Objective: This study aimed to investigate the effect of Cili extracts in the treatment of enteritis and diarrhea. Methods: The RAW 264.7 cells were treated with different concentrations (1.25, 2.50, 5.00, 10.00, 20.00 μg/mL) of Cili extracts, and then the cell viability was determined by MTT method. RAW264.7 cells were treated with different concentrations of Cili extracts, and then lipopolysaccharide (LPS) was used to induce the cell inflammation model. The content of NO in cell culture supernatant was detected by Griess method. The UC mice model was induced by 3% DSS. The changes of signs during the modeling and drug treatment were observed, and the disease activity index (DAI) was evaluated. The colon length, colon wet weight index and spleen weight were measured. HE staining was used to observe the pathological morphology of colon. The protein expression of Keap1 and Nrf2 in colon tissues were detected by western blot and immunohistochemistry. Results: The concentration of Cili extract ≤10 μg/mL had no significant effect on cell viability (P>0.05). Cili extract inhibited the release of NO, and the inhibitory effect was more obvious with the increase of drug concentration (P<0.05 or P<0.01). Compared with the model group, the colon length of Cili extract group was longer, and the colon wet weight index, spleen weight and DAI fraction were decreased (P<0.05 or P<0.01). HE staining showed that Cili extract had a better protective effect on the intestinal mucosa of UC mice. Western blot and immunohistochemistry showed that Cili extract down-regulated the expression level of Keap1 and up-regulated the expression level of Nrf2. Conclusion: Cili extract had good anti-inflammatory activity in vitro and expressed therapeutic effect on UC mice.
目的:基于谷氨酸钠诱导人神经母细胞瘤细胞(SH-SY5Y)细胞损伤构建的模型,观察从枫香槲寄生中提取得到丁香脂素的保护,并探讨其作用机制.方法:采用谷氨酸钠构建SH-SY5Y细胞损伤模型,实验分为正常组,损伤模型组(谷氨酸钠50 mmol· L-1,谷氨酸钠50 mmol·L-+ DMSO),丁香脂素组(6.25,12.5,25 μmol· L-),通过细胞计数法,细胞形态学观察,Annexin V-FITC/碘化丙啶(PI)细胞凋亡检测、活性氧(ROS)检测、线粒体膜电位以及蛋白免疫印迹法(Western blot)等方法,评价丁香脂素对谷氨酸钠诱导的神经兴奋性损伤的神经保护活性.结果:与正常组比较,模型组的细胞存活率明显降低(P<0.01),ROS显著升高(P<0.01),线粒体膜电位显著降低(P<0.01),聚腺苷二磷酸-核糖聚合酶(PARP),PARP1蛋白表达显著降低(P<0.01),细胞的凋亡率显著增大(P<0.01);与模型组比较,丁香脂素组(6.25,12.5,25 μmol· L-)呈浓度依赖性增加细胞的存活率(P <0.01),减少ROS的积累(P<0.01),显著恢复线粒体膜电位的变化(P<0.01),显著上调PARP,PARP1蛋白(P<0.01),显著降低细胞凋亡率(P<0.01).结论:提示丁香脂素对谷氨酸钠诱导的SH-SY5Y神经细胞的兴奋性损伤具有显著的保护活性,其作用机制可能是通过抗氧化应激,修复线粒体功能及DNA损伤等通路来显著降低谷氨酸钠诱导的神经细胞凋亡.
For the development and utilization of H.dulcis composition and content constituents of fatty acids,conjoint fatty acids and content of free fatty acids were identified by GC-MS.The antioxidant activities of the H.dulcis constituents of fatty acids in vitro were tested by the Fenton and DPPH free radical(DPPH).The results showed that 15fatty acids were identified in seed oil of H.dulcis,in which the main components were the unsaturated fatty acids with linoleic acid and(Z)-9-Octadecenoic acid(oleic acid).The weight rate of conjoint and free fatty acids was 48.5∶1,in which 15 fatty acids exist in free acids with linolenic acid as main constituent and 6conjugated type fatty acids with a major component oleic acid.The results of antioxidant acidities showed that seed oil had does-effect relationship at the scavenging activities of hydroxyl radical and DPPH radical in vitro.When the concentration of sample was 6mg/mL,the hydroxyl radical scavenging rate was 91.7%;the concentration of sample was 15 mg/mL,DPPH radical scavenging rate was 91.3%,and the above results were better than those of olive oil.
[Objective] To determine the chemical constituents of volatile oil from root of Gaultheria yunnanensis(Franch) Rehd,so as to provide evidence for the development of Gaultheria yunnanensis(Franch) Rehd root. [Method] By employing GC-MS,the compositions of volatile oil from root of Gaultheria yunnanensis(Franch) Rehd were analyzed. [Result] Seven compounds were obtained from the volatile oil from Gaultheria yunnanensis(Franch) Rehd root,of which,contents of three ones were relative higher,i.e. methyl salicylate(89.815%) ,methyl cyclopentene(6.258%) and cyclohexane(2.693%) . [Conclusion] Volatile oil from root of Gaultheria yunnanensis(Franch) Rehd is rich in methyl salicylate,and this study provides evidence for the development of Gaultheria yunnanensis(Franch) Rehd root.
目的研究刺梨单体CL1体外抗人子宫内膜腺癌(human adenocarcinoma endometrium cell.line JEC)作用。方法通过细胞形态学改变、MTT法和细胞生长曲线绘制法检测CL1的抗JEC作用。运用全自动分析仪检测细胞培养上清液中乳酸脱氢酶(Lactate dehydrogenase,LDH)含量;通过检测NBT还原法分析CL1对JEC细胞分化的影响;采用AO/EB荧光染色法和FCM检测CL1对肿瘤细胞周期和细胞凋亡的影响。结果生长曲线、细胞形态学观察和MTT检测显示,CL1对JEC细胞增殖的抑制作用呈时间和剂量依赖性;与阴性对照组比,随CL1剂量的增加,NBT阳性细胞比率增加,LDH含量逐渐降低;与阴性对照组比,CL1能诱导JEC细胞凋亡,并阻滞JEC细胞于G0-G1和G2-M期。结论 CL1具有体外抗JEC作用,其机制可能与其细胞毒性、诱导细胞分化、诱导细胞凋亡以及抑制细胞增殖有关。
Aroma components in seven kinds of pears were investigated.Aroma components in pear were determined and analyzed by method of solid-phase microextraction (SPME) coupled with gas chromatography-mass spectrometry(GC/MS).Fourteen components including ten esters and four alkenes were identified from seven kinds of pears.Major aroma components in pear were esters,minor were alkenes acetic acid hexyl ester and alpha-farnsene were the two main constituents.It was cleared that aroma components of esters and alkenes should be a guideline for quality evaluation of pears.
目的 对青风藤化学成分进行分离、鉴定.方法 青风藤干燥藤茎用95%乙醇溶液加热回流提取,减压回收乙醇,所得浸膏利用硅胶柱色谱、反相RP-18柱色谱及重结晶等方法进行分离及纯化,并通过~1H-NMR、~(13)C-NMR、EI-MS及理化常数对分离化合物进行结构鉴定.结果 从青风藤提取物中分离得到11个化合物,分别鉴定为尖防己碱(I)、蝙蝠葛波酚碱(II)、dauriporphinoline(III)、蝙蝠葛宁(Ⅳ)、dauricumine(V)、6-O-demethyl- menisporphine(VI)、acutuminine(Ⅶ)、(--)-8-oxotetrahydrothalifendine(Ⅷ)、(--)-oxoisocorypalmine(IX)、四氢表小檗碱(X)、四氢巴马亭(Ⅺ).结论 化合物VI、VII、Ⅺ为首次从青风藤中分离得到.
Qualitative analysis of flavoring compositions in Xijiu liquor was performed by use of supercritical fluid extraction (SFE) and gas chromatography-mass spectrometry (GC-MS). 23 kinds of flavoring compositions had been separated with their relative content as 99.3 % of total ion peaks. The main flavoring compositions covered ethyl caproate, ethyl lactate, hexanoic, acetic acid, ethyl acetate, isopentanol, and chavicol etc.

目的:建立便捷有效的筛选方法用于抗β淀粉肽损伤PC12细胞活性物质的筛选.方法:以不同浓度的β-淀粉肽与PC12细胞作用不同的时间,通过MTT法检测细胞存活率,确定合适的条件,建立淀粉肽损伤PC12细胞模型.以维生素E作为阳性对照,对选取的20个样品进行筛选.结果β-淀粉肽损伤PC12细胞模型稳定性较好,发现银杏叶粗提物具有抗β-淀粉肽损伤PC12细胞的活性.结论:该工作为开展抗β淀粉肽损伤PC12细胞活性物质的筛选提供了简便有效的筛选模型.

土党参Campanumoea javanica Bl.又名奶参、土羊乳、白洋参、对月参、土参、野党参、柴党参、浮萍参、孩儿葛、川人参,是桔梗科金钱豹属植物,生于山坡草地、灌丛中.分布于贵州松桃、遵义、毕节、兴义、织金等地苗乡.此外,我国南部和西南部也有分布.民间以根入药,有补气、止血、通乳的作用,用于虚劳内伤、肺虚咳嗽、脾虚泄泻、乳汁不多、小儿遗尿等症,为贵州苗族习用药材[1].本实验从土党参的根茎中分离出6个化合物,根据理化常数和波谱数据分析,分别鉴定为:党参苷Ⅰ(tangshenoside Ⅰ,Ⅰ)、丁香苷(syringin,Ⅱ)、5-羟基-4',6,7-三甲氧基黄酮(5-hydroxy-4',6,7-Trimethoxyflavone,Ⅲ)、5-羟基-4',7-二甲氧基黄酮(5-Hydroxy-4',7-dimethoxyflavone,Ⅳ)、蒲公英赛醇乙酸酯(taraxerol acetate Ⅴ)、无羁萜酮(friedelin,Ⅵ).

Objective: To study the chemical constituents of Stenoloma chusanum (L) Ching. Methods: The chemical constituents of AcOEt fraction from Ethanol Extraction were separated and purified with chromatographic techniques, and tested with mass spectrographic technique and nuclear magnetic resonance. Their structures were elucidated on the basis of special data analysis. Results: Three compounds were obtained as apigenin(1), syringic acid (2), and protocatechuic acid (3). Among them, apigenin (1) was reported in this plant for the first time. Conclusions: apigenin (1), syringic acid (2), and protocatechuic acid (3) are the main bioactive constituents that can inhibit bacteria. [
目的筛选贵州民族中草药对HBsAg的作用.方法选取211种贵州省民族中草药提取物分别与HBsAg 37℃作用1小时,通过ELISA双抗体夹心法分别检测HBsAg与药物作用前后的含量,计算HBsAg经药物作用后下降的百分率,筛选出有效的药物.结果211种药物中筛选出24种药物有抗HBsAg作用,占总数的11.4%.结论贵州民族中草药中有部分药物对HBsAg的抑制作用.
目的 结构修饰熊果酸 ,制备其衍生物以观察抗肿瘤活性。方法 对熊果酸的 1、2位和 3位进行氧化 ,制备其衍生物 ,并以Hela和HL - 6 0为模型进行了生物活性测试。结果 制备了 6个衍生物 ,其中 4个化合物为新化合物 ;熊果酸结构修饰物的活性均较熊果酸弱。结论 熊果酸经过结构修饰、改造后 ,生物活性受到影响。
