Kaposi's sarcoma (KS) is a locally aggressive, multicentric tumor. RNA-binding proteins (RBPs) are pivotal for post-transcriptional regulation in various tumors. However, the aberrantly expressed RBP genes and their regulatory patterns in KS remain unclear. This study aimed to identify relevant RBP genes in KS and assess the potential functions and molecular interactions of RPS27, a dysregulated RBP in KS tissues, Matched KS lesions and normal control tissues from ten patients were chosen for the study. Differentially expressed genes (DEGs) were first identified by RNA-sequencing, and results were validated through an independent public RNA-seq dataset (GSE147704). Among the DEGs, RBPs were selected for further analysis, with RPS27 chosen for detailed investigation due to its dysregulation in KS tissues. RT-qPCR and immunohistochemistry were employed to validate RPS27 expression. Cellular experiments were conducted for dysregulated RPS27 to explore its functions. Additionally, improved RNA immunoprecipitation (iRIP)-seq was performed to investigate potential binding interactions of RPS27 in KS. We identified 828 DEGs through RNA-seq, with 367 overlapping DEGs confirmed by the public RNA-seq dataset. We obtained 48 RBP genes from the overlapping DEGs, including 3 upregulated (PCBP3, L1TD1, and PEG10) and 45 downregulated RBP genes in KS. Notably, downregulated RBPs included TECR, PUSL1, DQX1, MAT1A, RACK1, EEF1A2, and EEF1B2, and the remaining downregulated RBPs were all ribosomal protein genes, including RPS27, which was selected for further exploration. Cellular experiments confirmed that RPS27 inhibition could promote cellular proliferation, migration, invasion, and angiogenesis of HUVECs, consistent with its downregulation in KS. iRIP-seq and RNA-seq analyses showed RPS27’s ability to selectively bind to 26 DEGs and showed correlation. The majority of RPS27-bound DEGs were ribosomal protein genes, including RPL8, RPL13, RPL13A, RPL18, RPL19, RPL23, RPLP1, RPL27A, RPL40, RPS2, RPS4X, RPS13, RPS18, RPS21, and RPS27, which were associated with viral transcription and gene expression. Our results identified dysregulated RBP genes in KS and explored the cellular functions and molecular targets of RPS27, indicating its potential regulatory role in KS development.
Viral infection hijacks host cell physiological processes to replicate and spread, in which the calcium ion (Ca2 +) signaling pathway plays a key role in the viral life cycle. Store-operated Ca2+ entry (SOCE), as the major Ca²⁺ influx pathway, is regulated by the endoplasmic reticulum (ER) Ca2+ receptor stromal interaction molecules 1 (STIM1) and plasma membrane channel protein Orai1. Recent studies have shown that viruses disrupt intracellular Ca2+ homeostasis by regulating the STIM1-Orai1-mediated SOCE pathway, thereby promoting viral entry, replication, and pathogenicity. This review systematically summarizes the mechanism of STIM1/Orai1 in viral infection and reveals the molecular basis of the Ca2+ signaling pathway hijacked by viruses, offering a new perspective for understanding the pathogenicity of viruses. To further advance this understanding, future studies should focus on the interaction between viral proteins and STIM1-Orai1 and its downstream signaling network, and aim to develop highly selective inhibitors targeting the SOCE pathway to provide new strategies for antiviral therapy and drug research and development. Collectively, these studies not only deepen the understanding of virus-host interactions but also lay the theoretical foundation for precision medicine and antiviral therapy.
T-cell receptor (TCR) diversity is crucial for adaptive immunity, yet baseline characterizations in pediatric populations remain sparse. We sequenced the TCRβ chain of 325 healthy Chinese children aged 0-18, categorized into six age groups. We also analyzed cellular composition and TCRβ associations using flow cytometry in 81 of these samples. Our results indicate a decrease in TCRβ diversity with age, characterized by an increase in high-frequency clonotypes and notable changes in CDR3 length and V(D)J gene usage. These changes are influenced by early life vaccinations and antigen exposures. Additionally, we found a significant association between reduced TCRβ diversity and a decrease in CD4+ T naïve cells. We also developed a predictive model that identifies specific TCRβ features as potential biomarkers for biological age, validated by their significant correlation with changes in the immune repertoire. These findings enhance our understanding of age-related variations in the TCRβ repertoire among children, providing resourceful information for research on pediatric TCR in health and disease.
皮肤科激光美容亚专业作为一门新兴的交叉学科,需要有针对性的培养学生,如何将皮肤科医学生培养为合格的激光美容亚专业医生是一种挑战.本文从皮肤科激光美容亚专业学科特点、学生特点、以及教学对策等方面进行教学方案的探索及实践.我们回顾性近20年来,皮肤科激光美容亚专业对医学专业研究生的培养,通过多种教学方法、学习途径及临床实践来提高激光美容亚专业医学生的专业知识及实践操作能力.我们认为皮肤科激光美容亚专业医学生的培养,以皮肤科知识学习为基础,结合激光专业理论知识,以及激光实践操作相结合的培养模式,同时培养医学生沟通能力,能促进皮肤科激光美容亚专业高层次人才培养.
目的 观察采用病灶内核切除联合局部放射治疗耳部不同部位瘢痕疙瘩的临床疗效.方法 自 2016 年 2 月至2021 年 6 月,新疆维吾尔自治区人民医院皮肤性病科收治 99 例(112 只耳)耳部瘢痕疙瘩患者,应用"梭形"切口手术切除瘢痕内核,术后辅助局部放射治疗.将耳部瘢痕根据不同部位分为耳垂组(57 只耳)和非耳垂组(55 只耳),比较术后两组复发率.结果 所有患者随访 1~4 年,治愈及有效 100 只耳,总有效率为 89.29%;复发 12只耳,复发率为 10.71%.耳垂组共 57 只耳,复发 6 只耳,复发率为 10.53%;非耳垂组共 55 只耳,复发 6 只耳,复发率为 10.91%.两组复发率相比,差异无统计学意义(P>0.05).结论 病灶内核切除联合局部放射治疗耳部瘢痕疙瘩可取得满意疗效,不同部位耳部瘢痕疙瘩的复发率无统计学意义.
目的 探讨线上以问题为导向的教学法(PBL)联合形成性评价在皮肤专业住培教学中的有效性及应用效果.方法 选取 2022 年 9-12 月新疆自治区人民医院皮肤性病科住培学员共 35 人作为研究对象,采用随机数字表法将研究对象分为观察组(n=18)和对照组(n=17).观察组采用线上PBL联合形成性评价的教学模式,对照组仅采用线上PBL教学模式,以终结性评价作为教学评价方式.结果 观察组学员的理论成绩、技能操作成绩、学习兴趣与主动性、理论知识记忆与掌握、文献检索与阅读能力、临床思维与分析能力、团队协作与沟通能力评分等均高于对照组,差异均有统计学意义(P<0.05).结论 线上 PBL 联合形成性评价的教学模式在皮肤专业住培教学中具有较好的培训和应用效果.
二甲双胍是2型糖尿病的一线治疗药物,目前也逐渐用于皮肤病的治疗,包括银屑病等.本文综述二甲双胍治疗银屑病的机制及临床研究进展,为皮肤科治疗银屑病尤其是银屑病合并代谢性疾病提供思路和方法.
乳糜泻是一种免疫介导的发生于遗传易感个体的慢性炎症性疾病,目前确切的病因尚未明确,遗传、免疫、环境因素均参与了乳糜泻的发生发展.目前研究发现乳糜泻与疱疹样皮炎、银屑病、白癜风、特应性皮炎、斑秃等皮肤病关系密切,故本文旨在总结与乳糜泻相关的皮肤表现.
Objective:To investigate the epidemiology, risk factors and biochemical indexes of nutritional status in children and adolescents aged 2~18 years in Southwest China.Methods:Children attending routine health checkups at the Children′s Hospital of Chongqing Medical University between April 2017 and March 2021 were enrolled in this study.Nutritional status was defined based on BMI cut-off values, and statistically analyzed based on gender, region and age.Its risk factors were analyzed by multivariate Logistic regression.Results:A total of 22 609 cases were recruited and the overall prevalence of wasting, overweight and obesity was 5.87%, 9.81% and 10.50%, respectively.The prevalence of obesity and wasting in boys was higher than that in girls ( χ2=24.79, 12.39, all P<0.05), and the prevalence of overweight in boys was lower than that in girls( χ2=4.32, P<0.05). The prevalence of overweight among boys in urban regions was higher than that in rural regions( χ2=4.68, P<0.05). Compared in three age groups, boys aged 12~18 years had the highest prevalence of obesity( χ2=12.49, P<0.01), while girls had the lowest prevalence of wasting( χ2=6.18, P<0.05). Compared with boys, girls had a lower risk of obesity and wasting ( OR=0.80, 95% CI: 0.73~0.87), ( OR=0.80, 95% CI: 0.71~0.90). Compared to children aged 12~18 years, children aged 2~<6 years had a lower risk of obesity ( OR=0.55, 95% CI: 0.46~0.67), while children aged 6~<12 years had a higher risk of wasting ( OR=1.70, 95% CI: 1.25~2.33). Significant differences were reported in ALT, AST, ALP, UA, UN, CRE, TG, TC, HDL-C and LDL-C levels in obese children (all P<0.05). Conclusion:The nutritional status of children and adolescents in Southwest China is comparable to that at the national level.The prevalence of obesity and wasting is associated with gender and age.Obese children are more likely to have biochemical abnormalities.
患者女, 64岁, 因全身丘疹、溃疡伴瘙痒7个月就诊。2020年2月患者无诱因前胸部出现散在红斑、丘疹, 米粒大小, 伴轻度瘙痒。后逐渐累及头面、颈、背、腹部及左侧大腿, 间歇自行涂抹软膏(具体不详), 经数日皮疹可消退并遗留色素沉着。2020年7月新发皮疹明显增多, 躯干、头面部均受累, 背部瘙痒剧烈, 部分皮疹中央轻度糜烂并结痂, 自行口服抗过敏药物, 病情未见改善。当地医院诊断"大疱性疾病可能", 给予抗过敏、调节免疫治疗, 用药数周效果不理想, 故至本院就诊。门诊以"类天疱疮?"收住入院。患者近期体力、体重未见明显变化。家族中无类似病史。既往体健, 否认糖尿病、心脏病、高血压等病史, 否认药物过敏史, 否认既往皮肤疾病史, 否认特殊物质接触史。体检:一般情况可, 各系统检查未见明显异常。皮肤科检查:头面部、躯干可见米粒至黄豆大小脐状丘疹、斑丘疹, 色泽暗红, 边缘清晰, 部分中央溃烂, 上覆褐色痂皮, 不易去除, 部分皮疹消退后遗留色素沉着, 未见大疱及水疱(图1A、1B)。实验室检查:总三碘甲状腺原氨酸0.72 ng/ml(参考值:0.8 ~ 2.0 ng/ml, 下同), 总甲状腺素4.78 μg/dl(5.1 ~ 14.1 μg/dl), 其他甲状腺功能指标未见异常。血脂系列:甘油三酯1.99 mmol/L(0.48 ~ 1.7 mmol/L), 极低密度脂蛋白0.91 mmol/L(0.18 ~ 0.78 mmol/L), 余未见异常。血尿粪常规、血糖、肝肾功能、电解质、体液免疫(补体C3、补体C4、IgG、IgA、IgM)、自身免疫性抗体(抗核抗体、抗双链DNA抗体、抗可提取性核抗原抗体)等均未见明显异常。肿瘤标志物均阴性。2020-09-21行间接免疫荧光(indirect immunofluorence, IIF)法检测抗棘细胞桥粒抗体阳性(1∶100)、抗桥粒芯糖蛋白(desmoglein, Dsg)1抗体186.00 Ru/ml(<20 Ru/ml);2020-09-28行IIF检测抗棘细胞桥粒抗体阴性, 抗Dsg1抗体>200.00 Ru/ml, 抗表皮基底膜带抗体、抗Dsg3抗体、抗大疱性类天疱疮(bullous pemphigoid, BP)180抗体、抗BP230抗体均阴性。头颅、肺部、腹盆腔计算机断层扫描(CT)均未见异常。
寻常痤疮是以毛囊皮脂腺单位的炎症过程为特征的疾病,会影响80%~90%的青少年[1].严重的痤疮炎症可导致遗留红斑或色素沉着,甚至永久性的痤疮瘢痕[2].与活动性痤疮或痤疮后瘢痕相比,座疮后红斑也称炎症后红斑(post-inflammatory ery-thema,PIE),是一种研究不足的座疮后遗症,其临床意义常被低估.目前很多激光设备已被应用于治疗PIE,但关于射频治疗PIE报道较少.因此,本文回顾性分析2019年3月至2021年3月,采用点阵微针射频(fractional microneedle radiofrequency,FMR)和强脉冲光(intense pulsed light,IPL)两种不同方法治疗痤疮炎症后红斑,比较其临床疗效和安全性,现报道如下.
替雷利珠单抗是由我国自主研发的一种新型人源化抗PD-1单克隆抗体,2019年12月27日获国家药品监督管理局批准,用于治疗复发或难治性经典型霍奇金淋巴瘤,此后陆续获批尿路上皮癌、晚期非小细胞肺癌、肝细胞癌、食管癌等适应证[1].随着替雷利珠单抗的广泛应用,其不良反应也逐渐被报道[2].目前尚无替雷利珠单抗致大疱性类天疱疮(bullous pemphigoid,BP)的报道,本文报道1例相关病例.
目的 论CO2点阵激光联合他克莫司软膏治疗白癜风的效果。方法 选取2020年1月至2021年12月我院白癜风患者58例,随机分成对照组、观察组(各29例)。对照组采用他克莫司软膏治疗,观察组增加CO2点阵激光治疗,比较两组临床疗效。结果 观察组治疗有效率高于对照组;白斑消退、色素消退时间均短于对照组;IgG、IgM、SICAM-1低于对照组;生活质量评分高于对照组;SAS、SDS评分低于对照组(P<0.05);两组的不良反应均较低(P>0.05)。结论 联合CO2点阵激光与他克莫司软膏治疗的疗效可
结节病眼部患病率 7%~60%不等,干眼症是结节病最常见的眼部症状,葡萄膜炎是眼结节病最常见的眼部受累形式.眼部症状可在出现全身症状前数月出现,也可作为孤立的症状出现,当患者出现眼部症状时,通常会忽略结节病的诊断.本文报道一例 65 岁女性患者.因双眼出现黑影 1 个月余就诊,右臀部可触及一 1 cm×1 cm大深在皮下结节.眼部超声提示双眼葡萄膜炎,肺部影像学表现为特征性的肺部结节及纵隔内淋巴结肿大.右臀部皮下结节组织病理示真皮内可见多个由上皮样组织细胞组成的裸结节.
目的:分析电离子联合5-氨基酮戊酸光动力在尖锐湿疣患者治疗中的应用效果.方法:选取2021年3月—2022年3月新疆维吾尔自治区人民医院尖锐湿疣患者86例,根据电脑随机分配为对照组与观察组各43例.对照组采用电离子治疗,观察组采用电离子联合5-氨基酮戊酸光动力治疗.治疗结束后,收集并统计两组患者数据的具体变化.结果:①观察组治疗优良率(95.43%)与对照组(74.41%)比较,差异有统计学意义(P<0.05).②两组治疗后IL-2、IL-4、IL-10、IFN-γ水平均低于治疗前,且观察组低于对照组,差
鲜红斑痣是一种体细胞基因突变疾病,这种突变可改变正常血管内皮细胞的功能,导致血管畸形.在大部分鲜红斑痣/Sturge-Weber综合征患者中存在鸟嘌呤核苷酸结合蛋白q多肽(GNAQ)基因体细胞突变位点,且突变与部分临床表现密切相关.推测GNAQ基因突变可作为早期鲜红斑痣的预测因子指导早期干预治疗.RAS p21蛋白激活因子1(RASA1)的体细胞"二次突变"与鲜红斑痣伴发综合征毛细血管-动静脉畸形的发病有关.基因突变引起下游通路中的促分裂原活化的蛋白激酶(MAPK)、磷脂酰肌醇-3-激酶(PI3K)等蛋白激酶活化,参与鲜红斑痣发生的不同阶段.深入研究鲜红斑痣相关GNAQ基因、RASA1基因和MAPK、PI3K等可为其早期诊断及分子治疗提供理论依据.
Vitiligo is a common acquired depigmenting disease characterized by the loss of functional melanocytes and epidermal melanin. Vitiligo has a long treatment cycle and slow results, which is one of the most difficult challenges for skin diseases. Oxidative stress plays an important role as an initiating and driving factor in the pathogenesis of vitiligo. Antioxidant therapy has recently become a research hotspot in vitiligo treatment. A series of antioxidants has been discovered and applied to the treatment of vitiligo, which has returned satisfactory results. This article briefly reviews the relationship between oxidative stress and vitiligo. We also describe the progress of targeted antioxidant therapy in vitiligo, with the aim of providing a reference for new drug development and treatment options for this condition.
目的 本研究对重庆市主动进行健康体检儿童的检验/检查结果进行分析,以进一步了解儿童健康状况,指导儿童健康管理工作.方法 选取2016年12月—2020年5月主动前往重庆医科大学附属儿童医院进行健康体检的34 219例儿童作为研究对象,对其健康体检资料进行收集及分析.结果 研究对象前10位体检结果异常的项 目依次为维生素D、超声骨密度、乙肝表面抗体、尿常规、血常规、腹部彩超、肾功、微量元素、肝功、血铅.12~17岁研究对象体检结果异常的项目主要包括维生素D、超声骨密度、尿常规、腹部彩超、肾功、肝功、乙肝表面抗体;2~5岁研究对象体检结果异常的项目主要包括血铅、血常规、微量元素.结论 儿童健康管理人员应根据儿童的年龄阶段及自身特点,制定个性化的动态健康管理方案,并积极进行卫生知识宣教,与家长和社会一起,一同为儿童健康保驾护航.
Objectives We investigated the effect and mechanism of miR-21-3p on the biological behavior of keratinocyte HaCaT cells. Methods HaCaT cells were transfected with miR-21-3p mimics and miR-21-3p inhibitor, respectively. Cell proliferation was detected by CCK-8, and apoptosis and cell cycle were analyzed by flow cytometry. The expression changes of Bax and Bcl-2 were measured with immunohistochemistry. The mRNA expressions of IL-17A and STAT3 were detected by real-time fluorescence quantitative PCR. Cytokeratin 17 protein was detected by Western blot. The relationship between miR-21-3p and STAT3 was verified by dual luciferase reporter gene assay. Results The miR-21-3p mimics significantly promoted the proliferation and cell cycle progression of HaCaT cells. However, miR-21-3p mimics significantly inhibited cell apoptosis, decreased Bax expression and increased Bcl-2 expression. Additionally, miR-21-3p mimics significantly increased the expressions of Cytokeratin 17 protein, IL-17A mRNA and STAT3 mRNA. Of note, the miR-21-3p inhibitor exhibited contrary effects to miR-21-3p mimics. Furthermore, STAT3 was a direct target of miR-21-3p . Conclusion miR-21-3p may promote the abnormal proliferation and inhibit apoptosis of HaCaT cells, possibly through regulating JAK/STAT pathway and the expression of Cytokeratin 17. 目的 探讨缺氧诱导因子-1α(HIF-1α)介导的缺氧相关通路在婴幼儿血管瘤中的作用.方法 采用免疫组织化学法分别检测婴儿血管瘤增殖期(30例)、消退期(30例)石蜡组织及正常组织(10例)中的HIF-1α及其靶基因血管内皮生长因子(VEGF)和B淋巴细胞瘤/白血病-2(bcl-2)/腺病毒E1B 19000相互作用蛋白3(BNIP3)的表达,采用酶联免疫吸附测定实验(ELISA)检测普萘洛尔治疗前及治疗后12周血管瘤患儿血清中HIF-1α的浓度,并进行统计分析.结果 HIF-1α、VEGF和BNIP3在增殖期血管瘤石蜡组织中阳性细胞的平均光密度值均显著高于消退期(P<0.05),消退期与正常组织相比差异无统计学意义(P>0.05).普萘洛尔治疗前和治疗后12周患儿血清中以及正常对照组血清中HIF-1α的浓度分别为(0.23±0.07)mg/L、(0.17±0.04)mg/L 和(0.15±0.02)mg/L,结果显示普萘洛尔治疗前HIF-1α的浓度显著高于治疗后12周,差异有统计学意义(P<0.05),治疗后12周与正常对照组相比差异无统计学意义(P>0.05).结论 HIF-1α及其靶基因VEGF、BNIP3在婴幼儿血管瘤的发生、发展及消退中发挥重要作用,且普萘洛尔治疗婴幼儿血管瘤的机制可能与HIF-1α介导的相关通路有关.
