BACKGROUND:ATP sulfurylase (ATPS) is a crucial enzyme for the selenate assimilation pathway in plants.RESULTS:In this study, genome-wide and comparative analyses of ATPS in Cardamine hupingshanensis, including sequence and structural analyses, were performed. The expression of ChATPS gene family members in C. hupingshanensis under selenium (Se) stress was also investigated, and our results suggest that ChATPS1-2 play key roles in the response to Se stress. Nine ATPS genes were found from C. hupingshanensis, which share highly conserved sequences with ATPS from Arabidopsis thaliana. In addition, we performed molecular docking of ATP sulfurylase in complex with compounds ATP, selenate, selenite, sulfate, and sulfite. ChAPS3-1 was found to have stronger binding energies with all compounds tested. Among these complexes, amino acid residues Arg, Gly, Ser, Glu, and Asn were commonly present.CONCLUSION:Our study reveals the molecular mechanism of C. hupingshanensis ATP sulfurylase interacting with selenate, which is essential for understanding selenium assimilation. This information will guide further studies on the function of the ChATPS gene family in the selenium stress response and lay the foundation for the selenium metabolic pathway in higher plants.
地膜对农业生产起着保墒增产、防草节肥、增温保湿等作用.恩施地区地膜的使用量及覆盖面积总体呈上升趋势,使用量每年2000 t以上,覆盖面积每年50千hm2以上,覆盖作物品种多样,对农林业发展及农民增产增收起到了重要的作用.但由于恩施地质地貌特点、土壤特性、农民环境保护意识、机械化程度底、劳动力成本增加、土地流转量增加等多种因素,导致恩施各地覆膜地块中都有一定量的地膜残留,平均残留量约为29.92 kg/hm2.需要通过提高农民环境保护意识、制定政策标准、加强经济调控、使用可降价地膜等多种措施来综合防治恩施地区的地膜残留污染.
研究生是高等教育人才培养的最高层次,是党和政府宝贵的人才资源.研究生教育是我国社会主义现代化建设拔尖创新人才培养的重要渠道.新时代背景下,随着教育改革的不断深入推进,研究生思想政治教育面临着新挑战.通过对省属地方民族院校M大学研究生的抽样、走访调查,对研究生思想政治教育当前面临的挑战进行分析,并从学校制度建设层面、课程改革方面以及对学生自我教育的引导等方面提出加强研究生思想政治教育的对策.
目的:研究壶瓶碎米荠叶片不同生长期挥发性成分和硒含量之间的关系.方法:使用顶空固相微萃取气相色谱-质谱联用仪(SPME-GC-MS)测定超聚硒壶瓶碎米荠叶片挥发性成分,通过石墨消解仪将叶片消解后,用双道原子荧光光度计测定硒的含量.结果:1)壶瓶碎米荠叶片在幼苗期、抽苔期、开花期、结荚期的挥发性成分分别有4、7、11、6种;2)异硫氰酸苄酯在不同时期壶瓶碎米荠叶片中相对含量较高,在幼苗期高达82.22%;3)随着壶瓶碎米荠的生长,叶片硒含量呈显著性增加,到开花期时,达到最高为1 103.55 mg/kg,随后变化不显著,幼苗期硒含量最低为339.00 mg/kg.结论:1)壶瓶碎米荠叶片挥发性物质主要为异硫氰酸苄酯、2,2,4,6,6-五甲基庚烷、苯乙腈、1-戊烯-3-醇,其相对含量在各个时期中均较高;2)壶瓶碎米荠叶片挥发性成分中的异硫氰酸苄酯含量和硒含量变化之间呈负相关.
The height of plant and the lodging resistance of rapeseed can be improved appropriately, and the crown width, branch angle and stem diameter of rape be not significantly affected when the rape was planted in super sparse planting pattern of 1 plants per m2, and covered with plastic film. Planting rape on rapeseed has a negative effect on rapeseed quality.The content of erucic acid, glucosinolates and protein content of rapeseed increased, and the oil content decreased. It showed that under the super sparse planting pattern, rape planting was not the best way of planting by comprehensive analysis.
为了培养生物工程创新应用型人才,针对目前生物工程专业人才培养存在的问题,以湖北民族学院生物工程专业为研究对象,通过构建生物工程专业创新应用型人才培养模式、优化生物工程专业专业课程教学体系、创立多层次的学生科研训练平台、建立生物工程专业实习实训基地库、构建双师型教学队伍等多种手段,进行生物工程专业创新应用型人才培养的探索和实践,成效明显:学生实验动手能力、创新能力均得到提高;学生专业技能、科研能力得到显著提高;毕业生受到社会各界的好评,学生就业率得到稳步提升.
本文以地方民族院校(湖北民族学院)生物工程专业为研究实例,探讨了"三轮驱动"导向式人才培养改革模式.依托校企共建平台,本着"厚基础、强能力、重技能、高素养、拓口径"的要求,明确专业培养目标和建设重点,按照"分类指导、准确定位、注重内涵、强化特色"的原则,坚持以"面向全体"、"面向全过程"、"面向全方位"为基准,为少数民族地区构建复合技能型生物产业高级人才和区域经济社会转型发展与产业结构调整需要的应用型人才的培养体系.
以西洋菜为原料,采用不同的提取温度、料液比、提取次数和提取时间进行单因素试验提取西洋菜多糖,然后在此基础上采用4因素3水平正交试验对提取条件进行优化.结果表明:提取时间对西洋菜多糖得率影响存在着显著差异(P<0.05),提取次数的影响呈极显著性差异(P<0.01),提取温度的影响呈极显著性差异(P<0.01),但料液比对西洋菜多糖得率的影响不显著.因此,西洋菜多糖提取的最优工艺参数为:提取次数为3次,西洋菜与提取液的比例为1∶25,在60℃条件下提取2h较为理想,为西洋菜的深加工和综合开发利用提供技术支撑.
师范生的教育实践能力是衡量师范生素质的重要标志.我国现有民族院校15所,其中多数院校都办有师范教育,几十年来培养了一大批立足本职岗位、服务民族地区教育事业的各级各类学校的合格教师.加强师范教育的教学改革,特别是实践教学的改革尤为重要.为此,我们以湖北民族学院师范专业为研究对象,深入分析新形势下我国高等教育和基础教育改革与发展形势对师范教育人才培养的要求,进行了一系列师范教育实践教学的改革与探索.

In order to improve the quality of the raw lacquer,we sprayed the lacquer tree nutrient solution with mineral elements(P,K,Cu,Mg,etc.) and plant growth regulators on the Maoba lacquer trees,then observed the growth of lacquer tree and tested the quality of raw lacquer.The results showed as follows:The lacquer trees in the treatment group flourish,barks are thicker,and the time of leaf shedding was postponed about half a month,with no side effect;the raw lacquer from the treated lacquer trees contained more urushiol,laccase and gum;the organoleptic index of the raw lacquer from the treated lacquer trees improved significantly;the surface drying time of raw lacquer from the treated lacquer trees was shortened.The main mechanism is that the lacquer tree nutrient solution changed the nutrient condition and improved metabolism.The nutrient solution can promote lacquer-tree and improve lacquer quatity.
In order to increase the production of the raw lacquer,the authors prepared the Toxicodendron verniciflum(Stokes) F.A.Barkley nutrient solution with mineral elements(P,K,Cu,Mg,etc.) and plant growth regulators,sprayed it on the Maoba lacquer tree were used to study.The result showed that the its flourish,bark thickening,the time of leaf shed were postponed about half a month,no side effect in the treat group.The raw lacquer production was increased 21.2% in the treat group.Production mechanism is that the lacquer enhances the metabolic synthesis after using nutrient solution.
采用垂直板不连续聚丙烯酰胺凝胶电泳对湖北省恩施州8个地区的续断进行过氧化物同工酶分析,并用NTSYS软件对谱带进行聚类分析.供试的8份续断材料共显示出5条谱带,并存在不同程度差异,具体表现在酶带数目、酶带强弱的不同.分析结果可以作为续断遗传差异及亲缘关系的基础.
With Dipsacus asper in three different habitats(Taiping Fownship of Hefen County,selenium-enriched area of Yutangba and Badong County) as materials,the TPS-1 portable photosynthesis system is used to study its dinrnal photosynthetic change and light response curves.The non-rectangular hyperbolic model and onon-linear regression combination of SPSS software are used to estimate the photosyntheetic pararneters.The results showed that Dipsacus asper diurnal variation of net photosynthetic rate of a typical single-peak cruve,the peak value occurred at 11:00 or so.Dipsacus asper light compensation point(LCP) is 21.120~66.52μmol/m2s and the light saturation point(LSP) is 300.793~689.710μmol/m2s.There were some differences among the three areas.Yutangba and Taiping Townshiphave better ability to adapt to light.
In order to increase yield and quality of raw lacquer,growth of lacquer tree was studied,by preparing nutrient solution added mineral elements(P,K,Cu,Mg,and so on) and plant growth regulators,and the nutrient solution with different concentrations was sprayed on Maoba lacquer tree.The results showed that the sprayed lacquer tree was more flourish,had thicker bark,and defoliation time was postponed about half month,compared with CK.Raw lacquer had no side effect,yield was obviously increased and quality was obviously improved.
Wu-He Dipsacus germination seeds and seedlings were used as the experiment materials to study its polyploid induction.The results showed that when 0.2% colchicine solution was used to dealthe seeding tips for 42h or 48h and 0.3% colchicine solution to deal with 36h,the rate of induction was highter.Plant material has obvious polyploid characteristics.The use of colchicine for different parts of Wu-He Dipsacus in polyploid induction is an effective method.
Genetic diversity of Dipsacus asper of eight different areas in Enshi by using RAPD molecular markers.5 among 50 primers were selected to amplify about 53 segments DNA.Cluster analysis using NTSYS software,a similar coefficient of 0.85 as the standard,all varieties can be divided into three categories.GroupⅠincluding Longping,Yuanbao,Yutangba embankments,the Se mine enriched area of Yutangba,Lvcongpo;GroupⅡincluding Taiping and Wuliping;Shuanghe of Enshi City is divided into a class of independent.This study can provide a reference and basis for the identification of Dipsacus asper.
Objective To provide the basic guarantee for studying the genetic diversion of genuine medicinal material,the species identification and the construction of gene fingerprint and so on.Methods With fresh and tender leaves of medicine source plant of Codonopsis pilosula and Dipsacus asperoides C.Y.cheng et T.M.Ai.as tested materials,the genomic DNA was extracted from medicine source plant of C.pilosula and D.asperoides by using the modified CTAB method and was tested.Results There are higher concentration of the genomic DNA extracted from medicine source plant of C.pilosula and D.asperoides by using the modified CTAB method,which satisfy demand of PCR.Conclusion It was effective to extract the genomic DNA from fresh leaves by modified CTAB method.
Extraction and stability of pigment from Magnolia denudata Desr.was studied using absorbance as the index by the single factor experiment and orthogonal experiment.The optimum extraction conditions were obtained through single factor analogic change method.The highest yield was obtained under the conditions of with ratios of solid to liquid by 1:15,extracted 2 times and for 60 min each time under 65℃.The pigment from Magnolia denudata Desr.was water soluble.The pigment could be significantly affected by pH and UV-light.Metallic ions,such as Al3+,Ba2+,Na+,Ca2+,K+ and Mg2+ had no effect on color of pigment,while Cu2+ and Fe3+ had negative effect.Temperature had little effect on pigment.The pigment had poor reduction tolerance and relatively high antioxidation capacity.Sucrose,salt and amylum had no effect on pigment.The pigment from Magnolia denudata Desr.can be widely used in the industry of food,medicine and cosmetic as a natural plant pigment.
