目的 建立子宫锭的质量控制标准.方法 采用薄层色谱法对处方中的血竭、蛇床子进行定性鉴别研究;分别采用高效液相色谱法和气相色谱法对子宫锭中的儿茶素和麝香酮进行含量测定.结果 薄层色谱可检出血竭、蛇床子的特征斑点;儿茶素在0.632 5 ~1.265 0 μg范围呈良好的线性关系(r=0.999 9),平均回收率为98.71%,RSD=1.56%;麝香酮在0.134 4 ~0.302 4 μg范围呈良好的线性关系(r=0.999 8),平均回收率为98.36%,RSD=1.82%.结论 本方法准确、灵敏、重复性好,为子宫锭的质量控制提供了依据.
目的 通过体外冷热循环模拟老化核树脂材料,比较不同核树脂的机械性能.方法 将A、B、C、D四种核树脂分别制备成25mm×2mm×2mm的6个棱柱状挠曲试样和高6mm,直径4mm的10个圆柱状抗压试样,通过万能试验机测试冷热循环前后试样的挠曲强度和抗压强度;用单因素方差分析和t检验进行统计分析.结果 未冷热循环的四组核树脂挠曲强度值和抗压强度值差异均没有统计学意义.经过5000次冷热循环后,自制核树脂与三种成品核树脂相比较,在挠曲强度和抗压强度上差异均没有统计学意义.A、B、C三组核树脂挠曲强度和抗压强度在循环后均有显著下降(P<0.05).结论 自制核树脂的挠曲强度和抗压强度良好,冷热循环对核树脂材料的老化作用明显.
Objective To control the quality of realgar in compound Huanglian Ointment.Methods As2O3which is the toxic component of realgar was carried limit test by Gutzeit's test; Potassium sulfate, ammonium sulfate and sulfuric acid were used to digest, and then titration method was used to determine the content of As2S2in compound Huanglian Ointment. Results The content of the soluble As in compound Huanglian Ointment was no higher than 15.6 μg/g. The content of As2S2in compound Huanglian Ointment was no less than 2.21 mg/g. Conclusion The method is simple and easy,which can be applied as the quantity control method of compound Huanglian Ointment.
目的:为香鳞毛蕨进一步开发利用提供参考.方法:以"香鳞毛蕨""化学成分""药理作用""Dryopteris fragrans""Dryofra-gin""Aspidin BB""Aspidin PB"等为关键词,组合查询1980年1月-2016年7月在PubMed、Web of Science、Elsevier、中国知网、万方、维普等数据库中的相关文献,对香鳞毛蕨的化学成分及药理作用进行综述.结果与结论:共检索到相关文献232篇,其中有效文献55篇.香鳞毛蕨对各种皮肤病及类风湿性关节炎有很好的疗效,被誉为"皮肤病的克星".迄今为止从香鳞毛蕨中分离得到了75个化合物,主要类型包括间苯三酚类、萜类、黄酮类及苯丙素类等,具有抗肿瘤、抗菌、止痒、抗过敏等多种药理作用.间苯三酚类是香鳞毛蕨的特征性化学成分,是发挥相关功效作用的主要有效成分,但也具有一定的毒性.除应对间苯三酚类成分进行深入探索外,还应对其有毒成分及其毒理机制进行深入系统地研究.
Objective To observe the intervention effects of Dryopteris fragrans extracts on psoriasis-like skin lesions induced by imiquimod in mice. Methods A psoriasis-like mouse model was made by applying 5% imiquimod cream daily on the back. Totally 726–8 week old male BALB/c mice were randomly divided into control group, model group, Dryopteris fragrans extracts groups and positive medicine group, 8 mice in each group. The model group and the control group were given distilled water for gavage. The mice in each administration group were given relevant medicine for gavage, 0.3 mL daily, for 6 days. The skin lesions were evaluated according to the psoriasis area and severity index (PASI). The histology and epidermal thickness were observed under light microscope. The spleen index was detected to reflect the changes of spleen in mice. Results Compared with control group, the erythema scales and skin lesions were thickened in the model group; lesions of the performance of the epidermis acanthosis was thickening, parakeratosis; spleen hypertrophy. Compared with model group, the cutaneous symptoms in Dryopteris fragrans extracts groups were alleviated; PASI scores decreased; epidermal parakeratosis was relieved; the spleen index was lower than that of model group. Conclusion Dryopteris fragrans extracts can significantly improve the psoriasis-like skin lesions induced by imiquimod in mice.
Objective To compare the mechanical properties of several kinds of self-made core resin with imported core resin. Methods Nano-SiO2and glass powder were added to different organic matrix by various proportions,to develop 3 groups of nanocomposite core resin. Group 1: matrix with BisEMA6,Nano-SiO2( diameter: 50nm); Group 2: matrix with BisEMA6,Nano-SiO2( diameters: 12nm and 50nm); Group 3: matrix with BisEMA,Nano-SiO2( diameters: 12nm and 50nm). The flexural strength and compressive strength of self-made and imported resin were evaluated and compared. Results The flexural strength of group 2 and 3 was not significantly different with that of the imported core resin( P 0. 05),but the compressive strength was much higher in group 2 and 3 than in imported resin( P 0. 05). The flexural strength and compressive strength of group 1 was significantly lower than that of group 2 and 3. Conclusion The self-made core resin containing BisEMA,BisEMA6 and 12nm SiO2presents as high flexural strength as the imported core resin but has higher compressive strength than imported core resin.
Objective To investigate the fracture resistance of maxillary incisors restored with different diameters of prefabricated Zirconia and fiber post with core resin material.Methods Twenty-four isolated maxillary central incisors which had received root cannl treatment were divided into 4 groups,which were then restored with 1.5mm,1.35mm diameters of Zirix post and 1.5mm,1.375mm diameters of fiber post,respectively.The fracture resistance of the samples was tested by universal test machine and the fracture modes were observed.Results The fracture resistance of 1.5mm and 1.35mm diameters of Zirix post was 508.5±143.3N and 434.9±141.7N,and of 1.5mm,1.375mm diameters of fiber post was 292.2±54.5N and 238.5±79.1N.There was a significant difference in resistance between prefabricated Zirconia post and the fiber post(P<0.05).50% of the prefabricated Zirconia post samples fractured at the root cervical portion and 83.3% of the fiber post samples fractured could be retreated.Conclusion Prefabricated Zirconia post could bear higher fracture load.
Objective:To investigate the effect of active part of spatholobus suberectus flavonoid(SSCE) on oxidative stress in human pulmonary adenocarcinoma cell A549.Method:Cells were divided into a high-dose group,a moderate-dose,a low-dose group and a normal control group.Cell A549 were interfered with different dosages of SSCE active part for 24 h(normal group with DMEM) level of malonic dialde hyde(MDA) and glutathione(GSH);reactive oxygen speiesROS was detected using fluorescence microplate reader and confocal fluorescence microscope,and using DCFH-DA as fluorescent probe;Antioxidant was added,CCK-8 was employed to detect cell proliferation of A549 and ROS was detected by fluorescence microplate reader.Result:After the addition of SSCE,MDA content in each group increased(P0.01) and GSH content in high density group decreased(P0.01);Cell ROS content in high and middle density groups increased(P0.01);Each group showed green fluorescence under confocal microscope,in which the high and middle density group showed higher;Addition of antioxidant made decrease of cell inhibition(P0.05) and ROS content(P0.01).Conclusion:SSCE can change the level of A549 MDA,GSH and ROS,and its mechanism of antitumor might be inducing oxidative stress reaction of tumor cells.
Objective:To define effect of chemical composition under decocting together with different group condition.Method: Different compatibility group was adopted,the content of target composition was determined after decocted,compared between two groups,and defined whether to decoct.Result:①Manis pentadactyla and Chinemys reevesii decocted with drug containing volatile oil,it would not extract volatile oil in this way;②M.pentadactyla and C.reevesii could reduce decocting ratio of alkaloids;③Rheum palmatum could decrease decocting ratio of gentiopicroside;④drugs containing alkaloids could decrease decocting ratio of anthraquinone from R.palmatum.Conclusion: Animal drugs could affect extraction ratio of volatile oil;drugs containing gentiopicroside alkaloids could not decoct with R.palmatum.
目的 优化清热除湿颗粒的提取工艺.方法 采用正交设计,以龙胆苦苷、黄芩苷、大车前苷的含有量为醇提工艺指标,考察含乙醇量、乙醇用量、提取时间等因素的最佳工艺条件;以总多糖含有量和浸膏得率为水提工艺指标,考察提取次数、提取时间、水的用量等因素的最佳工艺条件.结果 醇提最佳工艺为:加12倍药材量的70%乙醇提取3次,每次1.5 h;水提最佳工艺为:加8倍药材量的水提取3次,每次2h.结论 该提取工艺稳定、可行,为清热除湿颗粒的提取条件提供了实验依据.
目的 采用高速逆流色谱(HSCCC)技术分离纯化鸡血藤中的黄酮类抗肿瘤活性成分.方法 运用活性跟踪分离思路,优化制备型HSCCC分离条件,快速分离鸡血藤醋酸乙酯萃取物中的黄酮类单体化合物;根据理化性质和波谱方法(ESI-MS、UV、NMR等)鉴定化合物结构;采用MTT方法并结合细胞形态学观察评价化合物对人肺癌细胞A549的抗肿瘤活性.结果 利用正己烷-醋酸乙酯-甲醇-水四元两相溶剂体系,通过HSCCC法从鸡血藤醋酸乙酯萃取物中分离得到2个黄酮类成分,鉴定为甘草素(CSl)和刺芒柄花素(CS2),HPLC峰面积归—化法分析二者的质量分数分别为93.6%和94.3%.MTT法抗肿瘤活性测定发现CS1对A549细胞显示增殖抑制活性,当质量浓度为200 μg/mL时对A549细胞的增殖抑制率为49.9%.结论 可以采用HSCCC技术分离纯化鸡血藤提取物中的黄酮类抗肿瘤活性成分,该方法快速、简便、经济,为鸡血藤活性成分研究提供了技术依据.
Objective To determine berberine hydrochloride and palmatine hydrochloride in Haemorrhoid Lotion by HPLC.Methods The Kromasil 100-5C18(150 mm×4.6 mm,5 μm) column was used.The mobile phases consisted of 0.1% phosphoric acid solution-acetonitrile(50∶50,containing 0.1% sodium dodecyl Sulfonate) and the detection wavelength was at 265 nm.Results The linear range of berberine hydrochloride and palmatine hydrochloride was 0.02~1.0 μg and 0.017 2~0.861 μg respectively.The average recovery of berberine hydrochloride was 97.79%,and RSD was 0.95%.The average recovery of palmatine hydrochloride was 99.22%,and RSD was 0.86%.Conclusion The simple,sensitive and reliable HPLC identification method was established.The method can be used for quality control of Haemorrhoid Lotion.
OBJECTIVE:To establish a method for the content determination of β,β'-dimethylacrylalkannin in Radix Arnebiae.METHODS:Improved HPLC method was adopted and samples were prepared by mobile phase.Kromasil 100-5 C18(150 mm×4.6 mm,5 μm) column was used with mobile phase consisted of acetonitrile-water-formic acid(700:300:0.5)at flow rate of 1.0 mL· min-1.The detection wavelength was set at 275 nm.RESULTS:β,β'-dimethylacrylalkannin had good linearity in the range of 0.16-3.20 μg(r=0.999 9) with an average recovery of 99.46%(RSD=1.90%,n=6).CONCLUSION:The method is simple and feasible for content determination of Radix Arnebiae.Method verification is up to the standard.
Objective To investigate the effect of spatholobus suberectus column extract(SSCE) on apoptosis and mitochondrial transmembrane potential of non-small cell lung cancer A549 in vitro. Methods Cell counting kit-8 was used to investigate the effect of SSCE on the proliferation of A549,apoptotic morphological changes were observed by Annexin V-PI double-staining technique,nuclear changes were observed by Hochest33342 staining,mitochondrial changes were observed by Mitotracker red staining,and mitochondrial membrane potential was examined by JC-1 staining. Results The concentration of 40 mg/L,SSCE could suppress the proliferation of A549 cells in a dose and time-dependent manner.After cultured with SSCE for 24 h,A549 cells exhibited typical characteristics of apoptosis,such as phosphatidylserine(PS) externalization,pyknosis,dense-stained nucleus and cytoplasm,nucleus granulation,mitochondrias gathered to the nucleus and their shape changed from rod into granular form,mitochondrial membrane potential decreased. Conclusion SSCE induced apoptosis of A549 cells by changing the mitochondrial morphology and reducing the mitochondrial membrane potential.
目的建立洁阴洗剂的质量标准。方法采用薄层色谱法对洁阴洗剂中苦参、黄柏、虎杖、野菊花、淫羊藿进行定性鉴别;应用高效液相色谱法测定蛇床子素的含量,色谱条件为:Kromasil100-5C18(150mm×4.6mm)色谱柱,流动相:甲醇-水(65∶35),流速:1.0 mL.min-1,检测波长:322 nm,柱温:25℃。结果薄层色谱法能准确鉴别出制剂中的苦参、黄柏、虎杖、野菊花、淫羊藿,空白对照结果无干扰;蛇床子素与其他成分分离完全,峰形良好。蛇床子素在0.02~0.17μg范围内线性关系良好(r=0.9993)。平均加样回收率为100.4%,RSD为0.89%(n=6)。结论所建立的方法简便、准确、重现性好,可作为该制剂的质量控制方法。
Objective:To observe influence of Tiaomaiyin drug and its separate components on three kinds of experimental arrhythmia in animals.Method:The animals were randomly divided into a model group,a group treated by whole Tiaomaiyin,a group treated by main components for Liangxue Qingre function,a group treated by assisted components for Yiqi Yangxin function,a group treated by assisted components for Xingqi Tongmai function and a group treated by a known effective drug(Propafenone).By recording ECG,the effects of Tiaomaiyin and its separate components on ventricular arrhythmia induced by Aconitine in rats,that induced by Ouabain in guinea pigs,and that induced by ligating coronary artery were observed.Result: The whole Tiaomaiyin drug and the components for liangxueqingre function inhibited all the three kinds of ventricular arrhythmia in the three kinds of animals,respectively.Conclusion: The whole Tiaomaiyin drug and the components for Liangxue Qingre action provide the function against experimental arrhythmias.
目的:建立喘宁巴布剂中芥子碱硫氰酸盐的高效液相色谱测定方法.方法:采用Kromasil苯基色谱柱(150 mm×4.6 mm,5μm);以3%醋酸(A)-乙腈(B)为流动相,梯度洗脱;检测波长为326 nm.结果:芥子碱硫氰酸盐在0.04~0.40μg范围内呈良好线性关系,回归方程Y=3 931.3X-5.995 9,r=0.999 9,平均回收率为99.14%,RSD=1.69%(n=6).结论:建立的方法用于测定喘宁巴布剂中芥子碱硫氰酸盐的含量简便、可靠、准确,可用于该制剂的质量控制.
AIM: To establish the quality standard of Furong Gel(Rhizoma Coptidis,Radix Scutellariae,Radix et Rhizoma Rhei,etc.).METHODS: Rhizoma Coptidis,Cortex Phellodendri Amurensis,Radix Scutellariae,Ra-dix et Rhizoma Rhei,Herba Lycopi were identified by TLC.The contents of berberine hydrochloride,palmatin hydrochloride and baicalin were determined simultaneously by HPLC.RESULTS: The TLC spots were clear,and the blank test showed no interference.The condition of HPLC was C18 column,ACN-0.05 mol/L KH2PO4 as mo-bile phase,276 nm and 265 nm as detection wavelength.The linearities of berberine hydrochloride,palmatin hydrochloride and baicalin were good,and the average recoveries were 101.23%,100.49% and 101.49%,re-spectively.CONCLUSION: The method is reliable and accurate,and can be applied as the quantity control method of Furong Gel.
目的:筛选柿叶中对黑素细胞增殖及酪氨酸酶活性有明显抑制作用的部位.方法:柿叶70%乙醇提取后萃取得到不同的提取物;MTT法观察各提取物对小鼠B-16黑素瘤细胞增殖的影响,采用多巴氧化法研究柿叶提取物对黑素细胞酪氨酸酶活性的影响.结果:MTT法结果表明柿叶水提物和醇提物对黑素细胞的增殖均有抑制作用,且呈量效关系;70%醇提物的半数抑制浓度最低,与阳性对照相当;三氯甲烷和醋酸乙酯部分合并后对酪氨酸酶活性抑制作用明显.结论:本研究为柿叶的临床应用和进一步开发提供了一定的科学依据.
目的:优选芙蓉凝胶中药材提取工艺和凝胶基质配方.方法:设计L9(34)正交试验,采用HPLC法和分光光度法测定盐酸小檗碱含量和总黄酮及总蒽醌含量作为评价指标,优选药材提取工艺;以黏性、透皮速率、释药速率等理化性质作为评价指标,优化芙蓉凝胶基质配方.结果:黄连、黄柏饮片用12倍量70%乙醇浸泡1 h后提取2次,第1次1.5h,第2次1 h;黄芩等四药的提取工艺为药材饮片用12倍量65%的乙醇回流提取2次,第1次1.5 h,第2次1 h;芙蓉凝胶基质配方为CP9400.75%、PVA 7%、薄荷脑1%、三乙胺0.6%、甘油10%、聚山梨醇酯-80 1%.结论:优选出的提取工艺有效成分提取率高,重复性好,稳定可行;凝胶配方稳定性良好,黏度适宜,药物释放迅速,透皮吸收较快.
