Congrong-Shujing Granules (CRSJG) are known to protect dopaminergic neurons in Parkinson's disease (PD), and the mechanism may be related to the improvement of mitochondria-associated membranes (MAMs). To investigate the impact of CRSJG-medicated serum on MAMs in the 1-methyl-4-phenylpyridinium (MPP+)-induced PD cell model. Human neuroblastoma (SH-SY5Y) cells were treated with 1,000 μmol/L MPP+ for 24 h, resulting in three experimental groups: MPP+, CRSJG, and 2-APB. The MPP+ group received blank serum, CRSJG group was treated with CRSJG-medicated serum, and the 2-APB group was given 100 μmol/L 2-APB. An untreated control group was also included. Serum pharmacokinetics for the CRSJG group were analyzed using ultra-performance liquid chromatography-tandem mass spectrometry. Intracellular-Ca2+, reactive oxygen species (ROS), mitochondrial membrane potential (MMP), and apoptosis rates were measured using fluorescent probes and flow cytometry. Transmission electron microscopy was employed to examine the morphology of MAMs. Western blot was conducted to identify proteins related to apoptosis and Ca2+ transport. CRSJG-medicated serum identified by pharmacokinetic markers including echinacoside, paeoniflorin, salvianolic acid B, acteoside, and tanshinone IIa. The EC50 for MPP+-induced reduction in cell proliferation was 1,110 μmol/L. CRSJG-medicated serum, especially at 2.5%, significantly improved cell proliferation after 24 h. The serum effectively mitigated damage within the MAMs region and reduced both the mitochondrial-Ca2+ fluorescence intensity and the expression of the IP3R-VDAC-MCU complex in MPP+-induced neuronal cells. Additionally, it significantly decreased the levels of ROS, the decline in MMP, and apoptosis rates in these cells. The findings provide novel insights into the potential of CRSJG in treating neuronal loss in PD.
目的 研究苁蓉舒痉颗粒对鱼藤酮诱导的帕金森病(PD)模型大鼠黑质纹状体内质网应激蛋白激酶样内质网激酶(PERK)-真核翻译起始因子2α(eIF2α)通路的调节作用.方法 将100只健康SD大鼠随机分为正常组33只和造模组67只,造模组采用颈背部皮下注射鱼藤酮葵花油乳液建立PD模型,参照行为学评分标准鉴定造模成功后,随机分为模型组34只和治疗组33只.治疗组予苁蓉舒痉颗粒药液6.8 mL/(kg·d)灌胃,正常组、模型组予等量生理盐水灌胃,连续干预14 d.于灌胃第1、3、5、7、9、11、13、14天,采用平行反应监测(PRM)蛋白相对定量分析检测大鼠黑质纹状体eIF2α特异性肽段定量的变化;灌胃14 d后应用透射电镜观察3组大鼠黑质神经细胞超微结构;采用免疫组化法检测大鼠黑质PERK、p-PERK、eIF2α、p-eIF2α表达的平均光密度值.结果 与正常组比较,在灌胃第13、14天模型组黑质纹状体eIF2α特异性肽段定量明显增高(P均<0.05),灌胃14 d后黑质神经细胞肿胀,内质网扩张,黑质PERK、p-PERK、eIF2α、p-eIF2α的平均光密度值增高(P<0.05,P<0.01);与模型组比较,治疗组在灌胃第5天黑质纹状体eIF2α特异性肽段定量明显增高(P<0.01),灌胃第13、14天时明显减低(P均<0.05),灌胃14 d后黑质神经细胞肿胀、内质网扩张等形态异常有所改善,黑质PERK、p-PERK、eIF2α、p-eIF2α的平均光密度值明显降低(P<0.05,P<0.01).结论 苁蓉舒痉颗粒可能通过动态调节内质网应激PERK-eIF2α通路,起到恢复内质网稳态的作用.
目的:探讨归脾汤对老年慢性心力衰竭心脾两虚型患者心功能和肠道菌群的影响.方法:将60例老年慢性心力衰竭心脾两虚型患者随机分为治疗组和对照组,每组30例.对照组予常规抗心衰治疗,治疗组在对照组治疗的基础上加用归脾汤治疗,两组疗程均为4周.比较两组患者治疗前后N末端B型利钠肽前体(NT-proBNP)水平、左心室射血分数(LVEF)变化情况,并采用Illumina测序平台对各组粪便微生物进行16S rRNA高通量测序,分析两组肠道微生物群落的差异性.结果:治疗后,两组LVEF较治疗前升高,且治疗组高于对照组(P<0.05);两组NT-proBNP水平较治疗前降低,且治疗组低于对照组(P<0.05).测序结果显示,治疗后治疗组Chao1指数、ACE指数与对照组比较均显著升高(P<0.01或P<0.05),Simpson指数均显著下降(P<0.01);两组β-多样性分析无显著性差异(P>0.05);治疗组治疗后考拉杆菌属、经黏液真杆菌属、颤螺菌属、梭状芽孢杆菌属、霍尔德曼氏菌属、弯曲杆菌属水平与治疗前比较相对丰度相似性存在差异(P<0.05),其中拟杆菌、志贺菌和大肠埃希菌、普拉梭菌、双歧杆菌、韦荣氏球菌属相对丰度降低,普雷沃氏菌、粪副拟杆菌、乳酸菌、狄氏副拟杆菌治疗后相对丰度升高.结论:归脾汤可以明显改善老年慢性心力衰竭心脾两虚型患者心功能,可能通过调节肠道细菌结构改善肠道微生态,进而达到治疗效果.
Objective To investigate the effect of single nucleotide polymorphisms(SNPs)of EXT2gene on the susceptibility of metabolic syndrome(MS)with phlegm syndrome. Methods From November 2013to December 2020,a total of 849 patients with MS who were treated in the Pre Treatment Center and Physical Examination Center of the Third People’s Hospital Affiliated to Fujian University of Traditional Chinese Medicine,and the Endocrine Department of Jinjiang Hospital of Traditional Chinese Medicine Affiliated to Fujian University of Traditional Chinese Medicine were selected. MS patients were assigned to MS phlegm syndrome group(641cases)and non-phlegm syndrome group(208 cases)by using syndrome differentiation method.SNPscan multiple SNP typing technique was used to detect rs11037909,rs1113132 and rs3740878 genotypes of EXT2.Logistic regression analysis was performed on gene SNPs,body mass index(BMI)and triglyceride(TG).Results(1)BMI,TG and phlegm syndrome elements were positively correlated in MS phlegm syndrome group(P<0.05).(2)The risk of developing phlegm syndrome in patients with EXT2 rs11037909 CT/TC or CT+CC,rs1113132 CC or CG+CC,rs3740878 CC or CT+CC genotypes was 0.562 or 0.666 times,0.559 or 0.645 times,0.561 or 0.650 times than those with TT,GG and TT genotypes.(3)The risk of developing phlegm syndrome in overweight or obese individuals with EXT2 rs11037909 CT/TC+CC,rs1113132 CG/GC+CC,rs3740878 CT/TC+CC genotypes was 0.642 times and 0.618 times and 0.624 times than those with TT,GG,and TT genotypes.(4)The risk of phlegm syndrome in patients with high TG carrying EXT2 rs11037909 CT/TC+CC genotype was 0.544 times than that in high TG patients carrying TT genotype;The risk of developing phlegm syndrome in the non-TG patients with EXT2 rs1113132 CG/GC+CC,rs3740878 CT/TC+CC genotype was 0.524 times and 0.531times than those high TG patients with GG or TT genotype.(5)The risk of developing phlegm syndrome(mild)in patients with EXT2 rs11037909/rs1113132/rs3740878 CT/TC+CC or CG/GC+CC genotypes without overweight,obesity and high TG was 0.102 times than those with TT or GG genotypes who were overweight or obese and had high TG. Conclusion Taking T/G allele as reference,EXT2 gene rs11037909/rs1113132/rs3740878-C allele is a protective factor for phlegm syndrome compared with non-phlegm syndrome,EXT2 gene rs11037909/rs1113132/rs3740878-T allele,both BMI and TG are risk factors of phlegm syndrome,which will increase the susceptibility of phlegm syndrome.
目的:系统评价中药干预帕金森病伴睡眠障碍患者的疗效.方法:全面检索中英文数据库中涉及帕金森病伴睡眠障碍中医或中西医相关的临床随机对照试验研究,检索时间为建库至2021年8月,应用RevMan 5.4.1软件进行数据分析.结果:共纳入文献23篇,样本量为1483例.Meta分析结果示与单纯西药治疗比较,中药组的以下指标水平均更优:临床疗效(P<0.00001)、 中医证候积分(P<0.00001)、匹兹堡睡眠质量指数量表(Pittsburgh Sleep Quality Index,PSQI)评分(P<0.00001)、统一帕金森病评定量表第三部分(Unified Parkinson's Disease Rating Scale PartⅢ,UPDRSⅢ)评分(P<0.00001)、帕金森病睡眠量表(Parkinson's Disease Sleep Scale,PDSS)评分(P<0.00001)、爱泼沃斯嗜睡量表(Epworth Sleepiness Scale,ESS)评分(P=0.02)、39项帕金森病生存质量调查问卷(The 39-item Parkinson's Disease Quality of Life Questionnaire,PDQ-39)评分(P<0.00001).结论:中药干预较单纯使用西药可更好地改善帕金森病患者的睡眠障碍,且不良反应的报道较少.
目的 研究补肾活血颗粒对自然衰老大鼠骨骼肌组织的保护作用及其机制.方法 将20只12月龄SD大鼠随机分为自然衰老组及补肾活血颗粒组,以10只3月龄大鼠作为青年对照组.将补肾活血颗粒用100℃纯净水配置成371 mg/mL含药溶液,以10 mL/(kg·d)灌胃,每周连续6 d,停药1 d,持续6个月,青年对照组及自然衰老组正常饲养未予任何药物干预.6个月后光镜下观察各组大鼠骨骼肌组织形态学改变,Western blot检测腓肠肌组织沉默信息调节因子2相关酶1(SIRT1)、叉头框转录因子O3(FoxO3)蛋白相对表达量,ELISA法检测SIRT1去乙酰化酶活性.结果 与青年对照组比较,自然衰老组及补肾活血颗粒组大鼠腓肠肌组织SIRT1蛋白相对表达量显著增加,FoxO3蛋白相对表达量及SIRT1去乙酰化酶活性明显降低(P均<0.01或<0.05);与自然衰老组比较,补肾活血颗粒组大鼠腓肠肌组织FoxO3蛋白相对表达量及SIRT1去乙酰化酶活性明显增加,腓肠肌组织SIRT1蛋白相对表达量明显降低(P均<0.05).结论 补肾活血颗粒可调节SIRT1、FoxO3蛋白相对表达量及增加SIRT1去乙酰化酶活性,可能通过调节SIRT1/FoxO3信号通路,一定程度上对自然衰老大鼠骨骼肌组织具有保护作用.
The index weight coefficients were determined by comparing the analytic hierarchy process(AHP), the criteria importance through inter-criteria correlation(CRITIC), and the AHP-CRITIC mixed weighting method. The comprehensive scores of index components(echinacoside, salvianolic acid B, paeoniflorin, and ointment yield) of each group in the orthogonal test were compared to optimize the extraction process of Congrong Shujing Granules. The results showed that the AHP-CRITIC mixed weighting method scientifically optimized the extraction process. To be specific, the decoction pieces should be added with the 6-fold amount of water and extracted twice, 1 h each time. After three verification tests, the average mass fractions of echinacoside, salvianolic acid B, and paeoniflorin were 0.72, 9.34, and 5.92 mg·g~(-1), respectively, and the average ointment yield was 47.18%. As verified by the AHP-CRITIC mixed weighting method and the orthogonal test, the optimized extraction process of Congrong Shujing Granules was stable and feasible and could be applied to industrial production.
医学统计学是中医院校研究生教学中既重要又有难度的一门课程.文章从中医院校研究生医学统计学教学面对的困难出发,提出医学统计学课程体系设置、教学内容、教学方法及考核方式的教学改革思路.
目的 比较苁蓉舒痉颗粒在正常与帕金森病大鼠中的体内药动学.方法 大鼠随机分为正常组[皮下注射葵花油乳化液(1.5 mg/kg)]和模型组[皮下注射含鱼藤酮葵花油乳化液(1.5 mg/kg)],灌胃给予药物混悬液(10 g/kg),于0.083、0.167、0.25、0.5、0.75、1、2、4、6、8、12、24、36 h采血,UPLC-MS/MS法测定特征效应组分(松果菊苷、芍药苷、丹酚酸B、毛蕊花糖苷、丹参酮ⅡA)血药浓度,计算主要药动学参数.结果 与正常组比较,模型组松果菊苷、芍药苷、丹酚酸B、丹参酮ⅡA Cmax升高(P<0.05,P<0.01),毛蕊花糖苷Cmax降低(P<0.05);丹参酮ⅡA MRT0~t升高(P<0.01),丹酚酸B MRT0~t降低(P<0.01);芍药苷、丹参酮ⅡA MRT0~∞升高(P<0.01),丹酚酸B MRT0~∞降低(P<0.01);芍药苷、毛蕊花糖苷AUC0~t、AUC0~∞升高(P<0.01).结论 对于帕金森病状态下的大鼠,苁蓉舒痉颗粒口服吸收后进入其体循环的相对量更高,特征效应组分血药浓度、Cmax、生物利用度明显提高,从而发挥更好的作用.
目的:了解国内中医药院校课程思政开展的研究现状及发展趋势.方法:对2013—2021年中国知识资源总库(CNKI)收录的中医药院校发表的关于课程思政研究的文献,统计年发文量和发文机构,采用VOSviewer软件对学者分布聚类分析,CiteSpace对关键词进行共现、突现分析并绘制可视化知识图谱.结果:共纳入文献674条,中医药院校课程思政研究的中文文献发表量呈现逐年增长趋势,研究机构活跃的是长春中医药大学,研究热点主要围绕立德树人、教学改革.结论:中医药院校课程思政还处于理论及实践探索阶段,趋向建立具有中医药特色的课程思政相关研究.
Parkinson's disease (PD) is the second major progressive neurodegenerative disease, which critically impacts patients' quality of life. Based on genetics, animal models of genetic defects created by gene editing technology have clear advantages in reflecting PD's pathogenesis and pathological characteristics and exploring potential therapeutic targets for PD. In this review, we summarized animal models of genetic defects in various pathogenesis of PD, including α-synuclein abnormal encoding, autophagy-lysosome system defects, ubiquitin protease system defects, and mitochondria-related dysfunction, and discuss their respective advantages, limitations, and application directions to provide a reference for the application of animal models of PD and research on anti-PD therapy.
目的:利用七氯对斑马鱼多巴胺神经元的影响,建立斑马鱼帕金森病(PD)模型.方法:将斑马鱼鱼卵分为正常对照组(不予干预),750μmol/L 6-羟基多巴胺(6-OHDA)组和不同浓度七氯组(1 ng/mL、10 ng/mL、100 ng/mL、1000 ng/mL、10000 ng/mL),分别恒温孵育,每24 h观察胚胎发育情况并记录直到受精后120 hpf(h post fertilization,hpf),在120 hpf做行为学测定,然后实时荧光定量PCR(RT-qPCR)测定多巴胺通路酪氨酸羟化酶(TH2)的相对表达量.结果:与正常对照组比较,6-OH-DA组幼体出现发育迟缓、脊柱弯曲,运动距离缩短(P<0.05),TH2基因表达降低(P<0.05);与正常对照组比较,1~100 ng/mL七氯组幼体发育无明显变化,但运动距离缩短(P<0.05),TH2基因表达降低(P<0.05),1000~10000 ng/mL七氯组出现色素沉着分布和脊柱弯曲,而且运动距离更短(P<0.05)、TH2基因表达更低(P<0.05),变化具有浓度依赖性;与6-OHDA组比较,1000~10000 ng/mL七氯组运动距离更短,TH2基因表达更低,其中10000 ng/mL七氯组运动距离缩短和TH2基因表达降低有统计学差异(P<0.05).结论:传统PD模型造模药物6-OHDA和1000~10000 ng/mL七氯都可通过影响多巴胺神经元功能成功建立斑马鱼PD模型,其中10000 ng/mL七氯的造模效果比6-OHDA更显著.
目的 探讨松果菊苷(echinacoside,ECH)调控抗增殖蛋白(prohibitin,PHB)表达,及对MPP+诱导的SH-SY5Y细胞凋亡的效应及机制.方法 选取SH-SY5Y细胞分为con-trol组、MPP+组、MPP++ECH组、NC+MPP+组、NC+MPP++ECH组、PHB-RNAi+MPP++ECH组.CCK-8检测细胞存活率,倒置相差显微镜观察细胞形态,Hoechst33342荧光染色法观察凋亡细胞,流式细胞仪检测细胞凋亡率、活性氧含量及线粒体膜电位,Western blot检测PHB、Akt、p-Akt、Bcl-2、Bax、cleaved caspase-3相对蛋白表达.结果 与control组比较,MPP+组细胞存活率显著下降;细胞生长状态明显变差;细胞内活性氧含量增高、线粒体膜电位降低、凋亡相关蛋白表达增加、凋亡率升高.与MPP+组比较,MPP++ECH组细胞存活率显著上升;细胞生长状态明显好转;细胞内活性氧含量降低、线粒体膜电位升高、凋亡相关蛋白表达降低、凋亡率明显下降;PHB和p-Akt表达量明显上升.与NC+MPP++ECH组比较,PHB-RNAi+MPP++ECH组p-Akt水平降低,细胞凋亡率上升.结论 松果菊苷能减轻MPP+诱导的SH-SY5Y细胞凋亡,此效应可能是通过上调PHB表达,使Akt磷酸化,从而保护细胞线粒体功能来实现的.
目的 探讨苁蓉舒痉颗粒对帕金森病(PD)线粒体氧化应激及自噬相关SIRT1/FoxO通路的影响.方法 采用软件随机数字法将27只健康SD大鼠分为正常组9只、造模组18只,造模组采用颈背部皮下注射鱼藤酮葵花油乳化液建立PD模型,将造模成功的大鼠随机分为模型组和治疗组各9只,治疗组予苁蓉舒痉颗粒溶液灌胃,正常组、模型组予等量生理盐水灌胃,连续干预14 d.分别于灌胃第1、7、14天进行爬杆实验检测爬杆实验得分;灌胃第14天大鼠麻醉后取材,免疫组化法检测中脑酪氨酸羟化酶(TH)阳性细胞数,ELISA法检测中脑沉默信息调节因子1(SIRT1)、叉头蛋白O1(FoxO1)、FoxO3a蛋白水平,Western blot法检测中脑SIRT1、FoxO1、FoxO3a、微管相关蛋白轻链3-Ⅰ(LC3-Ⅰ)、LC3-Ⅱ蛋白表达,流式细胞术检测中脑活性氧(ROS)水平.结果 与正常组比较,模型组爬杆实验评分、ROS水平明显升高(P<0.01),TH阳性细胞数及SIRT1、FoxO1、FoxO3a、LC3-Ⅰ、LC3-Ⅱ蛋白表达明显降低(P<0.05,P<0.01).与模型组比较,治疗组TH阳性细胞数及SIRT1、FoxO1、FoxO3a、LC3-Ⅰ、LC3-Ⅱ蛋白表达明显升高(P<0.05,P<0.01),爬杆实验评分、ROS水平明显降低(P<0.01).结论 苁蓉舒痉颗粒能改善帕金森病大鼠行为障碍,降低中脑组织ROS水平,增加自噬相关LC3-Ⅱ蛋白表达,保护中脑多巴胺能神经细胞,其保护作用可能是通过调节线粒体氧化应激及自噬相关SIRT1/FoxO通路实现的.
Cerebral small vessel disease (CSVD) is a senile brain lesion caused by the abnormal structure and function of arterioles, venules and capillaries in the aging brain. The etiology of CSVD is complex, and disease is often asymptomatic in its early stages. However, as CSVD develops, brain disorders may occur, such as stroke, cognitive dysfunction, dyskinesia and mood disorders, and heart, kidney, eye and systemic disorders. As the population continues to age, the burden of CSVD is increasing. Moreover, there is an urgent need for better screening methods and diagnostic markers for CSVD, in addition to preventive and asymptomatic- and mild-stage treatments. Integrative medicine (IM), which combines the holistic concepts and syndrome differentiations of Chinese medicine with modern medical perspectives, has unique advantages for the prevention and treatment of CSVD. In this review, we summarize the biological markers, ultrasound and imaging features, disease-related genes and risk factors relevant to CSVD diagnosis and screening. Furthermore, we discuss IM-based CSVD prevention and treatment strategies to stimulate further research in this field.
目的 探究苁蓉舒痉颗粒对PD模型小鼠额叶保护作用及其机制.方法 腹腔注射MPTP复制PD模型.实验分组为正常组、模型组、苁蓉组和美多芭组.在造模第7天、给药14天后,对小鼠进行行为学测试;免疫组化检测小鼠黑质TH阳性细胞的表达;透射电镜观察小鼠额叶线粒体损伤情况;qPCR检测小鼠额叶中PPARγ和PGC-1αmRNA含量的表达;Western Blot检测小鼠额叶中TFAM、PPAR、NRf2、PGC-1α蛋白的表达.结果 (1)行为学:造模7d后,与正常组比较,模型组悬挂、爬杆评分、抓力值、自主活动次数降低,给药治疗14d后,与模型组比较,苁蓉组、美多芭组悬挂、爬杆得分、自主活动次数增加.(2)免疫组化:与正常组比较,模型组小鼠黑质TH阳性细胞数明显减少;与模型组比较,苁蓉组、美多芭组小鼠黑质TH阳性细胞数明显增多.(3)透射电镜:与正常组比较,模型组额叶线粒体出现肿胀、空泡样,嵴断裂甚至消失;与模型组比较,苁中组与美多芭组额叶线粒体数量有所增加,形态有所改善.(4)qPCR:与正常组比较,模型组小鼠PPARγ、PGC-1αmRNA表达水平明显降低;与模型组比较,苁蓉组、美多芭组小鼠PPARy、PGC-1αmRNA表达水平明显升高.(5)Western Blot:与正常组比较,模型组TFAM、PPAR、NRf2、PGC-1α蛋白表达量明显降低;与模型组比较,苁蓉组、美多芭组蛋白表达量明显升高.结论 苁蓉舒痉颗粒可以减少PD模型小鼠额叶损伤,这种保护作用可能是通过调节PPAR-γ/PGC-1α通路实现.
目的:建立HPLC多波长切换法同时测定苁蓉舒痉颗粒中松果菊苷、芍药苷、毛蕊花糖苷、丹酚酸B、丹皮酚和丹参酮Ⅱ A的含量.方法:采用Agilent Zorbax Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5 μm),以乙腈-0.1%磷酸溶液为流动相,梯度洗脱,流速1.0 mL·min-1,柱温25℃,进样量20 μL,检测波长分别为330 nm(松果菊苷和毛蕊花糖苷)、230 nm(芍药苷)、286 nm(丹酚酸B和丹皮酚)、270 nm(丹参酮ⅡA).结果:松果菊苷、芍药苷、毛蕊花糖苷、丹酚酸B、丹皮酚和丹参酮ⅡA 质量浓度分别在18.18~181.8μg·mL-1(r=0.999 8)、28.86~288.6μg·mL-1(r=0.999 8)、0.576 0~5.760 μg·mL-1(r=0.999 5)、32.96~329.6μg·mL-1(r=0.999 7)、31.97~319.7 μg·mL-1(r=0.999 0)、1.120~11.20 μg·mL-1(r=0.999 8)范围内与各峰面积呈良好的线性关系;平均回收率(n=6)分别为98.7%、99.4%、98.6%、99.0%、98.8%、99.1%,RSD分别为1.0%、0.92%、1.6%、0.85%、1.0%、1.7%;3批样品中上述各成分含量范围分别为3.571~3.702、6.692~6.911、0.071~0.089、7.343~7.947、2.201~2.259、0.041~0.055 mg·g-1.结论:该方法可作为苁蓉舒痉颗粒质量标准同时测定6个主要成分的含量测定法.
目的:立足整体探讨代谢综合征(MS)中医证候与微观理化指标的相关性.方法:采用标准化四诊信息采集量表收集MS患者中医证候信息,选取出现频率>10%的条目47项.同时,收集患者理化指标共21项.采用典型相关分析探讨MS中医证候信息与微观理化指标间的整体相关性.结果:共收集符合纳入标准对象450例,提取出4对MS中医证候与微观指标典型相关组合,相关系数分别为0.690、0.577、0.518、0.505(P<0.01).其中,脉弦与身高、心烦与体质量、脉沉与BMI、嗜烟与体质量等呈较明显的正相关关系,并且部分微观理化指标的组合与中医证候具有一定相关性.结论:微观理化指标与中医证候之间具有较为明显的典型相关关系,立足整体,将微观指标与中医证候变量结合,可为全面客观、中西医结合辨证提供一定参考.
目的:基于IRE1α/JNK通路研究苁蓉舒痉颗粒对帕金森病(parkinson's disease,PD)模型大鼠的影响.方法:将45只雄性SD大鼠随机分成正常组、模型组和苁蓉舒痉颗粒组(5.21 g·kg-1),除正常组外,其余大鼠采用连续14 d颈背部皮下注射鱼藤酮葵花油乳化液(1.5 mg·kg-1)(按1.5∶1将鱼藤酮、葵花油配为鱼藤酮葵花油乳液)制备PD模型,连续给予相应的药物14 d.免疫组化法检测大鼠脑黑质酪氨酸羟化酶(tyrosine hydroxylase,TH)表达;Western Blot法检测大鼠脑额叶中IRE1α、JNK蛋白及其磷酸化的表达.结果:与模型组比较,苁蓉舒痉颗粒组大鼠脑黑质TH阳性细胞数明显增加(P<0.05),脑额叶p-IRE1 α/IRE1α、p-JNK/JNK水平明显降低(P<0.05).结论:苁蓉舒痉颗粒通过增加大鼠脑黑质TH阳性细胞数,降低脑额叶p-IRE1 α/IRE1α、p-JNK/JNK水平来保护神经细胞,可能与IRE1α/JNK通路有关.
目的:观察松果菊苷对帕金森病(PD)模型大鼠中脑超微结构的影响.方法:将健康SD大鼠随机分为对照组、模型组和松果菊苷组,每组10只.对照组用生理盐水灌胃,模型组和松果菊苷组大鼠造模后,分别用生理盐水和2 mg/mL的松果菊苷水溶液灌胃,每天1次,连续14 d.灌胃结束后,采集各组大鼠中脑,制作超薄切片,在7000倍和50000倍电镜下观察并比较各组大鼠中脑神经细胞超微结构的差异.结果:对照组中脑神经细胞结构清晰,核膜光滑、完整,核内染色质分布均匀;线粒体数量较多,形态规则,双层膜结构、线粒体嵴结构清晰.与对照组比较,模型组的神经细胞胞体缩小,核染色质边聚;线粒体数量减少,双层膜结构不清,线粒体嵴排列紊乱.松果菊苷组与模型组比较,神经细胞胞体大小有所恢复,核染色质边集现象有所缓解;线粒体数量增加,双层膜结构清晰,线粒体嵴排列相对规律,形态基本正常.结论:松果菊苷能改善PD模型大鼠中脑神经细胞凋亡的超微结构,这一结果可能与缓解神经细胞线粒体功能障碍有关.
