目的:观察五味子乙素(Sch B)对慢性阻塞性肺疾病(COPD)急性加重期细胞模型的影响,并初步探讨其作用机制.方法:用香烟烟雾提取物(CSE)联合脂多糖(LPS)作用于肺上皮细胞株A549,制备COPD急性加重期体外模型,并给予相应浓度的五味子乙素进行干预,通过MTT法测定各组细胞存活率、RT-PCR法测定细胞炎症因子白细胞介素8(IL-8)、环氧合酶2(COX-2)的mRNA水平;ELISA法测定细胞上清中IL-8、COX-2水平;Western blot法测定细胞磷酸化NF-κB蛋白水平.结果:4%CSE联合0.1μg/ml LPS作用24 h能够促进肺上皮细胞增殖,炎症因子表达明显上升,给予Sch B能够明显抑制CSE联合LPS对细胞的增殖作用,改善细胞炎症状态;降低损伤细胞IL-8、COX-2 mRNA表达水平;减少细胞上清中IL-8、COX-2含量;降低细胞中磷酸化NF-κB的蛋白表达水平.结论:Sch B可改善CSE联合LPS引起的肺上皮细胞炎症,其作用机制可能与抑制肺上皮细胞增殖,抑制炎症因子表达,发挥抗炎作用有关.该研究为Sch B应用于COPD急性加重期的临床治疗提供实验依据.
化学药物治疗简称化疗,是癌症治疗的有效手段之一,阿霉素(doxorubicin,DOX)、紫杉醇(paclitaxel,PTX)等用于化疗的细胞毒性物质可显著改善癌症患者的生活质量[1].但多数成药的细胞毒性成分对肿瘤细胞选择性差,且存在非靶向性的全身副作用.迄今为止,这些问题仍然是化学治疗的主要瓶颈[2].为了提高肿瘤细胞选择性,改善抗癌疗效,多功能纳米系统研发受到了普遍关注[3].常见的药物递送系统包括:胶束[4]、脂质体[5]、金属纳米粒子[6]、二氧化硅粒子等[7].
脑胶质瘤严重威胁着人们的身体健康,治疗该病是一项巨大的挑战.为了改善脑胶质瘤的治疗,纳米粒子(NPs)成为一个很好的选择,它具有缓释性、靶向性和亲和性等特征,并且纳米材料在保持药物稳定性方面有明显的优势,可以成为抗肿瘤药物的最适载体.纳米材料作为药物载体为胶质瘤的治疗开辟了新的思路.目前已经成功构建了多种抗肿瘤NPs,每种NPs均有特定的性质,因而纳米给药系统及NPs在胶质瘤个体化治疗方面具有极大的应用优势.
肺癌是临床常见高发恶性肿瘤之一,邱志楠教授在长期的临床实践中积累了丰富的治疗经验,本文通过对邱志楠教授治疗肺癌的经验举隅,探讨邱志楠教授在肺癌中医治疗方面的特色经验,为提高肺癌的诊断、治疗提供一个值得借鉴的模式.
目的:研究不同脱色方法对全真一气汤多糖含量的影响,选择适宜的脱色方法,并初步探讨其对免疫抑制小鼠外周血免疫功能的影响.方法:分别采用大孔树脂静态吸附法、活性炭吸附法、过氧化氢氧化法进行脱色,以色素脱除率和多糖含量为指标,进行脱色效果比较;采用环磷酰胺造成免疫抑制模型,观察全真一气汤多糖对小鼠外周血白细胞、淋巴细胞及IL-2的影响.结果:大孔树脂静态吸附法处理后,多糖的色素脱除率为(48.32±1.05)%,多糖含量(39.63±1.08)%,相比粗多糖,含量有所提高.活性炭和过氧化氢处理后,色素脱除率虽然分别达到(68.49±1.02)%,(68.98±0.69)%,但多糖含量降低;全真一气汤多糖可对抗免疫抑外周血白细胞及淋巴细胞数目的降低,刺激外周血血清IL-2分泌的增加.结论:大孔树脂法适宜用于全真一气汤多糖脱色素;全真一气汤多糖可以提高免疫抑制小鼠的免疫功能.
Objective To establish a method for the extraction and determination of polysaccharides from Quanzhenyiqi Decoction. Methods Polysaccharide in Quanzhenyiqi Decoction was determined by the method of Spec-trophoto metry. Results The polysacharides in Quanzhenyiqi Decoction was 56.56%,the average recovery was 92.02%,RSD was 1.21%. Conclusion This method is simple,the result is stable,reproducible and satisfactory.
目的 探讨复方中药全真一气汤总多糖对正常小鼠及环磷酰胺免疫抑制小鼠的非特异性免疫功能的影响。方法取SPF级KM小鼠,按体重随机分为正常组、模型对照组及全真一气汤总多糖组。采用尾静脉注射印度墨汁,用分光光度法测定吸光度值,计算碳粒廓清指数K、吞噬指数α,观察正常小鼠和免疫抑制小鼠的腹腔巨噬细胞的吞噬功能及脾脏指数指标。结果全真一气汤总多糖提高正常小鼠和免疫抑制小鼠的碳粒廓清指数K值、吞噬指数α和脾脏指数(P< 0.05,P<0.01).结论复方中药全真一气汤总多糖对小鼠的非特异性免疫功能有不同程度的增强作用。
目的:探讨全真一气汤粗多糖对环磷酰胺所致免疫低下模型小鼠的体内免疫活性的影响.方法:取NIH小鼠,按体重随机分为正常组、模型组及全真一气汤粗多糖组.观察全真一气汤粗多糖对免疫低下小鼠的腹腔巨噬细胞吞噬功能、血清溶血素(HC50)、脾脏指数和耳肿胀率等多项指标.结果:全真一气汤粗多糖可显著提高正常小鼠和免疫低下小鼠的碳粒廓清指数K值、吞噬指数α、血清溶血素(HC50)、脾脏指数和耳肿胀率(P<0.05).结论:全真一气汤粗多糖对免疫低下小鼠的免疫功能有显著的增强作用.
Objective:To observe the effects of anticancer chinese medicine "Caning" on the anti-apoptosis gene bcl-2 of in vitro cultured human hepatic cancer cell line BEL-7402 cells,and investigates its anticancer therapeutic mechanism.Methods:In vitro cultured BEL-7402 cells in a logarithmic growth phase were randomly divided into 4 groups:control group,5-Fluorouracil treated group,serum group 1 containing "Caning"(10% pastille serum) and serum group 2 contain-ing "Caning"(15% pastille serum);then MTT assay was employed to detect the inhibitory ability of serums containing dif-ferent concentrations of "Caning" to in vitro cultured human hepatic cancer cell line BEL-7402 cells,and PCR was used to detect the effect of "Caning"on the anti-apoptosis gene bcl-2.Results:Results of in vitro experiment suggested that,the bcl-2 gene activity in serum group 1 and 2 containing "Caning" was significantly lower than that of control group(P=0.038);result of MTT assay indicated that the cell growth inhibitory rate was increased along with the increase of the action time and concentration of serum containing "Caning"(P0.05),and the 15% pastille serum showed the strongest inhibitory effect at 72 h.Conclusion:"Caning"could significantly inhibit the growth of hepatic cancer cells partly by inhibiting the anti-apoptosis gene bcl-2.
背景:研究证实,冻干和辐照灭菌的同种异体骨是理想的骨移植材料,不仅具有良好的生物相容性、生物力学特性,而且还保留骨基质中的骨形态发生蛋白和构成形态发生蛋白所必须的一些酶类,具有一定的骨诱导能力。目的:观察胎盘组织液-同种异体冻干骨修复犬的人工颌骨缺损效果,以同种异体冻干骨为对照。设计、时间及地点:对比观察性实验,于2007-12/2008-09在哈尔滨医科大学动物实验中心完成。材料:健康杂种成年犬8只;同种异体冻干骨由湖北联结生物材料有限公司提供;胎盘组织液为广东珠海丽珠制药厂产品,2mL/支。同种异体冻干骨∶胎盘组织液=(4.0~5.0)∶1。方法:在犬上、下中切牙、尖牙、第一磨牙根尖相对应的颌骨处分别建立直径1.0cm类半球型上下颌骨缺损模型共96个实验区,实验组选择大小合适的同种异体冻干骨及骨颗粒按比例浸泡在胎盘组织液呈饱和状,阳性对照组亦将大小合适的同种异体冻干骨及骨颗粒浸泡在生理盐水液呈饱和状,分别放入颌骨缺损处并添满,阴性对照组不植入任何材料。每只犬的实验区均由4个实验组材料、4个阳性对照组材料、4个阴性对照组材料组成。主要观察指标:术后2,4,8,12周取材进行放射线学和组织学观察。结果:实验组早期以软骨内化骨为主,晚期以膜内化骨为主,有明显的新骨形成,并与周围骨组织连接良好;阳性对照组可见新生骨,与周围骨组织连接欠佳;阴性对照组骨缺损区中央尚未被骨小梁充满。经组织学测定,术后2,4,8,12周实验组成骨量均高于其他两个对照组,差异有显著性意义(P<0.05,P<0.01)。结论:胎盘组织液-同种异体冻干骨复合材料能够积极地促进颌骨缺损的修复并有效缩短修复时间。
BACKGROUND: The success of obtaining seeding endothelial cell on artificial vascular graft by the mechanical method of scoraping autogenous vein provides guidance for vascular prothesis. However, the insufficient of vein leads to endothelial cell was in poor quantity and quality. OBJECTIVE: To investigate the endothelialization of luminal surface of allogenic vein after using bone marrow seeded vascular graft in venous vascular injury. DESIGN, TIME AND SETTING: The randomized control experiment on animals was performed in the Animal Experiment Center of Harbin Medical University between March 2007 and April 2008. MATERIALS: Eight New Zealand rabbits were randomly divided into the experimental and control groups, with 4 animals in each group. The Knitted double velour Dacron grafts were supplied by America Meadox Medicals Inc. METHODS: Rabbits in the experimental group were implanted Knitted double velour dacron grafts seeded by autogenous bone marrow into the infrarenal inferior vena caves, while, the rabbits in the control group were treated by autogenous plasma. The grafts were harvested at 10 days following transplantation. MAIN OUTCOME MEASURES: The condition of vascular patency, the thickness and endothelialization of neointima, as well as the level of thromboxane A2 (TXA2) were measured by light and electron microscopy and radioimmunology. RESULTS: A total of 8 animals were enrolled in the result analysis. All the bone marrow seeded grafts and 2 of the control group were patent. The thickness of neointima in the experimental group was significantly thinner than that of the control group (P < 0.01). Confluent endothelial cells only appeared on the neointima of seeded grafts. The lever of thromboxane B2, which was the main product of metabolism TXA 2, was lower than that in the control group (P < 0.05) CONCLUSION: Rapid endothelialization can be achieved by using bone marrow seeded vascular graft at 10 days, and the new endothelia cells can inhibit the hyperplasia and anti thrombus formation.
Objective To study the operation method to prevent complication in total knee arthroplasty(TKA) with severe valgus deformity.Methods 21 patients with severe valgus deformities were undergone TKA.The operations were done through lateral Z shape incisions.Prosthesis rotation positioning and soft tissue balancing were done,and the lower leg alignments were restored.When soft tissue was tight both in flexion and extension,releasing was done with 90° flexion in upper lateral collateral ligament of lateral condyle,articular capsule angulus posteriolateralis and popliteal tendon,and with extension in upper iliotibial tract in grid form.When soft tissue was fine in flexion and tight in extension,releasing was done in iliotibial track,lateral collateral ligment and posterior capsule;When soft tissue was tight in flexion and fine in extension,releasing was done in popliteal tendon,lateral collateral ligament and angulus posteriolateralis of the articular capsule.Results 21 cases were followed up for 9~28 months,averaged 26.7 months.Among them,2 cases had late onset peroneal nerve palsy,and recovered after 3 month.Operative incision got primary healing.Preoperative KSS score was(43.3±4.1) points,and was restored to(83.8±5.2) points in 12 months,and(87.4±4.5) points in 24 months.Conclusions It is more difficult to perform TKA with valgus than varus.Lateral approach and the operation is beneficial for prevention of complication.
[目的]探讨同期进行双侧人工全髋关节置换的临床疗效及其安全性。[方法]对2002年10月~2007年10月收治的69例行双侧人工全髋关节置换术患者的临床资料进行回顾性分析。[结果]经平均2年9个月随访,本组患者手术效果满意,术前Harris评分平均为29.6分,术后平均92.2分。术后出现早期并发症9例,经治疗后痊愈出院,无死亡病例发生。[结论]对双侧髋关节均有病变的疾患同期进行双侧全髋关节置换是安全可行的。手术适应证的严格掌握、充分的围手术期准备是取得手术成功的关键。
