dObjective: Comparison of the antibacterial activity of APS with four β-lactam antibiotics against different Staphylococcus aureus (SA) and speculation of the antibacterial mechanism of action of APS by network pharmacological approach. Methods: The MICs of APS and β-lactam antibiotics on MSSA-4 (non-membrane producing sensitive bacteria), MRSA-6 (non-membrane producing resistant bacteria), MSSA-11 (membrane producing sensitive bacteria) and MRSA-12 (membrane producing resistant bacteria) were determined by serial dilution method. The effects of drug resistance and biofilm on MIC were compared by one-way multivariate ANOVA. The structure of APS compound was obtained from PubChem database, and target fishing was carried out using PharmMapper database, and the screened targets were imported into STRING database to establish PPI network diagram and capture node information to establish “compound-target-target interaction” network. Metascape platform was used to perform GO enrichment analysis and KEGG pathway enrichment analysis on key targets to predict their antibacterial mechanism of action. Results: The MIC of MSSA-4, MRSA-6, MSSA-11 and MRSA-12 by APS were 125, 125, 62.5 and 62.5 μg/mL, respectively. Analysis of variance showed that bacterial drug resistance and biofilm had little effect on the antibacterial effect of APS, and had better antibacterial activity against membrane producing bacteria. And four β-Lactam antibiotics were easily affected by bacterial drug resistance and biofilm, resulting in the decrease of the sensitivity of the tested bacteria to antibiotics. 123 potential antimicrobial action targets of APS were obtained by network pharmacology, and the protein interaction network had suggested that ALB, AKT1, MMP9, MAPK1, CASP3, IGF1, MAPK8, HRAS, BCL2L1, ESR1 might be its core targets for the biological functions involved mainly include bacterial response, bacterial adhesion regulation, protein domain specific binding and so on, and mainly acted on focal adhesion, amino sugar and nucleotide sugar metabolism, drug metabolism and other pathways. Conclusion: Bacterial resistance and biofilm did not affect the antibacterial effect of APS. APS had the characteristics of multi-target and multi-pathway to affect the bacterial biofilm and exert antibacterial effects.

Objective: To investigate the antibacterial mechanisms of total flavonoids(TF) from Ampelopsis grossedentata on Staphylococcus aureus.Method: The antibacterial mechanisms were studied by scanning electron microscope for ultrastructural morphology of TF(155 mg.L-1).Microbial adhesion method was used for determination the effect of TF(155 mg.L-1) on cell-surface hydrophobicity of hydrocarbons.Cell permeability and conductivity colorimetric method were employed for the determination of dehydrogenase activity of S.aureus after being treated by different concentrations of total flavone(310-38.75 mg.L-1) from Ampelopsis grossedentata of Guangxi.Result: As the concentration increased and time prolonged,bacterial cell ultrastructural change was evident,characterized by cell surface uneven,warty protrusions,intercellular boundaries blurred,cell diameter increase.TF could significantly reduce the bacterial cell-surface hydrophobicity(P < 0.01) and increase in permeability of the bacterial cell.Activity of bacterial dehydrogenase was obviously inhibited by TF.Conclusion: The antibacterial mechanisms of TF from Ampelopsis grossedentata on S.aureus are related with reducing surface hydrophobicity,increasing permeability of the bacterial and inhibiting dehydrogenase activity of S.aureus.
Objective To observe the effect of serum containing Guantong Decoction and its separate prescriptions on C-myc gene expression in rat vascular smooth muscle cells(VSMCs) stimulated by angiotensin Ⅱ(Ang Ⅱ).Methods The rat VSMCs were stimulated by Ang Ⅱ and were treated by serum containing Guantong decoction and its separate prescriptions.C-myc gene expression was determined by reverse-transcription polymerase chain reaction(PCR) method.Results The C-myc / β-actin ratio in the AngⅡ stimulation group was increased,the difference being significant compared with that in the blank control group(P<0.01).The C-myc / β-actin ratio in Guantong Decoction group and three separate prescriptions groups was decreased,the difference being significant compared with that in Ang Ⅱ stimulation group(P <0.01).Guantong Decoction group had lower c-myc / β-actin ratio than the three separate prescription groups(P<0.05 or P<0.01),but the differences were insignificant among three separate prescription groups(P >0.05).Conclusion Inhibition of VSMCs proliferation by Guantong Decoction is probably related with down-regulation of C-myc gene expression,and Guantong Decoction has better inhibitory effect than the separate prescriptions.
Objective To investigate the activity of total flavonoids(TF) of Guangxi Ampelopsis grossedentata alone and in combination with β-lactam antibiotics against clinically important bacteria,including Staphylococcus aureus(SA),Methicilli-resistant Staphylococcus aureus(MRSA),Klebsiella pneumoniae and Beta paratyphoid bacilli.Methods The minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) of TF on these bacteria were tested by microdilution.Fractional inhibitory concentration index(FICI) and growth curve of TF combined with β-lactam antibiotics against SA and MRSA were measured by checkerboard method.Results The MIC of TF alone on SA and MRSA was 77.50-155.00 μg·mL-1.The combination of TF and β-lactam antibiotics such as oxacillin,tienam,cefoxitin showed synergistic effects with FICI less than 0.5.However,the additive effect of the combination was absent from cefazolin or cefepime,where the FICI ranged from 0.5 to 1.0.Conclusion TF in combination with β-lactam antibiotics presents strong antibacterial effect against MRSA in vitro.
Objective: To study the influence of serum containing Guantong Fang and its compositions on apoptosis in rat vascular smooth muscle cells.Method: SD rats in each group were administered corresponding doses: Guantong Fang group(23.75 g.kg-1),Huoxue Huayu(HXHY) group(8.75 g.kg-1),Yiqi Yangyin(YQYY) group(5 g.kg-1),Qingre Huatan(QRHT) group(10 g.kg-1) for 7 days.Then th serum containing drugs was prepared,VSMCs were divided into: blank control group(5% normal rat serum),model group(5% normal rat serum and 1 × 10-6mol.L-1 AngⅡ),Guantong Fang group(5% Guantong Fang Containing serum and 1 × 10-6mol.L-1AngⅡ),HXHY group(5% HXHY Containing serum and 1 × 10-6mol.L-1AngⅡ),YQYY group(5% YQYY Containing serum and 1 × 10-6mol.L-1AngⅡ),QRHT group(5% QRHT Containing serum and 1 × 10-6mol.L-1AngⅡ),After incubated 48h,cell apoptosis was evaluated by laser flow cytometry.Result: in the model group the early apoptosis rate of vascular smooth muscle cell was(0.36 ± 0.14) %,late apoptosis(1.02 ± 0.86) %,and in blank control group: early apoptosis(2.22 ± 0.43) %,late apoptosis(1.29 ± 0.57) %;In the Guantong Fang group: early(23.4 ± 0.95) %,late(9.1 ± 0.69) %,in HXHY group: early(8.08 ±0.54) %,late(18.6 ±1.48) %,in YQYY group early(16.9 ±0.79) %,late(4.38 ±0.90) %,in QRHT group early(5.9 ± 0.99) %,late(11.5 ± 0.7) %.Conclusion: Guantong Fang can inhibit the proliferation of VSMC and induce apoptosis;and effect of Guantong Fang group is better than its compositions.
Objective:To observe the effects of Guangxi curcuma extract on human hepatic stellate cells and to learn its impacts on the cell proliferation and apoptosis.Methods:To incubate the human hepatic stellate cells(HSC-LX2) with the drugs for 48h.Then to measure the cell proliferation inhibition rate by MTT,to detect the situation in different apoptosis phase by flow cytometry(ANNEXIN V-PE/7-AAD double staining).Results:Low,medium and high concentrations of Guangxi curcuma extract can inhibit proliferation of HSC-LX2 cells.The inhibition rates in 48h were 15.4%,49.1%,58.3% respectively;IC50 in 48h was 45μmol / L.All concentrations can induce early and late apoptosis of HSC-LX2 cells.The percentages of total apoptosis cells in 48h were 20.7%、29.8%、34.4%.Conclusion:Guangxi curcuma extract can inhibit the proliferation of HSC-LX2 cells,induce apoptosis so as to interfere with course of the liver fibrosis.This shows the dose-dependent effect and these effects of Guangxi curcuma extract in middle and high-dose groups were significantly stronger than that of colchicine group and shell liver softening tablets group.
Objective:In this study,Human gastric cancer cell line SGC-7901 cells were used as a model and treated with the aqueous extract of actinidia chinensis medicated sera,to investigate the antioxidant mechanism via inducing the produce of phase 2 enzymes.Method: The normal SD rats were randomly divided into 4 groups: blank control group,high,middle and low dose groups.The actinidia chinensis aqueous extract was gavaged in different doses,respectively and water was gavaged in the blank control group and twice a day for 14 days.The blood was collected from abdominal aorta to prepare serum containing drugs,then which were given to the SGC-7901 cells separately at different time.Immunohistochemistry staining was used to detect the localization of nuclear factor-E2 related factor2(Nrf2) in the SGC-7901cells,RT-PCR was used to detect the mRNA level of phase 2 enzymes.Result: The immunohistochemistry detecting showed that the expression of the nuclear Nrf2 was increased and the area broadened significantly(P0.05) in th groups after administration with different doses of serum containing drugs in the SGC-7901 cells.The results of RT-PCR showed that the mRNA expression levels of the phase 2 enzymes glutamate cysteine ligase catalytic subunit(GCLC) were induced(P0.05),and the mRNA expression levels of glutathione-S-transferase(GST)-P1 and GST-T1 were enhanced significantly(P0.01),which was no obvious effect on GST-M1 and heme oxygenase-1(NQO1).Conclusion: The Actinidia chinensis aqueous extract can increase nuclear translocation of Nrf2 and increase the phase Ⅱ enzyme mRNA expression.The Actinidia chinensis aqueous extract has the function of antioxidant.

[Objetive] To explore the antibacterial effect of aqueous extract of Veronicastrum caulopterum(Hance) Yamazaki in vitro.[Methods] Serial dilution method was used to MIC experiment.[Results] The aqueous extract of Veronicastrum caulopterum(Hance) Yamazaki had antibacterial effects on staphylococcus aureus and escherichia coli especially staphylococcus aureus but hardly have effect on candida albicans.[Conclusions] This research offers a experimental bases to the antibacterial effects of aqueous extrat from Veronicastrum Caulopterum(Hance) Yamazaki and provides a theoretical basis to seek pharmacodynamic antibacterial composition in it.
目的探讨三叶黄合剂对正常小鼠和免疫抑制小鼠细胞免疫功能的影响。方法用三叶黄合剂给正常小鼠和环磷酰胺(CTX)免疫抑制小鼠灌胃,然后用称重法检测小鼠胸腺、脾重量,用中性红比色法检测巨噬细胞活性,用R IA法检测白细胞介素-2(IL-2)含量,用MTT法检测NK细胞活性和T淋巴细胞增殖功能。结果三叶黄合剂对正常小鼠胸腺、脾重量和细胞免疫功能无显著影响,对CTX的免疫抑制作用有显著的抵抗作用,能增加免疫抑制小鼠胸腺和脾的重量,增强巨噬细胞的吞噬作用,促进IL-2分泌,增加NK细胞活性和T淋巴细胞增殖功能。结论三叶黄合剂对正常小鼠的细胞免疫功能无显著影响,但能抵抗CTX对小鼠细胞免疫功能的抑制作用。
Maf family proteins are a large number of transcriptional factors which include a characteristic basic region and a leucine zipper(b-Zip)motif.The Maf proteins form homodimers or heterodimers with other family transcription factors with b-Zip proteins present in the nucleus and bind to Maf recognition elements(MARE)in DNA.They play key roles in the proliferation,migration,differentiation of mammalian embryogenesis,development of lens,differentiation of Th2 lymphocyte,expression of the insulin and apoptosis.
OBJECTIVE To investigate the effect of Daganqing on the immunity in the normal or immunosuppressive mice.METHODS Daganqing was oral in normal or immunosuppressive mice and then the weight of immune organs,the numbers of hemolysin,antibody form cells,IL-2,the phagocytic activity of macrophage and the activities of T lymphocyte and NK cell were detected by the methods of weighting method,haematolysis experiment,QHS,RIA,MTT,respectively.RESULTS Daganqing could promote the weight of immune organs and the numbers of hemolysin and antibody form cells,and promote the IL-2 level in the serum,promote phagocytic activity of macrophage,T lymphocyte proliferation and NK cell activity in the immunosuppressive mice but did not have significant effect in the normal mice.CONCLUSION The results suggest that Daganqing has up-regulation on humeral immunity and cells-mediated immunity in the immunosuppressive mice but hasn't significant effects in the normal mice.
[目的]研究甘蔗叶水提物的降血糖、抑菌作用。[方法]采用连续稀释法对甘蔗叶水提物进行体外抗菌实验;降血糖试验采用四氧嘧啶诱导的化学性糖尿病模型,观察甘蔗叶水提物对小鼠血糖的影响。[结果]甘蔗叶水提物在体外对金黄色葡萄球菌、大肠埃希菌均有作用,其中对金黄色葡萄球菌的作用比较明显,但对白假丝酵母菌基本没有作用;药后1周甘蔗叶水提物高、中剂量组(与模型组比较)有延缓小鼠血糖的升高趋势。[结论]甘蔗叶水提物具有明显的抗菌作用;对四氧嘧啶诱导的糖尿病小鼠无明显的降糖作用,但对小鼠血糖有一定抑制作用。
医学微生物学作为基础医学中一门重要学科,是医学教育的主干学科,也是连接基础医学和临床医学的桥梁,其理论教学效果的好坏直接影响到后续课程的学习.这门课程涉及的知识量大,内容繁杂,不易记忆,学生极容易在学习上产生厌学情绪.如何改变这一现状,提高理论教学效果?为此我们进行了教学方法探讨,发现采用多元化教学方法,加强与生活、临床的联系,激发学生学习欲望是提高医学微生物学理论教学效果的有效方法.
该文认为在微生物学检验技能训练中,抓住细菌培养技术这个重点,整合实验教学内容,使技能训练呈连贯性,增强学生动手能力,加强实验报告管理,做到实验与考核相结合,综合主人学生实验技能,有利于提高教学质量。
目的:探讨绿素酊对正常小鼠和免疫抑制小鼠中性粒细胞免疫功能的影响.方法:用绿素酊给正常小鼠和环磷酰胺(CTX)免疫抑制小鼠灌胃,检测白细胞数、中性粒细胞游走功能和吞噬功能.结果:绿素酊对正常小鼠中性粒细胞免疫功能无显著影响(P>0.05),但能显著抵抗CTX对小鼠的免疫抑制作用,能增加白细胞数、促进中性粒细胞游走并增强其吞噬功能(P<0.01).结论:绿素酊对正常小鼠的中性粒细胞免疫功能无显著影响,但能抵抗CTX对小鼠中性粒细胞免疫功能的抑制作用.
目的:探讨百色灵芝对正常小鼠和免疫抑制小鼠非特异性免疫功能的影响.方法:用不同剂量赤灵芝液给正常小鼠和环磷酰胺免疫抑制小鼠灌胃,检测小鼠免疫器官重量和巨噬细胞吞噬功能.结果:赤灵芝水煎剂对正常小鼠非特异性免疫功能无显著影响,但能显著增加环磷酰胺免疫抑制小鼠的胸腺和脾脏重量及胸腺指数和脾指数,增加巨噬细胞吞噬功能.结论:赤灵芝对正常小鼠免疫功能无显著影响,对环磷酰胺免疫抑制小鼠的非特异性免疫功能具有正调节作用.
[Objective]To investigate the effect on nonspecific immunity of Lusu Tincture in the mice.[Methods] Lusu Tincture was perfused in normal and immunosuppressive mice, then the weight of immune organs and the function of phagocytes were detected .[Results]Lusu Tincture could increase the weight of immune organs and promote the phagocytic function of macrophage in the abdominal cavity in the CTX immunosuppressive mice; but couldn't produce a significant effect on the weight of immune organs and promote the phagocytic function of macrophage in the normal mice.[Conclusion]The results suggest that Lusu Tincture has a positive regulation on nonspecific immunity in the immunosuppressive mice, but no significant effect on nonspecific immunity in the normal mice.
