叶结构破坏,胸腺小体隐约可见,皮质髓质界限不清,其中放射+补肾解毒方预防组还表现出更多炎性细胞浸润.结论 补肾解毒方能减轻放射所致小鼠胸腺组织损伤,发挥了良好的放射防护作用.
目的 观察疏调平衡针刺对失眠患者头部缺氧缺血和疲劳状态的影响.方法 将2017年6—12月就诊于中国人民解放军火箭军总医院中医门诊的原发性失眠患者152例随机分为治疗组和对照组,每组76例.治疗组采用疏调平衡针刺治疗,对照组采用口服艾司唑仑片治疗,2组均治疗4周.观察2组患者治疗前后头部缺氧缺血和疲劳状态变化情况.结果 2组患者治疗后缺氧缺血状态和疲劳状态均有所改善(P均<0.05),且治疗组缺氧缺血和疲劳状态改善效果都明显优于对照组(P均<0.05).2组治疗后缺氧缺血和疲劳状态评分均明显降低(P均<0.05),且治疗组明显低于对照组(P均<0.05),就诊结束3个月后随访,治疗组缺氧缺血和疲劳状态评分仍明显低于治疗前及对照组(P均<0.05),对照组则与治疗前比较差异无统计学意义(P均>0.05).治疗组总有效率为86.8%(66/76),对照组为46.1%(35/76),2组比较差异有统计学意义(P<0.05).结论 疏调平衡针刺可显著改善失眠患者的头部缺氧缺血和疲劳状态.
It has been extensively verified that inflammation and oxidative stress play important roles in the pathogenesis of cardiovascular diseases (CVDs). Curcuminoids, from the plant Curcuma longa, have three major active ingredients, which include curcumin (curcumin I), demethoxycurcumin, and bisdemethoxycurcumin. Curcuminoids have been used in traditional medicine for CVDs' management and other comorbidities for centuries. Numerous studies had delineated their anti-inflammatory, antioxidative, and other medicinally relevant properties. Animal experiments and clinical trials have also demonstrated that turmeric and curcuminoids can effectively reduce atherosclerosis, cardiac hypertrophy, hypertension, ischemia/reperfusion injury, and diabetic cardiovascular complications. In this review, we introduce and summarize curcuminoids' molecular and biological significance, while focusing on their mechanistic anti-inflammatory/antioxidative involvements in CVDs and preventive effects against CVDs, and, finally, discuss relevant clinical applications.
目的:研究氨磷汀对急性放射性肠炎小鼠小肠组织中核转录因子kappaB(NF-κB)通路和血管间粘附分子-1(VCAM-1)、细胞间粘附分子-1(ICAM-1)基因表达的影响和意义.方法:将小鼠分成对照组、实验组、氨磷汀腹腔注射干预组,采用6Gy 60Coγ-射线全身辐射,于第1、7、14天分批处死小鼠观察病理改变.实时荧光定量PCR(Q-PCR)方法检测不同时间点对照组、实验组和氨磷汀干预组NF-κB、VCAM-1、ICAM-1基因mRNA表达水平的变化.结果:辐射后实验组、干预组均可见明显黏膜下层微血管损伤,干预组损伤较实验组轻.辐射后第1天实验组、干预组小鼠小肠组织中NF-κB、VCAM-1、ICAM-1基因mRNA表达水平均显著升高(P<0.05),干预组升高水平显著低于实验组(P<0.05);第7天实验组各基因mRNA水平均呈下降趋势但仍高于对照组(P<0.05),干预组达到或接近对照组水平;第14天实验组、干预组各基因mRNA表达水平与对照组比较无统计学差异(P>0.05).结论:急性放射性肠炎早期即发生NF-κB、VCAM-1、ICAM-1基因mRNA的表达上调,可能在辐射后微血管损伤中发挥作用,氨磷汀可能通过下调上述基因表达对微血管起到保护作用.
