Objective To develop an expert-assisted system for emergent food poisoning accidents in order to provide a tool for emergent treatment of and response to food safety incidents and make available remote professional assistance regarding food safety.Methods We used java software and designed seven function modules:the database of food safety,methods for emergent treatment of common food poisoning accidents,investigation and control of foodborne disease outbreaks,food safety during major events and sudden natural disaster,investigation and evaluation procedures,commonly used standards for food safety,and a remote expert-assisted system.Results All the software functioned well in activities to ensure food safety.Test data of each unit was queried and evaluated,and guidance was given via the website of military networks.Remote assistance was achieved.Conclusion This software is an accessory to the digitilized and rapid detection equipment for food safety.It can be used by grass -roots troops to deal with emergent food poisoning accidents and provide remote professional assistance.
To develop an up-converting phosphor technology based immunostrip assay for quantitative detection of estradiol.A composition contained up-converting phosphor (UCP) as a novel lable and estradiol antibody (E2-mAb) with glutaraldehyde method.The estradiol was detected by the fluorescent immunostrip assay based on indirect competitive principle which had this composition as a probe.Detection performance including stability,specificity and sensitivity was evaluated.The method showed well stability,specificity and sensitivity with the detection limit of 2.75 ng/mL,the linear range of 5 ng/mL to 2 000 ng/mL,the recovery of 88.88 % to 103.5 %,and the coefficient of variation (CV) less than 10 %.The established assay for E2 detection can be successfully used to fast-field analysis and has good economic value and broad application prospect.
•A lower detection limit is showed in this study.•AuNPs deposition extends the measurement range of the sensor.•The imprinted sensor exhibits good selectivity, stability, and reproducibility.•The developed sensor was applied to milk samples without any further treatment.
目的 改造百草枯(Paraquat PQ)半抗原,制备百草枯完全抗原及多克隆抗体,为建立酶联免疫吸附方法及其在食品安全快速检测中的应用奠定基础.方法 改造半抗原PQ-hap,混合酸酐法合成免疫原(PQ-BSA)和包被原(PQ-OVA),采用SDS-PAGE电泳和质谱对其鉴定,免疫家兔2只,制备多克隆抗体;间接ELISA对其进行效价、灵敏度和特异性检测.结果 成功合成百草枯免疫原和包被原,6次免疫后2只家兔血清的效价分别达到1∶40 000和1∶160 000,抗体的半数抑制浓度(IC50)分别为281.6 ng/ml和98.4 ng/ml,与百草枯类似物没有明显交叉反应.结论 合成的百草枯完全抗原具有较好的免疫原性,产生的抗百草枯多克隆抗体具有较好的灵敏度和特异性.
目的 总结第十一批赴刚果(金)维和医疗分队在中国二级医院的卫生防疫工作,为其他驻外执行维和任务部队提供参考.方法 回国后对从集训到赴非洲刚果(金)执行维和任务期间的卫生防疫工作进行回顾性总结.结果 总结出医疗分队在集训期间和执行维和任务期间卫生防疫工作的做法.集训期间:(1)强化体能,预防接种,增强身体免疫力;(2)了解任务区流行病信息,消除恐怖情绪困扰.执行维和任务期间:(1)科学做好流行病学调查并建立健全分队防疫方案;(2)适时做好健康教育和疫情通报;(3)主动做好个人防护工作;(4)扎实开展定期防疫消杀及营区环境卫生治理;(5)始终坚持水和食品安全监测工作;(6)积极开展对外服务和国际交流;(7)适时调整防疫工作重心.任务期内无霍乱、黄热病等传染病发生,疟疾防控达到零感染率.水质调查与保障涵盖了任务区8个分队,营院卫生综合治理有效达标,卫生防疫工作取得良好效果.结论 建立良好的卫生防疫工作机制,科学指导、迅速全面、坚持不懈,是做好维和部队卫生防疫工作的根本保证.
Fast detection is important in screening large-scale samples. This study establishes a direct competitive ELISA method (dcTELISA) based on an enzyme thermistor for fast atrazine (ATZ) detection. ATZ competes with β-lactamase-labeled ATZ (ATZ-E) for the binding sites on anti-ATZ monoclonal antibody (mAb). The mAb are covalently bound to Controlled Pore Glass (CPG) in an immunoreactor to form immunocomplexes with ATZ and ATZ-E. Several parameters of biosensor performance were optimized, such as the ATZ-E concentration, concentration and nature of the substrate, flow rate, and effect of temperature on the sensor response. After optimization, the assay time for a single sample was 12 min. The work process and result were compared with those of high-performance liquid chromatography (HPLC). The detection results exhibited a recovery rate of 88% to 107% in ATZ-spiked fresh cut corn stalks and silage samples. The results obtained via dcTELISA had good correlation with that of HPLC, and the biosensor response was reproducible and stable even when used continuously for over 4 months. All these properties suggested that the fast detection method, dcTELISA, may be used to detect pesticide residue in large-scale samples.
目的 制备抗日本血吸虫蛋白(SjP40)鸡卵黄抗体(egg yolk immunoglobulin Y,IgY),以期为日本血吸虫病的免疫学诊断提供一种特异性强、灵敏度高的优势抗体.方法 以纯化的SjP40抗原免疫健康SPF级产蛋鸡,初次免疫2周后开始每日收集的鸡蛋为样本,采用水稀释硫酸铵沉淀法从鸡蛋中初步提取抗Sjp40 IgY,进一步经透析袋、DEAE-FF阴离子交换柱纯化IgY.用SDS-PAGE、Western Blot及ELISA对纯化后的抗Sjp40 IgY进行分析鉴定.结果 纯化的特异性抗Sjp40IgY,抗体效价1∶106.相对分子质量为180 kDa,SDS-PAGE可见有2条主要蛋白带,分别约为66kDa和35 kDa,并可被Sjp40特异性识别.结论 成功制备具有良好免疫反应性、特异性强、灵敏度高的抗Sjp40特异性IgY抗体.
目的对联合国驻刚果(金)稳定特派团南基武旅部分分队驻地的水源水、饮用水进行检测,掌握当地旱季和雨季的饮用水水质状况,并进行安全评价,以有效防控水源传染病、提高各分队及联合国文职人员的身体健康水平。方法对水源地周边环境、使用人数进行调查;使用制式检水检毒箱、水质细菌检验箱,参照GB 5749-2006《生活饮用水卫生标准》对6个连续监测点的水样及其他分队送检水样进行21项指标的定期监测。结果 6个连续监测点的水质指标比较稳定,个别营地水源的感官指标不合格(有异味),部分山泉水菌群未达标(>100 CFU/ml),某前进营地水源六价铬、铅等重金属指标超标。在任务期检测的135个水样中,76.3%均达到了水源水质卫生学要求。对不符合饮用水卫生标准的水源及时给出使用建议。结论南基武旅部分偏远前进营地水源水质被污染,建议配备净水设备,以保障维和人员身体健康。
A piezoelectric crystal biosensor method has been established for detection of listeria monocytogenes (LM) by gold nanoparticles (GNPs) signal amplification. Based on specific recognition of single-stranded DNA (ssDNA) hybridization, a sandwich format of ssDNA probes/complimentary ssDNA probes modified by GNPs/ssDNA of LM was fabricated. This novel strategy allows the detection limit of LM as low as 10 cfu/mL. The biosensor showed friendly biocompatibility, good specificity, acceptable stability, which provided a potential alternative tool for detection of LM in real samples. Furthermore, the developed method could be easily extended to other gene analysis schemes.
Objective To establish colloidal gold immunochromatographic assay for the detection of Staphylococcus Enterotoxin B (SEB) by optimizing the main experimental conditions. Methods Colloidal gold particles of 16 nm in diameter were prepared by reduction of chlorauic acid (HAuCl4) with trisodium citrate,anti-SEB antibody was labelled with the colloidal gold,then the gold immunochromatography strip for SEB was assembled. Results The spheroidal colloidal gold particles prepared showed bright red color,and uniform size. The minimal stable concentration (MSC) of gold-SEB antibody was 15 μg/mL,and the suitable stable concentration was 18 μg/mL,appropriate pH 7.8. The detection limit of SEB was 0.2 μg/mL and the accuracy was 94.9%. Conclusion The colloidal gold immunochromatography strip for SEB was prepared.This method was simple,of high sensitivity and stability,suitable for rapid detection of SEB in foods.
Development and application of several fast detection techniques,including chemical chromatometry,enzyme linked immunosorbent assay(ELISA),immune colloidal gold technique,biochip,biosensor and mobile chromatography-mass spectrometer for rapid detection of food were introduced.The prospect of fast detection of food was discussed.59 references were cited.
雌二醇(17β-Estradiol)是天然雌激素的重要成分,为防止儿童性早熟等问题,食品中尤其是畜禽产品的雌二醇检测不容忽视.胶体金免疫层析技术(GICA)在检测小分子有机物方面越来越受到重视.其基本原理是胶体金颗粒能够稳定吸附蛋白质,而蛋白质的生物活性无明显变化,所以它可以取代酶标记抗体[1,2].本实验采用标记有雌二醇单克隆抗体的胶体金颗粒与相对应的固定在硝酸纤维膜上的雌二醇结合物相结合,固定有雌二醇结合物的检测线处就显现明显的颜色[3,4].本法具有灵敏度高、特异性强、简便快速、成本较低、结果容易判读等优点,适用于样品的初步筛选.
Objective To preparae man-made antigen and polyclonal antibody of nonylphenol(NP) and to make preparation for the immunoenzyme assay to detect nonylphenol.Methods Man-made antigens were made by DCC and it was tested by the method of UV-scanning.The rabbits were immunized with man-made antigen and the collected antiserum was evaluated by ELISA.Results UV-scanning showed that the link ratio of NP and keyhole-limpet hemocyanin(KLH) was 18∶1.ELISA revealed that the reactive titer of prepared antibody against NP-BSA was up to 1∶32 000.Conclusion The man-made antigen of nonylphenol can be made by using DCC and the polyclonal antibody serum has higher titer,and it takes less than 3 d to prepare the antibody.
