OBJECTIVE:To observe metabolomic changes in urine of chronic superficial gastritis (CSG) patients with Pi-qi deficiency syndrome (PQDS) or Pi-Wei dampness-heat syndrome (PWDHS), thereby providing scientific evidence for syndrome typing of them.METHODS:Urine samples were collected from CSG patients with PQDS/PWDHS and healthy volunteers, 10 in each group. Proton nuclear magnetic resonance spectroscopy (1H-NMR) based metabonomic analysis was performed on urine samples. Contents of related biomarkers were analyzed by principal component analysis (PCA), partial least square discriminant analysis (PLS-DA), and urivariate statistical analysis.RESULTS:PLS-DA analysis showed that metabolites among CSG patients with PQDS/PWDHS and healthy volunteers could be mutually distinguished. Seven differentially identified metabolites were screened from urines of CSG patients with PQDS and healthy volunteers included glutamate, methionine, α-oxoglutarate, dimethylglycine, creatinine, taurine, and glucose. Four differentially identified metabolites were screened from urines of CSG patients with PWDHS and healthy volunteers included 2-hydroxybutyric acid, trimethylamine oxide, taurine, and hippuric acid. Eleven differentially identified metabolites were screened from urines of CSG patients with PQDS and PWDHS included fucose, β-hydroxybutyric acid, alanine, glutamate, methionine, succinic acid, citric acid, creatinine, glucose, hippuric acid, and lactic acid.CONCLUSION:The metabolic differences of CSG patients PQDS and PWDHS mainly manifested in glycometabolism, lipid metabolism, and amino acids catabolism, and 1H-NMR based metabonomics may be used in classified study of Chinese medical syndrome typing.
Objective To observe the effect of astragalus injection of dexamethasone(Dex) proliferation induced immunosuppressed mice T cels.Methods Besides control group,the other five groups were injected Dex 5 days for immunodeficient mice model,two hours after the final injection,weight recorded.Results Compared with the control group, thymus index and IL-2 of serum and thymus homogenate in model group were significantly reduced(P<0.05),but protein expression of PCNA had no significant difference(P>0.05).Conclusion Immunity was impaired in mice exposed to Dex,AI could play an important role in recovering the immune function,the mechanisms would be further researched.
Objective To observe the treatment effect of Lumbrokinase on the neuron Nissl bodies in the gastric muscle plexus of diabetic rats. Methods An experiment on the SPF models established by using streptozotocin (STZ) and diabetic rats which were randomly put into the control and the I.umbrokinase trial groups was carried out. The effect was examined through the use of toluidine in 30 weeks. Results The amount of neuron Nissl bodies of the models was less than that of the control group (P〈0. 01) while the amount of neuron Nissl bodies of the Lumbrokinase trial group was more than that of the models (P〈0. 01). There was a difference between the control and the trial groups (P〈0. 05). Conclusion The decrease of the amount of neuron Nissl bodies of the models indicated the neuron was damaged and Lumbrokinase was effective in protecting neuron.
Background:Motilin(MTL) and somatostatin(SST) are the brain-gut peptides that have excitatory or inhibitory effect on gastrointestinal motility,respectively,and play an important role in gastrointestinal motility disorder.Aims:To study the change of gastrointestinal motility and possible mechanism in rats with spinal cord injury.Methods:One hundred and thirtyeight rats were randomly divided into model group and control group.The spinal cord injury model was established by weight drop method.Rats were sacrificed on the 1~(st),7~(th) day after establishment,HE staining was used to observe spinal cord tissue injury,while gastric emptying and propulsion rate of small intestine were measured.Expressions of MTL-RIA mRNA and SSTR_2 mRNA were determined by RT-PCR.Results:On the 1~(st) 7~(th) day after establishment,morphology of spinal cord tissue in model group obviously changed.On the 7~(th) day after establishment and compared with control group,gastric nuclide remnant rate in model group was significandy increased(76.66%±11.84%vs.55.32%±10.88%,P 0.01),propulsion rate of small intestine was significantly decreased(31.64%±5.47%vs.43.56%±7.00%,P 0.01);expression of MTLR1A mRNA in model group was obviously decreased(0.21±0.07 vs.0.47±0.13,P 0.01),while expression of SSTR_2 mRNA was significantly increased(1.12±0.21 vs.0.62±0.13,P 0.01 ).Conclusions:The occurrence of gastrointestinal motility disorder after spinal cord injury may be related to the down-regulation of expression of MTL-RIA mRNA in gastric tissue and up-regulation of expression of SSTR_2 mRNA in colonic tissue.
目的:探讨糖肾安对糖尿病肾病(DN)大鼠肾脏损害的保护作用及其机制。方法:采用链脲佐菌素(STZ)腹腔注射建立糖尿病(DM)大鼠模型,将SD大鼠随机分为正常对照组、模型组、辛伐他汀组、糖肾安组。治疗30周。观察治疗期间及治疗后大鼠的一般状况、血糖、肾质量/体质量(LKW)、内生肌酐清除率(CCr)和24小时尿蛋白改变;用免疫组化法检测细胞间黏附分子-1(ICAM-1)在DN大鼠肾小球及肾小管中的表达。结果:模型组LKW、CCr、24小时尿蛋白均高于正常对照组(P<0.01);肾小管中ICAM-1有明显表达,肾小球未见明显ICAM-1表达。糖肾安组血糖、LKW、CCr、24小时尿蛋白降低(P<0.01),ICAM-1表达明显下降。结论:糖肾安对糖尿病肾病有一定的保护作用。
目的:观察黄芪注射液(AI)对小鼠胸腺萎缩的干预作用。方法:将昆明小鼠随机分为正常对照组、模型组、IL-2组、黄芪注射液高(2 g/kg)、中(1 g/kg)、低(0.5 g/kg)剂量组,除正常对照组外,均采用地塞米松(25 mg/kg)腹腔注射复制小鼠胸腺淋巴细胞凋亡模型。观察黄芪注射液对小鼠胸腺指数的影响及胸腺的形态学改变,TUNEL及胸腺琼脂糖凝胶电泳测胸腺淋巴细胞凋亡,采用免疫组织化学与医学图像分析系统结合的方法检测小鼠胸腺淋巴细胞相关凋亡基因Bc l-2的表达。结果:黄芪注射液能明显改善萎缩的胸腺,增加胸腺指数(P<0.05);黄芪各剂量组小鼠胸腺皮质细胞明显增多,胸腺淋巴细胞凋亡数目及阳性细胞百分比与模型组比较显著减少(P<0.05或P<0.01),DNA琼脂糖凝胶电泳与模型组相比有显著变化,DNA的梯状图谱显色变浅甚至消失,同时还发现黄芪注射液能提高Bc l-2的表达(P<0.05或P<0.01)。结论:黄芪注射液有明显的抗胸腺萎缩及抑制胸腺淋巴细胞凋亡的作用,其作用机制可能与上调Bc l-2基因表达相关。
目的:观察黄芪注射液对小鼠急性胸腺萎缩后的T淋巴细胞凋亡及相关基因Caspase-3表达的影响.方法:昆明小鼠48只,随机等分为对照组、模型组、IL-2组、黄芪注射液治疗组.分别采用TUNEL法及免疫组织化学与医学图像分析系统结合的方法检测各组小鼠胸腺组织T淋巴细胞凋亡及Caspase-3蛋白的表达.结果:与对照组相比,模型组淋巴细胞凋亡数目及Caspase-3蛋白表达均显著增多(P<0.01).与模型组相比,黄芪注射液组小鼠胸腺组织淋巴细胞凋亡数目及Caspase-3蛋白表达明显减少(P<0.05).结论:黄芪注射液可通过抑制胸腺T淋巴细胞凋亡来发挥保护胸腺萎缩的作用,其抗凋亡机制可能与下调Caspase-3蛋白表达相关.
Objective To detect the information of gastric emptying and gastric mucosal membrane cell apoptosis for 30 weeks diabetic rats.Methods To establish 30 weeks diabetic rats model with Streptozotocin(STZ),then phenolsulfonphthalein test and optical density was used for gastric emptying and Tunel test was detected for gastric mucosal membrane cell apoptosis.Results The test showed the thirtieth week diabetic rats existed abnormal gastric emptying,the Chinese medicine group improved,compared with diabetic model group;the severe gastric mucosal membrane cell apoptosis was diabetic model group,compared with the traditional Chinese medicine group and the controlled group without significant deviation.Conclusions 30 weeks diabetic rats had abnormal gastric emptying,gastric mucosal membrane had different degree cell apoptosis,the apoptotic degree was significant,then the Chinese medicine healing group compared with the controlled group had no significant deviation,thus it showed the Chinese medicine could protect gastric mucosal membrane.

目的:研究三黄脂消饮对糖尿病胃轻瘫大鼠血糖和胃排空率的影响。方法:以三黄脂消饮治疗糖尿病胃轻瘫模型大鼠,并与正常组及模型组比较。结果:中药治疗后血糖下降,胃排空率增加。结论:三黄脂消饮不仅可以降低糖尿病血糖水平,而且增加了糖尿病胃轻瘫的胃排空率,明显地改善了胃排空延迟,所以三黄脂消饮是治疗糖尿病胃轻瘫的有效药物。
Objective:To investigate the effect of medicine interfering in blood flow Weichangshu(WCS)to rat’s diabetic model and rat’s diabetic gastroparesis model.Methods:One hundred thirty-one male SD rats who were SPF class were randomly divided into the control group(10 rats),model group(22 rats),ethamsylate group(33 rats),lumbrukinase group(33 rats)and WCS group(33 rats).Except for rats in the control group,surplus rats were injected streptozotocin(STZ)solution(60mg/kg)into their abdominal cavities.We measured blood sugar after seven days making diabetic mould and looked blood sugar more than or equal to 16.7mmol/L as experimental internalized standard.After 10 days,rats in the WCS group were treated by traditional Chinese medicineWCSapozem which were given by intragastric adminisreation one time per day.Rats in the ethamsylate group were treated by ethamsylate injection which were given by intragastric adminisreation one time per day.Rats in the lumbrukinase group were treated by lumbrukinase enteric-coated capsules which were injected into their abdominal cavities one time per day.Rats in the control group and rats in the model group were treated by tales doses distilled water which were given by intragastric adminisreation one time per day.After having been observed for 30 weeks,all rats were measured contents of blood sugar,FINS,C-P,TC,TG,HDL,LDL,rats’gastric fluid emptying rate.Subsequently,we got out the stomach and pancreas,and knew how pancreatic tissue and pancreatic islet were destructed,and observed pathological lesion of gastric neurotransmitter such as nNOS through light microscope.Results:after 30 weeks,blood sugar,FINS,C-P,TC,TG,HDL and LDL in rats of all made diabetic model group were significantly difference compared with ones in rats of the control group(P0.01),and there were no or few beta cells in rats’ pancreatic islets.However,in rats of the WCS group the biochemical indicators had amendment,hyperemia of pancreas were lessened,and there were more beta cells than ones in the model group.In rats of the WCS group gastric fluid emptying rate was obviously higher,and nNOS in gastric myenteric nerve plexus was significantly more than ones in rats of the model group、ones in rats of the lumbrukinase group or ones in rats of the ethamsylate group(P0.01).Conclusion:The results manifested that WCS could cut down blood sugar and speed up diabetic rats’ gastric fluid emptying,and there was some effect on medicine interfering in blood flow to rats’gastric fluid emptying.

Objective:To study the effect of Weichangshu(WCS) to hemorheology indexes and gastric micrangium pathology in rats of diabetic gastroparesis model.Methods: Male SD rats who were SΡF class were randomly divided into the control group、model group、ethamsylate group、lumbrukinase group and WCS group.Except for rats in the control group,surplus rats were injected streptozotocin(STZ) solution(60mg/kg) into their abdominal cavities.Rats in the control group were injected citric acid and citrate sodium buffer solution into their abdominal cavities with isasteric dosage one time.After seven days making diabetic mould,we measured blood sugar,we looked blood sugar more than or equal to 16.7mmol/L as experimental internalized standard.Rats consistented with experimental internalized criteria began to be treated after 10 days making diabetic mould.Rats in the WCS group were treated by traditional Chinese medicine WCS apozem which were given by intragastric adminisreation one time per day.Rats in the ethamsylate group were treated by ethamsylate injection which were given by intragastric adminisreation one time per day.Rats in the lumbrukinase group were treated by lumbrukinase enteric-coated capsules which were injected into their abdominal cavities one time per day.Rats in the control group and rats in the model group were treated by tales doses distilled water which were given by intragastric adminisreation one time per day.After having been observed for 30 weeks,all rats were measured contents of blood sugar、FINS、C-P、hemorheology indexes and rats' gastric fluid emptying rate.Subsequently,we got out the stomach and observed pathological lesion of gastro micrangium through light microscope and electron microscope.Results: After 30 weeks,in rats of the model group hemorheology indexes were obviously higher,gastric micrangium were deformed and narrowed,endotheliocyte were edema,intracytoplasm mitochondria were tumescent or vacuolization,micrangium lumens waxed.In rats of the WCS group and in rats of the lumbrukinase group hemorheology indexes took a turn for the better and pathological lesion of gastric micrangium were obviously lessened,microvessel count were significantly incremental compared with ones in rats of the model group.Nevertheless,in rats of the ethamsylate group hemorheology indexes were worse,pathological lesion of gastric micrangium were significantly aggratated,and microvessel count were less.Conclusion: WCS could treat Diabetic gastroparesis through losing blood sugar and improving hemorheology and microcirculation、adding volume of blood flow.

Objective:To identify the commercial specification of Pogostemon cablin by testing the volatile oil and con-stituent content of different market Pogostemon cablin. Methods: The volatile oil and constituent content of different mar-ket Pogostemon cablin were analyzed quantitatively by the volatile oil assay of pharmacopoeia and GC-MS method. Re-sults: The volatile oil content of different market Pogostemon cablin didn't show market difference. Comparing to Paixiang, Zhaoxiang and Nanxiang, most of the market Pogostemon cablin were Nanxiang. Conclusion: Reinforcing the planting of Paixiang and Zhaoxiang can provide medicinal materials of high quality for Pogostemon cablin.
目的:观察中医证型和结肠动力随年龄增高的改变。方法:观察普通成年人90例中医证型随年龄增长出现的频率改变和结肠电各高、中、低段的缺失率的比率的变化。结果:年龄36~50岁组受检者脾虚便溏的出现率显著高于18~35岁组,年龄>50岁组受检者脾虚便溏的出现率显著高于18~35岁组。说明,随着年龄增高正常大便的出现率降低,尤其是大便稀烂的出现率显著增高,而脾虚便溏型的出现率随年龄增高也增多。结肠频谱与年龄的相关分析表明,年龄较大者其乙状结肠高频段肠电缺失的比率显著降低,降结肠高频段肠电缺失的比率亦倾向降低。结论:脾胃气虚、脾虚便溏、气虚便秘构成比有随年龄增加而增加的趋势,而脾胃气虚、肝肠气滞构成比有随年龄增加而减小的趋势。年龄较大者其乙状结肠高频段肠电缺失的比率显著降低,降结肠高频段肠电缺失的比率亦倾向降低。
