The severity of ischemic injury was evaluated by densitometry of brain samples stained with 2,3,5-triphenyltetrazolium chloride (TTC) on a rat model of cerebral ischemia/reperfusion (common carotid artery occlusion) and the neuroprotective activity of an extract of Astragalus membranaceus, Scutellaria baicalensis, and Phlojodicarpus sibiricus was assessed. Occlusion of the common carotid arteries led to a weakening of TTC staining of the brain tissue: densitometric indicators of the staining intensity for the cortex and striatum were lower than the corresponding indicators of sham-operated rats by 18.3 and 10.4%. The mean intensity of staining of brain samples did not differ in rats treated with the extract and sham-operated animals, which attested to its neuroprotective effect. The applied method is convenient for evaluation of the severity of ischemic brain damage at the early stages and screening potential neuroprotective agents.
The goal of the study was to determine quantitatively the content of phenolic compounds in the extract from roots of Astragalus membranaceus, Scutellaria baicalensis, and Phlojodicarpus sibiricus and to evaluate the antioxidant activity and anti-inflammatory effect of the extract. The quantitative content of the flavonoid complex (baicalin, rutin, quercetin, epicatechin) was determined. The antioxidant activity of the dry EtOH extract was evaluated using the DPPH test (in vitro). The antioxidant activity was shown to be directly dependent on the concentration of the extract. The half-maximal inhibitory concentration IC50 was 210.99 μg/mL. The anti-inflammatory effect of the tested agent was studied in the model of formalin edema in white rats. Administration of the extract to rats at doses of 50, 100, and 200 mg/kg gave edema values of 31.4, 19.2, and 16.6
Polysaccharide fractions were isolated from the aerial part of Serratula centauroides . The monosaccharide compositions of the polysaccharide fractions were determined by HPLC-UV. The quantitative contents of free sugars in the aerial part and subterranean organs of S. centauroides growing in the Republic of Buryatia were determined by capillary electrophoresis. The effect of mechanically activated milling of the aerial part of S. centauroides on the sugar yield was studied.
Цель работы состояла в определении количественного содержания фенольных соединений в экстракте из корней Astragalus membranaceus, Scutellaria baicalensis, Phlojodicarpus sibiricus, а также в оценке антиоксидантной активности и противовоспалительного действия экстракта. В ходе исследований экстракта определено количественное содержание комплекса флавоноидов (байкалина, рутина, кверцетина, эпикатехина). Антиоксидантную активность сухого этанольного экстракта оценивали с помощью ДФПГ-теста (in vitro). Установлено, что антиоксидантная активность находится в прямой зависимости от концентрации экстракта; IC50 составила 210,99 мкг/мл. Противовоспалительное действие испытуемого средства изучали на модели формалинового отека у белых крыс. При введении экстракта крысам в дозах 50, 100 и 200 мг/кг величины отека составили 31,4, 19,2 и 16,6 % от показателей (толщины лапки) до введения раствора формалина, что в 2 – 4 раза меньше контроля (p < 0,05) и сопоставимы с показателями при применении индометацина.

A chromatography–spectrophotometry method is developed for determining total ecdysteroids in terms of 20-hydroxyecdysone in Serratula centauroides (L.) and Fornicium unflorum (L.) raw materials, based on the purification of extracts from the plant raw materials with aluminum oxide and subsequent spectrophotometric determination relative to a 20-hydroxyecdysone reference sample. Ecdysteroids were determined in plants by HPLC; rhapontisterone and turkesterone are found in F. uniflorum for the first time.
We determined promising sources of phytoecdysteroids - Rhaponticum uniflorum (L.) and Serratula centauroides (L.), growing in the Buryat Republic. Biologically active substances were founded: ecdysteroids, phenolic compounds, polysaccharides, flavonoids, tannins, saponins, vitamins.

The composition of amino acids isolated from the phytopreparation has been studied. A total of 19 amino acids, including seven essential, were identidied. The mist significant concentrations were found for asparaginic acid, glutaminic acid, prolyne, cysteine, methionine, leucine, γ-aminobutyric acid, alanine, and arginine.
