Coptisine (COP) is a natural protoberberine isoquinoline alkaloid that is isolated from Coptis chinensis and exhibits a variety of pharmacological activities, such as the inhibition of tumor growth, bacterial infection, inflammation and oxidative stress. In this study, COP penetrated and produced fluorescent signals in living tumor cell lines, primary MEF cells and polyformaldehyde-fixed cells. The fluorescent signal was detected at a wavelength of 488 nm. The fluorescent signal of COP was observed predominantly in the nucleoli and colocalized with nucleolus fibrillarin and B23. The fluorescence intensity of COP was associated with tumor malignancy. Compared with cells with high fluorescent signals, cells with low fluorescent signals were highly malignant. Taken together, these data suggest that COP can function as a nucleolus probe and a probe candidate for distinguishing tumor cell malignancy.
Cottonseed meal has been one of the predominant protein feedstuffs used in aqua feed, which contains gossypol. It is well known that gossypol, a natural polyphenolic compound found in seeds, can be toxic to fish. The toxicity of gossypol persists even after a recovery period; however, the underlying mechanism remains largely unknown. To investigate the toxic mechanism of gossypol after the recovery period, Cyprinus carpio were administered a continuous oral dose of 20 mg/kg·d gossypol (gossypol acetate dose/fish weight·d) for one week, followed by a two-week recovery period. The spleen index, spleen morphology, lysozyme (LZM) activity, acid phosphatase (ACP) activity and nitric oxide (NO) concentration were obviously changed in the spleens of C. carpio after two weeks of recovery. Then, miRNA sequencing identified 22 differentially expressed miRNAs in the spleens of C. carpio two weeks after the gossypol treatment. The mRNA expression levels of immune-associated genes were significantly increased in the spleens after recovery. MiR-194 was chosen for future study through a search of existing literature and an analysis of miRNA and mRNA expression levels. To further characterize the potential target of miR-194, we identified putative miR-194 binding sites in the 3′UTR of myeloid differentiation factor 88a (MyD88a). Dual-luciferase reporter assays revealed that miR-194 downregulated the expression level of MyD88a through binding to seed-matching sequence in the 3′ UTR of MyD88a. Moreover, C. carpio were intraperitoneally administered with miR-194 antagomir. Furthermore, down-regulation of miR-194 increased the expression levels of MyD88a and some pro-inflammatory cytokines and anti-inflammatory cytokines. Altogether, the results demonstrate miR-194 can target MyD88a to regulate the immune response of C. carpio after a two-week recovery period following gossypol administration.
目的:系统评价成人ICU病人专用压力性损伤风险评估工具的预测性.方法:计算机检索PubMed、EMbase、the Cochrane Library、CINAHL、中国知网、万方数据库、维普数据库关于ICU专用压力性损伤风险评估工具的研究,检索时限从建库至2022年5月31日.采用澳大利亚Joanna Briggs Institute(JBI)循证卫生保健中心推出的诊断性研究质量评价工具对纳入文献进行质量评价,使用STATA 16.0软件进行Meta分析.结果:共纳入18篇文献,涉及5642例病人,压力性损伤发生例数824例.5篇文献评估了COMHON量表,合并灵敏度为0.82,95%CI[0.46,0.96],特异度为0.83,95%CI[0.79,0.86],综合受试者工作特征曲线(SROC)下面积为0.85.10篇文献评估了Cubbin&Jackson量表,合并灵敏度为0.75,95%CI[0.66,0.83],特异度为0.81,95%CI[0.73,0.87],SROC下面积为0.85.结论:现有证据表明,ICU病人压力性损伤风险评估工具预测性尚可,但仍存在一些问题,有必要在借鉴现有评估工具的基础上,开发适合国情的ICU病人压力性损伤风险评估工具.
目的:系统评价非负重运动锻炼在糖尿病足溃疡病人中应用效果,为糖尿病足运动干预提供循证依据.方法:计算机检索PubMed、the Cochrane Library、EMbase、Web of Science、中国知网、万方数据库收录的关于非负重运动锻炼在糖尿病足溃疡病人中应用的随机对照试验,检索时限从建库到2022年8月31日.由2名研究人员进行筛选文献、提取资料和评价文献质量,采用RevMan 5.3软件进行Meta分析.结果:共纳入15篇随机对照试验,涉及1 296例糖尿病足病人.Meta分析结果显示,非负重运动锻炼可以改善糖尿病足溃疡病人的伤口愈合率[RR=2.76,95%CI(2.00,3.80),P<0.000 01],改善病人的踝肱指数(ABI)[RR=0.07,95%CI(0.04,0.10),P<0.000 1];两组运动时不良事件发生率比较,差异无统计学意义[RR=0.76,95%CI(0.54,1.09),P=0.14];非负重运动锻炼对血糖相关指标和生活质量的影响尚不明确.结论:现有证据表明,非负重运动锻炼对糖尿病足溃疡病人有较好的临床结局,可以提高病人的伤口愈合率,改善病人的ABI.但纳入的研究在质量和数量上有所限制,需要更多高质量和大样本的研究对此结论进行验证.
目的 研究血管内皮生长因子(VEGF)及低氧诱导因子-1α(HIF-1α)与糖尿病合并动脉狭窄病人支架植入术后再狭窄关系.方法 选取2016年9月至2018年2月华中科技大学同济医学院附属梨园医院糖尿病合并动脉狭窄病人188例,均行动脉支架植入治疗,支架植入前检测病人血清VEGFA及HIF-1α,糖、脂代谢指标及血清C反应蛋白(CRP),对病人追踪随访6个月,按是否发生冠脉再狭窄分为再狭窄组98例,未再狭窄组90例,比较两组病人VEGF及HIF-1α,糖、脂代谢指标及血清CRP水平差异,采用logistic分析糖尿病合并动脉狭窄病人支架植入术后再狭窄的风险因素.采用Pearson检验分析VEGF、HIF-1α与CRP的相关性.绘制受试者工作特征曲线(ROC曲线),分析VEGFA及HIF-1α对糖尿病合并动脉狭窄病人支架植入术后再狭窄的预测效能.结果 再狭窄组及未再狭窄组病人血清糖化血红蛋白(HbA1c)≥7.2%、总胆固醇(TC)≥5.7 mmol/L、VEGF≥106.4 pg/mL、HIF-1α≥217.4 ng/mL、小而密低密度脂蛋白胆固醇(sLDLc)≥3.4 mmol/L、CRP≥11.6 mg/L、病变在分支动脉,支架植入长度≥3.4 cm、支架数目为2个及以上、支架直径<4.2 mm所占比例差异有统计学意义(63/98比34/90,64/98比31/90,61/98比31/90,63/98比27/90,62/98比40/90,62/98比17/90,71/98比41/90,62/98比34/90,26/98比10/90,71/98比31/90,均P<0.05).多因素logistic回归分析结果显示,VEGF、HIF-1α、HbAH1c、CRP水平是动脉支架植入术后再狭窄的独立风险因素,支架直径是保护性因素(P<0.05).相关分析结果显示,VEGF、HIF-1α与CRP水平呈现正相关关系(P<0.05).VEGF、HIF-1α、CRP对糖尿病合并动脉狭窄病人支架植入术后再狭窄的评估效能:ROC-AUC(95%CI)分别为0.665(0.412~0.916)、0.761(0.552~0.971)、0.636(0.323~0.944).结论 糖尿病合并动脉狭窄病人血清VEGF、HIF-1α水平与支架植入术后再狭窄密切相关,检测血清VEGF、HIF-1α水平对冠脉再狭窄的发生具有较好的预测价值.
目的 探讨共享决策感知对糖尿病肾病患者血液透析依从性的影响.方法 2021年1—12月选取糖尿病肾病血液透析患者84例,根据信封随机分组法将患者分为观察组及对照组,每组42例.对照组行常规健康指导,观察组在对照组基础上实施共享决策感知护理干预,比较两组血液透析期间疾病管理依从性、透析相关并发症及患者生活质量情况.结果 干预后观察组透析管理依从性总评分及简明健康量表(SF-36)评分高于对照组(P<0.05).观察组血液透析期间并发症发生率低于对照组(P<0.05).结论 共享决策感知护理干预能有效提高糖尿病肾病患者血液透析期间疾病管理依从性,降低血液透析并发症,提高患者生活质量.
The major challenges of clean energy and environmental pollution have resulted in the development of photocatalysis technologies for energy conversion and the degradation of refractory pollutants. Herein, a novel CdSe/Se/BiOBr hydrangea-like photocatalyst was used to produce hydrogen peroxide (H2O2) and degrade ciprofloxacin (CIP). The Z-scheme heterojunction structure of the photocatalyst and the doping of selenium (Se) led to the efficient separation of electron-hole pairs and charge transfer. The optimized sample of 2 wt% CdSe/Se/BiOBr produced 142.15 mg center dot L-1 rate of H2O2, which was much higher than that produced by pure BiOBr (89.4 mg center dot L-1) or CdSe/Se (10.9 mg center dot L-1). Additionally, almost 100 % of CIP was degraded within 30 min, with a first order rate constant of nearly 5.35 times that of pure BiOBr and 81.44 times that of pure CdSe/Se. The excellent removal efficiency of CIP from natural water matrices confirmed that the composites are promising for the removal of contaminants from natural waterways. Based on trapping experiments, electron spin resonance spectra (ESR) spectroscopy, and density functional theory (DFT) calculations, the photocatalytic mechanisms of H2O2 and CIP degradation by the Zscheme CdSe/Se/BiOBr composites were proposed. Overall, the dual-functional CdSe/Se/BiOBr composite could potentially be applied for photocatalytic production of H2O2 and treatment of organic pollutants in water. (c) 2021 Elsevier Inc. Retinitis pigmentosa (RP) is the most common inherited retinal degenerative disease which is the major cause of vision loss. X-linked RP patients account for 5%-15% of all inherited RP cases and mutations in RP2 (Retinitis pigmentosa 2) were responsible for about 20% X-linked RP families. A majority of RP2 pathogenic mutations displayed a vulnerable protein stability and degraded rapidly through ubiquitin-proteasome system (UPS). Though the RP2 protein could be readily recovered by proteasome inhibitors, e.g., MG132, their applications for RP2-related RP therapy were limited by their nonspecific characterization. In the present study, we aimed to identify UPS-related factors, such as E3 ligases, which are specifically involved in degradation of RP2 pathogenic mutants. We identified several E3 ligases, such as HUWE1, and the co-chaperon BAG6 specifically interacting with RP2 pathogenic mutants. Knockdown of HUWE1 and BAG6 could partially rescue the reduced protein levels of RP2 mutants. BAG6 is required for recruitment of HUWE1 to ubiquitinate RP2 mutants at the K268 site. The HUWE1 inhibitor BI8622 could restore the levels of RP2 mutant and then the binding to its partner ARL3 in retina cell lines. This study revealed the details of UPS-related degradation of RP2 mutants and possibly provided a potential treatment for RP2-related RP.
坏死性筋膜炎(necrotizing fasciitis,NF)是一种进展迅速、以皮下组织及筋膜组织坏死为特征的严重软组织感染性疾病[1].NF虽然发病率不高,但病死率却高达约20%[2],且糖尿病是其常见的致病原因之一[3].随着我国糖尿病发病率的上升,糖尿病合并坏死性筋膜炎的患病人数有所增加[4].糖尿病患者合并心理障碍的现象十分普遍,发生率高达30%~70%[5].且糖尿病足病患者较无并发症的糖尿病患者更易发生焦虑和抑郁[6].心理共病可增加糖尿病患者的风险,导致其自我护理能力降低及预后不良.因此,糖尿病患者的严重心理问题亟须医护人员干预与治疗[7].目前,我国临床普遍采用心理测评及咨询等方式收集患者心理相关资料.但单纯的心理测量往往忽视了不同患者在文化、认知等方面的差异[8].房树人绘画测验(house-tree-person drawing test,HTPD)是一种心理投射绘画测验,通过分析被试者画出的房子、树和人,评估其心理特点及可能存在的心理障碍[9].HTPD目前较多用于1型青少年糖尿病患儿的心理健康咨询及干预[10].我科于2019年8月2日收治1例糖尿病合并坏死性筋膜炎伴重度焦虑的患者,通过采用HTPD疗法,患者的焦虑得到有效缓解,现将治疗过程报道如下.
目的:调查维持性血液透析患者营养状态,并探讨循证理论下营养管理护理对其预后的影响.方法:选择2018年2月1日~2020年2月29日收治的86例维持性血液透析患者,分析其临床资料,采用Logistic回归分析维持性血液透析患者发生营养不良的独立危险因素,并对发生营养不良者予以循证理论下营养管理护理,比较干预前后营养状态[采用主观综合性营养量表(SGA)]、血压、体质量指数(BMI)、血钙及血磷的变化,并随访记录并发症发生情况.结果:86例维持性血液透析患者发生营养不良共46例,营养不良率为53.49%;营养不良组在年龄≥60岁、无家属陪伴、无医保、病程≥9年、月收入<3500元、透析时间≥40个月、透析频率≤2次/周、促红细胞生成素使用频率≤2次/周占比高于营养正常组(P<0.05);根据Logistic回归分析显示,年龄≥60岁、无家属陪伴、透析时间≥60个月、透析频率≤2次/周、促红细胞生成素使用频率≤2次/周是维持性血液透析患者发生营养不良的独立危险因素(P<0.05).46例营养不良患者实施循证理论下营养管理护理发现,SGA评分、血压、BMI均较干预前改善(P<0.01),血钙、血磷均较干预前降低(P<0.01);46例患者中低血压、高血压、心律失常、失衡综合征等并发症发生率仅为8.69%.结论:维持性血液透析患者营养不良风险高,年龄、家属陪伴情况、透析时间频率、促红细胞生成素使用频率等均能影响患者营养状态;循证理论下营养管理护理能改善营养不良状态,对改善其钙磷代谢、降低并发症发生风险具有重要意义.
目的 观察分析预见性护理程序在脑出血护理中的效果.方法 选取本院2019年1月-9月收治的104例脑出血患者,按数字随机表法分为对照组52例用常规护理方法.实验组52例在对照组基础上应用预见性护理程序方法.分析两组患者的并发症,泌尿系统感染、口腔感染、肺部感染以及压疮等发生率、平均住院时间与护理操作时间、护理总满意率.结果 实验组脑出血患者的泌尿系统感染、口腔感染、肺部感染以及压疮等并发症发生率、和平均住院时间、护理操作时间以及护理总满意率均优于对照组,对比存在显著差异(P<0.05).结论 预见性护理程序在脑出血护理中的应用效果显著.
Reduced graphene oxide/ZnFe2O4 (rGO/ZnFe2O4) nanocomposite was facile prepared and applied as magnetic sorbent for the extraction of estrogens including 17 beta-estradiol, 17 alpha-estradiol, estrone and hexestrol from water, soil, and fish samples prior to HPLC analysis. The rGO/ZnFe2O4 nanocomposite was characterized by scanning electron microscope, Fourier transform-infrared spectroscopy, X-ray diffraction, and vibrating sample magnetometer. The experimental parameters affecting the efficiency of magnetic solid-phase extraction (MSPE) including the amount of material, extraction time, pH, temperature, desorption solvents, desorption time, and desorption solvent volume were investigated respectively. With the developed method, good linearity was observed in the range of 0.05-500 ng/mL with the correlation coefficients (R-2) between 0.9978 and 0.9993. The limits of detection (S/N = 3) and limits of quantification (S/N = 10) were achieved at 0.01-0.02 ng/mL and 0.05 ng/mL, respectively. The enrichment factors were calculated as the range of 241-288. Using rGO/ZnFe2O4 nanocomposite as the sorbent, the developed MSPE followed by HPLC analysis, was applied to analysis of estrogens in river water, soil and fish samples. The method has the potential application in the extraction and preconcentration ultra trace compounds in complex matrices, such as environmental and biological samples.
目的 研究小干扰RNA(siRNA)介导低表达基质细胞衍生因子-1(SDF-1)、血管内皮生长因子(VEGF)对血管内皮细胞增殖和凋亡的影响.方法 采用体外实验培养人血管内皮细胞,细胞分别经siRNA介导下调表达SDF-1、VEGF处理,随机分为空白对照组(未进行任何转染)、阴性对照组(转染非特异性对照)、转染SDF-1-siRNA组、转染VEGF165-siRNA组.各组分别经相应处理.通过免疫印迹法测定细胞SDF-1和VEGF165蛋白的表达水平,用MTT实验测定人血管内皮细胞增殖情况,用流式细胞仪检测细胞凋亡情况.结果 空白对照组与阴性对照组SDF-1和VEGF165蛋白表达水平的比较,无显著差异(P>0.05);相比空白对照组和阴性对照组,转染SDF-1-siRNA组细胞中SDF-1蛋白的表达水平显著下调(P<0.01),VEGF165蛋白表达并无显著差异(P>0.05);相比空白对照组和阴性对照组,转染VEGF165-siRNA组SDF-1和VEGF165蛋白表达水平均显著下调(P<0.01);转染VEGF165-siRNA组SDF-1蛋白表达水平显著高于转染SDF-1-siRNA组,VEGF165蛋白表达水平显著低于SDF-1-siRNA组(P<0.01).空白对照组与阴性对照组细胞细胞凋亡率的比较,并无显著差异(P>0.05);转染SDF-1-siRNA组、转染VEGF165-siRNA组细胞增殖抑制率和细胞凋亡率均显著高于空白对照组和阴性对照组(P<0.01),转染VEGF165-siRNA组细胞增殖抑制率和细胞凋亡率均显著高于转染SDF-1-siRNA组(P<0.01).结论 SDF-1和VEGF165基因均可增强人血管内皮细胞增殖能力,阻滞细胞凋亡,从而参与糖尿病血管病变的病理过程,同时该病理过程中VEGF165具有调节SDF-1表达水平的作用.
Here, we report a feasible strategy to construct a Z-scheme Bi2O3/SnO2 composite via a facile hydrothermal method. Photocatalytic activity of the as-prepared photocatalysts was evaluated through photocatalytic degradation of bisphenol A (BPA) under simulated sunlight irradiation. A photocatalytic efficiency of 93.42% was achieved by 1Bi(2)O(3)/SnO2 during 60 min of irradiation, which was 14.97- and 2.54-fold greater than the photocatalytic efficiency of pure SnO2 and Bi2O3, respectively. In addition, the composite was effective in the photocatalytic decomposition of several antibiotics. Degradation conditions, such as the catalyst dosage, the initial concentration, and pH of BPA aqueous solution, were also investigated. The enhanced photocatalytic degradation efficiency of Bi2O3/SnO2 composites was attributable to the increased absorption of light, as well as the effective separation of photo-induced carriers that resulted from the formation of a Z-scheme heterojunction. The main active species in photocatalytic degradation of BPA by Bi2O3/SnO2 composite were h(+) and O-1(2). This highly efficient and broad-spectrum photocatalyst renders it a promising candidate for water pollution control.
Objective:To observe the effect of stromal cell-derived factor-1 (SDF-1) on the proliferation of rat hair follicle keratinocytes cultured in vitro. Methods:From June 2019 to January 2020, immunofluorescence was used to detect the expression of keratin 14(K14) , a marker of rat hair follicle keratinocytes purchased from iCell Bioscience Inc.Cell count kit-8 (CCK-8) assay was used to detect the proliferation of hair follicle keratinocytes treated with 0, 5, 10, 25, 50 μg/L SDF-1 for 24 h, the proliferation of hair follicle keratinocytes treated with 10, 25 μg/L SDF-1 for 48 h, and the proliferation of hair follicle keratinocytes treated with 10, 25 μg/L SDF-1 combined with 50 nmol/L CXCR4 antagonist plerixafor (AMD3100) for 24 h. 5-Ethynyl-2’-deoxyuridine (EdU) detected the proliferation of hair follicle keratinocytes in control group (0 μg/L SDF-1), 25 μg/L SDF-1 group and 25 μg/L SDF-1 adding 50 nmol/L AMD3100 group for 24 h. T test was used to analyze the results. Results:Cell immunofluorescence showed that more than 95% of the cells were positive for K14 staining. CCK-8 experiment showed that compared with SDF-1 at concentration of 0 μg/L (103.04±0.99), 5 μg/L of SDF-1 had no effect on the proliferation activity of hair follicle keratinocytes (100.67±4.59, t=0.905, P>0.05). SDF-1 at concentrations of 10, 25, 50 μg/L promoted the proliferation of hair follicle keratinocytes (111.44±4.95, 124.59±2.08, 125.11±0.48, t10=3.221, P10<0.05; t25=8.624, P25<0.01; t50=8.464, P50<0.01). Compared with SDF-1 at concentration of 25 μg/L, the proliferation capacity of hair follicle keratinocytes is reduced at concentration of 10 μg/L ( t=5.043, P<0.01), and has no significant change at concentration of 50 μg/L ( t=0.199, P>0.05). The proliferative effect of SDF-1 at the concentrations of 10 μg/L and 25 μg/L for 48 h (98.26±5.24, 112.39±2.30) was reduced compared with the effect of 24 h (111.44±4.95, 124.59±2.08, t10=3.167, P10<0.05; t25=6.816, P25<0.01). After 10 μg/L and 25 μg/L of SDF-1 were added to 50 nmol/L AMD3100, the proliferation of hair follicle keratinocytes was obviously inhibited (84.20±1.06, 98.51±0.92, t10=9.325, P10<0.01; t25=19.900, P25<0.01). The results of EdU further verified that SDF-1 can promote the proliferation of hair follicle keratinocytes (SDF-1 group: 38.59±0.33, control group: 22.45±2.59, t=10.700, P<0.01), and AMD3100 can restrain the proliferative effect of SDF-1 (22.12±1.96, t=10.700, P<0.01). Conclusion:SDF-1 in a certain concentration range can promote the proliferation of hair follicle keratinocytes, and this proliferative effect can be suppressed by AMD3100.
目的 探讨miR-374a-3p对氧化低密度脂蛋白(ox-LDL)诱导的人脐静脉内皮细胞(HUVEC)损伤及炎症反应的影响及作用机制.方法 实验分为ox-LDL组、对照(Con)组、miR-NC组、miR-374a-3p组、anti-miR-NC组、anfi-miR-374a-3p组、ox-LDL+miR-NC组、ox-LDL+miR-374a-3p组、ox-LDL+si-NC组、ox-LDL+ si-Syk组、ox-LDL+ miR-374a-3p+ pcDNA3.1组、ox-LDL+miR-374a-3p+pcDNA3.1-Syk组.实时荧光定量PCR(RT-qPCR)检测miR-374a-3p表达水平;蛋白质印迹(WesternBlot)检测脾酪氨酸激酶(Syk)、Bcl-2相关X蛋白(Bax)、B细胞淋巴瘤/白血病-2(Bcl-2)蛋白表达水平;四甲基偶氮唑盐比色法(MTT)检测细胞活性;流式细胞术检测细胞凋亡;酶联免疫吸附试验(ELISA)检测肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量水平;荧光素酶报告实验检测miR-374a-3p和Syk的靶向关系.结果 ox-LDL作用的HUVEC中miR-374a-3p低表达,Syk高表达;细胞活性显著降低,细胞凋亡率显著升高,TNF-α、IL-6含量显著升高(P<0.05).过表达miR-374a-3p和抑制Syk表达抑制ox-LDL作用的HUVEC凋亡和炎症因子TNF-α、IL-6的释放.miR-374a-3p靶向调控Syk,过表达Syk逆转了miR-374a-3p对ox-LDL作用的HUVEC损伤及炎症因子释放的抑制作用.结论 过表达miR-374a-3p可抑制HUVEC凋亡和炎症因子的释放,保护ox-LDL引起的HUVEC损伤及炎症反应,其机制可能与Syk有关.
目的:探讨基于4R危机理论风险管理对颅内动脉瘤术后持续腰大池引流患者格拉斯哥昏迷量表(Glasgow coma scale,GCS)评分及脑脊液的影响.方法:选取2017年1月-2019年7月本院收治82例颅内动脉瘤手术后患者为研究对象,按随机数字表法分为对照组和观察组,每组41例.对照组予以常规干预,观察组在对照组的基础上予以基于4R危机理论风险管理.比较两组颈项强直时间、引流管留置时间、头痛持续时间及术后并发症发生情况.比较两组术后1、7、14 d的GCS评分及脑脊液压力.结果:观察组颈项强直时间、引流管留置时间、头痛持续时间均短于对照组(P<0.05).术后7、14 d,观察组GCS评分均高于对照组,而脑脊液压力均低于对照组(P<0.05).观察组并发症发生率低于对照组(P<0.05).结论:基于4R危机理论风险管理应用于颅内动脉瘤术后持续腰大池引流患者可以缓解症状及促进清醒,调控脑脊液压力,降低并发症.
Objective To investigate the effects of advanced glycation end products (AGEs) on the proliferation and migration of fibroblasts and its mechanism.Methods 20 diabetic foot wounds and 20 non-diabetic foot normal foot wounds were collected from the Department of Wound Repair,Liyuan Hospital Affiliated to Tongji Medical College,Huazhong University of Science and Technology from January 2018 to December 2018.Western blotting was used to detect the expression of AGEs,Vimentin and β-Catenin in non-diabetic and diabetic foot wound tissue.Prepuce fibroblasts were cultured and identified.Fibroblasts were treated for 48 hours with 0,100,300,500,1 000,2 000 μg/L AGEs,300 μg/L BSA as control group.Cell counting kit-8 (CCK-8),Transwell and Western blotting were performed after treatment and T-test analysis results.Results Compared with normal tissues,AGEs in diabetic foot tissues were significantly higher (t =3.564,P < 0.05),while Vimentin expression was lower (t =8.325,P < 0.05),but the expression of β-Catenin was higher than that in normal tissues (t =5.738,P <0.05).The morphology of the isolated and purified cells was spindle-shaped.Vimentin was expressed in more than 95% of the cells by immunofluorescence assay and identified as fibroblasts.When the concentration of AGEs increased from 100,300 μg/L to 500,1 000 μg/L,the proliferation (t =4.336,PA500/BSA <0.01;t =19.562,PA1000/BSA < 0.01;t =42.373,PA2000/BSA < 0.01;t =5.380,PA1000/A500 < 0.01;t =23.726,PA2000/A,000 < 0.01) and migration (t =12.117,PA300/BSA < 0.01;t =14.484,PA500/BSA < 0.01;t =21.940 PA1000/BSA < 0.01) ability of fibroblasts decreased gradually,showing a concentration-dependent.In addition,the expression of Wnt3α and Wnt7α in fibroblasts increased significantly after AGEs concentration was added more than 300 μg/L,and the expression of β-Catenin was significantly higher than that in control group,but not in BSA group.Conclusion AGEs can decrease the proliferation and migration of fibroblasts,which is related to Wnt/β-Catenin signaling pathway.
The efficient removal of heavy metals from aqueous environment is imperative and challenging. A novel ternary composite constructed of diaminopyridine polymers, graphene oxide, and ferrite magnetic nanoparticles was designed by a facile in situ polymerization strategy for the removal of Pb(II) from aqueous solution. Detailed characterization of morphological, chemical, and magnetic properties was employed systematically to confirm the formation of the composite material. Batch adsorption experiment studies suggested that the composite was an excellent adsorbent for Pb(II) which was easily collected after use via exposure to an external magnetic field for 30 s. The effects of different parameters such as solution pH, adsorbent dosage, contact time, initial Pb(II) concentration, temperature, and co-existing ions were examined. The maximum adsorption capacity at pH = 5 was estimated to be 387.2 mg g−1 at 298 K by the Langmuir isotherm model, accompanied by favorable adsorption recyclability according to the investigation of regeneration experiments. Thermodynamic studies revealed that the Pb(II) adsorption via our ternary composite was endothermic and spontaneous. The corresponding removal performance for effluent containing Pb(II) from the battery industry was successfully examined. The present results indicated that our designed adsorbent is beneficial to the practical Pb(II) removal in wastewater purification.
We report a facile hydrothermal method to synthesize a novel mesoporous graphitic carbon nitride (MCN)@NiCo2O4 nanocomposite, which can be used as a solid phase microextraction coating for high efficiency extraction and preconcentration of trace polychlorinated biphenyls and polycyclic aromatic hydrocarbons in human serum samples.
