The activation, injury, and dysfunction of endothelial cells are considered to be the initial key events in the development of atherosclerosis. Di (2-ethylhexyl) phthalate (DEHP), a prevalent organic pollutant, can cause damage to multiple organs. Polysaccharide of Atractylodes macrocephala Koidz (PAMK) is a bioactive compound extracted from A. macrocephala Koidz with various biological activities. This study investigates the protective effects of PAMK on porcine aortic valve endothelial cells (PAVEC) damaged by DEHP. PAVECs treated with DEHP alone or with PAMK showed reduced cell apoptosis and death in PAMK-pretreated cells. PAMK up-regulated Bcl-2 expression and down-regulated Bax protein, suppressing apoptosis. Flow cytometry analysis demonstrated that PAMK protected PAVECs from DEHP-induced damage. These findings suggest that PAMK inhibits cell apoptosis and protects against DEHP damage in endothelial cells.
Di(2-ethylhexyl) phthalate (DEHP) is a new environmental pollutant, which is widely used in plastic additives. DEHP and its metabolites pollute surface water and threaten the survival of fish. In order to investigate the mechanism of DEHP-induced apoptosis on grass carp hepatocytes, we treated grass carp hepatocytes with DEHP, and selected Atractylodes macrocephala Koidz (PAMK) to study its inhibitory effect on DEHP. The results showed that after DEHP exposure, apoptosis related proteins expression were increased significantly, leading to hepatocytes apoptosis. Moreover, AO/EB staining and Hoechst staining also showed that the number of apoptotic cells increased after DEHP exposure. It should be noted that PAMK simultaneous treatment could alleviate apoptosis induced by DEHP. The innovation of this study is that the application of Chinese herbal medicine (PAMK) to antagonize the damage of DEHP in fish was investigated for the first time. This study indicated that traditional Chinese medicine can also be used in fish production to reduce the accumulation of food-derived drugs.

药物靶点蛋白确定是研究开发新药的一个重要步骤.药物亲和反应的靶点稳定性(DARTS)技术是一种可以快速和直接识别小分子药物潜在靶点蛋白的技术,即靶蛋白与药物结合后,保护靶蛋白不被蛋白酶水解,进而维护靶蛋白的活性和功能.综述DARTS技术的起源、DARTS原理及优缺点、DARTS应用等方面研究进展,为进一步探索药物与靶蛋白的生物相关性、促进该方法在新药研发和靶标鉴定方面的应用提供参考.
猪呼吸系统疾病是猪场发病率和死亡率较高的疾病之一.四时阴阳是万物的根本,顺应四时阴阳的变化对猪体进行整体调节,将有利于猪场对呼吸系统疾病的防控.笔者从四时阴阳与猪呼吸系统疾病的关系及治未病的角度出发,探讨在生产实践中如何顺应四时气候的变化,应用中药方剂对猪呼吸系统疾病进行防控:春季扶正祛邪,解表理气;夏季清热泻火,抗菌消炎;秋季润肺止咳、化痰止喘;冬季帮助猪体御寒.
犬是恒温动物,体内的各种代谢反应是基于各种酶催化的生化反应.了解正常体温和正确处理许多与体温有关的并发症对临床医生有着重要的意义. 1 犬体温的变化 1.1 低体温 分娩之前(出现37.5℃左右低温期,10~24 h即分娩)、大多数中毒性疾病(有机磷中毒可能升高)、子宫蓄脓、大量输入低温液体、长期营养不良、麻醉、过低的环境温度、疾病后期、濒死期等都可能导致动物体温偏低.低温对机体既有益处也会产生复杂的甚至严重的问题.
To observe the effect of pIgR on Salmonella Enteritidis induced inflammation in jejunum and ileum in Chicken,7 day Hyline chickens were taken orally with Salmonella Enteritidis (SE) and killed after 1 d,3 d,7 d and 14 d.The mRNA expression of Toll-like receptor 4(TLR4),MyD88,TRAF6,NF-κB and pIgR was detected by real-time RT-PCR and pIgR protein level was detected by Western blot.The results showed that TLR4 signaling pathway was activated and mR-NA level of the pIgR in jejunum and ileum was enhanced (P<0.01) and protein level of the pIgR in jejunum and ileum was up-regulated by SE.The study proved that TLR4 signal pathway on mucosal cell surface of jejunum and ileum was activated and expression of pIgR was up-regulated and gut mucosal immunity of chicken was strenghtened.
子宫蓄脓症是一种常见的母犬生殖系统疾病.为建立子宫蓄脓症流行病学诊断与预防方案,笔者收集了2013-2014年前来东北农业大学动物医院就诊的50例病例,分析子宫蓄脓症的发生与年龄、发情情况、繁殖情况和品种之间的关系.结果表明:从年龄上看,开放型子宫蓄脓症的平均发病年龄为9.1岁,闭锁型为9.6岁;从生育史看,无生育史母犬的发病率高达84% (42/50),有生育史的母犬发病率相对较低为16% (8/50);从犬种上看,发病率最高的是中华田园犬,可达52% (26/50),京巴犬次之为16% (8/50);从体型上看,小型犬的发病率较高(86%,43/50),大型犬相对较低(14%,7/50).

根据GenBank中鸡pIgR基因序列和蛋白序列,设计引物,利用PCR技术扩增胞外配体结合区片段,构建原核重组表达质粒pET-32a (+)/pIgR,将重组质粒转化到大肠杆菌BL21(DE3),经IPTG的诱导表达融合蛋白,通过镍离子螯合柱纯化后免疫新西兰大白兔,获得兔抗鸡pIgR多克隆抗体,通过酶联免疫吸附试验(ELISA法)和Western Blot法检测抗体的效价和特异性.结果显示,成功制备特异性的鸡pIgR抗体,为研究鸡pIgR的功能提供有力依据.
Subclinical endometritis is a physiological inflammation that serves to clear persistent contaminants from the uterus. To investigate the alteration of antioxidant, such as vitamin E (VE) and vitamin C (VC), total oxidant capacity (TOC) and nitric oxide (NO) in cows with normal and subclinical endometritis (SCE), we examined the concentrations of NO, VC and VE, TOC and polymorphonuclear neutrophils (PMN) percentage in uterine secretion. The cows were divided into two groups, normal (n=20) and subclinical endometritis (SCE, n=60), based on endometrial cytology (presence of PMN±5%). Uterine secretion and blood were collected as described previously. Griess reaction was used to determine the concentration of NO. The concentrations of TOC, VC and VE were detected by a commercially available assay kit. The results showed that the concentrations of NO, TOC and PMN percentage were significantly higher (P<0.01, P<0.05, and P<0.01, respectively) in uterine secretion with SCE compared to those from normal; however, the levels of VC and VE were significantly lesser (P<0.01). In conclusion, the concentrations of NO, TOC, VC, VE and PMN percentage differed between normal and SCE cows. Meanwhile, the relationship between the concentration of NO and PMN percentage from uterine secretion in cows with subclinical endometritis were positively correlated. Consequently, these alterations in NO, TOC, VC, VE levels and PMN percentage contributed to as a diagnostic index of the uterine inflammation, with the aim to increase the reproduction of the cows and the decrease economic losses.
80只小鼠随机分为4组,采用背部皮下给药法,给药剂量分别为30 mg/kg·d,60 mg/kg·d和90 mg/kg·d,按0.01 mL/g体重连续给药一个月,对照组给予生理盐水.记录体重变化,检测血液生化指标,制作肝肾组织切片.结果表明,在高剂量组中,小鼠在第28天称重时,平均体重最轻,与其他3组比较,差异显著(P<0.05),GPT出现升高,部分小鼠的肝脏出现变性,甚至坏死,其他各组未见异常变化.
Pharmacokinetics of itraconazole injection was studied in healthy dogs.Subcutaneous injection was administrated to six dogs in a dose of 4mg/kg and the plasma concentration of itraconazole was detected by HPLC.The data was analyzed with 3p97 computer program.The results showed that the linearity was obtained over the range of 10~1000ng/mL and the recovery was 86.9%,88.4% or 89.2% when the addition concentration of itraconazole was 10 ng/mL/,100 ng/mL or 500 ng/mL respectively.The RSD was 2.8%,1.2% or 0.2% respectively.It was concluded that the method was simple,accurate and reliable,and could be used for the pharmacokinetic studies of itraconazole in dogs.
充气腹是进行腹腔镜手术操作时必要步骤之一.用于充气腹的气体种类比较多,如笑气、氦气、空气等是临床上曾经采用过的气体,众多学者对以上这些气体进行了大量的研究[1].结果表明,二氧化碳气体因其特有的优点被全世界众多腹腔镜外科医师们所采用,是目前临床上使用最广泛的充气腹气体.目前,关于二氧化碳气腹对机体呼吸系统、循环系统、肝肾功能及机体免疫方面影响的系列试验研究报道较多.
为了研究犬用抗真菌药伊曲康唑凝胶剂对皮肤的毒理学作用,探讨其使用安全性,试验采用一次给药的方法,将低、中、高3种浓度的伊曲康唑凝胶剂涂抹于大鼠完整皮肤及破损皮肤处,观察大鼠皮肤急性毒性反应;通过一次给药及多次给药方法将伊曲康唑凝胶剂涂抹于家兔完整皮肤及破损皮肤处,观察家兔皮肤刺激性反应;用伊曲康唑凝胶剂、空白凝胶及阳性致敏物涂抹于豚鼠完整皮肤处,观察比较皮肤过敏反应。结果表明:伊曲康唑凝胶剂对大鼠皮肤无毒性;对家兔皮肤刺激反应平均分值小于0.5,属于无刺激性药物;对豚鼠皮肤过敏反应分值为0,致敏率为0,具有弱致敏性。说明研制的伊曲康唑凝胶剂用于治疗犬皮肤真菌病是安全无毒的。
Using flow cytometry and ELISA method to detect the concentration of CD4+,CD8+ in blood and three immunoglobulins(IgA,IgG,IgM) in serm and in uterine secretion.The objective was to study the associativity between dynamic state change of each index and diagnosing endometritis.The results showed that CD8+ had the leading position in the later period,and CD4+/CD8+ was descent.The content of IgA,IgG and IgM in peripheral blood with infected dairy cows increased significantly(P0.05 or P0.01) and humoral immunity was activated in the infected cows when cell mediated immunity was lowered and that of CE group was variable more significantly than that of SE group.
This experiment was conducted to evaluate the concentration of NO in plasma and uterine secretions and ultrastructural change of uterine tissue in cows to explore the early diagnosis methods for endometritis cows.Five postpartum normal cows(N) as control group and ten subclinical endometritis cows(SE) and ten clinical endometritis cows( CE) were studied.Blood and uterine secretions samples were collected from a jugular vein and uterine cavity on 30 d after delivery.Endometrial samples were obtained from each group with endometrium sample collector.Transmission electronmicroscope was used to study the ultrastructural changes of the cows uterine tissue.The result showed that NO in CE and in SE were significantly higher than that of normal cows(N)(P0.01,P0.05);the array of endometrium cell,the integrality of cellular membrane and cellular structure displayed a corresponaing change.The higher the concentration of NO is,the severer the tissue changes are.The change of NO concentration in plasma and uterine secretions and ultrastructural changes of uterine tissue have some clinical significance for diagnosing endometritis.
To research the pathogenesis of WND the raccoon dog,the Blood Routine Test to WND and normal raccoon dog were done and analyzed by the method of Mann-whinney.Contrast to the normal raccoon dog,the Lymph%(P=0.049) and Gran%(P=0.031) of WND raccoon dog had declined and there were significant difference in those indexes between WND and normal raccoon dog(P0.05).The Lymph#(P=0.004),Min#(P=0.004),Min%(P=0.004),RBC(P=0.000),HCT(P=0.000),MCV(P=0.001),HGB(P=0.000),MCH(P=0.001),RBC(P=0.000),HCT(P=0.000),MCV(P=0.001),HGB(P=0.000) and MCH(P=0.001) of WND raccoon dog had declined and had extreme significant in those indexes between WND and normal raccoon dog(P0.01).WBC(P=0.130),Gran#(P=0.473),MCHC(P=0.915),RDW(P=0.831), RLT(P=0.915),PCT(P=0.852),MPV(P=0.202),PDW(P=0.749) of WND raccoon dog had no difference(P0.05) in those indexes between WND and normal raccoon dog.Based on the parameters of Red Cell Index,RBC,MCH,MCV and MCHV of WND raccoon dog,that the WND raccoon dog sickened for Hypochromic Microcytic Anemia was concluded.
To observe the changes of acute phase protein and IL-6 in the serum and uterine secretions in cattle with endometritis and search their significance on diagnose.Sixty-eight cows were selected and divided into three groups:N(Normal cows),SE(Subclinical Endometritis)and CE(Clinical endometritis).The concentration of acute phase proteins and cytokine in serum and uterine secretions of endometritis cows were detected.The results showed that the concentrations of CRP,Hp,SAA and IL-6 in the serum and uterine secretions of cows with clinical endometritis and subclinical endometritis were higher than that of control group(P0.01),and group CE was significant variable than group SE,but Hp and SAA were not detected in control group.The concentration of CRP,Hp,SAA and IL-6 were increased by different degree bacterial contamination in uterine and they can be the sensitive index for diagnosis with endometritis inflammation.
幼犬蜂窝织炎是3~16周龄幼犬的一种自发性皮肤病,也称为幼犬脓皮病或幼犬腺疫.本病的变化程度较大,可从浅表到严重,如果不予治疗,最终将留下大面积的皮肤疤痕甚至死亡(Rhea,2005).本病例是由用碘酒洗耳引起,最初表现过敏症状,之后逐渐严重,由耳部水肿、血疱、脓性渗出物,发展到颜面部肿胀,再到颈部、肩胛部淋巴结肿大,直至胸部、背部有完整脓疱,破溃流脓,为典型的幼犬蜂窝织炎.
