Anti-tuberculosis drug-induced liver injury (ATB-DILI) is a common serious adverse event observed during the clinical treatment of tuberculosis. However, the molecular mechanisms underlying ATB-DILI remain unclear. A recent study has indicated that ferroptosis and lipid peroxidation may be involved in liver injury. Therefore, this study aimed to investigate the role of ferroptosis in the molecular mechanisms underlying ATB-DILI. Our results showed that anti-TB drugs induced hepatocyte damage in vivo and in vitro and inhibited BRL-3A cell activity in a dose-dependent manner, accompanied by increased lipid peroxidation and reduced antioxidant levels. Moreover, ACSL4 expression and Fe2+ concentration significantly increased following anti-TB drug treatment. Interestingly, anti-TB drug-induced hepatocyte damage was reversed by ferrostatin-1 (Fer-1, a specific ferroptosis inhibitor). In contrast, treatment with erastin (a ferroptosis inducer) resulted in further elevation of ferroptosis indicators. Additionally, we also found that anti-TB drug treatment inhibited HIF-1α/SLC7A11/GPx4 signaling in vivo and in vitro. Notably, HIF-1α knockdown significantly enhanced anti-TB drug-induced ferroptotic events and the subsequent exacerbation of hepatocyte damage. In conclusion, our findings indicated that ferroptosis plays a crucial role in the development of ATB-DILI. Furthermore, anti-TB drug-induced hepatocyte ferroptosis was shown to be regulated by HIF-1α/SLC7A11/GPx4 signaling. These findings shed new light on the mechanisms underlying ATB-DILI and suggest novel therapeutic strategies for this disease.
Objective: To detect of gene expression and genotype of the ataxia telangiectasia mutated (ATM) from coal workers' pneumoconiosis (CWP) , It is explored whether CWP is related to ATM gene. Methods: In October 2020, the relevant information of 264 subjects who received physical examination or medical treatment in the Department of occupational diseases of Guiyang public health treatment center from January 2019 to September 2020 was collected. Through the occupational health examination, 67 healthy people with no history of exposure to occupational hazards were selected as the healthy control group; The coal miners with more than 10 years of coal dust exposure history and small shadow in the lung but not up to the diagnostic criteria were the dust exposure control group, a total of 66 people; The patients with the same history of coal dust exposure and confirmed stage I were coal worker's pneumoconiosis stage I group, a total of 131 people. The expression of ATM was detected by QRT PCR. ATM rs189037 and rs1801516 were genotyped by massarray. Results: There was significant difference in the expression of ATM among the groups (P<0.05) ; Compared with the healthy control group, the expression of ATM in the dust exposed control group was significantly increased (P<0.05) . With the occurrence and development of CWP, the GG of rs189037 wild type decreased, the GA of mutant heterozygote and AA of homozygote increased, but the difference was not statistically significant (P>0.05) ; Rs1801516 wild type GG and mutant heterozygote GA had no significant changes (P>0.05) . There were significant differences in age, neutrophils and basophils among rs189037 groups (all P<0.05) . There were no significant differences in blood pressure, eosinophils, lymphocytes, monocytes, smoking and drinking history among rs189037 groups (all P>0.05) . Compared with wild-type GG, the or of mutant heterozygotes and homozygotes increased, but the differences were not statistically significant (P>0.05) . Conclusion: ATM gene may be one of the early activation genes of CWP and rs189037 may be the functional loci which affects gene expression. ATM gene is related to inflammatory response, Neutrophils and basophils have an impact on the development of CWP.
目的:研究不同栓塞剂对咯血的靶血管行末梢栓塞的临床疗效。方法:研究样本为2019年3月至2021年4月期间在贵阳市公共卫生救治中心接受治疗的89例咯血患者,根据使用栓塞剂的不同分为A组、B组及C组,分析三组患者的治疗情况。结果:A、B、C组一周内的总有效率差异无统计学意义,但治疗1个月及12个月内A组的复发率最高,C组的复发率最低,B组介于两组之间。B组1例患者术后有脑梗塞的并发症,其余患者未见严重并发症发生。结论:栓塞剂以三种方式治疗咯血患者,短期内均可获得较理想的疗效,但中长期GSP的复发率更高,PVA可能导致短期疗效欠佳或异位栓塞,GSP联合PVA行末梢栓塞治疗更安全、更具优势。
目的 探讨乙肝肝纤维化患者肝组织局部粘着斑激酶(Focal Adhesion Kinase,FAK)、磷脂酰肌醇-3-激酶(Phosphatidylin-ositol-3-kinase,PI3K)的表达及其临床意义.方法 100例肝组织均来自于贵阳市公共卫生救治中心2017年11月30日-2018年9月30日期间收治的乙肝肝纤维化患者,采用免疫组化法及半定量分析方法对肝组织FAK、PI3K表达情况加以确定.比较不同肝纤维化病理分期下FAK、PI3K阳性表达积分、FAK、PI3K与肝纤维化分期之间的相关关系、乙肝肝纤维化组织中FAK、PI3K的阳性表达与临床病理之间的关系.结果(1)随着肝纤维化分期不断增大,FAK及PI3K阳性表达积分随之而显著增大,且S1-S4期FAK及PI3K阳性表达积分均显著高于S0期(P<0.05-0.01);(2)采用直线相关性分析方法,结果显示:FAK、PI3K与肝纤维化分期之间呈显著的相关性(P均<0.05);(3)乙肝肝纤维化组织中FAK、PI3K的阳性表达与卫星结节、淋巴转移之间呈显著的相关性(P均<0.05).结论 乙肝肝纤维化患者肝组织中存在FAK、PI3K过表达的情况,其可作为临床评估乙肝肝纤维化程度的重要基础因子,从而为临床诊疗乙肝肝纤维化提供一定的参考依据.
Objectives To estimate anaemia prevalence and the associated factors among hospitalised people living with HIV (PLHIV) receiving antiretroviral therapy (ART).Design A cross-sectional study.Setting PLHIV receiving ART and hospitalised in a specialised hospital for infectious disease in Guizhou Province, Southwest China, between 1 January 2018 and 31 March 2021.Participants A total of 6959 hospitalised PLHIV aged ≥18 years and receiving ART were included in this study.Primary and secondary outcome measures Anaemia was diagnosed as a haemoglobin concentration <120 g/L for non-pregnant females and <130 g/L for males. Mild, moderate and severe anaemia were diagnosed as below the gender‐specific lower limit of normal but ≥110 g/L, 80–110 g/L and <80 g/L, respectively.Results The prevalence of anaemia was 27.5%, and that of mild, moderate and severe anaemia was 9.2%, 12.2% and 6.1%, respectively. Results from multivariate logistic regression showed that females had increased odds of anaemia (adjusted OR (aOR)=1.60, 95% CI: 1.42 to 1.81) compared with males. Widowed or divorced inpatients (anaemia: aOR=1.26, 95% CI: 1.08 to 1.47; severe anaemia: aOR=1.52, 95% CI: 1.16 to 1.97) and thrombocytopenia inpatients (anaemia: aOR=4.25, 95% CI: 3.54 to 5.10; severe anaemia: aOR=4.16, 95% CI: 3.24 to 5.35) had increased odds of anaemia and severe anaemia compared with their counterparts. Hepatitis C was associated with increased odds of severe anaemia (aOR=1.80, 95% CI: 1.11 to 2.92).Conclusions Anaemia was prevalent among hospitalised PLHIV. Female sex, those widowed or divorced, and thrombocytopenia were associated with increased odds of anaemia, and those widowed or divorced, thrombocytopenia and hepatitis C were associated with increased odds of severe anaemia. Determination of anaemia predictors, early detection and timely management of anaemia are crucial to prevent anaemia progression.
目的 分析获得性免疫缺陷综合症(AIDS)合并隐球菌性脑膜炎(CM)患者预后影响因素.方法 137例AIDS合并CM患者根据随访结束时的生存状况分为生存组及死亡组,采用单因素方差分析两组患者的年龄、性别、影像学检查(胸部CT、头颅CT或MRI)、临床症状(头痛、发热、呕吐及意识障碍)、血常规、C反应蛋白、红细胞沉降率、降钙素原、血(1,3)-β-D葡聚糖、细菌内毒素、血液隐球菌抗原(CrAg)阳性率、CD4+T淋巴细胞、HIV病毒载量、脑脊液氯化物、脑脊液葡萄糖及脑脊液蛋白的差异,将有差异的因素(P<0.05)纳入COX比例风险回归模型,确定AIDS合并CM的独立预后影响因素;采用Kaplan-Meier法测定患者生存率,Log-rank检验各影响因素差异的显著性.结果 137例AIDS合并CM患者中,生存组111例、死亡组26例,单因素分析显示患者年龄、CD4+T淋巴细胞、HIV病毒载量、血(1,3)-β-D葡聚糖及血液CrAg阳性是AIDS合并CM患者预后影响因素(P<0.05);多因素COX回归分析显示患者年龄(HR=4.375,95%CI为1.855~10.319,P=0.001)、CD4+T淋巴细胞(HR=4.110,95%CI为2.351~12.553,P=0.013)、HIV病毒载量(HR=5.040,95%CI为1.119~22.699,P=0.035)及血液CrAg阳性(HR=8.135,95%CI为2.353~26.113,P=0.001)是AIDS合并CM独立预后影响因素;Log-rank检验及Kaplan-Meier生存曲线图显示患者年龄≥50岁、CD4+T淋巴细胞<50个/μL、HIV病毒载量≥1×107拷贝/L及血液CrAg阳性可导致AIDS合并CM患者生存率明显下降(P<0.05).结论 年龄、CD4+T淋巴细胞、HIV病毒载量、及血液CrAg阳性是影响AIDS合并CM独立预后影响因素.
目的:探讨血浆(1-3)-β-D葡聚糖对获得性免疫缺陷综合症(Acquired Immune Defi-ciency Syndrome,AIDS)合并隐球菌性脑膜炎(Cryptococcal meningitis,CM)患者预后评估的价值.方法:回顾性分析2015年1月1日至2020年12月31日贵阳市公共卫生救治中心收治的66例AIDS合并CM患者的临床病例资料,按照1年内生存状况分为生存组(56例)和死亡组(10例),收集两组患者性别、年龄、传播途径、临床症状(头痛、发热、呕吐及意识障碍),头颅影像学检查、入院时实验室检查(CD4+T淋巴细胞计数、血浆HIV-RNA病毒载量、外周血血红蛋白、外周白细胞计数、外周血中性粒细胞计数、C反应蛋白、血沉、PCT、脑脊液葡萄糖、脑脊液氯化物及脑脊液蛋白、脑脊液培养、脑脊液涂片、血液隐球菌培养、血浆(1-3)-β-D葡聚糖浓度)及抗真菌治疗后第一次血浆(1-3)-β-D葡聚糖浓度.多因素Logistic回归分析AIDS合并CM患者死亡危险因素.受试者工作特征曲线ROC分析血浆(1-3)-β-D葡聚糖浓度预测AIDS合并CM患者死亡的价值.结果:单因素分析显示PCT、CD4+T淋巴细胞、血浆(1-3)-β-D葡聚糖浓度是AIDS合并CM患者死亡危险因素(P均<0.05).多因素Logistic回归分析显示高浓度血浆(1-3)-β-D葡聚糖是AIDS合并CM患者独立死亡危险影响因素(P<0.05).ROC曲线分析显示,抗真菌治疗前血浆(1-3)-β-D葡聚糖为65.63pg/mL时患者死亡的曲线下面积最大(0.787,95%CI 0.665~0.910),其敏感度和特异度较高(分别为90.0%和62.5%,P<0.05).结论:血浆(1-3)-β-D葡聚糖对AIDS合并CM患者预后具有较高的评估价值,当血浆(1-3)-β-D葡聚糖>65.63pg/mL提示患者死亡风险高.

The micro-elimination strategy is an effective approach to rapidly reduce the incidence and mortality of specific populations infected with hepatitis C virus (HCV). This article combines the current status of hepatitis C prevention and treatment in Guizhou Province, and introduces the current domestic and foreign hepatitis C micro-elimination models. It is worth mentioning that the Guizhou Provincial Medical Quality Control Center for Infectious Diseases jointly formulated the "Guizhou Province Chronic Hepatitis C Health Education Standards" to improve the awareness of health care workers and patients about the disease, increase the screening rate, and increase the patients willingness to receive treatment and accomplish the World Health Organization’s goal of eliminating hepatitis C threat by 2030.
目的:探讨S100蛋白超家族A6(S100 family member A6,S100A6)在肝细胞癌(hepatocellular carcino-ma,HCC)中的表达及临床意义.方法:利用Oncomine生物信息学在线分析网站,挖掘分析大型癌症公共数据癌症基因组图谱(TCGA)及GEO(Gene Expression Omnibus)中肝癌S100A6 mRNA的表达信息,基于TCGA中的肝癌数据,利用GEPIA、Kaplan-Meier Plotter进行患者生存分析.结果:在Oncomine数据库中,前后共收集了337项不同类型的研究结果,关于S100A6基因表达有统计学差异的研究结果有30个,其中S100A6基因表达降低的研究有12项,表达增高的研究有18项.共有4项研究涉及S100A6在HCC癌组织和正常肝组织中的表达,与正常肝组织相比,S100A6在HCC中高表达(P=4.58E-4).此外,S100A6 表达量与HCC总体生存率存在相关性,高表达S100A6的患者总体生存率较差(P=0.048、P=0.005 1 ).进一步结合TNM分期进行分析发现,S100A6表达在Ⅳ期最高.S100A6表达与Ⅰ(P=0.31 )、Ⅱ(P=0.2)期患者预后无相关性,在Ⅲ期患者中,高表达S100A6的患者总体生存率较差(P=0.007 1 ).结论:本研究通过对基因芯片数据库中肿瘤相关基因信息的深入挖掘和剖析,发现S100A6 基因在肝细胞癌组织中呈现高表达,并且与肝细胞癌预后关系密切,尤其对HCC Ⅲ期患者有预后指导意义.
目的 观察邻苯二甲酸二丁酯(dibutyl phthalate,DBP)与苯并[a]芘[benzo (a) pyrene,BaP]染毒对大鼠肝脏毒性作用,并采用基准剂量(benchmarkdose,BMD)法估算DBP、BaP亚慢性染毒致肝功能指标改变的BMD、基准剂量下限值(benchmark dose lower bound,BMDL),为DBP、BaP亚慢性肝毒性的风险评估提供资料.方法 选择初断乳健康SD大鼠160只,分为空白对照组、溶剂对照组(玉米油)、50 mg/kg DBP、250 mg/kg DBP、1 mg/kg BaP、5 mg/kg BaP、50 mg/kg DBP+1 mg/kg BaP、250 mg/kg DBP+5 mg/kg BaP剂量组.连续染毒90d,采用高效液相色谱法检测尿液中主要代谢产物含量,观察大鼠肝脏组织病理学变化,测定大鼠血清中肝功能指标[丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、总胆红素(TB)、直接胆红素(DB)],分析尿液中主要代谢产物含量与肝功能指标相关关系,采用BMDS 3.2软件计算BMD和BMDL.结果 大鼠尿液中DBP和BaP的主要代谢产物分别为邻苯二甲酸单丁酯(mono-butyl phthalate,MBP)与1-羟基芘(1-hydroxypyren,1-OHP).各染毒组大鼠尿液中MBP及1-OHP含量均明显高于空白对照及溶剂对照组(P<0.05);DBP与BaP联合染毒组尿液中的代谢产物含量与相应单独染毒组比较,差异无统计学意义(P>0.05).病理学检查结果显示各染毒组大鼠肝脏组织均出现了不同程度的双核新生细胞增多、炎性浸润、小叶间胆管及纤维化增生等,且DBP与BaP联合染毒剂量组较相应单独染毒剂量组的损伤更为明显.肝功能检查显示,各染毒组的ALT、AST、ALP、TB水平均明显高于对照组,尤以联合染毒高剂量组的升高最为明显(P<0.05).DBP单独染毒组大鼠尿液中MBP含量与血清ALT、ALP及TB水平之间存在明显的正相关关系(P<0.05),BaP单独染毒组大鼠尿液中1-OHP含量与血清ALT及DB水平之间存在明显的正相关关系(P<0.05),DBP与BaP联合染毒组大鼠尿液中MBP和l-OHP含量均与ALT、AST、ALP和TB水平之间存在明显的正相关关系(P<0.05).以各染毒组大鼠尿液中MBP及1-OHP含量为暴露标志,ALT活力为效应标志,DBP单独染毒时,ALT的尿MBP的BMD和BMDL分别为1.87、1.83 mmol/mol Cr;BaP单独染毒时,ALT的尿1-OHP的BMD和BMDL分别为0.21、0.18 μmol/mol Cr.DBP和BaP联合暴露时,ALT的尿液中MBP的BMD和BMDL分别为1.85、1.83 mmol/mol Cr,ALT的尿液中1-OHP的BMD和BMDL分别为0.21、0.18 μ.mol/molCr.结论 DBP、BaP亚慢性染毒可导致肝组织损伤、肝功能指标异常;血清ALT活力可作为DBP与BaP暴露致肝损伤的早期监测指标.DBP、BaP暴露致大鼠肝脏毒性的生物接触限值分别为1.85 mmol/mol Cr、0.18μmol/mol Cr.
目的 探讨CHB肝纤维化的危险因素及FS检测肝硬度值(LSM)的影响因素.方法 选取CHB患者305例,测定LSM值与病理分期的相关性;进行肝纤维化多因素logistic回归分析及LSM值影响因素多元线性回归分析.结果 肝组织活检显示肝纤维化分期S1期至S3期分别为235例,28例,42例;肝纤维化各期LSM值与病理分期具有相关性;Logistis回归分析结果 显示性别、ALP、CHE、DBIL、FB、PLT、WBC、HGB、HBeAg是肝纤维化的独立危险因素;多元线性回归分析结果 显示CHE、DBIL、PTA、PLT是具有统计学意义的独立预测因子.结论 FS对慢性乙肝肝纤维化评估与肝组织活检评估肝纤维化程度具有较好的吻合性;性别、ALP、CHE、DBIL、FB、PLT、WBC、HGB、HBeAg是肝纤维化的危险因素;FS检测需考虑患者CHE、DBIL、PTA、PLT等因素的影响来判断其检测结果 .
目的 沉默乙型肝炎病毒(HBV) PS1反式激活蛋白2基因(PS1TP2)对肝癌细胞HepG2增殖和凋亡的影响.方法 荧光定量PCR (RT-PCR)用于检测PS1TP2基因在不同肝细胞系中基础表达水平.将PS1TP2小干扰RNA(siRNA PS1TP2)及对应的阴性对照小干扰RNA(siNC)作为干扰组和对照组分别瞬时转染至HepG2细胞中,培养48 h后,应用CCK-8试剂盒检测两组细胞的增殖活性水平;AnnexinV/7-AAD流式细胞术检测两组细胞凋亡程度;RT-PCR检测两组细胞中PS1TP2、B细胞淋巴瘤-2(Bcl-2)和Bcl-2相关X蛋白(Bax)基因mRNA表达水平;Western blot检测两组细胞中Bcl-2、Bax、人腺苷酸活化蛋白激酶(AMPK)、雷帕霉素靶蛋白(m-TOR)蛋白水平并计算Bcl-2与Bax的比例.结果 PS1TP2基因在肝癌细胞HepG2中的表达水平明显高于正常肝细胞L02.干扰组HepG2细胞的增殖速率明显慢于对照组.与对照组相比,干扰组的HepG2细胞中Annexin V+细胞数明显升高(P<0.05),HepG2细胞中Bcl-2 mRNA表达量明显下降(P<0.05),Bax mRNA表达量明显上升(P<0.05);同样在干扰组的HepG2细胞中Bcl-2、m-TOR蛋白水平明显下降(P<0.05),而Bax、AMPK蛋白水平则明显升高(P<0.05).结论 干扰掉PS1TP2基因可经过线粒体途径促进HepG2细胞凋亡,可能是通过活化AMPK-m-TOR途径而抑制其增殖.
目的 探讨静脉射频消融联合泡沫硬化剂在下肢静脉曲张上的临床治疗效果.方法 从2016年2月到2019年2月来我院进行下肢静脉曲张治疗的患者中随机选择98例作为基本研究对象,将其平均分成实验组和对照组,每组49例.对照组采取泡沫硬化剂注射治疗,实验组应用射频消融联合泡沫硬化剂进行治疗,在治疗一段时间之后分析比较两组患者的术后住院时间、术后并发症和一年后彩色多普勒随访情况.结果 在手术之后的三个月、六个月,实验组患者的VCSS评分、CIVIQ评分要比对照组优秀,差异显著(p<0.05),具有统计学研究意义.结论 射频消融联合泡沫硬化剂注射临床上安全、有效、创伤小、恢复快等优点,是一种可部分替代传统手术的有效方法,值得在临床上推广使用.
建立临床决策支持系统(CDSS)和多学科协作(MDT)模式是提高医疗效率和质量的重要措施.从国内外CDSS和MDT发展现状进行概述和分析,总结其特点与不足,同时结合"医疗大数据资源"及"大数据技术"的时代背景,对发展适合我国国情的肝胆肿瘤智能化诊疗平台—"联合CDSS和MDT"提出了展望.
Objective To evaluate the curative effect of recombinant human Interferon α2b (rhIFN α2b) spray in hand,foot and mouth disease (HMFD).Methods In total,313 HMFD children were enrolled from Beijing Ditan Hospital affiliated to Capital Medical University,Tianjing Second People's Hospital,Hebei Children's Hospital,the Second Hospital affiliated to Wenzhou Medical College,Kunming Maternal and Child Health Hospital and Guiyang Public Health Treatment Center from March,2015 to February,2017.They were divided into rhIFN α2b group (148 cases) and ribavirin group (165 cases).The children in rhIFN α2b group were given with the rhIFN α2b spray,and those in ribavirin group were given with the ribavirin spray.Meanwhile,the children were given unified standard interventions for basic treatment.The curative effect and safety between two groups was compared.The t test was used for intergroup comparison and the Wilcoxon rank test was used for non-normal quantitative data.Results At the end of the follow-up period,all kids reached the recovery level,with 144 cases in the rhIFN α2b group and 164 cases in the ribavirin group.Fever,herpes and rashes all disappeared with 7-day follow-up.The total efficiency of the rhIFN α2b group measured at the 72h after treatment was 74.15%,which showed significant differences compared with the ribavirin group with 49.09 % efficiency (Z=4.44,P<0.01).As the secondary outcome measures,the complete disappearance time of fever and the immediate disappearance time of fever in the rhIFN α2b group were significantly shorter than those in the ribavirin group ([27.03±21.99] vs [33.21±26.71],t=-2.13;[23.56±13.96] vs [28.51±18.84],t=-2.07,both P<0.05).The appetite improvement and the disappearance times of oral herpes and rashes in the rhIFN α2b group were shorter than those in the ribavirin group,with significant differences (x2 =4.94,3.17 and 3.55,respectively,all P=0.000).No adverse event in both groups.Conclusions rhIFN α2b spray treatment in HMFD is proved significantly effective,particularly,it can evidently relieve fever symptoms and promote the disappearance of oral herpes symptoms,and no adverse event is observed throughout the study,which indicates a good safety of the rhIFN α2b spray.Clinical trial registration Chinese Clinical Trial Registry,ChiCTR-OIN-17013182.
Background: A complex network has been studied and applied in various disciplines. As network analysis and image processing are based on matrices, this research analysed the changes in the chromatin image of lymphocyte nuclei in peripheral blood of humans using a network motif and static features (static parameters), so as to complete image classification with network method. Methods: Image processing technology was used to establish a chromatin image network of a cell nucleus; Network analysis tool Pajek was used to display the special motif of an isolated structural hole with different symmetric line values; afterwards, the frequency of occurrence of this structural hole in patients with nasopharyngeal carcinoma and AIDS, and healthy people was computed. Then by applying the network static features as variables, the chromatin images of stained lymphocytes from the three groups of people were classified and recognised by using an extreme learning machine (ELM). Results: The frequency of occurrence of the isolated structural hole with different symmetric line values was adopted to distinguish the structures of the chromatins of peripheral blood lymphocytes in patients with nasopharyngeal carcinoma and AIDS, and healthy people. Similarly, The static features of the chromatin image network of a cell nucleus were applied to classify and recognise the morphological and structural changes in chromatins for peripheral blood lymphocytes in the three groups of people. Conclusion: The surface chemical and physical characteristics, as well as the polymerisation link status of biomacromolecules such as DNA, RNA, and protein in the lymphocyte nucleus change under certain pathological conditions. The change influences the combination of small molecular staining materials and any associated biomacromolecules. Therefore, various macroscopic and microscopic changes were found in the chromatin images of the cell nucleus. The microscopic changes include the variations of the extent of staining of chromatin in the nuclei, coarseness and direction of the texture therein, the size of stained conglomerations, etc. These changes contribute to the differences in chromatin image networks among the same type of cells across the three groups. Based on this, the model can be used to classify and reorganise certain diseases. The results prove that using complex network to analyse the chromatin structure of a cell nucleus is of significance.
Objective To understand the enterovirus(EV) pathogenic composition of Coxsackie virus(CV)A6 and A10 causing hand foot and mouth disesse(HFMD) in Guiyang area during 2015 to provide a basis for the prevention and treatment of HFMD in local area.Methods The specific primers were respectively designed according to CVA6 and CVA10 sequence published in GenBank.The fluorescence quantitative RT-PCR method system was established.The gene sequencing method was used for conducting verification.Then this method was used to detect the clinical samples from 607 cases of HFMD.Results A total of 607 samples of suspected HFMD were detected,the overall positive rate was 59.47 % (361/607),in which EV71 accounted for 7.25 % (44/607),CVA16 for 11.37%(69/607),EV71+CVA16 double positive accounted for 0.16% (1/607) and other EV for 40.69% (247/607).The positive samples of CVA6,CVA10 and CVA6+CVA10 detected by the established real time fluorescence RT-PCR were 11 cases,71 cases and 1 case.Conclusion CVA6 and CVA10 are the main pathogens causing new onset HFMD in Guiyang area and the CVA10 monitoring should be strengthened.
目的 探讨在肝癌细胞系HepG2中过表达或干扰XTP4表达对细胞迁移和侵袭能力的影响.方法 实验分对照组、过表达组和干扰组,将构建成功的XTP4质粒和化学合成的XTP4小干扰RNA(siRNA)瞬时转染入HepG2,培养48 h后,实时荧光定量PCR(RT-q PCR)和蛋白印迹(Western blot)检测细胞内XTP4 mRNA和蛋白表达;Snail和NF-κB以及上皮间质转化(EMT)相关分子的表达.并通过细胞划痕愈合实验、Transwell迁移侵袭实验观察细胞迁移侵袭能力.结果 成功重培养并提取出XTP4质粒;过表达组XTP4的转录水平和翻译水平均明显高于对照组(P<0.05);迁移和侵袭能力明显增加(P<0.05);下游分子NF-κB、Snail和MMP-9表达均增加(P<0.05);干扰组XTP4的转录水平和翻译水平较对照组均明显降低(P<0.05);迁移和侵袭能力也明显降低(P<0.05);NF-κB、Snail和MMP-9表达减少(P<0.05).结论 乙型肝炎病毒反式激活因子XTP4可促进HepG2迁移和侵袭,与NF-κB、Snail和MMP-9调节相关,EMT相关分子E-cadherin、N-cadherin可能参与其中.
Hepatitis B virus X (HBx) protein trans-activate gene XTP4 is a new gene found in recent years, which is related to the development of tumor. Many studies showed that XTP4 gene is abnormally expressed in breast cancer, prostate cancer, gastric cancer, colorectal cancer, ovarian cancer, non-small cell lung cancer, hepatocellular carcinoma and other abnormal tumor. The abnormal expression of XTP4 gene is closely related to cell hyperplasia, metastasis, invasion, tumor grade, staging and chemotherapy resistance. In view of the XTP4 gene playing an important role in various types of tumors or different stages of tumors and in order to facilitate the latter part of its function in-depth study, this paper will clarify the relationship of XTP4 gene structure, function and tumor occurrence and development.
