Objective To examine the expression of Krüppel-like factor-6 (KLF6) in human hepatocellular carcinoma (HCC) and adjacent non-cancerous tissues (ANCT),and further explore the effects and molecular mechanisms of KLF6 on proliferation and invasion of HCC cells.Methods The expression of KLF6 in 50 cases of HCC and corresponding ANCT was detected by immunohistochemical assay.In vitro human HCC SMCC-7721 cells were stably transfected with lentivirus-mediated KLF6 overexpression vector,and the expression levels of proliferating cell nuclear antigen (PCNA) and metalloproteinase-9 (MMP-9) were detected by Western blotting assay.The proliferative activity and invasive capability of HCC cells were detected by MTT and Transwell assays.Results The expression level of KLF6 protein was significantly downregulated in 50% (25/50) of HCC tissues compared with than in 72% (36/50) of ANCT.Western blotting results showed that the expression of PCNA and MMP-9 protein in KLF6 group were reduced by 81.5% and 58.2%,respectively (P < 0.01);MTT assay showed that cell proliferation activity significantly decreased in KLF6 group (P < 0.01);Transwell assay results showed that invasion and metastasis cell number in KLF6 group was significantly reduced [(58.33 ±6.45) cells vs.(98.50±11.33) cells;(58.33 ±6.45) cells vs.(103.25 ±12.17) cells,P< 0.01).Conclusion KLF6 is lowly expressed in HCC,and KLF6 overexpression can suppresses HCC cell proliferation and invasion,which mechanism is related to down regulating the expression of PCNA and MMP-9.

Objective To study the effect of three kinds of warming interventions(thin quilt,heating blanket inflatable and comprehensive warming) for patients' temperature changes during radical gastrectomy. Method 120 patients with radical gastrectomy were randomly divided into three groups: 40 patients in group with thin quilt,40 patients in group with heating blanket inflatable and 40 patients in group with two warming intervention(heating blanket inflatable and fluid warmers.The core temperatures were recorded after entering the operating room,after intubation,during incision,difference time during operation(30,60,90,120,150,and 180 minutes,end of operation) respectively.Result The change trend of three groups of patients' core temperature were significant differences(P<0.05).The core temperatures at time pointes of intubation,incision and difference time during operation were significant difference between thin quilt group and heating blanket inflatable,thin quilt group and comprehensive warming group(P<0.05).there were significant difference in core temperature at difference operation time 60 minutes later after operation among group.Conclusion Thin quilt is less effective for thermal protection.Heating blanket inflatable can prevent perio-perative hypothermia,but it's unstable for temperature maintaining.Comprehensive warming can effectively maintain stable temperature and prevent perioperative hypothermia.
膜瘤占颅内肿瘤的第二位,而矢旁脑膜瘤占颅内脑膜瘤的50%之多[1].脑膜瘤血供丰富,矢旁脑膜瘤以颈外动脉系统供血为主,且常因肿瘤侵犯颅骨使矢状窦处颅骨增厚,开颅过程中出血较多.我院2001年7月~2007年9月手术治疗矢旁脑膜瘤392例,应用高速气钻开颅48例,效果满意.
Objective To discuss the influence of chronic hypoperfusion on the basal forebrain cholinergic system and cognitive function of rats.Methods The chronic hypoperfusion model rats were made by permanent occlusion of bilateral common carotid arteries. The learning and memory performance of rats were examined using Y type maze task. while the basal forebrain changes in pathology observed by microscope and the level of choline acetyltransferase estimated by immunohistochemistry after permanent occlusion 1.2 and 4 months.Results The learning ability of hypoperfusion group was severely impaired and ChAT positive cells and fibers decreased significantly comparing with sham-group(P0.01).while there were a severe loss and necrosis of neuron and glial in basal forebrain during the process. Moreover. as time went on.the learning and memory performance remarkably correlated with the density of cholinergic systems of basal forebrain in hyperperfusion group.Conclusion Cerebral chronic hypoper-fusion induced by permanent occlusion of bilateral common carotid arteries can cause impairment of cholinergic neurons and decline of learning and memory performance of rats.So acetylcholine may play more important role in the process of cognitive impairment. The further pathogenesis remains to be studied.
