Objective:To observe the expression of c-fos mRNA of rats with cerebral infarction in recovery period under supplementing qi and activating blood circulation drugs.Methods:Longa method was slightly modified in making the rat model of left middle cerebral artery occlusion.According to the random number table method,the rats were randomly divided into 7 groups,including a normal group,a model group,a sham operation group,a supplementing qi group,an activating blood circulation group,a supplementing qi and activating blood circulation group,and an ERK inhibitor group.Using RT-PCR technique,we observed the effect of supplementing qi and activating blood circulation drugs on c-fos mRNA of rats with cerebral infarction after 28 days' treatment in myocardial infarction cortex.Results:After 28 days' treatment,compared with normal group,c-fos mRNA expression in the model group was significantly decreased (P < 0.05);c-fos mRNA expression in the supplementing qi and activating blood circulation group and ERK inhibition group increased to a certain extent than that in the model group (P < 0.05);the activating blood circulation group bad no significant statistical difference with the model group(P > 0.05);c-fos mRNA expression in supplementing Qi group was lower than the other groups (P < 0.05).Conclusion:In the recovery period of cerebral infarction,supplementing qi and activating blood circulation drugs can improve cerebral ischemia in a certain degree,which is better than simply supplementing qi and activating blood circulation,and its therapeutic mechanism may be related to improving the expression of c-fos mRNA.
Objective:To study the effect of Xiongzhi Dilong decoction preadministration at different time on iNOS mRNA,c-fos mRNA expression in migraine animal.Methods:36 healthy male SD rats were randomly divided into six groups:saline group (n =6),migraine model group (n =6),sumatriptan group (n =6),1 day preadministration group (n =6),3 day preadministration group (n =6),7 day preadministration group (n =6).The drug was administered according to different groups,tissues were collected after subcutaneous injection of corresponding drug given 2h later.The c-fos mRNA,iNOS mRNA expression in trigeminal ganglion,spinal trigeminal nucleus,thalamus,dura mater was assayed by real-time PCR method.Results:Compared with saline group,the iNOS mRNA expression in trigeminal ganglion,spinal trigeminal nucleus,dura mater increased significantly,and the iNOS mRNA expression in thalamus decreased significantly in the model group (P< 0.01);after the drug intervention,the iNOS mRNA expression in trigeminal ganglion,spinal trigeminal nucleus,dura mater was decreased,and the preadministration of 7days group and sumatriptan group decreased significantly (P< 0.01).The iNOS mRNA expression in thalamus was increased,but there were no statistical significance compared with the model group (P> 0.05).Compared with saline group,the c-fos mRNA expression increased significantly in trigeminal ganglion(P< 0.01),and spinal trigeminal nucleus (P< 0.05),and the c-fos mRNA expression decreased significantly in thalamus (P<0.05) and dura mater (P<0.01);after the drug intervention,the c-fos mRNA expression in trigeminal ganglion,spinal trigeminal nucleus was decreased,and the preadministration of 7days group and sumatriptan group decreased significantly(P<0.01).The c-fos mRNA expression in thalamus and dura mater was increased,and the results in the preadministration of 7days group and sumatriptan group were statistically significant (P < 0.05).Conclusion:Preadministration of Xiongzhi Dilong decoction can reduce the c-fos mRNA,iNOS mRNA expression in trigeminal ganglion,spinal trigeminal nucleus which were induced by NTG,and can reduce the iNOS mRNA expression in dura mater.The prevention of preadministration of 7 days is better than the prevention of preadministration of 3 days and 1 day.
Objective To observe the infcuence of Xiongzhi Dilong decoction(XDD)on the expressions of c fos and calcitonin gene related peptide(CGRP)immunoreactive neurons in brain tissues of migraine model rats,and to explore its mechanism.Methods For-ty eight healthy male Sprague Dawley(SD)rats were divided randomly and equal y into 4 groups:saline group,migraine model group,sumatriptan group,XDD group.The rats were pretreated with sumatriptan,XDD or saline before received a subcutaneous injec-tion of 10 mg/kg nitroglycerin or saline.After the rats received a subcutaneous injection 4 h and 6 h,the brain tissue samples were col-lected.Immunohistochemistry was applied to detect c fos and CGRP immunoreactive neurons in trigeminal nucleus caudalis(TNC) and periaqueductal gray(PAG).Results Immunoreactive positive neurons in trigeminal nucleus caudalis:c fos immunoreactive neurons and CGRP immunoreactive neurons in XDD group were decreased at both 4 h and 6 h,and there was statistical difference comparing with the migraine model group(P<0.01).Immunoreactive positive neurons in periaqueductal gray(PAG):c fos immuno-reactive neurons and CGRP immunoreactive neurons in XDD group were decreased at both 4 h and 6 h,and there was statistical difference comparing with the migraine model group(P<0.01)as wel .Conclusion XDD can reduce the NTG induced c fos im-munoreactive neurons in trigeminal nucleus caudalis,and attenuate the activatin of trigeminovascular system,can reduce the CGRP immunoreactive neurons in periaqueductal gray as wel ,and inhibit nociceptive transmission.XDD can reduce the CGRP immunoreac-tive neurons in trigeminal nucleus caudalis and periaqueductal gray.
Objective To study the effects of Chaihu Shugan powder (CSD) on the ethological changes and levels of monoamine neu-rotransmitters in the hippocampus and hypothalamus in rats with liver qi stagnation syndrome.Methods The rat model with liver qi stagnation syndrome was established by chronic restraint stress and keeping them alone.Each rat was reviewed by Open field test, Sucrose test and determined the levels of monoamine neurotransmitters in the hippocampus and hypothalamus by high performance liquid chromatography (HPLC).Results The ethological manifestation of model rats was changed obviously,and the levels of 5 hydroxytryptamine (5 HT), 5 hydroxyindoleacetic acid (5 HIAA),dopamine(DA) in the hippocampus and hypothalamus were de-creased,while the level of norepinephrine (NE) was increased.CSD could regulate its trends.Conclusion CSD can improve the model rat’s ethological manifestation and have therapeutic effects of liver qi stagnation syndrome.The mechanism may be adjust the levels of 5 HT,5 HIAA,NE,DA in rat brain with liver qi stagnation syndrome,in order to effectively play the role of anti liver qi stagna-tion.
目的 研究柴胡疏肝散对肝郁证大鼠行为学和大鼠前额叶及海马单胺类神经递质的影响.方法 30只大鼠随机均分为空白对照组(对照组)、肝郁证模型组(模型组)、柴胡疏肝散组(治疗组)3组.用慢性束缚应激结合孤养法建立肝郁证大鼠模型,测评开野、蔗糖水实验,采用高效液相色谱法测定大鼠前额叶及海马单胺类神经递质.结果 模型组大鼠造模成功,与对照组比较,前额叶皮质及海马中的5-HT、5-HIAA、DA含量均下降,NE含量呈增加趋势,但差异无统计学意义;治疗组与模型组相比,5-HT、5-HIAA与DA含量均增加,差异有统计学意义(P<0.05,P<0.01),NE含量呈减少趋势,但差异无统计学意义(P>0.05).结论 柴胡疏肝散具有治疗肝郁证的作用,其作用机制可能与其能够调节肝郁证大鼠脑5-HT、5-HIAA、NE、DA有关,主要是通过增加海马中的5-HT、5-HIAA、DA水平,从而有效发挥抗肝郁作用.
目的:研究芎芷地龙汤不同时间预给药对偏头痛动物模型痛阈及血管活性物质的影响.方法:将健康雄性SD大鼠,随机分为生理盐水组、模型组、舒马普坦组、中药治疗组,每组8只,给药干预后,皮下注射硝酸甘油注射剂10 mg/kg制作偏头痛动物模型,在皮下注射药物后2h后测定,电子式机械测痛仪测定机械痛阈值,温度刺激仪测定温度痛阈值;硝酸还原法进行测定血浆NO浓度;ELISA法测定血浆CGRP、Neurokinin A、Endothelin-1(ET-1)、TXB2、6-Keto-PGF1α含量.结果:给予硝酸甘油后,模型组与生理盐水组比较,机械痛阈(1 d P<0.01,3 d P<0.05,7 d P<0.05)和温度痛阈(1dP<0.01,3 d P<0.01,7 d P<0.01)均降低,血浆NO浓度升高(1 d P<0.01,3 d P<0.01,7d P<0.01),血浆ET-1浓度降低(1 d P<0.05,3 dP<0.05,7 d P<0.01),血浆CGRP含量降低(1 d P<0.01,3dP<0.01,7 d P<0.05),血浆NKA含量升高(1 d P<0.01,3 d P<0.01,7 d P<0.01),血浆TXB2含量降低(1 d P<0.01,3 dP<0.01,7 d P<0.05),血浆6-Keto-PGF1α含量降低(1d P<0.05,3 d P<0.05,7 d P<0.01);预防给药后,芎芷地龙汤组和模型组比较,机械痛阈和温度痛阈均显著升高(3d P<0.01,7d P<0.05),NO浓度显著降低(3 d P<0.01,7 d P<0.01),血浆ET-1浓度升高(3 d P <0.01,7 d P<0.05),血浆CGRP浓度升高(7dP<0.01),NKA含量有所降低(1 d P<0.05,3 d P <0.05,7 d P<0.01),血浆TXB2含量有所增高(3d P<0.05,7 d P<0.01),血浆6-Keto-PGF1α有所增高(3d P<0.05,7 d P<0.05).结论:预防给药3天、7天,芎芷地龙汤均可以提高动物病理状态下的痛阈值,可减弱由NTG诱发的NO的升高水平,可升高由NTG诱发的TXB2降低水平,缓解由NTG诱发的6-Keto-PGF1α的降低水平,升高由NTG诱发的ET-1含量降低水平,减弱由NTG诱发的NKA的释放水平;预防给药7天后,芎芷地龙汤可以升高由NTG诱发的血浆CGRP降低水平.整体来分析,预防给药7天效果好于预防给药3天、1天.
目的:探讨偏头痛受风热侵袭后机体发生痛觉敏化的病理机制及芎芷地龙汤对模型的治疗机理.方法:使用热风及硝酸甘油协同诱导建立大鼠偏头痛风热证模型,观察2h内搔头次数,用免疫组化法测定三叉神经脊束核GLU、NR2B、nNOS表达情况.结果:偏头痛模型组、偏头痛风热证组2h内搔头次数明显增多,与正常组、风热组、中药治疗组相比,差异有统计学意义(P<0.01);偏头痛模型组、偏头痛风热证组三叉神经脊束核尾部GLU表达、NR2B表达、nNOS表达均明显上调,与正常组、风热组、中药治疗组相比,差异有统计学意义(P<0.01).结论:风热对正常机体无影响,而对硝酸甘油致偏头痛模型病理状态有显著影响,说明风热外袭致偏头痛痛觉敏化的发生,芎芷地龙汤能明显改善偏头痛动物模型的行为学症状,减少动物的阳性反应次数,减少三叉神经颈复合体中Glu、NR2B、nNOS表达.
目的 观察泻肝安神方改善广泛性焦虑症(GAD)肝郁化火证患者中文版健康调查简易量表(SF-36)生活质量评分情况及SF-36各维度与焦虑的相关性分析.方法 将130例GAD患者随机分为治疗组和对照组,治疗组100例,对照组30例,分别口服泻肝安神方和帕罗西汀治疗,疗程均为6周.以SF-36、汉密顿焦虑量表(HAMA)对两组GAD肝郁化火证患者治疗前后生活质量水平及焦虑程度进行评价.结果 两组在治疗前后SF-36各条目间比较均无统计学意义(P>0.05),但两组在治疗6周后与本组治疗前SF-36各维度比较有统计学意义(P<0.01).两组在治疗前后SF-36各维度中除生理机能外,其余各维度值与HAMA总分及因子分呈显著负相关(P<0.05).结论 泻肝安神方在改善GAD肝郁化火证患者的生活质量水平方面与帕罗西汀相当,且患者的焦虑情绪对其生活质量影响较大,即患者的焦虑情绪越严重其生活质量水平越低.
目的 观察泻肝安神方治疗广泛性焦虑症(GAD)的临床疗效.方法 将130例GAD患者按照3∶1比例随机分配到治疗组100例和对照组30例,治疗组给予泻肝安神方颗粒,每日1次,每次12g;对照组给予帕罗西汀片,每日1次,每次10或20 mg,两组患者均治疗6周后观察临床疗效.两组分别于治疗前、治疗后第2、4、6周进行汉密尔顿焦虑量表(HAMA)评分,治疗后第2、4、6周进行副反应量表(TESS)评分.结果 两组患者HAMA评分治疗第2、4、6周后与本组治疗前比较,差异均有统计学意义(P<0.05).治疗组临床疗效总有效率为94.0%,对照组为93.3%,两组总有效率比较差异无统计学意义(P>0.05).治疗组患者治疗2、4、6周后TESS评分低于对照组,差异均有统计学意义(P<0.05或P<0.01).结论 泻肝安神方治疗GAD与帕罗西汀片疗效相当,但泻肝安神方不良反应少、程度较轻.
川芎是治疗头痛的常用药物,临床上配伍得当,用量合适,可用于多种头痛的治疗,故有“头痛不离川芎之说”,如《神农本草经》谓川芎“主中风入脑头痛”,《本草衍义》言:“头面风不可阙也”;《本草纲目》云:“人头穹窿穷高,天之象也。此药上行,专治头脑诸疾”。然而对川芎用量、用法、适应症、禁忌症、药对配伍等方面各医家有多种见解,对于这些经验或观点应如何理解,让我们在临床应用川芎治疗头痛时达到心中有数。
《内经》中有关头痛的论述颇为丰富,提出了六经病变所致头痛及其伴随症状、针灸治疗方法,并根据头痛伴随的症状、症状所在的经络脏腑,而调其有关脏腑经络所归宿的腧穴.头痛外感病因:风、寒、湿、寒湿、热,内伤病因:脏腑气血功能紊乱,寸口脉短弱而出现头痛的脉诊诊断,提出治疗头痛的原则:调阴阳,补不足,泻有余;详细记述了厥头痛、头项部疼痛的治疗方法,同时还提出头痛治疗的宜忌及头痛的预后.为后世关于头痛病因、病机、辨证论治、治疗方法、针灸理论奠定了理论基础.
Objective To study the effect of Xiongzhi Dilong decoction(XDD)on vasoactive substance in migraine with wind heat syndrome.Methods To establish migraine with wind heat syndrome model by combining exterior pathogen (hot wind)and GTN ad-ministration,examine the frequency of scratching head within 2 h,and survey the content of thromboxane B2 (TXB2 ),6 Keto PGF1α,calci-tonin gene related peptide(CGRP),endothelin(ET)in blood plasma by radioimmunoassay technology.Results The content of TXB2 ,ET and CGRP in plasma of migraine model was significantly higher than normal group and wind heat group,and the content of 6 Keto PGF1αwas significantly lower than normal group and wind heat group.The content of TXB2 ,ET and CGRP in plasma of migraine with wind heat syndrome moldel persistently increased,and The content of 6 Keto PGF1αpersistently decreased,which is significant difference compared to normal group(P<0.01),and is significant difference compared to migraine model group(P<0.05). The content of TXB2 ,ET,CGRP and 6 Keto PGF1αin plasma of Chinese medicine treatment group is recovered,which was signifi-cant difference compared to migraine with wind heat syndrome model and migraine model group(P<0.05).Conclusion There was no significant effect on normal organism which sufferd from wind heat,But there was significant effect on migraine model induced by Nitroglycerin,The vasoactive substance of Migraine model which was invaded by wind and heat was disbalance severely.XDD could ameliorate remarkably disbalance state of vasoactive substance in wind heat syndrome migraine model.

中医将具有善行多变,动摇不定等特性的致病邪气,取类比象命名叫“风邪”,风的产生中医学分“外风”和“内风”两类.由于风为阳邪,其性轻扬,易袭阳位,而头为诸阳之会,位居高巅,因此易致风邪侵袭而引发头痛.肝为风木之脏,当情志刺激引起肝木失和,肝阳升而无制,肝阳化风,风阳上扰则可致偏头痛.风与头风(偏头痛)有密切的关系,对于头风(偏头痛)的治疗必用“风药”.“风药”具有以下功用:①祛风;②升阳(升发脾阳、升发肝胆、固肾助气化);③升清(升发清气、升精至脑);④疏肝解郁;⑤助行气血;⑥发散郁火;⑦除湿;⑧引经;⑨开通玄府;⑩熄风.风药与其他药物合理配伍可产生明显的增效作用.
Objective:To investigate intervention of Daotan decoction(DTD) in of intercellular adhesion molecule-1(ICAM-1) mRNA expression through the regulation of p53 in order to reveal the mechanisms of DTD in the treatment of atherosclerosis.Method: Neonatal umbilical cords were used to isolate endothelial cells,and they were subcultured for 1 to 3 generations in experiment.According to a dose of 0.9 g · kg-1 · d-1,DTD was given to SD rats by gavage.Blood samples were obtained via the abdominal aorta after 10 days and the serum was separated.The experiment was divided into 7 groups: blank control group,blank serum control group(HUVECs were treated with serum without DTD),tumor necrosis factor-α(TNF-α) induced group(200 U · mL-1 TNF-α,12 h),5% DTD group(5% DTD serum,6 h,and then 200 U · mL-1 TNF-α,12 h),10% DTD group(10% DTD serum,6 h,and then 200 U · mL-1 TNF-α,12 h),20% DTD group(20% DTD serum,6 h,and then 200 U · mL-1 TNF-α,12 h) and PFT-α group(p53 blocker) blocking groups(PFT-α 0.009 g · mL-1,30 min,and then 200 U · mL-1 TNF-α,12 h).The impact of DTD on ICAM-1 mRNA and p53 expression in human umbilical vein endothelial cells stimulated by TNF-α were observed with the PCR and Western blot.Result: ①The expression of ICAM-1 mRNA and p53 in the TNF-α induced group was significantly higher than the normal control and DTD control groups.The difference was statistically significant(P0.01);after being treated with DTD serum or PFT-α,the expressions of ICAM-1 and p53 mRNA were reduced(P0.05).②The protein expressions of ICAM-1 and p53 in the TNF-α induced group were significantly higher than the normal control and DTD control groups.The difference was statistically significant(P0.01);after being treated with DTD serum or PFT-α,the protein expressions of ICAM-1 and p53 were reduced(P0.05).③There was a significant positive correlation between the p53 and ICAM-1 mRNA level(r=0.981,P0.01),and protein level(r=0.854,P0.01).Conclusion:DTD inhibits TNF-α stimulation caused ICAM-1 mRNA expression in umbilical vein endothelial cells through the regulation of p53 expression,therefore,it may play a therapeutic role in the treatment of atherosclerosis.

Objective To establish a rat model of liver qi depression syndrome by using methods of chronic restraint stress and isolation rearing,and to review the animal model through determination of ethology,macroscopic manifestations,laboratory indexes and counter evidence of medicines.MethodsAll of the rats(n=30) were randomly divided into control group,model group and Chaihu Shugan San group.In each group,the changes of macroscopic manifestations and body weigh were observed.The levels of plasma adrenocoticotropic hormone(ACTH) and corticosterone(CORT) were detected by using enzyme linked immunosorbent assay(ELISA) open-field test and sucrose water test.Chaihu Shugan San was used as counter evidence.ResultsThe rats in model group were listless with dry and disheveled hair,waded interests,watery stool and slowly weight gaining.In model group the scores of horizontal movement and vertical movement decreased significantly,dejection amounts increased,preference for sucrose decreased,and levels of plasma ACTH and CORT increased.Chaihu Shugan San had significant curative effects.ConclusionThe rat model of liver qi depression syndrome is successfully established by using methods of chronic restraint stress and isolation rearing.

Objective:To observe the effects of CHSG powder on ERK and P-ERK in hippocampus of rats with liver-qi stagnation.Methods:The rats were divided into three groups:control,model and CHSG powder group.This animal model is made by the method of confining the rats'movement.The cell numbers of ERK1/2、P-ERK in CA1、CA3 and DG sections of hippocampus in different groups were observed by immunohistochemical method.Results:CHSG powder can significantly improve the cell numbers of ERK1/2、P-ERK in CA3 and DG sections of hippocampus.Conclusion:CHSG powder has antagonism against liver-qi stagnation by improving the cell numbers of ERK1/2、P-ERK in CA3 and DG sections of hippocampus.
