Objective To investigate the effects of Liancao Xieli Capsules on endoplasmic reticulum stress and inflammatory response in ulcerative colitis(UC) rats; To exploring its mechanism of treating UC. Methods Totally 40 SD male rats were randomly divided into blank group, model group, Liancao Xieli Capsules group and mesalazine group, with 10 rats in each group. Except for the blank group, the rat model of UC was established in the other groups by free drinking method of dextran sodium sulfate solution, during which the Liancao Xieli Capsules group and the mesalazine group were given corresponding drugs for intervention, for successive 7 d. The length of the colon in rats was measured, and the DAI score was performed, HE staining was used to observe the pathological morphology of colon tissue, ELISA was used to detect the contents of IL-1β, IL-17, IFN-γ in serum, Western blot was used to detect the expression of GRP78, CHOP, Bax, Bcl-2, Caspase-1, NF-κB p65, p-NF-κB p65, IκBα, p-IкBα, PERK, p-PERK,eIF2α, p-eIF2α, ATF4, IRE1α, p-IRE1α, xBP1s, and Cleaved ATF6 protein in colon tissue. Results Compared with the blank group, the length of colon was significantly shortened in the model group, the DAI score and colon tissue inflammation score significantly increased, the contents of IL-1β, IL-17 and IFN-γ in serum increased, the protein expression of GRP78, CHOP, Bax, Caspase-1, p-NF-κB p65, p-IкBα, p-PERK, p-eIF2α, ATF4, p-IRE1α, xBP1s and Cleaved ATF6 in colon tissue significantly increased, the expression of Bcl-2 protein and the ratio of Bcl-2/Bax significantly decreased, with statistical significance(P<0.01). Compared with the model group, the length of colon increased in the Liancao Xieli Capsules group, the DAI score and colon tissue inflammation score decreased, the contents of serum IL-1β, IL-17, IFN-γ decreased, the expression of GRP78, CHOP, Bax, Caspase-1, p-NF-κB p65,p-IкBα, p-PERK, p-eIF2α, ATF4, p-IRE1α, xBP1s and Cleaved ATF6 protein in colon tissue decreased, the expression of Bcl-2 protein and the ratio of Bcl-2/Bax increased, with statistical significance(P<0.01). Conclusion Liancao Xieli Capsules can improve intestinal inflammation in UC rats by inhibiting endoplasmic reticulum stress.
目的 观察溃疡性结肠炎(UC)患者大肠湿热证候评分与Mayo活动指数、Th17细胞、Treg细胞的相关性及连草泻痢胶囊对大肠湿热证UC患者的临床疗效.方法 分析UC大肠湿热证候评分与Mayo活动指数、Th17细胞、Treg细胞的相关性.美沙拉嗪组予美沙拉嗪缓释颗粒治疗,连草泻痢组则加用连草泻痢胶囊.对两组治疗前后大肠湿热证候评分、Mayo活动指数、Th17细胞、Treg细胞水平进行对比,并评价其临床疗效.结果 大肠湿热证候评分与Mayo活动指数、Th17细胞存在显著正相关(P<0.05),与Treg细胞相比存在显著负相关(P<0.05).治疗后连草泻痢组大肠湿热证候评分、Mayo活动指数、Th17细胞及Th17/Treg细胞比例降幅与Treg细胞升幅均高于美沙拉嗪组(P<0.05),总有效率亦高于美沙拉嗪组(P<0.05).结论 Th17/Treg细胞比例能反应UC大肠湿热证型的严重程度;连草泻痢胶囊联合西药治疗UC效果明显,可能与其调控Th17/Treg细胞比例平衡有关.
目的:观察芪参汤对非酒精性脂肪性肝炎(NASH)患者的临床疗效及对肠道菌群稳态的影响.方法:30例NASH患者随机分为治疗组(15例)和安慰剂组(15例),其中治疗组给予芪参汤治疗,安慰剂组给予芪参汤模拟剂治疗,疗程4周.分别观察2组患者治疗前后生化及纤维化指标的变化.另选取15例健康受试者作为对照,收集粪便样本通过16S rRNA检测各组肠道菌群的多样性和丰度.结果:与组内治疗前比较,芪参汤组治疗后AST、ALT、GGT、TC、TG、HA、LN、PⅢNP、CⅣ、LSM和CAP明显降低,HDL明显升高,差异有统计学意义(P<0.05),2组治疗后组间比较,芪参汤组均明显优于安慰剂组,差异均有统计学意义(P<0.05).相关性分析结果显示,Phascolarctobacterium、Lactobacillus、Bifidobacterium、Akkermansia的丰度分别与LN、HA、PⅢNP、CIV、CAP、LSM和GGT水平呈现显著负相关,而Fusobacterium、Coprococcus、Bilophila和Prevotella的丰度分别与LN、HA、PⅢNP、CIV、CAP、LSM和GGT水平呈现显著正相关.此外,芪参汤组治疗后OTU、Chaol和Shannon指数与同组治疗前及健康受试者比较均明显增加,Simpon指数明显降低,差异均有统计学意义(P<0.05).同时,与同组治疗前比较,芪参汤能够显著增加Clostridium、Roseburia、Eubacterium、Phascolarctobacteri-um、Lactobacillus、Parabacteroides、Pseudomonas、Bifidobacterium、Corynebacterium和Akkermansia菌属丰度,降低Faecalibacterium、Peptostreptococcus、Erysipelatoclostridium、Flavobacterium、Bilophila和Escherichia菌属丰度,差异均有统计学意义(P<0.05).结论:芪参汤能通过增加NASH肠道菌群多样性和稳态,从而改善肝功能、血脂异常和纤维化水平,达到治疗NASH的作用.
目的:观察连草泻痢胶囊对溃疡性结肠炎(UC)小鼠模型STAT信号通路及其介导的T细胞向Th17和Treg细胞分化的影响.方法:40只BALB/c小鼠随机分为空白组、模型组、美沙拉嗪组和连草泻痢胶囊组.除空白组外,余下3组采用3%葡聚糖硫酸钠自由饮水构建溃疡性结肠炎模型.造模期间各组给予相应的药物进行干预,而空白组和模型组给予等体积的蒸馏水灌胃作为对照.实验结束后通过HE染色观察结肠组织病理形态的改变,流式细胞术检测脾脏和肠系膜淋巴结中Th17和Treg细胞的比例,ELISA法检测结肠组织中TGF?β、IL?6和IL?17A的含量,Western blot法检测结肠组织中STAT3/ROR?γt和STAT5/Foxp3信号通路中相关蛋白的表达水平.结果:与模型组相比,实验结束后美沙拉嗪组小鼠体重、结肠长度和结肠组织中TGF?β的含量明显增加,DAI评分、结肠组织病理学评分和IL?6、IL?17A的含量明显降低,差异均具有统计学意义(P<0.01).同时,美沙拉嗪组能够降低UC小鼠脾脏和肠系膜淋巴结组织中Th17细胞的比例和Th17/Treg的比值,增加Treg细胞的比例,与模型组比较,差异均具有统计学意义(P<0.01).此外,与模型组相比,实验结束后美沙拉嗪组小鼠结肠组织中p?STAT3和ROR?γt蛋白的表达水平明显降低,p?STAT5和Foxp3蛋白的表达水平明显增加,差异均具有统计学意义(P<0.01),而结肠组织中STAT3和STAT5蛋白的表达水平未见明显改变,差异均无统计学意义(P>0.05).结论:连草泻痢胶囊能够调控Treg/Th17免疫平衡从而改善UC肠道炎症,其作用机制主要通过抑制STAT3/ROR?γt和促进STAT5/Foxp3信号通路的活化.
Objective: To observe the effect of Liancao-Xieli capsule on intestinal mucosal inflammatory factors and TLR4/PI3K/Akt/mTOR signaling pathway in mice with ulcerative colitis (UC); Methods: 40 male C57BL/6 mice were randomly divided into the control group, model group, Liancao-Xieli group and mesalazine group, with 10 mice in each group. In addition to the control group, the remaining three groups of mice were induced by 3% dextran sulfate sodium (DSS) to induce acute UC model. During the modeling period, mice in each group were given corresponding drugs and normal saline by gavage. At the end of the experiment, HE staining was used to observe the pathological changes of colonic tissue in each group, and ELISA was used to detect the inflammatory factors (TNF-α, IL-6, IL-1β, IL-8, IL-17, and INF-γ) in serum and colonic tissue. The expression levels of TLR4/PI3K/Akt/mTOR signaling pathway related proteins were also detected by Western blot; Results: Compared with the model group, Liancao-Xieli capsule could significantly increase the colon length and decrease the score of colon histopathology in UC mice (P<0.01). In addition, the levels of TNF-α, IL-6, IL-1β, IL-8, IL-17, and INF-γ were significantly reduced in serum and colon tissue, and the expressions of TLR4, PI3K, p-Akt and p-mTOR were significantly down-regulated in Liancao-Xieyi group when compared with the model group (P<0.01).While the expressions of Akt and mTOR were not significantly affected in Liancao-Xieyi group (P>0.05); Conclusion: Liancao-Xieli capsule can reduce the secretion of inflammatory factors, improve the intestinal mucosal damage and inflammatory response in UC by inhibiting the activation of TLR4/PI3K/Akt/mTOR signaling pathway.
目的 观察疏肝利胆颗粒治疗肝胆湿热型慢性胆囊炎的临床疗效.方法 选取2018年3月-2019年3月黑龙江省中医药科学院南岗分院消化科门诊收治的符合诊断为慢性胆囊炎,且辨证属于肝胆湿热型的患者78例.患者口服疏肝利胆颗粒2个月后,观察患者临床疗效;观察患者胆囊和胆囊管壁厚、毛糙、透声等积分情况;观察上腹痛,上腹胀满,恶心等中医证候积分情况;血中谷草转氨酶、谷丙转氨酶、谷氨酰转肽酶、总胆红素变化情况以及不良反应发生情况.结果 治疗后患者痊愈占35.9%;显效占44.9%;有效占12.8%;无效占6.4%,总有效率93.6%;患者胆囊积分、中医证候积分较治疗前明显降低,P<0.05;患者血中谷草转氨酶、谷丙转氨酶、谷氨酰转肽酶、总胆红素未出现异常,无明显不适症状发生.结论 疏肝利胆颗粒可缓解肝胆湿热型慢性胆囊炎患者临床症状,提高临床总有效率,改善彩超下胆囊和胆囊管壁厚、毛糙、透声差等情况,且不良反应少,安全性强,值得临床推广.
幽门螺杆菌(Helicobacter pylori,Hp)感染是慢性胃炎、胃、十二指肠溃疡及胃癌的重要致病因素[1].Hp在人群中传播广、感染率高,呈全球性分布,据统计我国Hp平均感染率高达58. 07%[2].近几年,随着人群对抗生素耐药率普遍提高,Hp首次治疗根除率逐年下降,多次联合应用抗生素使得患者依从性差,故而临床急需新的有效治疗 Hp的药物[3].本研究观察连草颗粒联合三联疗法治Hp阳性慢性胃炎的疗效,报道如下.
目的:通过16S rRNA技术探讨芪参汤对非酒精性脂肪性肝纤维化(NAFLF)的作用机制.方法:采用四氯化碳腹腔注射联合高脂饮食的方法构建NAFLF大鼠模型,造模期间各组给予相应的药物干预治疗,疗程8周.分析比较各组大鼠治疗后肝脏组织病理、血清肝功能、血脂及肝纤维化指标的改变,并收集盲肠末端内容物,通过ⅢuminaMiseq平台进行肠道菌群测序.结果:与模型组相比,芪参汤能够显著改善NALFL大鼠肝脏组织病理学改变,降低NAS评分、油红染色面积、胶原染色面积、ALT、AST、TC、TG、HA、LN、PⅢNP和C-Ⅳ的水平,差异均具有统计学意义(P<0.05).此外,与空白组大鼠相比,模型组大鼠肠道菌群丰富度和多样性显著降低(P<0.05),芪参汤能够显著增加NAFLF大鼠肠道菌群的丰富度和多样性(P<0.05),并且上调拟杆菌目S24-7群、双歧杆菌属、乳酸杆菌属、苏黎世杆菌属、副杆状菌属、考拉杆菌属、毛螺菌科NK4A136群、红蝽菌科_UCG-002、毛螺旋菌属、臭味菌属、Anaerostipes菌属、瘤胃菌属、艾克曼菌属、Romboutsia菌属、霍尔德曼菌属和嗜血杆菌的表达丰度,差异均具有统计学意义(P<0.05).结论:芪参汤通过恢复NAFLF大鼠肠道菌群的多样性,增加肠道中益生菌的丰度,从而实现抗肝纤维化的作用.
目的:观察疏肝利胆颗粒治疗肝胆湿热型胆结石的临床疗效.方法:观察48例肝胆湿热型胆结石患者口服疏肝利胆颗粒2个月后临床疗效、血中总胆固醇、碱性磷酸酶、谷丙转氨酶、总胆红素变化,以及不良反应发生情况.结果:治疗后,89.9%患者临床症状明显改善,胆结石变小或者消失;血液中胆固醇水平治疗后较治疗前降低,差异有统计学意义(P<0.05);不良反应较少.结论:疏肝利胆颗粒治疗肝胆湿热型胆结石有较好疗效,值得推广.
目的 探讨定性风险评估法在职业病危害预评价的适用性、实用性和优越性.方法 采用定性风险评估模型综合评价建设项目各种危害因素,得出风险等级,判定拟采取的防护设施是否能满足防护要求.结果 原料上料、粉碎及输料过程、成品粉碎、包装过程采取2级风险控制对策;炭化炉炭化过程和秸秆燃烧过程采取3级风险控制对策;化验作业中使用氢氧化钠、硫酸、盐酸时分别采取1、1、3级风险控制对策.结论 在实际工作中将风险评估法运用到建设项目的职业病危害预评价中是完全可行的,该方法弥补了类比法的不足.
目的:观察五龙消癥丸对脾胃虚弱、血瘀内阻证Ⅲ/Ⅳ期胃癌的临床疗效.方法:将64例Ⅲ/Ⅳ期胃癌患者,随机分为对照组32例与治疗组32例.对照组采用消癌平片+五龙消癥丸模拟剂口服治疗,治疗组采用五龙消癥丸+消癌平片模拟剂口服治疗,每日3次,两组患者疗程均为24周,并随访24周.分别观察两组患者生存率及治疗前后中医症状积分、Karnofsky评分、QLQ-C30评分的改善情况.结果:治疗组中位生存期为289d,对照组中位生存期为232d,治疗组明显优于对照组(P<0.05).治疗组中医症状积分、Karnofsky积分及QLQ-C30积分较治疗前明显改善(P<0.01),且明显优于对照组(P<0.05,P<0.01).结论:五龙消癥丸能够改善脾胃虚弱、血瘀内阻证Ⅲ/Ⅳ期胃癌患者的临床症状及体征,增加其体力状况和生活质量,增加生存率,疗效显著.
[目的]观察连草泻痢胶囊对活动期溃疡性结肠炎大肠湿热证患者炎性因子及肠道黏膜屏障的影响;方法:选取符合诊断及纳入标准的溃疡性结肠炎患者60例,随机分为对照组(n=30)和治疗组(n=30).其中对照组给予美沙拉嗪肠溶片口服治疗,治疗组在对照组的基础上给予连草泻痢胶囊口服治疗,2组患者疗程均为8周,分别观察2组患者治疗前后中医症状积分、Sutherland疾病活动指数和Baron评分的改善情况,同时检测炎性因子(TGF-α、IL-10和IL-17)、基质金属蛋白酶(MMP2和MMP9)及氧化三甲胺(TMAO)水平的变化,并评价其临床疗效;结果:治疗组总有效率为90%,对照组总有效为73.3%,治疗组优于对照组(P<0.05);治疗组治疗后中医症状积分、Sutherland疾病活动指数和Baron评分与本组治疗前比较显著降低(P<0.01),明显优于对照组(P<0.01);此外,治疗组治疗后TNF-α、IL-17、MMP-2、MMP-9和TMAO水平较治疗前显著降低,IL-10水平显著升高(P<0.01);2组治疗后组间比较,治疗组明显优于对照组(P<0.01);[结论]连草泻痢胶囊联合美沙拉嗪肠溶片能够改善炎性因子的失衡和肠道黏膜屏障功能的异常,缓解大肠湿热证活动期溃疡性结肠炎患者临床症状,疗效显著.
Objective:To observe the clinical efficacy of Jiedu Huayu Decoction on patients with active ulcerative colitis and its effect on Th17/Treg cells and related cytokines.Methods:48 patients with active ulcerative colitis of accumulated damp-heat type were divided into the treatment group(24 cases) and the control group(24 cases) according to random number table.The control group was treated with Mesalazin enteric-coated tablets and the treatment group was treated with Mesalazin enteric-coated tablets combined with Jiedu Huayu Decoction.The course of treatment was 12 weeks in both groups.The clinical efficacy were observed;ELISA method was used to detect the levels of plasma IL-17,IL-10 and TGF-β.Real-Time PCR was used to detect the expression of nuclear transcription factor RORγt/Foxp3 mRNA in Th17/Treg cells.Results:The total effective rate of the treatment group was 91.67%,significantly better than that of the control group (P< 0.05);IL-17 was reduced significantly after treatment compared with before treatment in both groups (P< 0.01) and IL-10 and TGF-β were increased significantly after treatment compared with before treatment in both groups (P<0.01).After treatment,the treatment group was obviously better than the control group(P< 0.01).RORγt mRNA and RORγt/Foxp3 ratio was reduced significantly after treatment compared with before treatment in both groups(P<0.01) and Foxp3 mRNA was increased significantly after treatment compared with before treatment in both groups.(P < 0.01).After treatment,the treatment group was obviously better than the control group (P< 0.05 or P< 0.01).Conclusion:Jiedu Huayu Decoction can obviously improve the clinical symptoms of patients with active ulcerative colitis of accumulated damp-heat type,decrease IL-17,increase levels of IL-10 and TGF-β,regulate the expression of RORγt and Foxp3 mRNA,and promote the balance of Th17/Treg cells,with a low recurrence rate and obvious curative effect.

目的 探究五龙消癥丸对胃癌BGC-823细胞侵袭与迁移能力的影响及其作用机制.方法 分别用五龙消癥丸、PI3K激动剂胰岛素样生长因子l (IGF-1)及PI3K特异性抑制剂LY294002作用于BGC-823细胞,采用MTT法检测不同质量浓度五龙消癥丸对BGC-823细胞增殖的影响;Transwell小室模型研究五龙消癥丸对BGC-823细胞垂直侵袭和迁移能力的影响;采用ELISA法检测五龙消癥丸对血管内皮生长因子(VEGF)、基质金属蛋白酶2(MMP-2)和MMP-9表达的影响;采用Western blotting及RT-PCR法检测脂酰肌醇3-激酶(PI3K)/蛋白激酶B(Akt)/核转录因子-κB(NF-κB)信号通路中相关蛋白及mRNA的表达.结果 五龙消癥丸可显著抑制BGC-823细胞的增殖、黏附、侵袭和迁移能力,显著抑制BGC-823细胞中VEGF、MMP-2和MMP-9蛋白的表达,显著抑制p-PI3K、p-Akt、磷酸化IkB激酶-α(p-IKK-α)、p-NF-κB p65Ser276蛋白及PI3K、Akt、IKKα、NF-κB mRNA的表达,并且呈现一定的时效和量效关系.结论 五龙消癥丸通过抑制PI3K/Akt/NF-κB信号通路相关蛋白及mRNA表达,从而达到抑制BGC-823细胞的增殖、迁移、侵袭作用.
介绍张雅丽教授治疗肝郁脾虚型非酒精性脂肪肝经验.非酒精性脂肪肝多以肝郁脾虚为主要分型,治以疏肝健脾,佐以化痰、化瘀.
Objective:To study the effect of Qishen Erlian Decoction combined with Lamivudine at virology,serology,liver function,immune factors and drug resistance on chronic viral hepatitis B patients with high viral load.Methods:A total of 87 patients with chronic hepatitis B were divided into treatment group (44 cases) and control group (43 cases) according to the random number table.The treatment group were treated with Qishen Erlian Decoction combined with lamivudine and the control group received lamivudine treatment.Two groups were treated for 48 weeks.To observe and compare the symptoms and signs,viral serological and liver function changes,immune factors and drug resistance rate of two groups patients before and after the treatment.Results:The total effective rate in the treatment group was 84.09% and 67.44% in the control group.The difference between the two groups was statistically significant (P < 0.05).After treatment,HBeAg negative and HBeAg conversion number,HBsAg negative number,HBV-DNA negative cases and the number of treatment were statistically significant compared with those before treatment (P < 0.05).ALT,AST,TBIL and ALB were lower in two groups and the difference was statistically significant after 12 and 48 weeks compared with those before treatment in treatment group(P < 0.01).CD3+,CD4+,CD4+/CDs+ and NK in the two groups after treatment were increased.There were no significant difference compared with those before treatment after 12 weeks treatment and the difference was statistically significant after 48 weeks in treatment group(P < 0.05).There were no significant changes in the two groups after 12 weeks treatment (P > 0.05) and there was no significant difference between the two groups (P > 0.05).CD8+ decreased compared with that before treatment in the two groups after 48 weeks treatment and the difference was statistically significant (P <0.05).The total drug resistance rate was 11.31% in the treatment group and 32.56% in the control group.The difference of drug resistance rates between the two groups was statistically significant (P < 0.05).Conclusion:Qishen Erlian Decoction combined with lamivudine not only can significantly suppress HBV-DNA replication,improve the HBsAg,HBeAg negative conversion rate and effectively improve liver function,but also can regulate immune function and reduce the variation of HBV drug resistance,worthy of further popularization and application.
Objective:To observe the curative effect of Danqi Quyu Zhitong Granule on patients with PLGC and influence on expressions of CDX1,CDX2 in gastric mucosa.Methods:64 Patients of PLGC were randomly divided into control group (34 cases) and observation group (34 cases) according to the diagnostic criteria and inclusion criteria,the observation group was given Danqi Quyu Zhitong Granule,the control group treated with Weifuchun tablet,they were treated for three months.To observe the clinical symptoms changes and the change of endoscopic mucosa and the positive expression rate of CDX1,CDX2 proteins in gastric mucosa by immunohistochemical methods at the end of the course.Results:The total effective rate of observation group was 88.2%,the control group total effective rate was 64.7%,difference was statistically significant between the two groups (P <0.05).The total effective rate of gastroscopy in the two groups were respectively 85.3% and 58.8%,the difference was statistically significant between the two groups (P < 0.05).The positive expression rates of CDX1 protein in gastric mucosa of the two groups patients were 82.35% and 79.41% before treatment (P >0.05) and the positive rates were 41.18% and 67.65% after treatment(P <0.05),the difference was statistically significant in the observation group compared with pre-treattment (P < 0.01);The positive expression rates of CDX2 protein in gastric mucosa were 88.24% and 91.18% before treatment (P > 0.05) and the positive rate were 20.59% and 55.88% respectively(P < 0.01) after treatment,the difference was statistically significant in both groups after treatment compared with pre-treattment (P < 0.01),the effect of observation was more significant (P < 0.05).Conclusion:Danqi Quyu Zhitong Granule can improve the clinical symptoms and the changes of gastric mucosa,down-regulation the expressions of CDX1 and CDX2 proteins.
Objective:To observe the effect of Danqi Quyu Zhitong Granule on the PTEN and RUNX3 gene methylation and expressions of PTEN and RUNX3 mRNA on gastric mucosa in rats with PLGC.Methods:Eighty rats were divided into blank group,model group,vitacoenzyme group and Danqi Quyu Zhitong granule group randomly.Except the blank group,the other groups were created PLGC rat model.After modeling,the vitacoenzyme group and Danqi Quyu Zhitong granule group were fed with corresponding drugs.The blank group and model group were treated with 0.9% NaCl solution orally,once a day.After 12 weeks of intragastric administration,the pathological changes of gastric tissue were observed.The expressions of PETN and Runx3 gene methylation were detected by MSP method and the expressions of PTEN and Runx3 mRNA were detected by RT-PCR method.Results:The vitacoenzyme group and Danqi Quyu Zhitong Granule group can improve gastric mucosa inflammation,atrophy,intestinal metaplasia and dysplasia,and the Danqi Quyu Zhitong Granule group was the best.After modeling,the PTEN and RUNX3 gene methylation in the model group were significantly higher than those of the blank group (P <0.01).After drug intervention,the methylation rate of the vitacoenzyme group was decreased and the difference was statistically significant compared with that of the model group (P < 0.05).And the difference was also statistically significant compared with that of Danqi Quyu Zhitong Granule group(P < 0.01),while PTEN and RUNX3 gene methylation of Danqi Quyu Zhitong Granule group were significantly lower than those of the vitacoenzyme group (P < 0.01).The results of RT-PCR showed that the PTEN mRNA and RUNX3 mRNA in the model group decreased significantly compared with those of the blank group (P < 0.01).PTEN and RUNX3 mRNA were significantly up-regulated after drug intervention.The Danqi Quyu Zhitong Granule group had the best effect and the difference was statistically significant (vitacoenzyme group P < 0.05,Danqi Quyu Zhitong Granule group P < 0.01).Conclusion:Danqi Quyu Zhitong Granule can significantly improve the atrophy,intestinal metaplasia and dysplasia of gastric mucosal tissue.Its mechanism may through the reduction of gene methylation ratio and up-regulation of the expression of PTEN RUNX3 mRNA implementation.
[Objective]To observe the clinical effect of Jiedu Huayu decoction on active ulcerative colitis of accumulated damp-heat type.[Methods]70 patients with active ulcerative colitis of accumulated dampheat type were divided into the treatment group(35 cases)and control group(35 cases)according to random number table.The control group was treated with Mesalazin enteric-coated tablets and the treatment group was treated with Mesalazin enteric-coated tablets combined with Jiedu Huayu decoction.The course of treatment was 12 weeks in both groups.The clinical efficacy was observed.ELISA method was used to detect the levels of plasma COX-2,PGE2 and MMP-1,and flow cytometry was used to detect the contents of CD8+ CD28+ and CD8+ CD28-T lymphocyte subsets in peripheral blood.[-Results]The total effective rate was 93.3% in the treatment group and 80.6% in the control group.The COX-2,PGE2 and MMP-1 in treatment group were significantly lower after treatment(P<0.01).After treatment,the difference of COX-2 and MMP-1 level were statistically significant compared with the control group(P<0.05)and the difference of PGE2was statistically significant compared with the control group(P<0.01).CD8+ CD28+,CD8+ CD28 and CD8+ CD28+/CD8+ CD28-were improved after treatment in the treatment group,the differences were statistically significant(P<0.01).Differences of CD8+ CD28+,CD8+ CD28+/CD8+ CD28-were statistically significant compared with the control group(P<0.01)and difference of CD8+ CD28-was statistically significant compared with the control group(P<0.05).[Conclusion]Jiedu Huayu decoction can obviously improve the clinical symptoms of patients with active ulcerative colitis of accumulated damp-heat type,reduce the level of COX-2,PGE2 and MMP-1,regulate the balance of CD8 + CD28+/CD8+ CD28-,with a low recurrence rate and obvious curative effect.
